Abstract
본 발명은 기능성 혈관이 도입된 인공피부 및 그 제조방법에 관한 것으로, 구체적으로는 인공피부 피하조직 층의 혈관구조를 모사하고, 모사한 혈관이 진피층에 미세혈관으로 도입되어 결과적으로 진피층 내에 모세혈관 모사가 가능하게 제조하는 것을 특징으로 한다. 이는 피부 상태에 따라 유도되어지는 혈관화된 새로운 구조의 인공피부에 관한 것이다.
이로써 인체피부에서 혈관 거동에 영향을 주는 노화, 상처, 혈행개선 등의 평가가 가능한 동물대체 시험법의 시료로 사용 가능하다.

Description
본 발명은 기능성 혈관이 도입된 인공피부 및 그 제조방법에 관한 것으로, 구체적으로는 인공피부 피하조직 층의 혈관구조를 모사하고, 모사한 혈관이 진피층에 미세혈관으로 도입되어 결과적으로 진피층 내에 모세혈관 모사가 가능하게 제조하는 것을 특징으로 한다. 이는 피부 상태에 따라 유도되어지는 새로운 구조의 혈관화 된 인공피부에 관한 것이다.
피부는 신체의 외부를 덮고 있는 기관으로 기능적으로 다양하며 구조적으로 복잡한데, 기본적으로 표피층(epidermis), 진피층(dermis), 피하지방층(hypodermis)의 3개의 층으로 구분된 입체적인 3차원 층상 구조를 가진다.
표피는 수분 손실의 조절, 자외선에 대한 방어, 감염을 포함한 조절과 방어 기능을 가진 계층화된 상피로 구성된 혈관이 없는 최외각층이다. 표피는 대부분 분화 정도에 따라서 층층을 이루는 각질세포로 구성된다.
진피는 표피와 기저막 사이를 이어주는 중간층이다. 섬유아세포에서 분비되는 세포외기질이 풍부한 부위로 혈관과 림프관, 신경, 땀샘, 모낭 등이 위치한다. 피부의 가장 하부에는 지방을 저장하는 지방세포로 구성된 지방조직과 느슨한 연결조직들로 이루어진 피하지방층이 존재하며, 신체의 에너지 보관 및 단열 등의 기능을 가진다.
인공피부(artificial skin)는 피부 세포와 피부 구성 물질인 콜라겐, 엘라스틴 등을 이용하여 3차원적으로 상기와 같은 피부를 재구성한 것으로서, 살아있는 섬유아세포와 각질형성세포로 구성되어 실제피부와 유사한 구조적, 기능적 특성을 나타내기 때문에 피부모사체(skin equivalent 또는 reconstructed skin)라고도 불린다.
이러한 인공피부는 화상, 외상 등 손상을 입은 피부의 대체(영구 생착형) 또는 재생(일시 피복형)을 위해 개발되었지만, 현재는 화장품과 같은 생명과 무관한 제품 개발에 동물실험이 금지됨에 따라, 동물실험 대체법으로도 활발하게 연구되고 있다. 특히 인공피부 배양용기 및 이를 이용한 인공피부 제조기술에 대한 연구가 진행되고 있으며(특허문헌 1~3 참조), 최근에는 연령별에 따른 인공피부의 구조 및 물성에 대해서도 연구되고 있다.
인공피부는 주로 피부와 탄력, 강도, 물질 투과 등에 있어 비슷한 물성을 나타내는 고분자 복합체이지만, 단순피부의 구조를 모사한 진피-표피가 구현되어 있을 뿐이어서 피부와 같은 생명현상을 나타내지 않는다는 점에서 차이점이 있다. 실제피부(인체피부)와 다른 점은 피하조직 층이 없는 경우가 대부분이고 특히 진피층 내부로 혈관구조가 구현된 모델은 아직까지 개발되어 있지 않다.
