この遺伝子は、MAGEA遺伝子ファミリーのメンバーである。このファミリーのメンバーは、互いに50?80%の配列同一性を有するタンパク質をコードする。MAGEA遺伝子のプロモーターおよび第1のエクソンは、かなりの変動性を示し、この遺伝子ファミリーの存在が、異なる転写制御下で同じ機能を発現できるようにすることが示唆される。MAGEA遺伝子は、染色体位置Xq28でクラスター化する。それらは、先天性角化異常症などのいくつかの遺伝性疾患に関与するとされている。同一タンパク質をコードする少なくとも4つの変異型が、この遺伝子について判明している。(RefSeq,Jul 2008によって提供される)。
第I相臨床試験では、食道がん患者において、HLA−A*24:02に結合したMAGEA4(143−151)に対して反応性のTCR操作自己CTLの養子免疫伝達が調査された。患者には、予備調節治療なしでTCR形質導入リンパ球が1回投与され、それに続いて2週間および4週間後に、MAGEA4ペプチドで皮下免疫化された。おそらくは、リンパ枯渇レジメンおよびIL2の投与の欠如のために、客観的な腫瘍退縮は観察されなかった(Kageyama et al.,2015)。マウスにおける前臨床試験は、移入されたT細胞が、マウスに接種されたMAGEA4発現腫瘍細胞株の増殖を阻害し、さらなるペプチドワクチン接種がこの抗腫瘍活性を増強することを実証した(Shirakura et al.,2012)。
健常細胞と比較した腫瘍細胞上のTAAの過剰提示または特異的提示は、免疫療法におけるその有用性にとって十分であり、いくつかのペプチドは、それらの起源タンパク質が健常組織にもまた存在するにもかかわらず、腫瘍特異的である。それでもなお、mRNA発現プロファイリングは、免疫療法のためのペプチド標的の選択において、安全性のレベルを高めることができる。特に、アフィニティ成熟TCRなどの安全性リスクが高い治療選択肢では、理想的な標的ペプチドは、腫瘍に特有で健常組織上には見いだされないタンパク質に由来する。
外科的に除去された組織標本は、告知に基づく同意書が各患者から入手された後に、上述の通り提供された。腫瘍組織標本が手術直後にスナップ凍結され、その後、液体窒素下で乳鉢と乳棒を用いて均質化された。TRI試薬(独国ダルムシュタットのAmbion)を使用して、これらのサンプルから全RNAが調製され、RNeasy(独国ヒルデンのQIAGEN)による精製がそれに続き;どちらの方法も製造業者のプロトコルに従って実施された。
腫瘍および健常組織RNAサンプルの遺伝子発現解析は、CeGaT(独国チュービンゲン)によって、次世代配列決定(RNAseq)によって実施された。配列決定ライブラリーは、RNA断片化、cDNA転換、および配列決定アダプターの付加を含む、Illumina HiSeq v4試薬キットを使用して、販売業者(米国カリフォルニア州サンディエゴのIllumina Inc.)のプロトコルに従って作成される。複数のサンプルに由来するライブラリーは等モル混合され、Illumina HiSeq 2500配列決定装置上で、製造会社の使用説明書に従って配列決定され、50bpのシングルエンドリードが生成される。処理された読み取りは、STARソフトウェアを使用して、ヒトゲノム(GRCh38)にマッピングされる。発現データは、ensembl配列データベース(Ensembl77)の注釈に基づいて、RPKM(100万個のマッピングされた読み取り当たりキロベース当たり読み取り、ソフトウェアCufflinksによって作成される)として転写物レベルで、そしてエクソンレベルで(全読み取り、ソフトウェアBedtoolsによって作成される)提供される。エクソン読み取りは、エクソン長さおよびアライメントサイズについて正規化されて、RPKM値が得られる。

