Description
本特許文書は、参照により本明細書に組み込まれる、2015年5月5日に出願した米国仮特許出願第62/157,286号の合衆国法典第35巻第119条(e)による出願日の利益を主張するものである。
本発明は、米国科学財団EAGERにより授与された契約第CMMI-1548261号の下で政府の支援により行われた。
改変された小さく、高度に安定なタンパク質であるアフィマー(affimers)を含む。
変色性インク
図6Aについて述べると、印加せん断応力が降伏応力(τy)を超える場合に(例えば、印刷中に)強いずり減粘応答、並びに印加せん断応力がτyを下回る場合に(例えば、印刷後に)せん断粘性係数(G'')を超えるプラトーせん断弾性係数(G')を示す、高度に濃縮された(40重量%)Pluronic F127Aを例示システム用の変色性インクとして選択する。変色性インクの弾性は、図6Bに示すように22℃で約2×104 Paであることが見い出される。CMT(約4℃)未満では、インクが液化し、その弾性が数桁低下し、それにより組織構築物からのその除去が容易になる。
上述のように、天然ECMを模倣し、組織構築物の細胞外マトリックス組成物及び/又は材料に用いることができるゼラチン及びフィブリンに基づく相互貫入ポリマー網状構造が開発された。
Pluronic F127-GelMA系について上述した相補的な熱的挙動を利用して、複数の犠牲フィラメントを含む代表的な血管パターンを印刷し、次いでこれを無細胞性細胞外マトリックス組成物(純GelMA)に封入する。図7A〜7Kに各血管網設計の概略図及び対応する光学像により1D、2D及び3D血管網の形成並びに通路の内皮化を示す。変色性インクを除去した後、各血管網を視覚化の助けとするために蛍光赤色染料で潅流する(図7C、7F及び7I)。各組織構築物内で、犠牲フィラメントの直径は、印刷圧力、速度及び/又はノズル高を変更することによって所望の通りに変化させることができる。例えば、45μmから500μmに漸増する直径を有する1D微小通路アレイは、各印刷通路(feature)の間で印刷圧力及びノズル高を段階的に単に増加させることにより単一30μmノズルを用いて印刷する(図7A〜7B)。
多種類の流体を埋め込み血管網に流して、それらの潅流可能な性質を実証することができる。例えば、2D階層的分岐網状構造にヒト臍静脈内皮細胞(HUVEC)懸濁液を注射した後、緩やかに揺り動かす。しに成功を収めることができることが示された(図7J参照)。48時間後に、代表的な分岐微小通路内の共焦点撮像と組み合わせた生/死染色により判定したとき、細胞が95%を超える生存率を保持し、ほぼ融合層に集合していることが見いだされる。
単一インクフィラメント内の2つ以上の細胞型の送達を可能にする細胞共培養インクの印刷を実証する。図20A〜20Cにおいて明らかなように、この例では、ゼラチン-フィブリンマトリックス材料中HNDF及びHUVECの分散体を含む細胞負荷インクが、印刷フィラメントにおける自発的新生血管系の形成をもたらすことが観察される。これは、HUVECの単一培養に基づく印刷フィラメントにおいては認められない。このアプローチは、組織特異的細胞型(例えば、肝細胞、膵島、足細胞、ニューロン等)又は幹細胞(例えば、iPSC、MSCs等)を含む1つ以上の細胞型を含む細胞負荷フィラメントを用いて、所望の不均質性を達成すること、且つ機能を増強することもできることを示唆するものである。
先の例と同様に、HNDF及びHUVECを細胞外マトリックス組成物(具体的には、ゼラチン-フィブリンマトリックス材料)内に分散させて、細胞負荷マトリックスを形成する。変色性インクを細胞負荷マトリックス上に直接印刷し、次いでゼラチン-フィブリンマトリックス材料により封入する。変色性インクを排出して、血管通路を形成し、血管通路にHUVECを播種する。時間の経過につれて、HUVECが印刷細胞負荷フィラメント内の毛細管構造中に集合する。図19A〜19Dに内皮細胞が血管通路に付着した状態になり、融合層を形成することを示し、図19E〜19Fに本方法が細胞リモデリング及びより高レベルの生物学的過程をもたらすことを示す融合性血管からの小毛細血管の血管新生発芽の証拠を示す。
血管、複数の種類の細胞及び細胞外マトリックス組成物を十分に備えた組織構築物の製作を実証するために、様々な設計の3D不均質性構造を印刷する。
顕微鏡法を用いて、独立に染色されている3種の細胞型(緑色GFP HNDF, 青色10T1/2及び赤色HUVEC)の位置を特定する。この人工組織構築物の半織性が図10B〜10Dに示す概略図及び画像に明確に見える。共焦点レーザー走査顕微鏡法を用いて、この3D組織構築物を十分にインタロゲイトし、各細胞の正確な位置を決定することが可能である。2日間の培養の後のXY、XZ及びYZにおける3D印刷構造の共焦点顕微鏡像を図10Gに示す。このアプローチの多用途性を実証するために、他の3D組織構築物も設計し、印刷した。細胞負荷フィラメントが高密度で、相互貫入性であるため、共焦点像を得ることは困難であるが、GelMA中の緑色蛍光タンパク質発現HNDF及び3D血管網の内面を覆う赤色HUVECの両方が見える。
