技術分野
本開示は一般に、幹細胞および神経前駆細胞のin vitroにおける分化、障害および疾患を研究するための、in vitroにおける細胞ベースのモデル系の開発、ならびにニューロン細胞の方向付けられた分化を介してin vitroで発生させた細胞の投与による障害および疾患の処置に関する。より具体的に、本開示は、例えば、皮質介在ニューロン、視床下部ニューロン、および視索前野コリン作動性ニューロン、ならびにこれらの前駆体細胞を含む、ニューロン細胞およびニューロン細胞の前駆体細胞を発生させるためのin vitroにおける方法、およびこれらを含む組成物を提供する。このようなニューロン細胞は、ニューロン細胞を発生させるために幹細胞を含む多分化能、多能性、および/または全能性細胞を、SMADおよびWntシグナル伝達阻害剤および/またはアンタゴニストと接触させ、かつ、1つまたは複数のSHHシグナル伝達経路の活性化因子と、皮質介在ニューロン、視床下部ニューロン、および/または視索前野コリン作動性ニューロンのマーカー1つまたは複数を産生するニューロン細胞を発生させるのに要求される時点において、これに要求される持続期間にわたり接触させることにより発生させる。本明細書で開示されるin vitroにおける方法により発生させた、皮質介在ニューロン、視床下部ニューロン、および視索前野コリン作動性ニューロンは、神経変性障害および神経精神障害ならびに神経変性疾患および神経精神疾患を処置するのに治療的に使用することができ、このような神経変性障害および神経精神障害ならびに神経変性疾患および神経精神疾患を研究するためモデル系として使用することができ、かつ、このような障害および疾患を処置するための治療用候補化合物をスクリーニングおよび同定するために使用することができる。
神経学的障害の処置は従来、1つまたは複数の低分子のin vivoにおける投与を含んだ。これらの治療アプローチがin vivoにおいて神経学的障害に罹患した患者にもたらす有効性は、残念ながら、極めて微弱なものであった。
マウス皮質介在ニューロンは、Lhx6::GFPレポーターマウスESC系を使用して導出されているが、皮質介在ニューロン発生の効率は低かった(Maroofら、J Neurosci、30巻:4667〜4675頁(2010年))。ヒト皮質介在ニューロンの発生上の起源は、いまだ確立されておらず、介在ニューロン特化は、哺乳動物の種間で異なりうることが報告されている(Letinicら、Nature、417巻:645〜649頁(2002年);ならびにYuおよびZecevic、J Neurosci、31巻:2413〜2420頁(2011年))。さらに、長期間を要するin
vivoにおける皮質介在ニューロンの発生は、ヒト細胞による、in vitroにおける分化の達成に対して、難題を提示してきた。したがって、Maroofらにより記載された条件を、ヒトESCおよび/または他の多能性幹細胞からのヒト皮質介在ニューロンの発生に適用しうるのかどうかは、依然として不確定である。
vitroにおける方法であって、ニューロン細胞を、全能性細胞、多能性細胞、および/または多分化能細胞の分化により発生させ、(a)1つもしくは複数の全能性細胞、1つもしくは複数の多能性細胞、および/または1つもしくは複数の多分化能細胞を、(i)1つ、2つ、またはそれより多くのSMAD阻害剤、および(ii)1つまたは複数のWntシグナル伝達アンタゴニストと接触させ、これにより、ニューロン前駆体細胞の発生を誘導するステップと、(b)ニューロン前駆体細胞を、1つまたは複数のSHHシグナル伝達活性化因子と接触させ、これにより、ニューロン前駆体細胞内で、視床下部ニューロンおよび/または視床下部ニューロン前駆体細胞のマーカー1つまたは複数の産生を誘導するステップとを含むin vitroにおける方法を提供する。
a.多能性細胞、多分化能細胞、およびこれらの混合物からなる群から選択される、1つまたは複数の出発細胞を、少なくとも2つのSMAD阻害剤および少なくとも1つのWntシグナル伝達アンタゴニストと接触させて、該多能性細胞もしくは多分化能細胞またはこれらの混合物からの該前脳前駆体細胞の発生に助力するステップと、

vitroにおける方法であって、ニューロン細胞を、多能性細胞または多分化能細胞の分化により発生させ、(a)1つもしくは複数の多能性細胞および/または1つもしくは複数の多分化能細胞を、(i)2つまたはそれより多くのSMAD阻害剤、および(ii)1つまたは複数のWntシグナル伝達アンタゴニストと接触させ、これにより、ニューロン前駆体細胞の発生を誘導するステップと、(b)ニューロン前駆体細胞を、1つまたは複数のSHHシグナル伝達活性化因子と接触させ、これにより、ニューロン前駆体細胞内で、視床下部ニューロンおよび/または視床下部ニューロン前駆体細胞のマーカー1つまたは複数の産生を誘導するステップとを含む方法を提供する。
ある特定の実施形態では、本開示は、腹側前駆細胞を発生させるための方法であって、細胞を、ソニックヘッジホッグ(SHH)などの腹側化因子へと曝露するステップを含む方法を提供する。代替的に、細胞は、Shhを模倣する薬剤またはSHHのアゴニストに曝露される。