告知に基づく同意を得た後、HLA−A*02陽性およびHLA−A*02陰性の健常ドナーからのPBMCを使用した。組換えビオチン化HLA−A2クラスIモノマーと、MAG−003を含有するA2蛍光性四量体をMBLI(マサチューセッツ州ウーバン)から得た。リン酸緩衝食塩水(PBS)で希釈した抗CD20SAと共に、PBMCを室温で1時間培養し、洗浄して、ビオチン化A2/MAG−003モノマーと共に室温で30分間培養し、洗浄して、24ウェルプレート内の10%ヒトAB血清添加RPMIに、3×106細胞/ウェルで播種した。インターロイキン7(IL−7;R&D systems、ミネソタ州ミネアポリス)を1日目に10ng/mLで添加し、IL−2(英国ヘアフィールドのChiron)を4日目に10U/mLで添加した。5週間にわたり、細胞を新鮮なPBMCで毎週再刺激し、応答性細胞と1:1の比で混合して、24ウェルプレートに3×106/ウェルで播種した。
MAG−003を使用して、そのT細胞がマウスTCR欠損を補う多様なヒトTCRレパートリーを発現する、全ヒトTCRαβ遺伝子座(1.1および0.7Mb)について遺伝子組換えであるマウスを免疫化する(Li et al.2010)。高親和性T細胞を得るために、遺伝子組換えマウスから得られたPBMCを四量体フィコエリトリン(PE)と共に培養し、上記のような細胞選別がそれに続く。次に、当該技術分野で記載された方法を使用して、アミノ酸/DNA配列決定および発現ベクターへのクローニングのために、このようにして得られた高結合活性T細胞を使用してTCRを同定し、単離する。

T細胞は、高結合活性TCR(いわゆるTCR療法)、またはMHCI/MAG−003複合体またはMHCII/MAG−003複合体に対する抗原特異性が増強されたタンパク質融合由来キメラ抗原受容体(CAR)を発現するように、遺伝子操作し得る。一態様では、このアプローチは、中枢および末梢寛容に関連するいくつかの制限を克服し、患者における新生T細胞活性化の必要なしに、腫瘍の標的化においてより効率的なT細胞を生じる。

MAG−003/MHC複合体に対するTCRの結合モチーフを判定するために、MAG−003ペプチドの9個のアミノ酸のそれぞれでポジショナルアラニンスキャニング分析を実施した。アラニン置換MAG−003ペプチドは、表12に示される。
病原体に対する防御の第一線に関与する非従来的なTリンパ球エフェクターであるガンマデルタ(γδ)Τ細胞は、受容体、特に、TCR−γおよびTCR−δ鎖を活性化することで、MHC非依存的様式で腫瘍細胞と相互作用して、腫瘍細胞を根絶し得る。これらのγδT細胞は、活性化に際して、迅速なサイトカイン産生(IFN−γ、TNF−α)および強力な細胞毒性応答細胞傷害性応を可能にする前活性化表現型を示す。これらのT細胞は多くのがんに対して抗腫瘍活性を有し、γδT細胞媒介免疫療法が実行可能であり、客観的な腫瘍応答を誘導し得ることが示唆される。(Braza et al.2013)。
それぞれ腫瘍特異的TCR−αおよびTCR−β鎖をコードする3つのMAG−003特異的TCR(R7P1D5、R10P2G12、R20P1H7、表2を参照されたい)を健常ドナーのT細胞から単離し増幅した。健常ドナーからの細胞を、以前記載された方法(Walter et al.,2003 J Immunol.,Nov 15;171(10):4974−8)に従って生体外刺激し、HLA−A*02多量体を用いて標的特異的細胞を単細胞ソートし、次に引き続くTCR単離のために使用した。例えば、Molecular Cloning a laboratory manual fourth edition by Green and Sambrookに記載されるように、標準法によって5’RACEを通じてTCR配列を単離した。TCR R7P1D5、R10P2G12、およびR20P1H7のαおよびβ可変領域を配列決定し、さらなる機能特性解析のためにクローン化した。TCR R7P1D5、R10P2G12 およびR20P1H7は、HLA−A*02陽性ドナーに由来する。

結果:
TCR R7P1D5、R10P2G12またはR20P1H7を発現するCD8+T細胞では、健常組織由来のHLA−A*02陽性細胞型との共培養で時に活性化は観察されなかった(図13を参照されたい)一方で、MAG−003ペプチドの供給源遺伝子としてHLA−A*02およびMAGEA4を発現する腫瘍細胞株A−375およびNCI−H1755に対する活性があった。同様の反応性パターンがNYESO1特異的TCR 1G4を発現するCD8+T細胞で観察され、これはNYESO1発現HLA−A*02陽性腫瘍細胞株に対して反応性であるが、健常組織細胞の示されたパネルには反応しないことが判明した。