最終ステップとして、1週間にわたる印刷10T1/2線維芽細胞の生存率を検討した。0日目に、細胞生存率は、61%であったが、7日後に82%に増加した。対照(0日目に78%)と比較して低い初期生存率が存在するが、印刷細胞は、増殖し、時間の経過につれて広がり、1週間後に培養中の同様なレベルの生存率に至った。初期の生存率の低下は、印刷工程中に細胞が経験したせん断又は伸長応力により生じた可能性がある。印加圧力、ノズル径、細胞の種類及び環境条件が印刷後の細胞生存率に影響を及ぼす可能性がある。他の重要なパラメーターは、所望の人工組織構築物を印刷するために必要な総構築時間である。細胞負荷インクを劣化する前にインク貯槽に保存することができる最長時間が存在し得る。しかし、高処理多材料印刷用の以前に報告された多ノズルプリントヘッドの実装(J. A. Lewisら、「Multinozzle Deposition System for Direct Write Applications」、国際特許公開番号PCT/US2012/044794、2012年6月29日出願、参照により組み込まれる)により、単一ノズル印刷と比較して特性構築時間が2桁短縮し得る。例えば、一般的なヒト成人の肝臓と同等な1000cm3の容積を有する人工組織構築物を印刷するには一般的な印刷速度で単一200μmノズルを用いて約72時間必要である得る。しかし、64多ノズルアレイの実装により、各構築時間が約1時間に短縮し得る。
厚い生組織の3Dバイオプリンティング
厚い生組織の製作に重要なことは、多材料3Dバイオプリンティング用の生物学的、犠牲及びエラストマーインクの設計である。
溶液の調製
人工組織構築物を作製する前にマトリックス及びインク前駆体溶液を調製した。15重量/容積%ゼラチン溶液(A型、ブタ皮膚からの300ブルーム(bloom)、Sigma)は、DPBS(カルシウム及びマグネシウム不含有の1Xダルベッコのリン酸緩衝生理食塩水)を70℃(特記しない限り)に加温し、次いで激しく撹拌しながらゼラチン粉末を溶液に加えることにより調製した。70℃(特記しない限り)で12時間撹拌することによりゼラチンを完全に溶解し、次いで1M NaOHを用いてpHを7.5に調整した。温ゼラチン溶液を濾過滅菌し、後(<3ヵ月)の使用のために分割して4℃で保存した。
ゼラチン-フィブリンIPNsを調製するために、フィブリノーゲン、ゼラチン、カルシウム及びTGを37℃で様々な濃度で一緒に混合した。一般的な最終濃度は、10mg mL-1フィブリノーゲン、7.5重量%ゼラチン、2.5mM CaCl2及び0.2重量%TGであった。大規模組織(図33)を印刷するために、印刷フィラメントへの拡散及び希釈を補償するために1重量%TGを用いた。トロンビンと混合する前にこれらの組成物を放置する時間がマトリックスの光学的透明度を決定づけるものであった(図24A〜24D)。この待機ステップは、「TGプレインキュベーション時間」と呼び、一般的に15〜20分であった。TGプレインキュベーション時間の後、溶液をトロンビンと500:1の比で速やかに混合し、1UmL-1の最終濃度が得られた。混合した後、フィブリノーゲンのフィブリンゲルへの不可逆的な酵素により促進される重合が急速に起こるので、マトリックスを30秒以内に注型した。
厚い血管新生組織の3Dバイオプリンティングのためのいくつかのインクを調製した。
印刷フィラメント及び注型マトリックス中のフィブリン網状構造を視覚化するために、フィブリノーゲンを2つの発蛍光団にコンジュゲートした。具体的には、1gのウシフィブリノーゲンを100mlの50mMホウ酸緩衝液pH 8.5(Thermo Scientific)に溶解して、10mg ml-1溶液を調製した。この溶液に、フルオレセイン又はローダミンとコンジュゲートしたN-ヒドロキシスクシンイミド(NHS)を10:1染料:フィブリノーゲンのモル比で加えた。室温で2時間の反応の後、浴中PBSを1日2回交換した、PBSに対する2L浴中10kDa MWCO透析チューブを用いた3日間にわたる透析により、標識フィブリノーゲンを非コンジュゲート染料から分離した。透析が完了した後、蛍光コンジュゲートフィブリノーゲンを-80°Cで凍結し、凍結乾燥し、使用前に-20℃で保存した。
インクのレオロジーは、直径40mm、2°円錐及び平板形状を有する応力制御レオメーター(DHR-3、TA Instruments、New Castle、DE)を用いて測定する。せん断貯蔵(G')及び損失(G'')係数は、1Hzの振動数及び0.01の振動歪み(γ)で測定する。温度掃引は、ペルチエプレートプレートを用いて-5℃〜40℃の範囲にわたって実施した。試料は、試料全体の温度勾配を最小限にするために試験前に5分間、後のそれぞれの温度で1分間平衡化した。時間掃引は、特に明期しない限り、37℃又は22℃に保持した温度制御ペルチエプレート上に予混合溶液を速やかにのせることにより実施した。溶液が注型後に急速にゲル化するため、混合中の気泡の発生を最小限にすることが重要であった。
