Abstract

7からなる融合タンパク質またはHPV−16およびHPV−18に由来するE6、E7およびL2からなる融合タンパク質、キメラL1−E7 VLP、HPV−16およびHPV−18のE6およびE7またはウシパピローマウイルス(papilloma virus)E2のいずれかを発現する組換えワクシニアウイルス(vaccinia virus)、HPV−16およびHPV−18のE6およびE7のCTLエピトープを発現するDNAワクチン、HPV−16 E7抗原を分泌する弱毒化生リステリア菌(Listeria monocytogenes)(Lm)、ならびにHPV−16 E6およびE7ペプチドを含む合成ロングペプチド(SLP)を用いた臨床試験が実施されている。これらの手法の一部が限定はされないが幾つかの臨床有効性を示す一方、大部分が奏功しておらず、これは現行の方法の改善が必要であることを示している。

野生型HPV16 E6およびE7タンパク質は、特定のアッセイにおける形質転換活性(例えば軟寒天アッセイにおけるコロニー形成)として明白な腫瘍化の可能性を有する(Massimi and Banks,2005,Methods Mol Med 119:381−395)。実施例1に記載のE6E7SHポリペプチドは、再整理された様式でのE6およびE7タンパク質の断片を含む。これは、例えばかかるアッセイにおける野生型E6およびE7タンパク質のいずれかと比べて有意に低下した形質転換活性により測定可能である通り、腫瘍化の可能性を排除することが期待される。

