Abstract
Description
果糖をプシコースに転換する菌株を分離するために1%(w/v)プシコースが添加されたMineral salt broth(KH2PO4 2.4g/L、K2HPO4 5.6g/L、(NH4)2SO4 2.6g/L、MgSO47H2O 0.1g/L、yeast extract 1g/L)を使用した。
実施例1で分離された菌株を同定するために16SリボソームRNAの塩基配列を確認した。分離菌株の16SリボソームRNAの塩基配列(5’→3’)はそれぞれ配列番号1および2の塩基配列を有することを確認した。
前記で分離された菌株を、多様な温度条件下で菌体と基質を反応させてそれによるプシコース転換活性を比較した。
反応時間による最大生産性を確認した。前記実施例1で分離された2つの菌株の菌体濃度20mg/mL、果糖濃度400g/L、温度70℃およびpH7.0条件下で反応時間ごとに活性を確認した。前記反応は12時間行い、2時間間隔でプシコース生産性を、HPLC分析により確認した。その結果を下記表と図4および図5に示す。
Claims (16)
- 果糖からプシコースを生産するプシコース転換活性を有するマイクロバクテリウム属菌株を用いて果糖含有基質からプシコースを生産する方法であって、
前記マイクロバクテリウム属菌株は、マイクロバクテリウムオキシダンス(Microbacterium oxydans)およびマイクロバクテリウムフィロスファエラエ(Microbacterium phyllosphaerae)からなる群より選択された1種以上である、プシコースを生産する方法。 - 前記マイクロバクテリウム属菌株は、前記菌株の菌体、前記菌株の培養物、および前記菌株の破砕物からなる群より選択された1種以上である、請求項1に記載のプシコースを生産する方法。
- 前記マイクロバクテリウム属菌株を果糖含有基質と反応させる工程を含む、請求項1に記載のプシコースを生産する方法。
- 前記マイクロバクテリウム属菌株を果糖含有基質と反応させる工程は、前記マイクロバクテリウム属菌株の菌体、前記菌株の培養物、および前記菌株の破砕物からなる群より選択された1種以上を果糖含有基質と混合して行われる、請求項3に記載のプシコースを生産する方法。
- 前記マイクロバクテリウム属菌株を果糖含有基質と反応させる工程は、前記マイクロバクテリウム属菌株の菌体、前記菌株の培養物、および前記菌株の破砕物からなる群より選択された1種以上が固定化された担体に果糖含有基質を接触させる工程である、請求項3に記載のプシコースを生産する方法。

- 前記マイクロバクテリウムオキシダンス(Microbacterium oxydans)菌株は寄託番号KCCM12033Pを有する菌株であり、前記マイクロバクテリウムフィロスファエラエ(Microbacterium phyllosphaerae)菌株は寄託番号KCCM12034Pを有する菌株である、請求項1に記載のプシコースを生産する方法。
- 果糖からプシコースを生産するプシコース転換活性を有するマイクロバクテリウム属菌株を含む、果糖からプシコースを生産するプシコース生産用組成物であって、
前記マイクロバクテリウム属菌株は、マイクロバクテリウムオキシダンス(Microbacterium oxydans)およびマイクロバクテリウムフィロスファエラエ(Microbacterium phyllosphaerae)からなる群より選択された1種以上である、組成物。 - 前記マイクロバクテリウム属菌株は、前記菌株の菌体、前記菌株の培養物、および前記菌株の破砕物からなる群より選択された1種以上である、請求項11に記載のプシコース生産用組成物。
- 前記マイクロバクテリウム属菌株は、果糖と70℃の温度で18時間反応時、9〜30%の相対転換活性を有する、請求項11に記載の組成物。
- 前記マイクロバクテリウムオキシダンス(Microbacterium oxydans)菌株は寄託番号KCCM12033Pを有する菌株であり、前記マイクロバクテリウムフィロスファエラエ(Microbacterium phyllosphaerae)菌株は寄託番号KCCM12034Pを有する菌株である、請求項11に記載の組成物。
- 果糖をプシコースに転換する活性を有するマイクロバクテリウム属菌株であって、前記菌株はマイクロバクテリウムオキシダンス(Microbacterium oxydans)またはマイクロバクテリウムフィロスファエラエ(Microbacterium phyllosphaerae)である、菌株。
- 前記菌株は、寄託番号KCCM12033Pを有するマイクロバクテリウムオキシダンス菌株または寄託番号KCCM12034Pを有するマイクロバクテリウムフィロスファエラエ菌株である、請求項15に記載の菌株。