ヒト骨髄由来間葉系幹細胞(hMSC)(Rooster Bio)をブースター培地(Booster Media)(Rooster Bio)中で培養し、2継代を超えて用いなかった。
すべての血管新生組織は、ビルド容積725mm×650mm×125mmを有する3軸運動制御ガントリ上に取付けた4つの独立に制御可能なプリントヘッドを装着した特注設計の多材料3Dバイオプリンター(AGB 10000、Aerotech Inc.、Pittsburgh、PA USA)で作製した。各インクは、様々なサイズ(すなわち、直径100μm〜410μm)のノズルをルアロック(EFD Inc.、East Providence、RI、USA)を介して取付けた別個の注射筒の内部に収容した。インクは、1mm s-1〜5cm s-1の印刷速度に対応する10〜140psiの範囲の空気圧を印加することによって各堆積ノズルを介して押し出した(800Ultra dispensing system、EFD Inc.、East Providence、RI、USA)。印刷の前に、直角に取付けた光学顕微鏡(LS-7600 series、Keyence、Japan)を用いて4つのプリントヘッドの相対X-Yオフセットを決定した。
チップ出口から蠕動チューブを除去して、50〜500μLのHUVEC懸濁液(1×107細胞mL-1)をピペットにより注入して、血管網を満たした。次いで、シリコーンチューブをチップ出口に戻し、出口及び入口ピンチクランプの両方をシールした。潅流チップを37℃でインキュベートして、無流動条件下で通路への細胞の接着を促進した。30分後に、チップを180°反転させて、通路の他の側への細胞の接着を促進し、通路における細胞の円周播種を達成した。最後に、能動的潅流を開始する前に細胞を37℃で5時間から一夜までの間にさらにインキュベートした。
内皮細胞の播種の後、蠕動チューブを24チャンネル蠕動ポンプ(Ismatec)に取付け、その後、入口及び出口ホースのクランプを外して、潅流の準備をした。次いで、蠕動ポンプを所望の潅流速度で始動させた。単一血管通路については、潅流速度を13μL/分に設定したが、厚く、大規模な組織については、27μL/分に設定した。
細胞生存率は、各条件について2×106細胞mL-1でインクを印刷することによって印刷後に測定した。印刷細胞の生存率については、印刷フィラメントをガラス基板上に堆積させ、共焦点顕微鏡法による撮像(n=3固有の試料、撮像n=10回)の前に、カルセイン-AM(「生」、1μL mL-1、Invitrogen)及びエチジウムホモダイマー(「死」、4μL mL-1、Invitrogen)を用いて染色した。ImageJソフトウエアにおける3Dオブジェクツカウンタープラグイン(3D objects counter plugin)を用いて生及び死細胞数を得た。各試料について結果を平均し、標準偏差を算定した。
組織の印刷及び集合の顕微鏡写真及びビデオをDSLRカメラ(Canon EOS、5D Mark II、Canon Inc.、USA)を用いて収集した。蛍光染料を用いて、Pluronic F127(赤、Risk Reactor)及びゼラチン-フィブリンインク(フルオレセイン、Sigma Aldrich)の視覚化を改善した。Keyence Zoom(VHX-2000、Keyence、Japan)、倒立蛍光(Axiovert 40 CFL、Zeiss)及び正立共焦点顕微鏡(LSM710、Zeiss)を含む様々な顕微鏡を用いて、印刷組織構造を視覚化した。ImageJを用いて、蛍光チャンネルを組み合わせることによって複合顕微鏡像を得た。共焦点スタックの3Dレンダリング及び視覚化は、Imaris 7.6.4、Bitplane Scientific Software及びImageJソフトウエアを用いて行った。細胞の計数は、Imarisにおける半自動ネイティブアルゴリズム並びにImageJ計数及び追跡アルゴリズムを用いて行った。
免疫染色及び共焦点顕微鏡法を用いて、3D視覚化組織を評価した。印刷組織を最初に潅流によりリン酸緩衝生理食塩水(PBS)で数分間洗浄した。次に、10%緩衝ホルマリンを3D組織を通して10〜15分間潅流した。組織を潅流チップから除去し、10%緩衝ホルマリンに浸した。ホルマリンによる固定時間は、組織構築物の厚さによって異なっており、1cmの厚さの組織については約2時間の固定が必要であった。次いで3D組織をPBSで数時間洗浄し、PBS中1重量%のウシ血清アルブミン(BSA)を用いて一夜ブロックした。目的の細胞タンパク質又はバイオマーカーに対する一次抗体を0.5重量% BSA及び0.125重量% TritonXの溶液中で下の表1に示す希釈度で構築物と共に2日間インキュベートした。