発明者は、実施例1に記載のようなDNAベクターおよびアデノウイルスベクターを調製している。
L/6マウスまたはBalb/cマウスよりも広範な細胞免疫応答がCB6F1で誘導される場合の初期実験に基づき、免疫応答を測定するのにCB6F1マウス株を用いた。別々の実験において、マウスを本発明の分子をコードするDNAベクターで免疫し、細胞免疫応答を測定した。HPV16 E7に特異的な免疫応答は、E6E7SHを発現するDNAプラスミドで免疫したマウスにおいて測定することができた(図5)。
本発明のデザイナー配列を発現するアデノウイルスベクターが非ヒト霊長類における免疫応答を誘導する能力を評価するため、E2E6E7SHを発現するアデノベクター(Ad26)または導入遺伝子をコードしないアデノベクター(Empty)を1×1011vpの用量で筋肉内注射することにより、アカゲザル(rhesus macaques)を免疫した。免疫の8週後、同じ抗原を発現するAd26ベクターによる免疫により免疫応答をブーストした。16週後、動物は、同じ抗原を発現するAd35ベクターのもう1回の注射を受けた。血液試料は数回の時点で採取し、単離した白血球をHPV16 E2、E6もしくはE7に対応するペプチドプールで一晩刺激した。特異的応答をIFNγELISPOTにより測定した。データを図11に示す。さらにプライム免疫の10週後および18週後、本発明における新規な接合に及ぶペプチドに特異的な細胞免疫応答を評価した。IFNγ応答の誘導は、すべての動物において<50SFU/1×106PBMCの検出限界未満であった(データは示さず)。
本発明のポリペプチドは、動物においてHPV16に特異的な細胞免疫応答を誘導することができ、HPV16 E6および/またはE7を発現する細胞に対して治療効果を発揮し得る。治療的免疫、すなわち腫瘍増殖が開始してからの免疫は、治療用HPVワクチン候補の有効性を実証するのに用いることができる。Ad26およびAd35ベクターの治療効果を、TC−1細胞が注射されたマウス(HPV16 E6およびE7を発現する
マウス細胞)において試験した(Lin et al.,1996,Cancer Res 56:21−6)。TC−1細胞は、マウスにおける皮下注射後、数日から数週以内に固形腫瘍を形成することになる。ワクチンの不在下では、腫瘍は急速に増殖し、30日以内に1000mm3の所定のサイズに達した(パネルDおよびE)。このサイズに達したら、倫理的理由でマウスを屠殺する。
強力で構成的に活性なプロモーターの制御下でアデノウイルスベクターに挿入される導入遺伝子が、導入遺伝子産物の特性に応じてベクター産生に負の影響を与え得ることは先行的に報告されている(Yoshida & Yamada,1997,Biochem
Biophys Res Commun 230:426−30;Rubinchik
et al.,2000,Gene Ther 7:875−85;Matthews
et al.,1999,J Gen Virol 80:345−53;Edholm et al.,2001,J Virol 75:9579−84;Gall et
al.,2007,Mol Biotechnol 35:263−73)。導入遺伝子依存性ベクターの生産性での課題の例として、非効率的なベクターの救出および増殖、低い最終ベクターの収量、また深刻な場合では欠陥のある導入遺伝子カセットを有するウイルス突然変異体の迅速な増殖が挙げられる。これらの課題を解決するため、複数の試験において、ベクター・導入遺伝子の発現をプロデューサー細胞内でのベクター複製中に抑制するための可能性が探索された(Matthews et al.,1999,J Gen Virol 80:345−53;Edholm et al.,2001,J Virol 75:9579−84;Gall et al.,2007,Mol Biotechnol 35:263−73;Cottingham et al.,2012,Biotechnol Bioeng 109:719−28;Gilbert et al.,2014,J Virol Methods 208:177−88)。これに関して、異なる抑制系がAdベクターとの関連で先行的に装備されており、当然ながら、異なるタイプの(阻害性)導入遺伝子をコードするベクターにおいてベクターの生産性および遺伝的安定性を改善することを示している。
用により、本明細書に記載されるようなHPV由来の抗原を発現するAdベクターの生成特性が改善され得るか否かを検討しようと努力した。この目的のため、発明者は、2つの既存のリプレッサー−オペレーター系、すなわちTetR/TetO(Yao & Eriksson,1999,Hum Gene Ther 10:419−22、欧州特許第0990041B1号明細書)およびCymR/CuO(Mullick et al.,2006,BMC Biotechnol 6:43)を我々のアデノウイルスベクタープラットフォームに装備した。TetR/TetO系およびCymR/CuO系の双方が、アデノウイルスベクターの生産性をベクター複製中でのベクター・導入遺伝子のサイレンシングを介して改善するため、他者により先行的に用いられている(Gall et al.,2007,Mol Biotechnol 35:263−73;Cottingham et al.,2012,Biotechnol Bioeng 109:719−28;Gilbert et al.,2014,J Virol Methods 208:177−88)。これら2つの系の装備は、TetOまたはCuO配列のいずれかを有するCMVプロモーターの制御下での目的の遺伝子を発現するアデノウイルスベクターの作製を含んだ。さらに、その装備は、各々の同族のリプレッサータンパク質(すなわちTetRまたはCymR)を安定的に発現する細胞株の作製を必要とした。
をコードする配列番号16)によって置換される。安定的細胞株の作製は、主に、CMVTetOまたはCMVCuO駆動遺伝子の発現を抑制する能力がある細胞クローンについてスクリーニングするための一過性遺伝子導入に基づくアッセイを用いて、pcDNA(商標)6/TRの供給業者による説明に従って実施した。得られたPER.C6/TetRおよびPER.C6/CymR細胞株では、これらの細胞内でのベクター複製中に導入遺伝子発現を抑制するそれらの能力について分析した。オペレーター含有CMVプロモーターの制御下でGFP−Lucを発現するベクターを用いて実施された実験によると、各々のオペレーター配列に対応するリプレッサーを発現する細胞株中での完全なウイルス複製サイクル全体を通じて、ルシフェラーゼ遺伝子発現の少なくとも10倍の低下が示された(データは示さず)。これにより、PER.C6/TetRおよびPER.C6/CymR細胞株がアデノウイルスベクターの複製との関連でベクター・導入遺伝子の発現を抑制する能力があることが確認された。
イルスベクターの遺伝的安定性に関する主要な課題について認めている。例えば、PER.C6上でこのベクターが数継代経た後、ベクター集団の大半がHPVAgコード配列内に大規模な欠失を保有する突然変異体ベクターからなることが認められた(データは示さず)。
