Abstract
Description
前記人工皮膚培養容器の内部に無細胞コラーゲン(Acellular Collagen)溶液を導入するステップ;
前記無細胞コラーゲン溶液を培養して、前記導入された無細胞コラーゲン溶液を、前記人工皮膚培養容器の寒天内部へ進入(invasion)させるステップ;
前記人工皮膚培養容器の内部に、皮膚線維芽細胞(Human dermal fibroblast)溶液を導入し、前記コラーゲン溶液をゲル化させるステップ;および
前記皮膚線維芽細胞溶液を培養しつつ、前記人工皮膚培養容器の外部から培養液を供給するステップ;
を含む人工皮膚の製造方法を提供してよい。
本発明の一実施例として、疎水性に改質されていない寒天(Agar,extra pure reagents,Yakuri pure chemicals Co., Ltd,Japan)とオクタデシル(octadecyl,Sigma Aldrich)疎水性分子で改質された寒天を10:2の質量比で含み、寒天を人工皮膚培養容器に対して3(w/v)%で含む寒天ゲルからなる人工皮膚培養容器を準備した。前記寒天培養容器の厚さは2mm、内部底部の径(diameter)は12mmであり、内部微細気孔の大きさは約0.5μmであった。
本発明の比較例として、寒天(Agar,extra pure reagents,Yakuri pure chemicals Co., Ltd,Japan)を、人工皮膚培養容器に対して3(w/v)%で含む寒天ゲルからなる、純粋寒天で構成された人工皮膚培養容器を準備した。前記寒天培養容器の厚さは2mm、内部底部の径(diameter)は12mmであり、内部微細気孔の大きさは約0.5μmであった。
本発明のまた別の比較例としては、既存に一般的に使用されている人工皮膚培養容器である、径12mm、底面細孔サイズ0.4μmを有するトランズウェル(Transwell,Corning,NY,14831,USA)を準備した。
前記製造した実施例1、比較例1および2の人工皮膚培養容器を使用して、人工皮膚を下記の方法に従ってそれぞれ培養した。

Claims (11)
- ゲル形態の寒天(agar)を含み、
前記寒天は、一部が疎水性に改質されたものである、人工皮膚培養容器。 - 前記寒天のうち疎水性に改質された寒天(Hydrophobically‐modified agar)は、寒天に含まれるアガロース(agarose)の一つ以上の水素が疎水性分子で置換されたものである、請求項1に記載の人工皮膚培養容器。
- 前記疎水性分子は、
炭素数12〜18のアルキル(alkyl)、アルケニル(alkenyl)およびアシル(acyl)鎖のうち一つ以上;
ポリプロピレングリコール(Polypropylene glycol);および
ポリカプロラクトン(polycaprolactone)からなる群より選択されたものである、請求項2に記載の人工皮膚培養容器。 - 前記寒天がゲル化される前の寒天粉末100個を1ユニット(unit)とし、1ユニットあたりの寒天粉末に置換された疎水性分子数を置換度(Degree of substitution,%)とするとき、前記人工皮膚培養容器の寒天に対する疎水性分子置換度は、5.0%〜20%である、請求項1に記載の人工皮膚培養容器。
- 前記人工皮膚培養容器の総寒天質量に対する、疎水性に改質された寒天の割合は、5〜50質量%である、請求項1に記載の人工皮膚培養容器。
- 前記寒天は、微細気孔を含むものである、請求項1に記載の人工皮膚培養容器。
- 前記微細気孔の平均粒径は、0.1〜10μmである、請求項6に記載の人工皮膚培養容器。

- 請求項1〜8のいずれか一項に記載の人工皮膚培養容器で人工皮膚を製造する方法であって、
前記人工皮膚培養容器の内部に無細胞コラーゲン(Acellular Collagen)溶液を導入するステップ;
前記無細胞コラーゲン溶液を培養して、前記導入された無細胞コラーゲン溶液を、前記人工皮膚培養容器の寒天内部へ進入(invasion)させるステップ;
前記人工皮膚培養容器の内部に、線維芽細胞(Human dermal fibroblast)溶液を導入し、前記コラーゲン溶液をゲル化させるステップ;および
前記線維芽細胞溶液を培養しつつ、前記人工皮膚培養容器の外部から培養液を供給するステップ;
を含む人工皮膚の製造方法。 - 前記導入された無細胞コラーゲン溶液を、前記人工皮膚培養容器の寒天内部へ進入(invasion)させるステップは、前記コラーゲン溶液のコラーゲン線維が人工皮膚培養容器に含まれている微細気孔に進入して、人工皮膚培養容器の疎水性に改質された寒天と結合して固定されるステップを含む、請求項9に記載の人工皮膚の製造方法。
- 前記人工皮膚培養容器の外部から培養液を供給するステップは、前記人工皮膚培養容器をチャンバ内部に位置させてからチャンバ内に培養液を供給して、前記培養液を前記人工皮膚培養容器の側面部および下部から供給するステップを含む、請求項9に記載の人工皮膚の製造方法。