ファストブルー(Sigma Aldrich)及びアリザリンレッド(SigmaFast、Sigma Aldrich)を用いて、AP活性及びカルシウム沈着を視覚化した。ファストブルーの1つの錠剤を10mLのDI水に溶解した。この溶液を暗所に保存し、2時間以内に用いた。細胞を上述のようにカルシウム及びマグネシウム不含有のDPBS中0.05% Tween 20を用いて洗浄した。次いで、試料をファストブルー溶液で覆い、暗所で5〜10分間インキュベートし、PBS-Tween緩衝液を用いて洗浄した。鉱物化を評価するために、2%アリザリンレッド溶液をDI水に溶解し、激しく混合し、濾過し、24時間以内に用いた。試料をDI水中で平衡化し、アリザリンレッド溶液と共に2〜5分間インキュベートし、次いで染色溶液を除去し、試料をDI水で3回又はバックグラウンド染料が観察できなくなるまで洗浄した。
印刷血管系のバリア機能を評価するために、血管通路に、生存中に、25μg/mLのFITCコンジュゲート70kDaデキストラン(FITC-Dex、Sigma product 46945)を含む培養培地を3分間にわたり20μL/分、その後の約33分間にわたり1μL/分の速度で潅流した。広視野蛍光顕微鏡(Zeiss Axiovert 40 CFL)を用いてFITC-Dexの拡散パターンを検出した。蛍光画像は、潅流の前及び後に3〜5分ごとに33分間にわたり取得した。FITC-Dexの拡散透過率は、以下の式を用いて時間の経過に伴う蛍光強度の変化を定量化することによって計算した。
加工性、異種集積化、生体適合性及び長期安定性の付随要件を満たすために、本発明者らは、最初に新たな印刷可能な細胞負荷インク並びにゼラチン及びフィブリノーゲンの混合物を含む注型可能な細胞外マトリックス(ECM)を開発した(Lee KY, Mooney DJ (2001) Hydrogels for Tissue Engineering. Chem Rev 101(7):1869-1880)。
潅流可能なチップ上の曲3D腎近位尿細管のバイオプリンティング
細胞外マトリックスの調製及びレオロジー
ECMは、ゼラチン及びフィブリンの網状構造からなるものであり、上文の実施例2で述べたように調製した。
潅流可能PTモデルの3Dバイオプリンティングには2種のインクが必要である。2種のインクを上文の実施例2で述べたように調製した。
3D PT構築物は、725mm×650mm×125mmのビルド容積を有する3軸運動制御ガントリ上に取付けた4つの独立に制御可能なプリントヘッドを装着した特注設計の多材料3Dバイオプリンター(AGB 10000、Aerotech Inc.、Pittsburgh、PA USA)を用いて作製した。インクは、様々なサイズ(すなわち、直径50μm〜410μm)のノズルをルアロック(EFD Inc.、East Providence、RI、USA)を介して取付けた別個の注射筒に収容した。インクは、1mm/s〜5cm/sの印刷速度に対応する10〜90psiの範囲の空気圧を印加することによって堆積ノズルを介して押し出した(800 Ultra dispensing system、EFD Inc.、East Providence、RI、USA)。
ヒト不死化PTEC(RPTEC/TERT1、ATCC CRL-4031)をATCCの指示に従って培養し、すべてのPTモデル試験に20継代まで用いた。遺伝子発現解析のために、ヒト初代RPTEC(Cell Science)、不死化PTEC(RPTEC-TERT1、Evercyte)及びA498(ATCC HTB-44)腎臓がん細胞を用い、供給業者の指示に従って培養した。GFPを発現する、ヒト新生児皮膚線維芽細胞(HNDF)(Angio-Proteomie)を供給業者の指示に従って培養し、15継代まで用いた。
ヒト初代RPTEC(Cell Science)、不死化RPTEC-TERT1(Evercyte)及びA498(ATCC HTB-44)腎臓がん細胞を供給業者の指示に従って96ウエルにおいて増殖させ、融合後3日目に培養培地を100μl/ウエルの1×RNA溶解混合物(QuantiGene(登録商標)試料処理キット、QS0101)と交換することにより収集した。次いで40μlの溶解物をmRNA捕捉磁気ビーズセット(Panomics QuantiGene(登録商標) Plex Set 12631、カタログ番号312631)と混合し、一夜インキュベートし、分岐DNA増幅のために処理し、製造業者の指示(Panomics QuantiGene(登録商標) Plexアッセイキット、QP1015)に従って解析した。PPIBプローブを標準化のためのハウスキーピング遺伝子として用いた。蛍光強度(FI)データは、3回の生物学的反復実験の平均値及び標準偏差として示した。