T細胞産物は、小規模なACTengine製造プロセスに続いて、健常ドナーから単離されたバルク全末梢血単核細胞(PBMC)から出発して生成された。ACTengineT細胞を製造するための本発明のR7P1D5 TCRを発現するレンチウイルスベクター骨格は、以前の研究および複数の臨床試験に基づいてデザインされた(Porter et al.,2011;Kalos et al.,2011;その他)。簡潔に述べると、解凍し休ませたPBMCをCD3/CD28抗体で活性化し、α鎖およびβ鎖の異なる配向を有するR7P1D5TCR導入遺伝子と、プロモーターおよびエンハンサー配列の様々な組み合わせとを保有する種々のレンチウイルスベクターを用いて生成された濃縮ウイルス上清で、形質導入した(コンストラクトはR71−R78と表記される)。8つのコンストラクト(R74、R75)のうち2つは、低い力価および/または低い生産性のために排除された。残りの6つのウイルス上清で形質導入された細胞を増殖させ、フローサイトメトリーによって導入遺伝子発現について評価した。

Claims (38)
- 配列番号44、52、60、68、76、および84から選択されるアミノ酸配列と少なくとも80%の配列同一性を有する、少なくとも1つの相補性決定領域(CDR)3を含んでなる抗原認識コンストラクト。
- 前記抗原認識コンストラクトが、本発明の抗原ペプチドのTAAに特異的におよび/または選択的に結合できる、請求項1に記載の抗原認識コンストラクト。
- 前記抗原認識コンストラクトが、抗体、またはその誘導体もしくは断片、またはT細胞受容体(TCR)、またはその誘導体もしくは断片である、請求項1または2に記載の抗原認識コンストラクト。
- 前記抗原認識コンストラクトがTAA抗原ペプチドを提示するヒト白血球抗原(HLA)に結合し、前記HLAが任意選択的にA*02型である、請求項1〜3のいずれか一項に記載の抗原認識コンストラクト。。
- 前記コンストラクトが、配列番号1〜25、好ましくは配列番号1から選択されるアミノ酸配列を有するエピトープに、特異的におよび/または選択的に結合する、請求項1〜4のいずれか一項に記載の抗原認識コンストラクト。
- 前記コンストラクトがα/β−TCRまたはその断片もしくは誘導体であり、または前記コンストラクトがγ/δ−TCRまたはその断片もしくは誘導体である、請求項1〜5のいずれか一項に記載の抗原認識コンストラクト。
- 前記コンストラクトがヒト起源であり、TAA抗原ペプチドを特異的におよび/または選択的に認識することを特徴とする、請求項1〜6のいずれか一項に記載の抗原認識コンストラクト。
- 前記抗原認識コンストラクトが、対象において免疫応答を誘導でき、任意選択的に、前記免疫応答がインターフェロン(IFN)γレベルの増加によって特徴付けられる、請求項1〜7のいずれか一項に記載の抗原認識コンストラクト。
- 前記TCRαまたはγ鎖が配列番号44、60、および76から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するCDR3を含んでなり、および/または前記TCRβまたはδ鎖が配列番号52、68、および84から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するCDR3を含んでなる、TCRα鎖またはγ鎖および/またはTCRβまたはδ鎖を含んでなる、請求項1〜8のいずれか一項に記載の抗原認識コンストラクト。
- 前記TCRαまたはγ鎖が、配列番号42、58、および74から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するCDR1;および/または配列番号43、59、および75から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するCDR2をさらに含んでなる、請求項9に記載の抗原認識コンストラクト。
- 前記TCRβまたはδ鎖が、配列番号50、66、および82から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するCDR1;および/または配列番号51、67、および83から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するCDR2をさらに含んでなる、請求項9または10に記載の抗原認識コンストラクト。
