Abstract
Description
8.ナチュラルキラー細胞の分泌物を含むアトピー性皮膚炎、脱毛、傷または皮膚しわ改善用薬学組成物。
本発明のアトピー性皮膚炎、脱毛、傷または皮膚しわ改善用化粧料組成物は、ナチュラルキラー細胞の分泌物を含む。
抗−CD3抗体とは、T細胞受容体(TCR)と結合して抗原認識複合体を形成する分子群であるCD3抗原に特異的に結合する抗体であり、CD3分子はTCRと結合して抗原認識信号を細胞内に伝える役割を担当する。
サイトカインは、成長因子であり、インターロイキン類から選択される一つ以上のものが望ましいが、これに限定されるものではない。
本発明の薬学組成物は、薬学的に許容可能な担体、希釈剤または賦形剤を含み、退行性神経疾患の予防及び治療用の医薬製剤に製剤化することができる。
実施例.ナチュラルキラー細胞の分泌物の取得
1.ナチュラルキラー細胞の分泌物の取得
1−1.CD3(−)PBMC播種細胞の用意
健康ドナーから採取したPBMCにPBS(phosphate buffered saline、LONZA、17−516Q)を1:1の割合で加え、1500rpm、4℃で10分間遠心分離した。PBMCペレットを10×106cells/mLでMACSランニングバッファ(running buffer、2%FBSと2mM EDTAを含むPBS)に懸濁し、自動細胞計数装置を用いて細胞数を測定した。
末梢血PBMC支持細胞は、PBS(phosphate buffered saline、LONZA、17‐516Q)を1:1の割合で加え、1500rpm、4℃で10分間遠心分離した。PBMCペレットを10×106cells/mLでMACSランニングバッファ(running buffer、2%FBSと2mM EDTAを含むPBS)に懸濁し、自動細胞計数装置を用いて細胞数を測定した。PBMC支持細胞は、凍結培地に懸濁し、1バイアル(vial)あたり1×108cellsを凍結して液体窒素に保管した。
ナチュラルキラー細胞の培養時、Cellgro SCGM培地に500IU/mLのIL−2(Proleukin Injection、韓国ノバルティス)、10ng/mLのOKT−3(eBioscience、16‐0037‐85)と1〜2%ドナー血漿を入れて培養培地を用意する。培養0日目にCD3(−)PBMC播種細胞とガンマ線照射された支持細胞とを1:5の割合で混ぜた後、総細胞数を基準に2×106cells/mLで細胞濃度を合わせて、37℃のインキュベーターで4〜5日間静置培養した。
ヒト繊維芽細胞におけるNK細胞培地とNK−CMの増殖能の差異を評価するために、セルカウンティングキット−8(CCK−8)を用いて細胞増殖能を測定した。
ヒト繊維芽細胞におけるNK−CMの細胞移動能及び傷の回復能を評価するために、セルスクラッチアッセイ(Cell scratch assay)方法を用いて測定した。
NK−CMのプロコラーゲン(procollagen)合成能を調べるために、毒性を示さない濃度2.5、5、10%で処理した後、24時間でプロコラーゲンの合成能を評価した。
細胞生存率の測定方法の1つであるセルカウンティングキット−8(CCK−8)を用いて、一般的な状況における角質形成細胞に対するNK−CMの効果を確認した。

RT−PCR(Real time−Polymerase Chain Reaction)を用いて、細胞内親炎症性サイトカインの分泌に関与するメッセンジャーRNA(messenger RNA、mRNA)の発現量を測定することにより、炎症誘発状況におけるNK−CMの役割を確認した。

Claims (14)
- ナチュラルキラー細胞の分泌物を含むアトピー性皮膚炎、脱毛、傷または皮膚しわ改善用化粧料組成物。
- 前記ナチュラルキラー細胞の分泌物は、IL−2、TIMP2、VEGF、PDGF−AAおよびIL−10からなる群より選択される少なくとも一つを含む、請求項1に記載の組成物。
- 前記ナチュラルキラー細胞の分泌物は、ナチュラルキラー細胞を培養し、それを分離精製し、残存する培養液から得られたものである、請求項1に記載の組成物。
- 前記培養は、播種細胞を支持細胞、支持細胞刺激因子および成長因子を含む培地中で培養することである、請求項3に記載の組成物。
- 前記播種細胞は、ナチュラルキラー細胞に分化できるヒト由来の細胞である、請求項4に記載の組成物。
- 前記ヒト由来の細胞は、末梢血、末梢血白血球細胞、PBMC(末梢血単核細胞)、濃縮された(enriched)ナチュラルキラー細胞、分離されたナチュラルキラー細胞、臍帯血、造血幹細胞および誘導多能性幹細胞からなる群より選択される少なくとも一つである、請求項5に記載の組成物。
- 前記成長因子は、インターロイキンである、請求項4に記載の組成物。
- ナチュラルキラー細胞の分泌物を含むアトピー性皮膚炎、脱毛、傷または皮膚しわ改善用薬学組成物。
- 前記ナチュラルキラー細胞の分泌物は、IL−2、TIMP2、VEGF、PDGF−AAおよびIL−10からなる群より選択される少なくとも一つを含む、請求項8に記載の組成物。
- 前記ナチュラルキラー細胞の分泌物は、ナチュラルキラー細胞を培養し、それを分離精製し、残存する培養液から得られたものである、請求項8に記載の組成物。
- 前記培養は、播種細胞を支持細胞、支持細胞刺激因子および成長因子を含む培地中で培養することである、請求項10に記載の組成物。
- 前記播種細胞は、ナチュラルキラー細胞に分化できるヒト由来の細胞である、請求項11に記載の組成物。
- 前記ヒト由来の細胞は、末梢血、末梢血白血球細胞、PBMC(末梢血単核細胞)、濃縮された(enriched)ナチュラルキラー細胞、分離されたナチュラルキラー細胞、臍帯血、造血幹細胞および誘導多能性幹細胞からなる群より選択される少なくとも一つである、請求項12に記載の組成物。
- 前記成長因子は、インターロイキンである、請求項11に記載の組成物。