특허문헌 4는 진피, 표피, 피하조직으로 구성된 인공피부에 관한 것이지만 피하조직에 혈관 채널이 포함되어 있을 뿐이지, 진피층으로의 모세혈관 형성은 전혀 없다. 해당 혈관 채널이 관류 흐름이 가능할 수 있어도, 진피나 표피층으로의 유효물질 등을 전달하기는 어렵다. 또 스캐폴드가 없는(scaffold-free) 사전 혈관화 3D 피부 대체물에 대한 연구가 시도되고 있을 뿐이다(비특허문헌 1 참조).
따라서 실제피부처럼 진피 및 표피의 변화뿐 아니라 혈관까지의 효과적인 분석이 가능한, 피하조직의 큰 혈관으로부터 진피층으로의 모세혈관 형성이 유도된 혈관화 된 인공피부의 개발이 필요하다.

본 발명은 상기와 같은 문제점을 해결하기 위하여, 진피와 표피의 변화뿐만 아니라 혈관 거동에 영향을 주는 노화, 상처, 혈행개선 등의 평가가 가능한 새로운 구조의 인공피부를 제공하고자 한다.
상기 목적을 달성하기 위하여, 본 발명은 (1) 3D 바이오프린팅 기술을 사용하여 vessel guide가 도입된 피하조직 층을 제조하는 단계; (2) 피하조직층 위에 콜라겐(collagen)과 섬유아세포(HDF)를 사용한 진피층을 제조하는 단계; (3) 혈관내피세포의 bridge gel로 스프라우팅(sprouting)하는 단계; (4) 진피층 위에 단일층 표피세포(HEK)를 시딩(seeding)하고 진피층으로 혈관이 유도되는 단계; 및 (5) 표피층의 증식 및 분화하는 단계;를 포함하는 혈관화 된 인공피부의 제조방법을 제공한다.
상기 (1) 단계의 피하조직 층에 도입된 vessel guide의 형상은 원형 모양이고, 개수는 가로와 세로가 n x m 으로 설계된 것을 특징으로 한다.
바람직하게는 상기 vessel guide의 가로와 세로의 수는 동일하게 설계된 것을 특징으로 한다.
상기 피하조직 층은 히알루론산, 젤라틴, 피브리노겐, 트롬빈, 및 HUVEC(혈관내피세포, Human Umbilical Vein Endothelial Cells)로 구성되어 있는 것을 특징으로 한다.
상기 피하조직 층은 HUVEC를 이용한 blood vessel을 제조하고, blood vessel에 z축으로의 vessel guide가 제조되며, 나머지 공간은 피브린 패턴(fibrin pattern)으로 덮어 채워지는 것을 특징으로 한다.
상기 진피층은 콜라겐(collagen) 이외에도 엘라스틴, 키토산, 글리코사미노글루칸, 하일루론산 등과 같은 세포외기질을 더 함유하는 것을 특징으로 한다.
다른 일 실시예에 따르면, 상술한 제조방법으로 제조된 혈관화 된 인공피부를 제공한다.
본 발명에 의하면, 피하조직의 큰 혈관구조 및 진피층으로의 모세혈관을 함께 형성시킴으로써 새로운 구조의 혈관화 된 인공피부를 개발하였다. 이로 인해 인체피부에서 혈관 거동에 영향을 주는 노화, 상처, 혈행개선 등의 평가가 가능한 동물대체 시험법의 시료로 사용 가능하다.

이하, 본 발명의 실시예에 대하여 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 쉽게 실시할 수 있도록 상세히 설명한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
본 명세서에서 “혈관화 된 인공피부(vascularized reconstructed skin)”라 함은 진피, 표피, 피하조직이 모두 포함되고 모세혈관을 가진 구조를 3차원적으로 피부를 재구성한 것을 의미한다.
이하, 본 발명을 상세히 설명한다.
