皮膚透過促進ペプチドを選別するためにファージライブラリと経皮製剤の溶出試験方法を組み合わせた、ファージディスプレイ法を実施した。
実施例2−1:神経伝達物質放出調節ペプチドに皮膚透過促進用ペプチドが結合された融合タンパク質の製造
上記実施例1で得られた配列番号1のアミノ酸配列を有する、皮膚透過促進用ペプチドと配列番号2のアミノ酸配列を有する神経伝達物質放出調節ペプチドが結合された配列番号32の融合タンパク質、配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドと配列番号3のアミノ酸配列を有する神経伝達物質放出調節ペプチドが結合された配列番号33の融合タンパク質、配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドと配列番号4のアミノ酸配列を有する神経伝達物質放出調節ペプチドが結合された配列番号34の融合タンパク質及び配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドとボツリヌストキシン(botulinum toxin)の神経伝達物質放出調節ペプチドが結合されたボツリヌストキシン+配列番号1の融合タンパク質のアミノ酸配列を含むペプチドを合成し、分離及び精製して神経伝達物質放出調節融合タンパク質を製造した。
上記神経伝達物質放出調節融合タンパク質をペプチド自動合成器(Applied Biosystems Model 431A)を用いて、固相(solid phase)ペプチド合成法で合成した。
上記実施例2−1−1で合成された神経伝達物質放出調節融合タンパク質を下記のような過程を経て分離及び精製した。
上記実施例1で得られた配列番号1のアミノ酸配列を有する、皮膚透過促進用ペプチドのC末端と配列番号35のアミノ酸配列を有する血小板由来成長因子aサブユニット(PDGFa)のN末端が2個のアミノ酸(GG)で構成されたリンカーを媒介として連結された形態の融合タンパク質であるT−PDGFa(配列番号36)を生産した。
上記実施例1で得られた配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドのC末端と配列番号38のアミノ酸配列を有する血管内皮細胞成長因子(VEGF)のN末端が2個のアミノ酸(GG)で構成されたリンカーを媒介として連結された形態の融合タンパク質であるT−VEGF(配列番号39)を生産した。
上記実施例1で得られた配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドのC末端と配列番号41のアミノ酸配列を有するインスリン様成長因子−1(IGF−1)のN末端が2個のアミノ酸(GG)で構成されたリンカーを媒介として連結された形態の融合タンパク質であるT−IGF−1(配列番号42)を生産した。
上記実施例1で得られた配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドのC末端と配列番号44のアミノ酸配列を有する角質細胞成長因子(KGF)のN末端が2個のアミノ酸(GG)で構成されたリンカーを媒介として連結された形態の融合タンパク質であるT−KGF(配列番号45)を生産した。
上記実施例1で得られた配列番号1のアミノ酸配列を有する皮膚透過促進用ペプチドのC末端と配列番号47のアミノ酸配列を有するチモシンベータ4(Tβ4)のN末端が2個のアミノ酸(GG)で構成されたリンカーを媒介として連結された形態の融合タンパク質であるT−Tβ4(配列番号48)を生産した。
実施例3−1:神経伝達物質放出調節融合タンパク質の効果検証
上記実施例2−1で製造した神経伝達物質放出調節融合ペプチドの皮膚改善用途を確認するために下記のように筋収縮抑制効果、皮膚透過性、皮膚残留性及びしわ改善効果を確認する実験を行った。