培地潅流液を細胞播種後25日間の期間にわたって尿細管から採取し、分析の前に-80℃で保存した。サイトカインのプロファイリングのために、上清を氷上で解凍し、試料希釈緩衝液(BioRadカタログ番号M60-009RDPD)で2倍に希釈し、製造業者の指示に従って、Bio-Plex Pro(商標)Human Chemokine IL-6(Set #171BK29MR2)、IL-8(Set #171-BK31MR2)及びMCP-1(Set #171-BK36MR2)並びにBio-Plex(登録商標)200システム(BioRad)を用いてLuminex技術ベースのELISAにより分析した。データは、技術的3反復実験の平均サイトカイン濃度及び標準偏差として報告した。
各印刷3D PT構築物を、細胞負荷/播種の前にインキュベーター中でPTEC培地により数時間潅流した。TECs(PTEC/TERT1、ATCC)をそれらの培養皿からトリプシン処理し、培地中で約2×107細胞/mLに濃縮した。次いで細胞懸濁液を出口を介して潅流チップ中に負荷した(図39B及び39C)。負荷済み構築物をインキュベーター中に横方向に数時間配置し、次いで180°反転し、無流動で尿細管中で一夜インキュベートした。翌日、重力による流動下で非接着細胞を尿細管から洗い流した。次いで新鮮な培地の潅流を開始したところ、残存細胞が密集し、次いでそれらが播種後約3週目に融合性の状態に達するまで(図39K)それらのコロニーから増殖した(図39F)。増殖期中に、PTECにATCCガイドラインに従って調製したPTEC培地と、さらに1%アプロチニン(EMD Millipore、ECMの分解を遅くするために用いた)、1%ウシ胎児血清(FBS)及び1%抗生物質-抗真菌薬(Gibco)を供給した。成熟後、FBSを除去し、PTECは、緊密な上皮単層に詰まった(示さず)。播種後1日目に、PTECを、尿細管断面よって0.1〜0.5ダイン/cm2に変化するせん断応力と同等と見なされる、1μl/分の連続した一方向流に曝露した。培地は、閉ループ回路で蠕動ポンプにより供給し、2日ごとに交換した。
アルブミン取込みは、印刷3D PTモデル並びに2D対照について評価した。第1の対照は、組織培養プラスチック上で増殖したPTECからなっているが、第2の対照は、本発明者らのECM上で増殖したPTECからなっている。各場合に、PTECを融合性を示すまで増殖させ、血清不含有培地中で成熟させた。FITCとコンジュゲートしたヒト血清アルブミン(HSA-FITC、Abcam ab8030)をPTEC培地に50μg/mLで懸濁する。すべての試料をそれらの培地中でHSA-FITCと共に2時間インキュベートする(潅流の場合、それを開放内腔を通して潅流する)。曝露後、すべての試料を3倍量で洗浄し、次いで10×トリプシンを用いてトリプシン処理して、個々の細胞を収集する。細胞を固定し、メガリンについて一次及び二次抗体を用いて対比染色する。表2に用いた特異抗体を示す。
2D対照及び印刷3D PTの両方におけるCysAの毒性作用を探究した。2Dにおいて、細胞を組織培養プラスチック上の96ウエルフォーマットに播種し、融合性を示すまで増殖させた。それらにATCCのガイドラインに従って培地を供給した。CysA(Sigma-Aldrich、SML1018)をそれらの培地に様々な濃度で懸濁し、細胞と共に24時間インキュベートした。フェナジンメトスルフェート(MTS)の存在下における(3-(4,5-ジメチルチアゾール-2-イル)-5-(3-カルボキシメトキシフェニル)-2-(4-スルホフェニル)-2H-テトラゾリウム)を用いた生存率アッセイを曝露後24時間目に行った。このアッセイは、細胞が融合性に達した当日に細胞にCysAを投与することにより、初期融合性時に、並びに細胞が融合性に達した数日後にCysAを投与することにより、後期融合性時にPTECにおいて完了した。特に、毒性結果は、各場合について同様であった(図40N)。3D PTについて、CysAは、血清を最低限10日間培地に含めなかった、融合性に達した後に(約3週目に)成熟尿細管の開放内腔を通して様々な濃度で供給した。CysA曝露後24時間目に、FITC-デキストラン漏出試験(以下で述べる)を実施して、PTECのバリア機能に対する擾乱を評価し、定量化した。直後に、PTを10%緩衝ホルマリンを用いて1時間固定し、上の表2に示すようにアクチン及びDAPIについて対比染色した(特殊染色)。