인체의 피부구조에서 혈관구조는 피하조직의 동맥, 정맥과 같은 큰 혈관으로부터 진피층으로 모세혈관이 뻗어있는 형태로 형성되어 있다. 혈관은 피하조직 층에서 진피 및 표피로의 영양성분 공급과 피부의 온도 조절에 중요한 역할을 한다. 또한, 실제 피부에서의 노화는 진피 및 표피의 변화도 있지만, 혈관의 구조도 변화가 있다. 진피층에 존재하는 모세혈관의 크기 및 개수도 노화가 진행됨에 따라 줄어들고, 따라서 인체 피부를 전층 모사하고 기능적인 영향의 평가를 위해서는 혈관 도입은 필수적이다.
이러한 혈관 도입은 3D 바이오프린팅을 사용하여 제조할 수 있으며, 피하조직 층에 특별한 형상의 혈관 구조를 다양하게 도입할 수 있는 장점이 있다. 구체적으로는 vessel guide의 형상과 개수를 달리 형성할 수 있으며, 가능하면 원형 모양이거나 가로와 세로의 수가 동일하게 설계하는 것이 바람직하다.
구체적으로 본 발명에서 개발한 혈관화 된 인공피부는, 피하조직 층에 큰 blood vessel이 포함되고, 해당 blood vessel로부터 vessel guide로 혈관이 형성(연결)되고, 최종적으로 진피층으로의 미세혈관 도입(또는 유도)되어 진피층과 피하조직 층의 문합(grafting)이 가능하도록 설계 제조되는 것이 특징이다.
실시예 1. vessel guide가 도입된 피하조직 층 제조
피하조직 층은 히알루론산, 젤라틴, 피브리노겐, 트롬빈, 그리고 HUVEC로 구성되어 있으며, 농도(함량)과 구성성분은 표 1과 같고, 트롬빈은 2.5U 조건으로 동일하게 하였다. 제조된 피하조직 층은 EGM-2 배양액 상에서 3일간 배양 후, 진피와 표피를 피하조직 층 위에 도입하게 된다.

HUVEC의 blood vessel 및 vessel guide 부분은 히알루론산과 젤라틴을 각각 3 mg/mL와 40mg/mL로 구성되고, guide form일 때에만 추가적으로 피브리노겐이 5mg/mL 농도로 포함된다. 이는 구조 내에 매트릭스(matrix)가 가교(cross-linking)되어 혈관내피세포가 blood vessel을 채우는 형태로 자라도록 하기 위함이다.
세포는 모두 8×106 cells/mL 농도로 제조되었고, 도 1에서 볼 수 있듯이 vessel guide 및 blood vessel은 x축, y축 방향으로 각 3개씩 형성시켰으며(3 X 3), 이는 서로 교차하게 설계되었고, 교차지점에서 진피층으로 혈관 형성이 되도록 z축 방향으로 vessel guide를 printing 하였다.
정리해보면, 피하조직 층은 ① HUVEC를 이용한 blood vessel을 제조하고, ② blood vessel에 z축으로의 vessel guide가 제조되며, ③ 나머지 공간은 피브린 패턴(fibrin pattern)으로 덮어 채워지는데, 이는 3D 바이오프린팅으로 제조한다. 이러한 피하조직 층은 필요에 따라 vessel guide의 형상과 개수 등을 쉽게 설계할 수 있는 장점이 있다.
실시예 2. 혈관화 된 인공피부 제조
먼저, 진피와 표피로 이루어진 인공피부 제조는 종래 알려진 기술로도 가능하지만, 본 발명은 ① 콜라겐(collagen)과 섬유아세포(HDF)를 사용한 진피층 제조하고, ② 진피층 위에 표피세포(HEK) 시딩(seeding)한 후, ③ 표피층의 증식 및 분화과정을 거쳐 배양하는 방법을 채택하였다. 이에 대해서는 본 발명자의 선행특허인 국내공개특허 제2016-0116982호 또는 국내출원특허 제2020-0163297호에 기재된 제조방법을 참고할 수 있다.