上記実施例2−1で製造した神経伝達物質放出調節融合タンパク質の筋収縮抑制効果を確認するためには、まずC2C12細胞をプレートに10%(v/v)のウシ胎児血清と1%(v/v)の抗生物質が入ったDMEM培地で培養した後、神経芽細胞を同じプレートに追加的に共同培養した。その後、C2C12細胞の細胞収縮が始まるとき、30秒間C2C12細胞の収縮回数を測定し、培地をすべて除去した後、PBSで3回洗浄した後、子牛血清が入っていない培地と、上記融合ペプチド50ppmを入れて2時間反応させた。その後、C2C12細胞の収縮回数を再び30秒間測定し、筋収縮抑制程度を確認した。
上記実施例2−1で製造した神経伝達物質放出調節融合タンパク質の皮膚透過性を確認するために、Franz glass cell(Standard直径9mm、Receiver 5ml、Permegear)を用いた。
上記実施例2−1で製造した神経伝達物質放出調節融合タンパク質の皮膚残留性を確認するために、Franz glass cell(Standard直径9mm、Receiver 5mL、Permegear)を用いた。
上記実施例2−1で製造した神経伝達物質放出調節融合タンパク質のしわ改善効果を確認するためには、一般的な水中油型乳化剤形(oil in water emulsion)のクリームにArgirelineTMと上記神経伝達物質放出調節融合タンパク質をそれぞれ含浸してしわ改善効果を比較し、それぞれの含有クリームに含まれる成分及び含量は、下記表3の通りである。
上記実施例2−2で製造したT−PDGFaの皮膚改善用途を確認するために下記のように皮膚弾力性の改善、皮膚しわの改善、脱毛の改善、皮膚透過性及び皮膚残留性の効果を確認する実験を行った。
上記実施例2−2で合成されたT−PDGFaをPDGFaと比較してコラーゲンの生成に及ぼす効果を検証した。
上記実施例2−2で合成されたT−PDGFaをPDGFaと比較してヒアルロン酸(HA)の生成に及ぼす効果を検証した。
上記実施例2−2で合成されたT−PDGFaをPDGFaと比較して皮膚細胞の増殖に及ぼす効果を検証した。
上記実施例2−2で合成された融合タンパク質のT−PDGFaをPDGFaと比較して皮膚透過性を検証した。
上記実施例2−2で合成された融合タンパク質のT−PDGFaをPDGFaと比較して皮膚残留性を検証した。
上記実施例2−3で製造したT−VEGFの皮膚改善用途を確認するために、下記のように皮膚弾力性の改善、皮膚しわの改善、脱毛の改善、皮膚透過性及び皮膚残留性の効果を確認する実験を行った。
上記実施例2−3で合成されたT−VEGFをVEGFと比較してヒアルロン酸の生成に及ぼす効果を検証した。
上記実施例2−3で合成されたT−VEGFをVEGFと比較して内皮細胞増殖に及ぼす効果を検証した。
上記実施例2−3で合成されたT−VEGFをVEGFと比較して皮膚細胞の増殖に及ぼす効果を検証した。
上記実施例2−3で合成された融合タンパク質のT−VEGFをVEGFと比較して皮膚透過性を検証した。
上記実施例2−3で合成された融合タンパク質のT−VEGFをVEGFと比較して皮膚残留性を検証した。
上記実施例2−4で製造したT−IGF−1の皮膚改善用途を確認するために下記のように皮膚弾力性の改善、皮膚しわの改善、脱毛の改善、皮膚透過性、皮膚残留性の効果を確認する実験を行った。
上記実施例2−4で合成されたT−IGF−1をIGF−1と比較して角質細胞(Keratinocyte)の成長に及ぼす効果を検証した。
上記実施例2−4で合成されたT−IGF−1をIGF−1と比較して皮膚細胞の増殖に及ぼす効果を検証した。
上記実施例2−4で合成された融合タンパク質のT−IGF−1をIGF−1と比較して皮膚透過性を検証した。
上記実施例2−4で合成された融合タンパク質のT−IGF−1をIGF−1と比較して皮膚残留性を検証した。
