Abstract

本发明涉及一种基于重组减毒单增李斯特菌的宫颈癌治疗性疫苗。本发明以减毒单增李斯特菌(Listeria monocytogenes,在下文中简称Lm)作为载体,利用李斯特菌独特的能在宿主吞噬细胞胞内生长,是天然的T细胞免疫活化佐剂的特性,有效提高在肿瘤微环境中的特异性免疫应答,打破机体的免疫耐受,从而清除机体持续性病毒感染,达到清除病灶,遏制肿瘤发展的良好治疗效果。其优势在于弥补了核酸类及蛋白质多肽类疫苗免疫原性较低的缺点,减毒的李斯特菌保留了原始菌株能在胞内生长,完成抗原递呈的特性的同时又具有更高的安全性。

技术领域
本发明属于生物技术领域,涉及一种基于重组减毒单增李斯特菌的宫颈癌治疗性疫苗。
背景技术
宫颈癌严重威胁着全球女性的健康,是死亡率仅次于乳腺癌的妇科恶性肿瘤。研究表明,HPV是宫颈癌的主要危险因素,有超过90%的宫颈癌患者伴随着高危型HPV的感染,其中以16型和18型为主。宫颈癌90%的癌症类型为宫颈鳞癌,其主要致病型为HPV16型。HPV的主要致癌因子为E6和E7蛋白,E6通过抑制肿瘤抑制因子p53的活性破坏正常的细胞周期调控,促使细胞永生化;E7蛋白通过结合并降解pRb蛋白使其失去抑癌作用。E6、E7自HPV感染早期持续高表达,且仅表达于HPV感染组织中使其成为制备宫颈癌治疗性疫苗的理想靶标。
治疗性疫苗与传统的治疗手段,如外科手术,放化疗相比具有伤害小、毒性低的特点。目前治疗性疫苗的存在形式主要有三种:蛋白质多肽疫苗、病毒(细菌)疫苗以及核酸疫苗。其成功的关键是引起足够的免疫应答,但是目前的核酸类及蛋白质多肽类疫苗免疫原性较低,缺陷明显。
发明内容
有鉴于此,本发明的目的在于提供一种基于重组减毒单增李斯特菌的宫颈癌治疗性疫苗。
为达到上述目的,本发明提供如下技术方案:
基于重组减毒单增李斯特菌的宫颈癌治疗性疫苗,其有效成分为携带HPV16型E6E7融合抗原表位肽基因的重组减毒单增李斯特菌。
优选的,所述减毒单增李斯特菌为单增李斯特菌完整敲除actA和plcB这两个毒力基因而得。
优选的,所述重组减毒单增李斯特菌的制备方法,是以减毒单增李斯特菌为载体,携带HPV16型E6E7融合抗原表位肽基因而得。
进一步优选的,具体步骤如下:
(1)合成HPV16型E6E7融合抗原表位肽基因片段;
(2)构建打靶质粒;
(3)制备感受态细胞LmΔactAplcB-lacZ;
(4)利用步骤(2)制备的打靶质粒对步骤(3)制备的感受态细胞进行电转化;
(5)LmΔactAplcB-lacZ与打靶质粒同源重组杂交培养、筛选验证。
更进一步优选的,步骤(1)的具体方法是:从NCBI上查询HPV 16型E6蛋白和E7蛋白氨基酸序列,再通过软件(如使用CTLPred和NetMHC 4.0软件分析序列中可能的CTL表位;ProPred和NetMHCIIpan 3.1软件分析序列中可能的Th表位),从中筛选出优势T细胞抗原表位依次排列,然后根据李斯特菌偏好密码子表进行密码子优化得到优化后的序列,再在其上游加入HindⅢ酶切位点(AAGCTT),下游加入XhoⅠ酶切位点(CTCGAG),将上述设计好的序列最后通过合成直接获得。其中,密码子优化方法是将氨基酸序列提交到网站(如http://www.jcat.de/),然后选择单增李斯特菌偏好密码子优化选项即可实现;DNA序列的合成方法是:使用ABI 3900-Thermo Fisher Scientific等型号的基因合成仪直接合成DNA序列,共866bp,如SEQ ID NO.1所示。