皮質介在ニューロン発生については、マウス胚MGE由来の前駆細胞を、主にマウス皮質錐体ニューロンおよびグリアから構成される「フィーダー培養物」上に播種した共培養系を使用して研究されている(Xuら、J Neurosci、24巻:2612〜2622頁(2004年))。ヒト介在ニューロンの成熟の態様が、この系では、異種移植片と比べて加速されるのかどうかを決定するために、NKX2.1::GFP+細胞を、分化32日目において、FACSにより回収し、解離させたマウス胚皮質の培養物上に再播種した(図7A)。皮質細胞は、腹側由来の皮質介在ニューロンの背側新皮質への遊走の前の段階である、胚生13.5日目のマウス胚から単離した(Andersonら、Science、278巻:474〜476頁(1997年))。

さらなる実施形態では、本開示は、皮質介在ニューロン細胞および/または皮質介在ニューロン前駆体細胞のマーカー1つまたは複数を産生する1つまたは複数のin vitroで分化させたニューロン細胞を含む組成物であって、in vitroで分化させたニューロン細胞が、(a)多分化能細胞または多能性細胞を、2つまたはそれより多くのSMADシグナル伝達阻害剤と接触させるステップ、(b)多分化能細胞または多能性細胞を、1つまたは複数の阻害剤と接触させるステップ、(c)多分化能細胞または多能性細胞を、1つまたは複数のSHHシグWntシグナル伝達ナル伝達活性化因子と接触させるステップにより産生されている、組成物を提供する。
in vitroにおける、ニューロン細胞の、幹細胞または前駆細胞からの導出は、著明な臨床的含意を有し、また、疾患のモデル化および薬物スクリーニングのためにも重要である。本発明のこの方法に従う処置のために適切な状態として、てんかんまたは乳児けいれんなどの発作障害;自閉症、統合失調症、不安障害、および摂食障害などの神経精神障害;全前脳症または小頭症などの神経発達障害;ならびにパーキンソン病が挙げられるが、これらに限定されない。hPSCを使用した初期研究は、パーキンソン病における中脳ドーパミンニューロン(PD;Kriksら、Nature、480巻:547〜551頁(2011年);Soldnerら、Cell、136巻:964〜977頁(2009年);およびSoldnerら、Cell、146巻:318〜331頁(2011年))、または筋萎縮性側索硬化症(ALS;Dimosら、Science、321巻:1218〜1221頁(2008年))および脊髄筋萎縮症(SMA;Ebertら、Nature、457巻:277〜280頁(2009年))における運動ニューロンなど、特定のニューロン型に影響を及ぼすことが公知の神経変性障害に主に向けたものであった。より最近の研究では、統合失調症(Brennandら、Nature、473巻:221〜225頁(2011年))または自閉症関連症候群(Marchettoら、Cell、143巻:527〜539頁(2010年);およびPascaら、Nat

ある特定の実施形態では、本開示は、例えば、パーキンソン病(PD)およびアルツハイマー病(AD)を含む神経変性と関連する障害および疾患を処置するための方法であって、本明細書で開示される方法により発生させた皮質介在ニューロンを、PDまたはADに罹患した患者へin vivoにおいて投与するステップを含む、方法を提供する。
他の実施形態では、本開示は、例えば、統合失調症および自閉症関連障害を含む、精神障害および疾患を処置するための方法を提供する。

10〜18プロトコールに由来するMGE様細胞(図4P)とは著しく対照的に、SHH 2〜18プロトコール(図4O)も、SHH 6〜18プロトコール(図示しない)も、移植部位からの広範な遊走を結果としてもたらさなかった。移植の4週間後において、本発明者らは、マウス皮質内の放射面および接平面の両方において、広範な遊走を観察したが、10〜18日目群(図4Oおよび4P)に限られた。

USA、106巻:15472〜15477頁(2009年))、パーキンソン病の態様を処置すること(Martinez−Cerdenoら、Cell Stem Cell、6巻:238〜250頁(2010年))、および生後脳内で学習および可塑性を誘導すること(Southwellら、Science、327巻:1145〜1148頁(2010年))における、皮質介在ニューロン移植片の使用の潜在的可能性を踏まえると、疾患についての複数の成体CNSモデルへの移植研究は、特に興味深い。このような移植研究についての1つの具体的な難題は、移植されたhPSC由来の皮質介在ニューロン前駆体細胞のin vivoにおける長期間を要する成熟である。長期にわたる予備的な、成体マウス皮質への移植研究により、それらの持続的な緩徐な成熟速度が確認され、NKX2.1+推定介在ニューロン前駆体細胞は、移植の3カ月後において、未成熟な成長円錐を保持し、限定的な統合を示す(データは示さない)。これらのデータは、迅速な機能的な統合および表現型の成熟がたやすく達成された、本発明者らのin vitroにおける共培養物による研究と対照的である。まとめると、本研究は、発達の多様な態様を研究するのに使用される可能性があり、多種多様な神経精神疾患に関与している特定のニューロン型の機能不全について研究するための強力なin vitroプラットフォームとして用いることができる、異なる腹側前脳ニューロン型の発生についての枠組みを提供する。