上記実施例2−5で製造したT−KGFの皮膚改善用途を確認するために下記のように皮膚弾力性の改善、皮膚しわの改善、脱毛の改善、皮膚透過性、皮膚残留性の効果を確認する実験を行った。
上記実施例2−5で合成されたT−KGFをKGFと比較して角質細胞の成長に及ぼす効果を検証した。
上記実施例2−5で合成されたT−KGFをKGFと比較して皮膚細胞の増殖に及ぼす効果を検証した。
上記実施例2−5で合成された融合タンパク質のT−KGFをKGFと比較して皮膚透過性を検証した。

上記実施例2−6で製造したT−Tβ4の皮膚改善用途を確認するために下記のように皮膚弾力性の改善、皮膚しわの改善、脱毛の改善、皮膚透過性及び皮膚残留性の効果を確認する実験を行った。
上記実施例2−6で合成されたT−Tβ4をTβ4と比較して内皮細胞増殖に及ぼす効果を検証した。
上記実施例2−6で合成されたT−Tβ4をTβ4と比較して皮膚細胞の増殖に及ぼす効果を検証した。
上記実施例2−6で合成された融合タンパク質のT−Tβ4をTβ4と比較して皮膚透過性を検証した。

Claims (16)
- 配列番号1のアミノ酸配列を含む皮膚透過促進用ペプチド及び生理活性タンパク質を含む融合タンパク質。
- 前記皮膚透過促進用ペプチドは、生理活性タンパク質のN末端に結合されたものである、請求項1に記載の融合タンパク質。
- 前記生理活性タンパク質は、神経伝達物質放出調節ペプチド、血小板由来成長因子aサブユニット(PDGFa)、血管内皮細胞成長因子(VEGF)、インスリン様成長因子−1(IGF−1)、角質細胞成長因子(KGF)及びチモシンベータ4(Tβ4)からなる群から選択されたいずれか一つである、請求項1に記載の融合タンパク質。
- 前記神経伝達物質放出調節ペプチドは配列番号2〜4からなる群から選択されたいずれか一つのアミノ酸配列を含み、前記血小板由来成長因子aサブユニット(PDGFa)は配列番号35のアミノ酸配列を含み、内皮細胞成長因子(VEGF)は配列番号38のアミノ酸配列を含み、インスリン様成長因子−1(IGF−1)は配列番号41のアミノ酸配列を含み、角質細胞成長因子(KGF)は配列番号44のアミノ酸配列を含み、前記チモシンベータ4(Tβ4)は配列番号47のアミノ酸配列を含むものである、請求項2に記載の融合タンパク質。
- 前記神経伝達物質は、ドーパミン、セロトニン、ヒスタミン、アセチルコリン、アドレナリン、ノルアドレナリン、ガンマアミノ酪酸、L−グルタミン酸、及びグリシンからなる群から選択されたいずれか一つ以上である、請求項4に記載の融合タンパク質。
- 前記融合タンパク質は、配列番号32〜34、36、39、42、45、及び48からなる群から選択されたいずれか一つのアミノ酸配列を含むものである、請求項1に記載の融合タンパク質。
- 前記融合タンパク質は、前記生理活性タンパク質の皮膚透過性及び皮膚残留性が増大したものである、請求項1に記載の融合タンパク質。
- 前記融合タンパク質は、前記皮膚透過促進用ペプチド及び生理活性タンパク質との間にリンカーを含むものである、請求項1に記載の融合タンパク質。
- 請求項1〜8のいずれか一項に記載の融合タンパク質をコードするポリヌクレオチド。
- 請求項1〜8のいずれか一項に記載の融合タンパク質を有効成分として含む皮膚改善用化粧料組成物。
- 前記皮膚の改善は、皮膚しわの改善または皮膚弾力性の増進である、請求項10に記載の皮膚改善用化粧料組成物。
- 請求項10に記載の化粧料組成物を有効成分として含む皮膚改善用機能性化粧品。
- 前記皮膚の改善は、皮膚しわの改善または皮膚弾力性の増進である、請求項12に記載の皮膚改善用機能性化粧品。
- 請求項1〜8のいずれか一項に記載の融合タンパク質を有効成分として含む脱毛改善用化粧料組成物。
- 請求項1〜8のいずれか一項に記載の融合タンパク質を有効成分として含む皮膚改善用医薬部外品組成物。
- 請求項1〜8のいずれか一項に記載の融合タンパク質を有効成分として含む脱毛改善用医薬部外品組成物。