更进一步优选的,步骤(2)的具体方法是:用限制性内切酶HindⅢ和XhoⅠ双酶切步骤(1)获得的融合抗原表位肽基因片段,同时用相同的限制性内切酶酶切质粒pCW203(专利CN103074361B),经连接、转化等技术将融合抗原表位肽基因片段插入pCW203HindⅢ和XhoⅠ酶切位点,得到中间质粒pCW203-E6E7,如SEQ ID NO.2所示,通过PCR验证、提质粒验证、酶切验证证实中间质粒的成功构建。切下中间质粒pCW203-E6E7的BamH I酶切位点与Xho I酶切位点之间的片段插入重组质粒pCW180的BamH I酶切位点与Xho I酶切位点之间,得到打靶质粒pCW180-E6E7,如SEQ ID NO.3所示;其中,所述重组质粒pCW180的基因序列为序列表中SEQ ID NO.13所示。
更进一步优选的,重组质粒pCW180的制备方法如下:
(2-A)将上游携带SpeⅠ、下游携带NotⅠ的Lm mpl基因(Genbank ID:DQ054595.1)插入质粒pCW154(专利CN103074361B)的SpeⅠ和NotⅠ酶切位点之间,得到第一中间重组质粒pCW160,所述第一中间重组质粒pCW160的基因序列为序列表中SEQ ID NO.4所示;
(2-B)将上游携带XbaⅠ、下游携带NotⅠ的Lm orfBAldh基因(Genbank ID:M82881.1)插入上述步骤(2-A)得到的第一中间重组质粒pCW160的XbaⅠ和NotⅠ酶切位点之间,得到第二中间重组质粒pCW170,所述第二中间重组质粒pCW170的基因序列为序列表中SEQ ID NO.5所示;
(2-C)用NotⅠ酶切质粒pCW154,得到两头为NotⅠ酶切位点的一段基因,插入上述步骤(2-B)得到的第二中间重组质粒pCW170的NotⅠ酶切位点,得到重组质粒pCW180;所述重组质粒pCW180的基因序列为序列表中SEQ ID NO.13所示。
更进一步优选的,步骤(3)的具体方法是:将LmΔactAplcB-lacZ用含0.5mol/L蔗糖的BHI液体培养基进行培养,定期测定A600,当A600=0.4时,加入盘尼西林G,使其终浓度为12.5μg/ml继续培养;当A600=0.7时,离心,洗涤,分装,保存即可。

更进一步优选的,步骤(3)的具体方法是:接种LmΔactAplcB-lacZ于15ml含0.5mol/L蔗糖的BHI液体培养基,37℃220rpm培养12~16小时;将上述菌液转移至250ml含0.5mol/L蔗糖的BHI液体培养基,定期测定A600,当A600=0.4时,加入盘尼西林G,使其终浓度为12.5μg/ml继续培养;当A600=0.7时,倒入50mL离心管,4℃10000rpm离心5分钟,弃上清;以0.5mol/L冰冷蔗糖溶液反复洗涤沉淀,每次加入20ml 0.5mol/L蔗糖溶液,4℃10000rpm离心10分钟,弃上清,洗涤3次;每管加0.5mol/L蔗糖的300μl重悬,按50μl/支分装预冷无菌EP管,-80℃保存。
进一步优选的,步骤(4)的具体方法是:取打靶质粒电转至LmΔactA/plcB-lacZ感受态细胞中,电转完成后加入BHI肉汤培养2小时;转化液涂布于红霉素BHI琼脂平板,通过蓝白斑筛选出单个蓝色菌落,纯培养后进行质粒提取和PCR验证。
更进一步优选的,步骤(4)的具体方法是:将制备的感受态细胞从-80℃冰箱中取出置于冰盒中,待其完全融化后,取5μl打靶质粒按1/10的比例用-20℃预冷枪头缓慢加至感受态细胞LmΔactAplcB-lacZ,同时作ddH2O阴性对照;温和混匀后,冰浴5分钟,再全部转入电转杯,冰浴5分钟,于电转仪中进行电转;电转完成后,冰浴5分钟,用37℃预热的枪头加入37℃预热的BHI肉汤750μl,混匀后全部转至EP管,摇床30℃,150rpm培养2小时;用无菌L形玻棒将菌液全量涂布于含红霉素1~5μg/ml、X-gal 20~60μg/ml的BHI琼脂平板(以下简称BHI-Ery-X-gal,BEXI平板),30℃培养48~72小时后,阳性菌为蓝色菌落,于BEXI平板上进行纯培养,30℃培养24小时,进行提质粒及PCR验证。