즉 섬유아세포와 collagen으로 이루어진 진피층(dermis layer)를 제조하고 7일 간 배양한 이후, 진피층 위에 표피세포를 시딩(seeding)하여 액침(submerged) 배양하였다. 이때 표피세포가 진피층 표면에 mono-layer가 형성될 때까지 배양해주어야 한다. mono-layer가 형성된 후에 air-liquid interface 배양을 수행하여 표피세포의 각질로의 분화를 진행시켜 주었다.
상기 진피층은 콜라겐(collagen) 이외에도 엘라스틴, 키토산, 글리코사미노글루칸, 하일루론산 등과 같은 세포외기질을 더 함유할 수 있다. 세포외기질 물질은 인공피부의 기계적 물성을 조절하는 기능을 수행한다. 인체 내에서 세포가 부착하여 성장할 수 있는 물리적 환경을 제공하는 세포외기질은 단순히 세포 지지체로서 구조적인 역할뿐 아니라, 세포가 성장, 증식 및 분화하는데 매우 중요한 세포 생리 조절인자로 역할을 한다.
사용되는 콜라겐은 소 기원, 레트 꼬리로부터 또는 어류로부터, 또는 천연 콜라겐 또는 유전 공학에 의해 생성된 콜라겐 중 임의의 다른 기원의 콜라겐일 수 있다.
상기 표피세포(HEK)는 상용되는 어떠한 각질세포라도 사용될 수 있으며, 인간으로부터 직접 분리 또는 분리된 후 배양된 각질형성세포뿐만 아니라 다른 세포로부터 유도된 각질형성세포도 사용 가능하다. 상업적으로 입수 가능한 인간 각질형성세포로는 터모피셔(ThermoFisher)에서 제공하는 HEKn이 바람직하다.
상기 각질형성세포는 예건대 시딩(seeding)에 의해 상기 진피층 위에 적용될 수 있으며, 배양 기간은 예건대 1일 내지 4일일 수 있으나, 이에 한정되는 것은 아니다.
상기 인공피부에, 앞서 실시예 1에서 제조한 피하조직 층을 도입하는 개념으로 본 발명을 완성하였다.
따라서 본 발명의 혈관화 된 인공피부의 제조 단계를 구분하여 보면, (1) vessel guide가 도입된 피하조직 층을 제조하는 단계, (2) 피하조직층 위에 콜라겐(collagen)과 섬유아세포(human dermal fibroblast, HDF)를 사용한 진피층을 제조하는 단계, (3) 혈관내피세포의 bridge gel로 스프라우팅(sprouting, 싹트다)하는 단계, (4) 진피층 위에 단일층 표피세포(human epidermal keratinocyte, HEK) 시딩(seeding)하고 진피층으로 혈관이 유도되는 단계, 및 (5) 표피층의 증식 및 분화하는 단계로 이루어지며, 이는 도 2에 자세하게 도시하였다.
한편 도 3은 vessel guide와 blood vessel 개수와 크기에 따른 혈관화 된 인공피부의 단면에 대한 모식도이다. 개수가 증가하거나 크기가 커질수록 진피층으로 유도되는 혈관 구조가 많아지는 경향이 보인다(도 3의 1, 2). 상기 vessel guide를 3 x 3, 4 x 4, 또는 5 x 5 타입으로 테스트를 한 결과이지만, 이에 한정되는 것은 아니다.
참고로 진피층의 특성에 따라(도 3의 3), 예를 들면 콜라겐만 존재하고 섬유아세포가 없는 경우에는 혈관 구조가 유도되지 않으며, 섬유아세포의 수가 증가하면 혈관 구조가 더 많이 유도됨을 알 수 있다. 즉 본 발명의 혈관화 된 인공피부는 진피층과 매우 밀접한 상관관계가 있으며, 따라서 실제피부의 변화뿐만 아니라 혈관 기능까지 분석이 가능한 장점이 있다.
평가 1. 인공피부 모델의 혈관 형성 관찰
상기 방법으로 제조된 인공피부의 혈관도입 여부를 확인하기 위하여, 형광 현미경(fluorescence microscope)으로 관찰해 본 결과(도 4 참조), 혈관세포가 진피층을 향하여 z축 방향으로 혈관 형성이 되고 잘 유지되고 있는 것을 확인하였다.