3Dの上皮のバリア機能を評価するために、25μg/mLのFITCコンジュゲート70kDaデキストラン(FITC-Dex、Sigma product 46945)を含むPTEC培地を3分間にわたり15μL/分、その後約30〜45分間にわたり1μL/分の速度で開放内腔に潅流することによって、拡散透過率を定量化した。全試験は、視野内の尿細管及び周囲ECMの両方についてライブセルイメージングのもとで実施した(図41A〜41E)。FITC-Dexの拡散パターンは、広視野蛍光顕微鏡(Zeiss Axiovert 40 CFL)を用いて検出した。蛍光像は、潅流の前及び30〜45分間にわたり3〜5分ごとに取り込んだ。FITC-Dexの拡散透過率は、以下の式を用いて蛍光強度の経時的変化を定量化することによって計算した(Price, G. & Tien, J. in Biological Microarrays, Vol. 671. (eds. A. Khademhosseini, K.-Y. Suh & M. Zourob) 281-293 (Humana Press, 2011))。
透過型電子顕微鏡観察(TEM)のために、2D又は3D構造のPTECを0.1Mカコジル酸ナトリウム緩衝液(pH 7.4)中2.5%グルタルアルデヒド、1.25%パラホルムアルデヒド及び0.03%ピクリン酸を用いて固定した。小試料(1mm×1mm)を除去し、0.1Mカコジル酸緩衝液で洗浄し、1%四酸化オスミウム(OsO4)(EMS)及び1.5%フェロシアン化カリウム(KFeCN6)(Sigma)中に1時間浸し、水で3回洗浄し、その後、様々な度数のアルコール(それぞれ10分;50%、70%、90%、2×10分100%)で脱水した。次いで試料をプロピレンオキシド(EMS)中に1時間入れ、プロピレンオキシド及びTAAB Epon(Marivac Canada Inc.、St. Laurent、Canada)の1:1混合物中で一夜インキュベートした。翌日、試料をTAAB Eponに包埋し、60℃で48時間重合させた。超薄切片(約60nm)をReichert Ultracut-Sミクロトームで切り出し、銅グリッド上にのせ、クエン酸鉛で染色し、JEOL 1200EX透過型電子顕微鏡で観察し、像をAMT 2k CCDカメラで記録した。画像解析は、ImageJソフトウエアを用いて実施した。
免疫染色の後に共焦点顕微鏡法を用いて、2D及び3D PTECモデルにおけるタンパク質の細胞局在性を評価した。免疫染色の前に、各構築物をPBSで洗浄し、次いで10%緩衝ホルマリンを用いて20分間から1時間固定した。固定剤を数時間にわたるPBSによる数回の洗浄を用いて除去し、次いでPBS中1重量%ウシ血清アルブミン(BSA)を用いて一夜ブロックした。目的の細胞タンパク質又はバイオマーカーに対する一次抗体を0.5重量% BSA及び0.125重量% Triton X-100の溶液中で上の表2に示した希釈度で構築物と共に1日間インキュベートした。非結合抗体の除去は、PBS又はPBS中0.5重量% BSA及び0.125重量% Triton X-100の溶液に対する1日間の洗浄ステップを用いて達成された。二次抗体は、PBS中0.5重量% BSA及び0.125重量% Triton X-100の溶液中で上の表に示した希釈度で構築物と共に1日間インキュベートした。試料をNucBlue又はActinGreenで2時間対比染色し、次いで撮像の前にPBSで1日間洗浄した。
位相差顕微鏡法は、1.25X〜40Xの範囲の対物レンズ付きの倒立Leica DM IL鏡を用いて実施した。共焦点顕微鏡法は、405、488、514、561及び633nmの波長のスペクトルレーザーを用いた5X〜40Xの範囲の水浸対物レンズ付きの正立Zeiss LSM 710を用いて実施した。zスタックの画像再構成は、最大ピクセル強度設定のz投影機能を用いてImageJで実施した。輝度の増加は、全z投影画像にわたって均一に行った。3D画像の再構成は、Imarisソフトウエアを用いて実施した。インキュベーターシステムにおける新たなCytoSMART(Lonza)を用いて、微速度撮影画像を取り込んだ。拡散透過性の定量化のための画像解析は、以前に報告された方法を用いたカスタムMATLABスクリプトを用いて実施した(Price, G. & Tien, J. in Biological Microarrays, Vol. 671. (eds. A. Khademhosseini, K.-Y. Suh & M. Zourob) 281-293 (Humana Press, 2011))。TEM像解析は、各条件の少なくとも3つの独立した試料における細胞高さ(n≧50)、微絨毛密度(n≧25)及び微絨毛長(n≧200)を測定するためにImageJソフトウエアを用いて実施した。
データは、平均値±標準偏差として表した。統計解析は、MATLABを用いて行い、統計的有意性は、チューキーの多重対比較検定を用いてANOVAにより判定されるp<0.05の値で判定した。異なる有意水準(p値)をアステリスクにより示し、個別のp値を各図の説明文に示す。
3D近位尿細管の印刷、播種及び縦方向培養