进一步优选的,步骤(5)的具体方法是:将携带打靶质粒的单增李斯特菌,在42℃和30℃连续传代,利用同源重组杂交原理和基因打靶技术,将打靶质粒携带的目的片段整合至李斯特菌中,利用蓝白斑及红霉素敏感性筛选出可疑菌株;然后进行PCR筛选和基因测序验证。
更进一步优选的,传代培养的具体方法是:将已电转入打靶质粒的LmΔactAplcB-lacZ划线接种BEXI平板,42℃传代培养,挑取单个蓝色菌落接种BHI肉汤,30℃传代培养,待传到肉汤第3代开始,每代取培养物加入BHI稀释,分别涂布稀释菌液100μl于BEXI和BXI(含X-gal 20~60μg/ml的BHI琼脂平板)平板,30℃培养24小时。
更进一步优选的,传代培养的具体方法是:将已电转入打靶质粒的LmΔactAplcB-lacZ划线接种BEXI平板,42℃传代培养2~3代(每代培养时间为48小时),挑取单个蓝色菌落接种5ml BHI肉汤,30℃,200rpm传代培养6代(每代培养时间为24小时);待传到肉汤第3代开始,每代取40μl培养物加入360μl BHI肉汤(1:10稀释),多次连续1:10稀释直到稀释106倍,分别涂布106倍稀释的菌液100μl于BEXI和BXI平板,30℃培养24小时;阳性菌落LmΔactAplcB-HPV在BXI平板上长成白色菌落,在BEXI平板上不生长。
更进一步优选的,PCR筛选的具体方法是:对蓝白斑及红霉素筛选结果正确的上述可疑重组菌进行以下三组基因片段的扩增:抗红霉素基因、靶抗原基因HPV16E6E7以及actA基因,鉴定相应疫苗菌株。

本发明所制备的宫颈癌治疗性疫苗携带HPV16型E6E7融合抗原肽表位基因,以减毒单增李斯特菌(Listeria monocytogenes,在下文中简称Lm)作为载体,利用李斯特菌独特的能在宿主吞噬细胞胞内生长,是天然的T细胞免疫活化佐剂的特性,有效提高在肿瘤微环境中的特异性免疫应答,打破机体的免疫耐受,从而清除机体持续性病毒感染,达到清除病灶,遏制肿瘤发展的良好治疗效果。其优势在于弥补了核酸类及蛋白质多肽类疫苗免疫原性较低的缺点,减毒的李斯特菌保留了原始菌株能在胞内生长,完成抗原递呈的特性的同时又具有更高的安全性。
本发明的来源菌是LmΔactAplcB-lacZ,最后导致的减毒是actA和plcB这两个基因的全部基因的缺失减毒,缺失的基因与细菌在胞间的转移和细菌逃离次级溶酶体有关,缺失后细菌毒力减低,与仅缺失溶血素部分基因的减毒方式相比,具有更好的安全性,同时本发明缺失的基因不影响细菌在吞噬细胞内的抗原递呈作用。本发明插入的HPV基因除E7蛋白编码基因外,还加入了E6蛋白编码基因,并且挑选出E6和E7蛋白编码基因中的抗原表位肽基因融合而成,且经过了李斯特菌密码子优化,具有独特性。本发明插入的HPV融合抗原基因可以提高疫苗菌对肿瘤细胞的识别,攻击更多的肿瘤细胞,达到更好的治疗效果。本发明对阳性菌的筛选是通过温度、红霉素和蓝白斑筛选。本发明的蓝白斑筛选通过肉眼识别,简单易行,筛选成功率高。HPV基因插入位点是在mpl基因下游,该位点不影响细菌其他重要蛋白的表达,尽量保留了原菌株的生物活性。
具体如下:
1)针对性:诱导特异性针对宫颈癌细胞的细胞因子,靶向治疗癌组织,不损伤正常细胞。
2)高效:疫苗菌株免疫活性高,诱导高效的细胞因子表达。李斯特菌株本身具有高效活化机体细胞免疫功能的作用,因此携带抗原的重组菌株能激活多种抗肿瘤免疫机制,激活特异的肿瘤杀伤性T细胞,打破免疫抑制重新激活有效的肿瘤杀伤机制。