혈관구조를 도입하고 진피 및 표피층까지의 전층 인공피부를 제조 배양하였음에도, 혈관구조가 무너지지 않고 유지되고 있었다. 이는 섬유아세포의 혈관신생인자(angiogenic factor)가 피하조직 층에 전달이 되고 이로 인해 혈관내피세포가 vessel guide로 sprouting되었기 때문이다. 즉 피하조직 층과 진피층이 접하게 제조되었다는 것이다(도 5 참조). 만약 진피층이 없다면, 즉 접하게 제조되지 않는다면, 본 발명의 기능성 혈관은 형성되지 않는다. 따라서 본 발명의 인공피부는 피부 상태에 따라 혈관이 유도되는 특징이 있다.
평가 2. 인공피부 모델의 혈관 기능성 관찰
또한 이렇게 제조된 혈관이 혈액뿐만 아니라 배양액 등을 통과시켜 혈관을 통해 영양분, 유효물질, 약물 등을 주입할 수 있는지를 확인하기 위하여, 혈관의 기능성을 알아보는 실험을 하였고, 그 과정과 결과는 도 6에 도시되어 있다.
제조한 혈관화 된 인공피부를 ex-vivo실험인 CAM(chorioallantoic membrane) assay를 통해 관찰하였으며, 실제로 CAM에 도입된 혈관화 된 인공피부는 문합(grafting)이 되어, 형성된 혈관 속으로 혈액이 흐르고 다른 물질들도 주입할 수 있음을 확인할 수 있었다.
앞에서 살펴본 바와 같이, 본 발명은 진피층으로의 구조적인 혈관을 도입한 새로운 전층 인공피부를 설계 또는 제조하였고 이의 기능성도 모두 확인하였다. 혈관화 된 인공피부를 이용한다면, 혈류의 흐름, 피부 온도의 변화, 유효성분으로 인한 혈관 변화, 수분 변화, 및 세포외기질의 재조합 또는 생성 등의 영향을, 실제피부와 거의 동일한 수준으로 평가할 수 있으며, 나아가 동물대체 시험법의 시료물로도 사용 가능하다.
비록 본 발명이 상기에 언급된 바람직한 일 실시예로서 설명되었으나, 발명의 요지와 범위로부터 벗어남이 없이 다양한 수정이나 변형을 하는 것이 가능하다.
Claims (8)
- (1) 3D 바이오프린팅 기술을 사용하여 vessel guide가 도입된 피하조직 층을 제조하는 단계;
(2) 피하조직층 위에 콜라겐(collagen)과 섬유아세포(HDF)를 사용한 진피층을 제조하는 단계;
(3) 혈관내피세포의 bridge gel로 스프라우팅(sprouting)하는 단계;
(4) 진피층 위에 단일층 표피세포(HEK)를 시딩(seeding)하고 진피층으로 혈관이 유도되는 단계; 및
(5) 표피층의 증식 및 분화하는 단계;를 포함하는 혈관화 된 인공피부의 제조방법 - 제 1항에 있어서, (1) 단계의 피하조직 층에 도입된 vessel guide의 형상은 원형 모양이고, 개수는 가로와 세로가 n x m 으로 설계된 것을 특징으로 하는 제조방법
- 제 2항에 있어서, 바람직하게는 상기 vessel guide의 가로와 세로의 수는 동일하게 설계된 것을 특징으로 하는 제조방법
- 제 1항에 있어서, 피하조직 층은 히알루론산, 젤라틴, 피브리노겐, 트롬빈, 및 HUVEC(혈관내피세포)로 구성되어 있는 것을 특징으로 하는 제조방법
- 제 1항에 있어서, 피하조직 층은 HUVEC를 이용한 blood vessel을 제조하고, blood vessel에 z축으로의 vessel guide가 제조되며, 나머지 공간은 피브린 패턴(fibrin pattern)으로 덮어 채워지는 것을 특징으로 하는 제조방법