述べたバイオプリンティング法を用いて、図36Aに示すような、ネフロンの3D近位曲尿細管セグメントを構築した。最初に、図36B及び36Cに示すように、3D組織チップの外縁の境界を定めるシリコーンガスケットをガラススライド上に印刷した。ゼラチン-フィブリンヒドロゲル(Kolesky, D.B.ら、Proc. Natl. Acad. Sci. U. S. A. (2016))からなる人工細胞外マトリックス(ECM)の層をガスケット内に均一に堆積させた。次に、ピンク色で示した、変色性インクをECM層上に印刷した。「変色性インク」という用語は、最終的に液化し、最終3D PT構築物から除去される印刷材料を意味する。印刷後、変色性インクを、ガスケット壁を介して連結した中空金属ピンに接続し、追加のECMを印刷構造上に注型した。次いで、3D組織モデルを潅流可能なチップ内に収容し、それを4℃に冷却して、液化し、その後、変色性インクを除去して、ECM内に埋め込まれた開放屈曲管状通路を得た。最後に、細胞培地を外部蠕動ポンプによりチップ上の3D屈曲管状構造を通して潅流した。特に、述べた方法は、正確に制御されたサイズ、曲率及び位置を有する多種多様な立体配置の3D近位尿細管モデルを作製することができる。例えば、アッセイの統計的妥当性を増大させるために又は基底側アクセス通路を備えるために複数の尿細管が必要である場合、それらは、互いに並行して印刷することができ(図42A及び42B参照)、独立に又は単一の入口を介して潅流することができる。図42A及び42Bに示すように、複数のPTを並行して印刷し、融合性の状態まで増殖するPTEC細胞により内面を覆うことができる。
PTECを尿細管に播種し、成熟するまで増殖させた後、光学顕微鏡法、走査型電子顕微鏡法(SEM)及び透過型電子顕微鏡法(TEM)の組合せを用いて、印刷され、潅流された3D PTの特徴付けを行った(図45A〜46K及び46A〜46G)。
PTEC細胞による受容体媒介エンドサイトーシスは、体液恒常性のために必須である。糸球体濾液からの血漿タンパク質の再吸収は、刷子縁にあるメガリン-キュビリン複合体に部分的に依拠しており(Cui, S.ら Megalin/gp330 mediates uptake of albumin in renal proximal tubule. American Journal of Physiology-Renal Physiology 271, F900-F907 (1996); Gekle, M. Renal proximal tubular albumin reabsorption: daily prevention of albuminuria. Physiology 13, 5-11 (1998);及び Norden, A.G.ら Urinary megalin deficiency implicates abnormal tubular endocytic function in Fanconi syndrome. J. Am. Soc. Nephrol. 13, 125-133 (2002))、PTECによるアルブミンの取込みをモニターすることによりin vitroでモデル化することができる。FITC標識ヒト血清アルブミン(HSA)を取り込む、潅流3D PT構築物又は2D対照において増殖した、PTECの能力を試験した。FITC-HASへの2時間の曝露後に、PTECを採取し、メガリン発現について染色し、フローサイトメトリーにより解析した。アルブミンの取込みに関する結果を図48Aに示す。2D対照における大集団は、非蛍光対照と同様の蛍光強度を示しているのに対して、3D PT構築物の内面を覆う細胞は、FITC-HSA強度の有意な増加を示している。アルブミンの輸送体の1つである、メガリンに関する結果は、その発現も3D PTにおいて最高であったことを示すものである(図48B)。フローサイトメトリーにより解析した集団の蛍光強度の平均値を表3に示す。
シクロスポリンAは、拒絶反応を予防するために移植手術の後に一般的に投与される薬物であり、腎近位尿細管細胞に損傷を与える公知の腎臓毒素であwpる。潅流3D PTに対するその作用を試験するために、3D PTを様々な濃度のシクロスポリンA(CysA)に曝露し、アクチンフィラメントの免疫染色により、細胞形態及び細胞骨格形成の変化をモニターした。尿細管の明視野像(図40A〜40D)及びアクチン染色の対応する3Dレンダリング(図40E〜40H及び図40I〜40L)から、CysA誘発性損傷の用量依存的徴候が明らかである。細胞間結合の軽微な切断(図49A〜D)及びアクチンの再構成(図40J)が10μM CysAで認められたのに対して、細胞を欠いている離散部位が50μM CysAで容易に明らかであり(図40G及び40K)、100μM CysAで悪化した(図40D、40H及び40L)。本発明者らは、50及び100μM CysAで細胞層が締まり、曲がったことも確認している(図40G、40K及び39)。最後に、処理した尿細管におけるFITC-デキストラン(70kDa)の拡散透過率を定量化することによって上皮バリア機能のCysA誘発性破綻を評価した。図40Mに示すように、50及び100μM CysAへの曝露により、上皮バリアの透過率がそれぞれほぼ4倍及び6倍増加した。2D培養プラスチック皿上で増殖したPTECのそれぞれの細胞生存率が50及び100μM CysAによる処理後に40%及び60%低下したことが見い出された(図40N)。全体的にみて、これらの結果から、3D PT構築物を用いて、腎臓毒性を定性的に(免疫染色)且つ定量的に(拡散透過率測定)評価することができることがわかる。