3)安全:减毒菌株不会在体内长期存在,不会引起感染,在我们的临床前试验中表现了很好的耐受性,并经其他类似临床试验证实了其安全性,且能跟其他肿瘤治疗手段很好的兼容,如鸡尾酒式免疫抑制剂、外科手术、放疗和化疗。
4)稳定:本发明制备的疫苗菌株携带的抗原基因是整合到菌株基因组中的,抗原基因在菌株内稳定存在,不会丢失。
5)低成本:疫苗的生产成本低、快速、经济,使用标准的细菌发酵技术就可完成疫苗的大规模生产,无需细胞培养、冷链运输和储藏。

图5为重组菌LmΔactAplcB-E6E7的基因组中插入的外源基因及前后基因结构示意图。由此图可知外源基因的插入位点,重组后的细菌基因组敲掉了actA和plcB基因。插入的外源基因在自带的启动子phly的作用下表达融合蛋白,融合蛋白具有分泌信号肽、GP33和GP61抗原表位肽标签、HA和VSV-G western blot检测标签。
图6为C57BL/6小鼠接种重组菌株后的生存曲线。为C57BL/6小鼠分别尾静脉接种不同剂量的LmΔactAplcB-E6E7,即2.7×107CFU/只、2.4×108CFU/只和2.6×109CFU/只3个接种剂量,连续10d观察小数的生存情况。计算各组生存率,根据改良寇氏法计算得LmΔactAplcB-E6E7的LD50为1.3×108CFU/只。
图7为宫颈癌模型C57BL/6小鼠经尾静脉注射重组单增李斯特菌(LmΔactAplcB-E6E7)免疫治疗后不同时间点的肿瘤体积。C57BL/6小鼠第0天右侧腹皮下接种TC-1细胞1×105个/只造模,对造模小鼠于TC-1细胞接种后第7天、第14天以及第21天进行三次免疫治疗,记录各组小鼠的肿瘤体积。图中a为模型小鼠自TC-1细胞接种后第7天各组肿瘤体积;b为各组模型小鼠免疫治疗后不同时间点的肿瘤体积。其中A为生理盐水对照组,B为Lm载体菌(LmΔactAplcB-lacZ)对照组,C为疫苗治疗组。第一次免疫治疗时(第7天)各组小鼠肿瘤体积均无统计学差异。经第二次(第14天)及第三次肿瘤治疗(第21天)后观察至第42天(当小鼠肿瘤长径为20mm时处死小鼠),生理盐水对照组和载体菌对照组均无小鼠存活,治疗组存活小鼠肿瘤大小得到有效控制,其中有两只小鼠完全治愈。
图8为宫颈癌模型小鼠经尾静脉注射免疫治疗后各组的生存情况。对宫颈癌模型小鼠在TC-1细胞接种后第7天、第14天以及第21天进行三次免疫治疗后各组小鼠的生存情况,其中A为生理盐水对照组,B为Lm载体菌对照组,C为疫苗治疗组。治疗组与各组进行生存分析,生理盐水与治疗组相比(P<0.001),载体对照组与治疗组相比(P<0.001),治疗组明显延长了宫颈癌模型小鼠的生存时间。

具体实施方式
下面将结合附图,对本发明的优选实施例进行详细的描述。
实施例1.疫苗菌LmΔactAplcB-HPV的制备
(1)HPV16E6E7基因片段合成
从NCBI上查询HPV 16型E6蛋白和E7蛋白基因,利用网上在线软件分析预测其优势T细胞抗原表位,选择并组合成融合抗原表位肽基因,然后根据李斯特菌密码子进行相应优化,并在上游加入HindⅢ酶切位点,下游加入XhoⅠ酶切位点,最后通过合成直接获得DNA序列(以公司提供克隆质粒pUC57-E6E7的形式获得),片段长度为866bp(如SEQ ID NO.1所示)。
(2)构建打靶质粒
①中间质粒pCW203-E6E7的构建
按照质粒说明书提取质粒pUC57-E6E7和pCW203,以30μL Elution Buffer洗脱。