- 配列番号41、49、57、65、73、および81から選択されるアミノ酸配列と少なくとも80%の配列同一性を有するTCR可変鎖領域を含んでなる、請求項1〜11のいずれか一項に記載の抗原認識コンストラクト。
- 前記コンストラクトが、ヒト化、キメラ化および/またはマウス化されている、請求項1〜12のいずれか一項に記載の抗原認識コンストラクト。
- TCR結合断片を含んでなり、前記結合断片が、任意選択的に、配列番号42、43、44;または50、51、52;または58、59、60;または66、67、68;または74、75、76;または82、83、84のアミノ酸配列を有するCDR1〜CDR3配列から選択されるCDR1〜CDR3を含んでなる、請求項1〜13のいずれか一項に記載の抗原認識コンストラクト。
- 前記コンストラクトが、少なくとも1つのTCRα鎖および1つのTCRβ鎖配列から構成されるTCRまたはその断片であり、前記TCRα鎖配列が、配列番号42〜44のアミノ酸配列を有するCDR1〜CDR3配列を含んでなり、前記TCRβ鎖配列が、配列番号50〜52のアミノ酸配列を有するCDR1〜CDR3配列を含んでなり;または前記TCRα鎖配列が、配列番号58〜60のアミノ酸配列を有するCDR1〜CDR3配列を含んでなり、前記TCRβ鎖配列が、配列番号66〜68のアミノ酸配列を有するCDR1〜CDR3配列を含んでなり;または前記TCRα鎖配列が、配列番号74〜76のアミノ酸配列を有するCDR1〜CDR3配列を含んでなり、前記TCRβ鎖配列が、配列番号82〜84のアミノ酸配列を有するCDR1〜CDR3配列を含んでなる、請求項1〜14のいずれか一項に記載の抗原認識コンストラクト。
- 前記コンストラクトが、少なくとも1つのTCRα鎖および1つのTCRβ鎖配列からなるTCRまたはその断片であり、前記TCRα鎖配列が、配列番号41のアミノ酸配列を有する可変領域配列を含んでなり、前記TCRβ鎖配列が、配列番号49のアミノ酸配列を有する可変領域配列を含んでなり、または前記TCRα鎖配列が、配列番号57のアミノ酸配列を有する可変領域配列を含んでなり、前記TCRβ鎖配列が、配列番号65のアミノ酸配列を有する可変領域配列を含んでなり、または前記TCRα鎖配列が、配列番号73のアミノ酸配列を有する可変領域配列を含んでなり、前記TCRβ鎖配列が、配列番号81のアミノ酸配列を有する可変領域配列を含んでなる、請求項1〜15のいずれか一項に記載の抗原認識コンストラクト。
- 前記コンストラクトが、配列番号46、54、62、70、78、および86から選択されるアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有するTCR定常領域をさらに含んでなる、TCRまたはその断片であり;好ましくは前記TCRが、少なくとも、1つのTCRαおよび1つのTCRβ鎖配列から構成され、前記TCRα鎖配列が、配列番号46、62、および78から選択されるアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する定常領域を含んでなり;前記TCRβ鎖配列が、配列番号54、70、および86から選択されるアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する定常領域を含んでなる、請求項1〜16のいずれか一項に記載の抗原認識コンストラクト。
- 配列番号39のアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する第1のTCR鎖、配列番号47のアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する第2のTCR鎖を含んでなる、請求項1〜17のいずれか一項に記載の抗原認識コンストラクト。
- 配列番号55のアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する第1のTCR鎖、配列番号63のアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する第2のTCR鎖を含んでなる、請求項1〜17のいずれか一項に記載の抗原認識コンストラクト。