バイオプリンティングの最近の進歩により、人工細胞外マトリックス内の広範囲に広がり、相互に連結した通路の作製が可能となった(Kolesky, D.B.ら、Three-dimensional bioprinting of thick vascularized tissues. Proc. Natl. Acad. Sci. U. S. A. (2016); Kolesky, D.B.ら 3D Bioprinting of Vascularized, Heterogeneous Cell-Laden Tissue Constructs. Adv. Mater. 26, 3124-3130 (2014);及びMiller, J.S.ら Rapid casting of patterned vascular networks for perfusable engineered three-dimensional tissues. Nature materials 11, 768-774 (2012))。これらの通路の内面を内皮細胞により被覆し、潅流して、埋め込まれた血管系を有する組織を作製することができることが先に示された(Kolesky, D.B.ら(2016)及びKolesky, D.B.ら(2014))。in vitroでの上皮化により潅流可能で、屈曲した3D近位尿細管を製作するための経路が現在開発されている。重要なことに、本発明者らの3D PTモデルにおいて生理的に適切なせん断応力で縦断的研究を可能とするための制御された一方向流が組み込まれた。3D細胞培養、チップ上組織及びバイオプリンティング法を同時に取り入れることによって、チップ上の3D組織及び臓器モデルを製造するための容易で、カスタマイズ可能なプラットフォームが創製された。尿細管のサイズ及び屈曲を含む形状をプログラム可能に規定する能力は、ゲル中ピン引抜き生成直尿細管に依拠している既存の3D PTモデルにまさっている。さらに、述べた人工の3DバイオプリンティングされたPTは、解析のための莫大な数の細胞、例えば、フローサイトメトリーによる正確なサンプリングに必要な10,000個の細胞をはるかに超える細胞の収集を可能にする。それに反して、高細胞集団の定量的な研究は、平面的なマイクロ流体力学ベースのデバイスにおいて達成することが以前には困難であった(Adler, M.らA quantitative approach to screen for kidney toxic compounds in vitro. J. Am. Soc. Nephrol. (2015);及びAdler, M.ら A quantitative approach to screen for kidney toxic compounds in vitro. J. Am. Soc. Nephrol. (2015))。

- 管状組織構築物を印刷する方法であって、
1つ以上の犠牲フィラメントを基板上又は基板内に堆積させて、機能性通路パターンを形成するステップであって、各犠牲フィラメントが変色性インク及び複数の所定の種類の生細胞を含み、各所定の種類の生細胞が犠牲フィラメントの長さに沿った異なる所定の位置に堆積されている、ステップ、
機能性通路パターンを細胞外マトリックス組成物によって少なくとも部分的に取り囲むステップ、
変色性インクを除去して、細胞外マトリックス組成物中の1つ以上の機能性通路を作るステップであって、各異なる所定の種類の生細胞の少なくとも一部が、変色性インクの除去の後に異なる所定の位置に残存し、それにより、管状組織構築物を形成する、ステップ

第2の変色性インク製剤が変色性インク及びヘンレ係蹄細胞を含み、
第3の変色性インク製剤が変色性インク及び腎遠位尿細管細胞を含む、ステップ、
機能性通路パターンを細胞外マトリックス組成物によって少なくとも部分的に取り囲むステップ、
変色性インクを除去して、細胞外マトリックス組成物中の1つ以上の機能性通路を作るステップであって、第1のインクの除去の後に腎近位尿細管細胞の少なくとも一部が1つ以上の機能性通路の第1の長さに沿って残存し、第2のインクの除去の後にヘンレ係蹄細胞の少なくとも一部が1つ以上の機能性通路の第2の長さにおいて残存し、第3のインクの除去の後に腎遠位尿細管細胞の少なくとも一部が1つ以上の機能性通路の第3の長さにおいて残存し、それにより、ネフロンを形成する、ステップ