pUC57-E6E7和pCW203用HindⅢ和XhoⅠ双酶切,酶切体系20μL:pUC57-E6E7或pCW203<1ng,HindⅢ1μL,XhoⅠ1μL,10×NEB Buffer 2μL,ddH2O将体系补足20μL。37℃水浴1h酶切质粒,载体片段需加0.5μL CIAP,37℃水浴30min去磷酸化。将酶切混合物与6×Loading Buffer混合,电泳(1%琼脂糖,94V)并胶回收相应长度的片段(pUC57-E6E7回收酶切后的小片段即融合抗原表位肽基因片段,长度为866bp;pCW203回收酶切后的大片段即载体片段,长度约>10Kb)),以30μL Elution Buffer洗脱。
分别取融合表位肽抗原基因片段与载体片段按照连接体系:载体50ng,插入片段摩尔数:载体片段摩尔数=5:1,T4Ligase 1μL,10×Ligase Buffer 5μL,ddH2O将体系补足10μl,22℃连接1h。将连接产物按体积1:10与大肠杆菌DH5α的感受态细胞轻柔混匀,冰浴30min,42℃热击45s,冰浴3min,加入预热的SOB肉汤500μL,混匀后于37℃,180rpm培养1h,涂布于LA平板(LB-Amp平板:含100μg/mL Amp的LB平板),37℃培养16-20h。
PCR筛选:以煮沸法提取的细菌基因为模板,扩增目的条带HPV16E6E7,引物HPV-f/r(f:5’-CATCAAAAACGTACAGCAATG-3’,r:5’-TGGTTTTTGAGAACAAATTGG-3’),反应循环条件为:94℃3min→(94℃1min,47℃30s,72℃1min)×30个循环→72℃10min→4℃;电泳观察PCR结果(电泳条件:1%琼脂糖凝胶;90V,上样量:5μl PCR产物),预期结果:HPV16E6E7片段约852bp。
酶切验证:PCR筛选验证的阳性菌提取质粒,按照上述酶切体系以HindⅢ和XhoⅠ进行酶切,将酶切混合物与6×Loading Buffer混合进行电泳(1%琼脂糖,94V)。
测序验证:提取PCR及酶切验证符合预期的质粒送测序公司测序。将测序验证后完全正确的携带阳性质粒的大肠杆菌保存于-80℃。
②打靶质粒的构建
提取质粒pCW180,以30μL Elution Buffer洗脱。将质粒pCW180及中间质粒pCW203-E6E7分别用限制性内切酶BamHⅠ和XhoⅠ按照(2)①的体系混匀(将HindⅢ酶替换为BamHⅠ酶),进行酶切和去磷酸化,电泳后胶回收pCW203-E6E7酶切后的小片段即插入片段(931bp)及pCW180载体骨架(酶切后长片段,长度约8621bp)。按照(2)①的体系混匀进行连接并转化入大肠杆菌DH5α,涂布于LA平板,对长出的单个菌落进行PCR筛选、BamHⅠ和XhoⅠ双酶切验证以及测序验证,具体操作同实施例(2)①(将HindⅢ酶替换为BamHⅠ酶)。将测序验证后携带阳性质粒的大肠杆菌保存于-80℃。打靶质粒的结构图谱见图1。
(3)电转化用LmΔactAplcB-lacZ感受态细胞的制备
接种LmΔactAplcB-lacZ于15ml含0.5mol/L蔗糖的BHI液体培养基,37℃220rpm培养12~16h。将上述菌液转移至250ml含0.5mol/L蔗糖的BHI液体培养基,定期测定A600,当A600=0.4时,加入盘尼西林G,使其终浓度为12.5μg/ml继续培养。当A600=0.7时,倒入50mL离心管,4℃10000rpm离心5min,弃上清。以0.5mol/L冰冷蔗糖溶液反复洗涤沉淀,每次加入20ml 0.5mol/L蔗糖溶液,4℃10000rpm离心10min,弃上清,洗涤3次。每管加0.5mol/L蔗糖的300μl重悬,按50μl/支分装预冷无菌EP管,-80℃保存。
(4)打靶质粒电转化LmΔactAplcB-lacZ
