Description
〔1〕リン酸カルシウムを含むことを特徴とする、植物親和性材料。
〔2〕前記リン酸カルシウムがハイドロキシアパタイトであることを特徴とする、〔1〕に記載の植物親和性材料。
〔3〕前記ハイドロキシアパタイトが焼結体であることを特徴とする、〔2〕に記載の植物親和性材料。
〔4〕前記ハイドロキシアパタイトがナノ粒子であることを特徴とする、〔2〕または〔3〕に記載の植物親和性材料。
〔5〕前記リン酸カルシウムが、基材表面に付着してなることを特徴とする、〔1〕〜〔4〕の何れかに記載の植物親和性材料。
〔6〕前記植物親和性材料は、シート状であることを特徴とする、〔5〕に記載の植物親和性材料。
〔7〕〔1〕〜〔6〕の何れかに記載の植物親和性材料を用いることを特徴とする、植物組織の接合方法。
本明細書における「植物親和性材料」とは、植物細胞と親和性を有し、植物細胞と複合体を形成でき、かつ、植物細胞と植物細胞との接着を補助する材料のことを意図する。それ故に、植物親和性材料は、植物の繁殖技術または育種技術に適用される。植物細胞と植物細胞との接着を補助するとき、互いに接着する植物細胞は、同種の植物に由来するものであってもよいし、異種の植物に由来するものであってもよい。
本発明の一実施形態における植物親和性材料では、リン酸カルシウムが、基材表面に付着していてもよい。なお、本発明の一実施形態における植物親和性材料では、リン酸カルシウムが、基材表面の全体に付着していてもよいし、基材表面の一部に付着していてもよい。
本発明の一実施形態における植物組織の接合方法は、上記植物親和性材料を用いることが好ましい。
アンモニア水にてpHを12.0に調節したCa(NO3)2水溶液(42mM、800mL)を、コンデンサーおよび半月状攪拌翼が設置された1Lフラスコに注ぎ込み、80℃に保持した。
ポリ乳酸不織布膜(生分解性シート)(ユニチカ製、商品名:テラマックス、厚さ100μm(実測値)、目付20g/m2、引裂強力3N)をアンモニア水溶液(pH12.0)に30分間浸漬することによって、ポリ乳酸不織布膜の表面にカルボキシル基を導入した。上記処理後のポリ乳酸不織布膜を、水およびエタノールにて洗浄した。
〔3.植物細胞と植物親和性材料との接着性試験〕
栽培期間7〜10日のトマト(品種:マイクロトム(Micro−Tom)、カリフォルニア大学デービス校から分譲)の子葉を用いて、カルス由来細胞を採取し、MS液体培地で調製し、トマト懸濁細胞を得た。さらに、2cm×2cmのHApシートを加えて培養した。
〔4.接ぎ木試験〕
栽培期間20〜25日のトマト(品種:マイクロトム(Micro−Tom)、カリフォルニア大学デービス校から分譲)の茎をメスで切断し、茎の切断上部および茎の切断下部をそれぞれ、穂木および台木とした。その後、穂木の切断面を斜めに切断し、台木の切断面に縦に切れ目を入れた。上記穂木の切断面に1cm×1cmのHApシートを付着させ、余分なシート部分を切除し、上記HApシートを付着させた穂木を、上記台木の切れ目にはめ込んだ(図5参照)。台木と穂木とのはめ込み部分が完全に覆われるように、台木および穂木をパラフィルム(Bemis Flexible Packaging社製)で巻いて固定し、HApシート処理区とした。上記シートを挟んでから毎日、HApシート処理区において、台木と穂木とが活着しているかどうかを観察した。

- リン酸カルシウムを含むことを特徴とする、植物親和性材料。
- 前記リン酸カルシウムがハイドロキシアパタイトであることを特徴とする、請求項1に記載の植物親和性材料。
- 前記ハイドロキシアパタイトが焼結体であることを特徴とする、請求項2に記載の植物親和性材料。
- 前記ハイドロキシアパタイトがナノ粒子であることを特徴とする、請求項2または3に記載の植物親和性材料。
- 前記リン酸カルシウムが、基材表面に付着してなることを特徴とする、請求項1〜4の何れか1項に記載の植物親和性材料。
- 前記植物親和性材料は、シート状であることを特徴とする、請求項5に記載の植物親和性材料。
- 請求項1〜6の何れか1項に記載の植物親和性材料を用いることを特徴とする、植物組織の接合方法。
