Abstract
Description
本発明で使用するコオロギはフタホシコオロギ(Gryllus bimaculatus)であって、フタホシコオロギを熱風乾燥するか、または凍結乾燥後に粉碎機(ball millまたはhammer mill)を用いて粉砕(100〜300メッシュ)した。凍結乾燥粉砕することがビタミンなどの栄養素の破壊を防ぐことができるので好ましい。本実験で使われたコオロギ粉末は(株)239BIOのコオロギ粉末を使用したものであり、その商標名であるD&Dと表記した。
蕎麦粉末、タマネギ粉末、カカオニップス粉末を各々50:25:25の重量比で混合して植物性混合粉末(mixture)を供給する。前記植物性混合粉末は凍結乾燥粉砕を用いて粉末で供給することがビタミンなどの栄養素の破壊を防ぐことができるので好ましい。
前記実施例1を通じて得たコオロギ粉末と、前記実施例2を通じて混合した植物性混合粉末を各々1:1の重量比で混合したものを使用する。
実施例1で製造したコオロギ粉末と関連して説明すると、9週齢の実験動物(ネズミ)を12時間絶食させた後、膵臓ベータ細胞のみに特異的に作用して他の器官に影響を及ぼさないと知られたStreptozotocin(STZ、S0130、Sigma−Aldrich、USA)を0.1 M citrate buffer(ph4.5)に溶解させて65mg/kg濃度でIPして糖尿を誘発した。
体重増加量及び食餌摂取量は実験開始日を始めて十分な量の飼料と水を給与しながら一週間間隔で測定した。食餌摂取量は給与量から残量を減らして計算した。食餌効率(Food efficiency ration、FER)は実験期間の間体重増加量を同じ期間の食餌摂取量で割って換算した。
1)C−peptide
1−1)分析概要
C−peptideはproinsulinの分解過程で生じる物質であって、インスリンの分泌程度を反映した。
1−2)実験方法
測定のために採血血液から血漿を分離した後、C−peptide ELISA kitを用いて分析し、その分析方法はkit会社のマニュアルに従って測定した。
2−1)分析概要
糖化血色素は長期間に亘る血糖の変化を測定するバイオマーカーである。
2−2)実験方法
測定のために採血血液で分析し、病院の臨床病理実験室を用いて分析したものであり、分析方法はkit会社のマニュアルに従って測定した。
3−1)実験方法
測定のために採血血液から血漿を分離した後、Insulin ELISA kitを用いて分析した。
血糖は自由に水を摂取するようにしながら12時間絶食した後、毎週同一の時間帯に尻尾静脈血から血液を採取して血糖計で測定した。
コオロギ粉末(D&D)及び/又は植物性混合粉末(mixture)の持続的な投与によって肝や腎臓に如何なる損傷をもたらすことがあるという可能性、即ち毒性実験を進行した。図7及び図8は、本願発明が糖尿疾患を改善する過程で発生することがある肝または腎臓への影響を調べたものである。この写真から見る時、本願発明が肝や腎臓の組織に留意的な変化がないと観察されるように、特別に悪影響を及ぼすことと見なされない。
1)膵臓組織染色
白ネズミの膵臓を摘出して3.7ホルマリン溶液に固定した後、自動組織過程処理器を用いて脱水した。パラフィン包埋器を用いてパラフィンブロックを製作し、これを組織切片機(mircotome)で薄切して4μm厚さの切片を製作し、その製作された切片をまたxyleneとalcoholを用いて脱パラフィン及び脱水過程を経た後、 hematoxylin & eosin(H&E)染色し含水させた後、光学顕微鏡で観察した。
糖尿誘発実験ネズミでコオロギ粉末(D&D)、植物性混合粉末(mixture)、そしてコオロギ粉末と植物性混合粉末の混合物(D&D+mixture)の投与によるブドウ糖耐性効果を確認するために、実験4週経過後、耐糖能とインスリン耐性を測定した。
Claims (3)
- 蕎麦粉末、タマネギ粉末、カカオニップス粉末を含有する植物性混合粉末(mixture)及び/又はコオロギ(Gryllusbimacalatus)粉末を含有する、糖尿疾患の治療用組成物。
- 前記植物性混合粉末(mixture)は蕎麦粉末30〜50%、タマネギ粉末25〜40%、カカオニップス粉末10〜25%の重量比で混合して形成する、請求項1に記載の糖尿疾患の治療用組成物。
- 前記植物性混合粉末(mixture)とコオロギ粉末は各々1:1の重量比で混合した、請求項2に記載の糖尿疾患の治療用組成物。