将制备的感受态细胞从-80℃冰箱中取出置于冰盒中,待其完全融化后,取5μl打靶质粒按1/10的比例用-20℃预冷枪头缓慢加至感受态细胞LmΔactAplcB-lacZ,同时作ddH2O阴性对照。温和混匀后,冰浴5min,再全部转入电转杯,冰浴5min,于电转仪中进行电转(电转条件:电压=1500V,持续时间=5ms,电转杯内径=1mm)。电转完成后,冰浴5min,用37℃预热的枪头加入37℃预热的BHI肉汤750μl,混匀后全部转至EP管,摇床30℃,150rpm培养2h。用无菌L形玻棒将菌液全量涂布于含红霉素1~5μg/ml、X-gal 20~60μg/ml的BHI琼脂平板(以下简称BHI-Ery-X-gal,BEXI平板),30℃培养48~72h后,阳性菌为蓝色菌落,于BEXI平板上进行纯培养,30℃培养24h,进行PCR验证及提质粒验证,如图2和图3。
PCR验证:以煮沸法提取细菌全部基因为模板,扩增目的条带E6E7,引物HPV-f/r(f:5’-CATCAAAAACGTACAGCAATG-3’,r:5’-TGGTTTTTGAGAACAAATTGG-3’),反应循环条件为:94℃3min→(94℃1min,47℃30s,72℃1min)×30个循环→72℃10min→4℃;电泳观察PCR结果(电泳条件:1%琼脂糖凝胶;90V,上样量:5μl PCR产物),预期结果:HPV16E6E7片段约852bp。如图2所示,提示打靶质粒已转入细菌。
按照质粒提取试剂盒说明提取质粒(李斯特菌为革兰阳性菌,SolutionⅠ使用前需加入溶菌酶,溶菌酶终浓度为20mg/L,加入含溶菌酶的SolutionⅠ后重悬细菌,37℃水浴45min),最后以30μl Elution Buffer洗脱后电泳。凝胶成像,观察有无条带以判断重组质粒导入是否成功,质粒大小为9540bp。如图3所示,表明质粒提取成功,长度符合预期,提示打靶质粒已成功转入LmΔactAplcB-lacZ。
(5)LmΔactAplcB-lacZ与打靶质粒同源重组及筛选
①LmΔactAplcB-lacZ与打靶质粒同源重组杂交培养
将已电转入打靶质粒的LmΔactAplcB-lacZ划线接种BEXI板,42℃传代培养2-3代(每代培养时间为48小时),挑取单个蓝色菌落接种5ml BHI肉汤,30℃,200rpm传代培养6代(每代培养时间为24小时)。待传到肉汤第3代开始,每代取40μl培养物加入360μl BHI作1:10稀释,连续稀释106倍,分别涂布106倍稀释的菌液100μl于BEXI和BXI(含X-gal20~60μg/ml的BHI琼脂平板)平板,30℃培养24h。阳性菌落LmΔactAplcB-E6E7在BXI平板上长成白色菌落,在BEXI平板上不生长。
②PCR鉴定LmΔactAplcB-E6E7
细菌基因组提取试剂盒提取LmΔactAplcB-E6E7的基因组DNA作为模板,进行以下三组基因片段的扩增:抗红霉素基因、靶抗原基因盒以及actA基因,根据扩增结果对重组菌LmΔactAplcB-E6E7进行鉴定。结果如图4所示,从左到右依次为A、B、C,其中,A中的1为以LmΔactAplcB-E6E7基因组DNA为模板扩增融合表位肽抗原基因E6E7,其引物及反应条件见(2)①,B中的1为扩增目的条带抗红霉素基因Ery,约1471bp,引物Ery-f/r(f:5’-GATAAGTCGACGATTCACAAAAAATAG-3’,r:5’-AAAACTAGTCCCGGGGCGAATTG-3’),反应循环条件为:94℃3min→(94℃1min,60℃30s,72℃2min)×30个循环→72℃10min→4℃;C中的1为扩增Lm actA基因,约950bp,引物Lm-actA-f/r(f:5’-GCTATAAATGAAGAGGCTTCAGG-3’,r:5’-CTCTTAAATCAGCTAGGCGATC-3’),反应循环条件为:94℃3min→(94℃1min,50℃30s,72℃1min)×30个循环→72℃10min→4℃。经PCR鉴定,LmΔactA/plcB-E6E7携带有E6E7融合抗原表位肽基因,不携带抗红霉素基因,且actA基因被剔除。鉴定结果与预期吻合,表明LmΔactAplcB-E6E7构建成功。
