本発明の範囲内において、式(I)の化合物またはその塩は、互変異性の現象を示す場合があり得ることは理解されよう。互変異性は、2つの原子間で水素原子が交換され、該2つの原子のいずれかと該水素原子が共有結合を形成することによって容易に相互変換し得る2種類の化学物質化合物によるものである。互変異性体化合物は、互いに動的平衡の状態で存在するため、同じ化合物の異なる異性体形態とみなされ得る。本明細書における式の図は、考えられ得る互変異性体形態のうちの1つだけを表したものであり得ることは理解されよう。しかしながら、また、本発明は任意の互変異性体形態を包含しており、式の図において使用したいずれか1つの互変異性体形態だけに限定されないことも理解されよう。本明細書における式の図は、考えられ得る互変異性体形態のうちの1つだけを表したものであり得、本明細書には、本明細書において便宜的に図式的に示している形態だけでなく、図示した化合物の考えられ得るすべての互変異性体形態が包含されていることは理解されよう。例えば、互変異性は、波線で図示のように結合されるピラゾリル基によって示され得る。どちらの置換基も4−ピラゾリル基と称され得るが、各構造において異なる窒素原子が水素原子を担持していることは明白である。
本発明の化合物が1つ以上のキラル中心を含むものである場合、該化合物は、1種類の実質的に純粋なエナンチオマー形態もしくはジアステレオマー形態またはラセミ混合物で存在するものであり得、該形態または該混合物として単離され得ることは理解されよう。したがって、本発明は、本発明の化合物の考えられ得る任意のエナンチオマー、ジアステレオマー、ラセミ化合物またはその混合物を包含している。
本明細書において開示し、請求項に記載した任意の化合物を、スフィンゴシン−1−リン酸受容体の阻害における有効性および種々の細胞アッセイにおける有効性について、上記の手順または科学文献にみられる手順を用いて評価することは、通常の技能の範囲内である。したがって、当業者は、必要以上に実験を行なうことなく、請求項に記載の任意の化合物を調製し、評価することができよう。
公開されたツール化合物は有益な概念実証をもたらすものであるが、アミノリン酸エステルであるスフィンゴ脂質アナログは、一般に、最適な有用性に必要な速度論および安定性を有していない。本発明者らの最近の刊行物には、S1PR1をS1PR3結合ポケットから分離する鍵となる態様が記録されている(7,46)。S1PR1モジュレーターの全身性「免疫抑制性」作用は、理論的には、局所環境における炎症、例えば、EAEにおけるCNSの炎症またはインフルエンザ感染による肺の炎症を減弱させるのに有用であり得る(25,47)が、S1PR1アゴニストは、徐脈(6,7)およびその肺の微小血管透過性を増大させる可能性(48)(49)のため敗血症のリスクをもたらす可能性があり得る。したがって、敗血症における全身性の炎症を、S1P1親和性のない選択的S1PR3アンタゴニストにより減弱させることが望ましい。

- XがNRであり、Lが結合であるか、またはC(=O)である、請求項12に記載の化合物。
- Ar1およびAr3がフェニルであるか、Ar2がピリジルもしくはキノリルであるか、またはその任意の組合せである、請求項12に記載の化合物。
- XがNRであり、Lが結合であるか、またはC(=O)である、請求項15に記載の化合物。
- Ar2がフェニルであるか、Ar1がピリジルもしくはキノリルであるか、Ar3がピリジルもしくはキノリルであるか、またはその任意の組合せである、請求項15に記載の化合物。
- 請求項1に記載の化合物および薬学的に許容され得る賦形剤を含む医薬組成物。
- 有効量の請求項1に記載の化合物を投与することを含む、心肺疾患をこれに罹患している患者において処置する方法。
- 患者に有効量の表1の化合物を投与することを含む、心肺疾患をこれに罹患している患者において処置する方法。
- 患者に有効量の表2または3の化合物を投与することを含む、心肺疾患をこれに罹患している患者において処置する方法。
- 前記疾患が喘息または慢性閉塞性肺疾患である、請求項19に記載の方法。
- 前記疾患が敗血症を含む、請求項19に記載の方法。
- 前記疾患が冠動脈アテローム性硬化である、請求項19に記載の方法。
- 前記疾患が、気管支収縮、肺線維症、冠動脈収縮、樹状細胞によるサイトカイン増幅または播種性血管内凝固障害の発生を特徴とする臨床症候群を含む、請求項19に記載の方法。
- 前記疾患がインフルエンザ感染による炎症を含む、請求項19に記載の方法。