- 配列番号71のアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する第1のTCR鎖、配列番号79のアミノ酸配列と少なくとも50%、60%、70%、80%、90%、95%、98%、99%、または100%の配列同一性を有する第2のTCR鎖を含んでなる、請求項1〜17のいずれか一項に記載の抗原認識コンストラクト。
- 請求項1〜20のいずれか一項に記載の抗原認識コンストラクトをコードする核酸。
- 請求項21に記載の核酸を含んでなるベクター。
- 請求項1〜20のいずれか一項に記載の抗原認識コンストラクト、または請求項21に記載の核酸、または請求項22に記載のベクターを含んでなる宿主細胞。
- 前記宿主細胞が、リンパ球、好ましくはTリンパ球またはTリンパ球前駆体、より好ましくはCD4またはCD8陽性T細胞である、請求項23に記載の宿主細胞。
- 請求項1〜20のいずれか一項に記載の抗原認識コンストラクト、または請求項21に記載の核酸、または請求項22に記載のベクター、または請求項23または24に記載の宿主細胞、および薬学的に許容可能な担体、安定剤および/または賦形剤を含んでなる医薬組成物。
- 医療で使用するための請求項1〜20のいずれか一項に記載の抗原認識コンストラクト、または請求項21に記載の核酸、または請求項22に記載のベクター、または請求項23または24に記載の宿主細胞、または請求項25に記載の医薬品組成物。
- 悪性または良性の腫瘍疾患を含んでなる増殖性疾患の診断、予防および/または治療における使用のための、請求項26に記載の使用のための抗原認識コンストラクト、または核酸、またはベクター、または宿主細胞、または医薬組成物。
- 前記腫瘍疾患が、前記腫瘍疾患の腫瘍細胞内のTAAの発現によって特徴付けられる、請求項27に記載の使用のための抗原認識コンストラクト、または核酸、またはベクター、または宿主細胞、または医薬組成物。
- 医学における使用が、任意選択的に養子細胞移入を含んでなる免疫療法における使用であり、前記免疫療法が、養子自己または異種T細胞療法を含んでなる、請求項26〜28のいずれか一項に記載の使用のための抗原認識コンストラクト、または核酸、またはベクター、または宿主細胞、または医薬組成物。
- a.適切な宿主細胞を提供するステップと、
b.請求項1〜20のいずれか一項に記載の抗原認識コンストラクトをコードするコード配列を含んでなる遺伝子コンストラクトを提供するステップと、
c.前記適切な宿主細胞に前記遺伝子コンストラクトを導入するステップと、
d.前記適切な宿主細胞によって前記遺伝子コンストラクトを発現させるステップと
を含んでなる、細胞株を発現するTAA特異的抗原認識コンストラクトを製造する方法。 - 前記抗原認識コンストラクトの細胞表面提示をさらに含んでなる、請求項30に記載の方法。
- 前記遺伝子コンストラクトが、前記コード配列と作動可能に連結されたプロモーター配列を含んでなる発現コンストラクトである、請求項30または31に記載の方法。
- 前記抗原認識コンストラクトが哺乳類起源、好ましくはヒト起源である、請求項30〜32のいずれか一項に記載の方法。
- 前記適切な宿主細胞が、任意選択的にヒト細胞またはヒトTリンパ球から選択される哺乳類細胞である、請求項30〜33のいずれか一項に記載の方法。
- 前記抗原認識コンストラクトが修飾TCRであり、前記修飾が、標識を含んでなる機能ドメインの付加、または膜アンカードメインを含んでなる代替ドメインの付加を含んでなる、請求項30〜34のいずれかに記載の方法。
- 前記抗原認識コンストラクトがα/βTCR、γ/δTCR、または一本鎖TCR(scTCR)である、請求項35に記載の方法。
- 前記遺伝子コンストラクトが、レトロウイルス形質移入によって前記適切な宿主細胞に導入される、請求項30〜36のいずれか一項に記載の方法。
- 前記適切な宿主細胞からの前記抗原認識コンストラクトの単離および精製と、任意選択的に、T細胞内の前記抗原認識コンストラクトの再構成とをさらに含んでなる、請求項30〜37のいずれか一項に記載の方法。