③基因测序验证
PCR鉴定后以细菌基因组DNA作为模板,PCR扩增靶抗原基因HPV16E6E7,对扩增产物测序验证,对测序正确的菌株进行菌种保存。最终得到的重组菌基因组插入的外源基因及插入位点前后的基因结构示意图如图5所示。
(6)LmΔactAplcB-E6E7疫苗候选株LD50的测定
将-20℃保存的LmΔactAplcB-E6E7菌株置37℃水浴融化后,分别取10μl接种于5ml BHI肉汤中,37℃200rpm培养过夜。第二日,取复苏一次的菌液40μl于20ml BHI肉汤中,37℃200rpm培养过夜二次复苏。
取17.5ml二次复苏的各菌液接种350ml BHI肉汤,于37℃200rpm摇床培养,培养至A600值为0.3~0.7之间时,根据李斯特菌生长曲线,确定此时A600值所对应的菌量,并根据预计的细菌接种剂量进行浓缩或是稀释处理:取一定量菌液离心,13000rpm 2min,用生理盐水重悬后,再次离心,再用生理盐水将菌液重悬至染菌剂量,并置于冰上备用。
6~8周龄C57BL/6雌鼠适应性喂养3d后,随机分为3组,每组7只,尾静脉分别接种2.7×107CFU/只、2.4×108CFU/只和2.6×109CFU/只;连续观察10天,统计小鼠死亡、存活情况,绘制各剂量组生存曲线,如图6所示;利用改良寇式法计算LD50,并以0.1×LD50作为最适后续免疫剂量。
实施例2:疫苗菌株对宫颈癌治疗效果评价
(1)TC-1肿瘤细胞培养及宫颈癌模型的建立
用含10%胎牛血清、1%青霉素、链霉素的RPMI1640Medium在37℃ 5%CO2培养箱内培养TC-1细胞,待其密度达到80%左右,用Hank’s液润洗细胞2~3次后,胰酶消化并用PBS重悬计数,将细胞配制成浓度为1×106/ml的细胞悬液,6~8周龄的C57BL/6雌鼠右侧腹皮下注射100μl细胞悬液,建立小鼠HPV感染肿瘤模型,大约7d形成肉眼可见的肿瘤。肿瘤体积根据公式1/2×(a×b2)计算(a,b分别为肿瘤的最长直径和最短直径)。使用SPSS软件对数据进行统计学分析。两样本均值比较采用t检验,P<0.05为差异有统计学意义。如图7a所示,第7天各组小鼠肿瘤体积均匀,各组间无统计学差异。
(2)对建模小鼠进行免疫治疗
实验设立阴性对照组:分别于造模第7、14、21天注射与治疗组同等体积生理盐水(NS);Lm载体菌对照组:分别于造模第7、14、21天注射0.1×LD50的LmΔactAplcB-lacZ(注射剂量为5×106CFU/只),治疗组:分别于造模第7、14、21天注射0.1×LD50的LmΔactAplcB-E6E7(注射剂量为1.3×107CFU/只)。
将33只造模小鼠随机分为3组,每组11只。分别于TC-1细胞注射后第7天进行第一次免疫治疗,按实施例1(6)配制菌液,尾静脉接种菌液量为100μl,NS对照组尾静脉接种等体积NS。第14天进行第二次加强免疫治疗,第21天进行第三次加强免疫治疗,方法和剂量同第一次免疫治疗。每隔2天观察并记录小鼠体重,肿瘤生长情况及生存状况(当肿瘤长径达20mm时处死小鼠),并根据公式1/2×(a×b2)计算肿瘤体积(a,b分别为肿瘤的最长直径和最短直径)。生存期比较采用对数秩检验,P<0.05为差异有统计学意义。结果如图7中的a和b所示,加强免疫治疗后观察至42天,治疗组与Lm载体菌对照组及生理盐水组相比,肿瘤大小得到了有效控制,治疗组有两只小鼠治愈(图7)。治疗组小鼠比Lm载体菌对照组(P<0.001)及阴性对照组小鼠(P<0.001)生存率显著提高。实验制备疫苗株对宫颈癌起到有效的控制和治疗作用。(图8、图9)
最后说明的是,以上优选实施例仅用以说明本发明的技术方案而非限制,尽管通过上述优选实施例已经对本发明进行了详细的描述,但本领域技术人员应当理解,可以在形式上和细节上对其作出各种各样的改变,而不偏离本发明权利要求书所限定的范围。

Claims (3)

1.基于重组减毒单增李斯特菌的宫颈癌治疗性疫苗,其特征在于,其有效成分为携带HPV16型E6E7融合抗原表位肽基因的重组减毒单增李斯特菌;
所述减毒单增李斯特菌为单增李斯特菌完整敲除actAplcB这两个毒力基因而得;
所述重组减毒单增李斯特菌的制备方法,是以减毒单增李斯特菌为载体,携带HPV16型E6E7融合抗原表位肽基因而得;具体步骤如下:
(1)合成HPV16型E6E7融合抗原表位肽基因片段;
(2)构建打靶质粒;
(3)制备感受态细胞LmΔactAplcB-lacZ
(4)利用步骤(2)制备的打靶质粒对步骤(3)制备的感受态细胞进行电转化;
(5)LmΔactAplcB-lacZ与打靶质粒同源重组杂交培养、筛选验证;
所述步骤(1)的具体方法是:从NCBI上查询HPV 16型E6蛋白和E7蛋白氨基酸序列,再通过软件,从中筛选出优势T细胞抗原表位依次排列,然后根据李斯特菌密码子进行相应优化得到优化后的序列,再在其上游加入HindⅢ酶切位点,下游加入XhoⅠ酶切位点,将上述设计好的序列最后通过合成直接获得,共866bp,如SEQ ID NO. 1所示;
所述步骤(2)的具体方法是:用限制性内切酶HindⅢ和XhoⅠ双酶切步骤(1)获得的融合抗原表位肽基因片段,同时用相同的限制性内切酶酶切质粒pCW203,经连接、转化等技术将融合抗原表位肽基因片段插入pCW203 HindⅢ和XhoⅠ酶切位点,得到中间质粒pCW203-E6E7,如SEQ ID NO. 2所示,通过PCR验证、提质粒验证、酶切验证证实中间质粒的成功构建;切下中间质粒pCW203-E6E7的BamH I酶切位点与Xho I酶切位点之间的片段插入重组质粒pCW180的BamH I酶切位点与XhoI酶切位点之间,得到打靶质粒pCW180-E6E7,如SEQ IDNO. 3所示;其中,所述重组质粒pCW180的基因序列为序列表中SEQ ID NO.13所示;
所述步骤(3)的具体方法是:将LmΔactAplcB-lacZ用含0.5mol/L蔗糖的BHI液体培养基进行培养,定期测定A600,当A600=0.4时,加入盘尼西林G,使其终浓度为12.5μg/ml继续培养;当A600=0.7时,离心,洗涤,分装,保存即可;
LmΔactAplcB-lacZ的制备方法如下:
(3-A)将上、下游分别带Not I酶切位点的lacZ基因片段插入第二中间重组质粒pCW170的Not I酶切位点之间,得到一个打靶质粒pCW190,所述质粒pCW190的基因序列为序列表中SEQ ID NO.6所示;
(3-B)将步骤(3-A)所得打靶质粒pCW190电转化Lm,然后将转化得到的菌经单、双同源重组杂交培养,即得。
2.根据权利要求1所述的疫苗,其特征在于,所述步骤(4)的具体方法是:取打靶质粒电转至LmΔactAplcB-lacZ感受态细胞中,电转完成后加入BHI肉汤培养2小时;转化液涂布于红霉素BHI琼脂平板,通过蓝白斑筛选出单个蓝色菌落,纯培养后进行质粒提取和PCR验证。
3.根据权利要求1所述的疫苗,其特征在于,所述步骤(5)的具体方法是:将携带打靶质粒的单增李斯特菌,在 42℃和30℃连续传代,利用同源重组杂交原理和基因打靶技术,将打靶质粒携带的目的片段整合至李斯特菌中,利用蓝白斑及红霉素敏感性筛选出可疑菌株;然后进行PCR筛选和基因测序验证。
