図4は、図3に示されているピーク2のデコンボリューションされたスペクトルでの、配列番号119のマススペクトルであり、4544.8838の質量値を有する。
ペプチド並びにピーク1及びピーク2によって示されるその形態は単離され、その活性が比較された。以下の実施例に、ペプチドのピーク1、形態1、ネイティブ、酸形態、ペプチド酸、元の、「変換」前、未変換、または「変換」されてない形態、のいずれかとして呼ばれる元々の形態の活性の比較が示されている。形態1は、パート2で規定されているラクトンのような形態2あるいはラクトン形態あるいはペプチドラクトンへと変わるために熱をかけられるか酸性化される形態あるいは酸形態である。これらの実施例のいくつかでは、熱処理は21psi、約121℃で20分のオートクレーブ処理である。あるいは、ペプチドが液体の形態の場合には、これはペプチドを、ピーク2、形態2、ラクトン形態、ペプチドラクトン(ラクトンはパート2で定義される)、ペプチドの脱水形態、またはペプチドの「変換」された形態、のうちのいずれかとして呼ばれるものへと「変換」するために7.0未満のpHに下げることを意味する。
飼料混入試験。図5のグラフは、ピーク2で示されるラクトン形態のペプチドの、あるいはペプチドラクトンの、処理後の、あるいは「変換」された、ペプチドの毒性と比べた、元の形態、ピーク1、ペプチド酸、未変換、のペプチドの毒性の比較を示している。両方の形態は対照とも比較される。図5は、空腹の幼虫に対照の飼料または処理した飼料を与えた後の1日目、2日目、3日目、及び4日目の幼虫死亡率(100%は16匹すべての幼虫が死亡)を示している。この試験で使用されたペプチドは配列番号119であり、これらは粉末618または618ハイブリッド粉末(両方の語は同じ意味である)と呼ばれる噴霧乾燥粉末に製剤された。飼料中に2ppt(1000分の1)相当の用量割合で昆虫に投与した。ピーク1は「変換」または処理前の元のペプチドである。これは、「従来型618」、または単に618粉末もしくは乾燥粉末とも呼ばれる。ピーク2は、「変換」あるいは処理後の、この場合では121℃、21psi(すなわち高温、蒸気、及び圧力)で20分間のオートクレーブ処理後のペプチドである。ピーク2は、図5で「オートクレーブ処理された6−18乾燥粉末」と名付けられている。図5は、水平軸すなわちX軸上の各数字に対して3つの組のデータすなわち棒についての、棒グラフの形態でのデータを示している。この数字は試験に使用した昆虫(ジュウイチホシウリハムシ(southern corn rootworm(SCR))と呼ばれる実際の昆虫)に餌を与えた後の日数であり、試験は、幼虫期に対して行われた。各試験を始めるために16匹の昆虫の幼虫が使用された。凡例が図5に示されており、これは、4日目の上の3つ組の棒の右にある4日目の上に見られる大きい暗い棒が、形態2のペプチドラクトンを昆虫に与えた結果であることを説明している。マススペクトルのピーク2は、ペプチドの「変換」された形態2、ペプチドラクトンの形態である。図5において、4日目のピーク2である黒い棒は、ペプチドの「変換」された形態2を幼虫に摂取させたことによる死亡率を示している。この事例では、形態2は形態1をオートクレーブ処理することによって「変換」された。4日目では、形態1、ペプチド酸、またはネイティブのもしくは未変換のペプチドについては約22%の死亡率なのに対し、形態2、ペプチドラクトンは95%のレベルのウリハムシ死亡率である。4日目の対照は5%未満の死亡率である。幼虫には、未処理の昆虫飼料すなわち対照も与えられた。これは、図5の細かいグレーのクロスハッチであるそれぞれの日の1番目の棒である。より大きい白と黒のクロスハッチ模様の2番目の棒は、マススペクトルのピーク1によって示される「変換」前のペプチドである形態1のペプチドの餌が与えられた幼虫についてのデータを示している。微細な暗色の棒の3番目の棒は、マススペクトル分析のピーク2によって示される形態2の餌が与えられた幼虫についてのデータを示している。餌を与えた後の1〜4日目が示されており、ほとんどの死亡が4日目に生じている。Y軸は死亡した幼虫の割合を示しており、初めに16匹の生きている幼虫が使用された。
培地:SCRの幼虫の飼料は、Bioserve(製品番号F9800B、ニュージャージー州フレンチタウン)から購入した。100mLの飼料を作るため、100mLの脱イオン水を1.44gの準備された寒天と共に沸騰させる。寒天が完全に溶解するまで溶液を沸騰させる。その後、13.28gの飼料と460μlのKOHを添加し、培地を温かい撹拌プレート上で均一になるまで混合する。その後、培地を20mLずつ分割し、水浴中で65Cまで冷却する。
バイオアッセイの比較。バイオアッセイ比較の結果は表6及び7に示されている。ピーク1及びピーク2は、図1〜4に示されている試験を行うために使用されたものと同様に、別個に用意され、別個に液体クロマトグラフィーカラムから単離された。配列番号119のペプチドはこの比較のために使用された。「変換」前ピークであるピーク1、または「変換」後ピークであるピーク2のいずれかを取り出して測定された濃縮物を作り、その後、これを注射によってイエバエに投与した。LD50すなわちハエの50%致死量は、pmol/gの濃度として決定された。ハエは12〜20mgの重さであった。各試料に10匹のハエが存在した。ピーク1の形態とピーク2の形態の分子量の差は、基準のpmol/g溶液を調製する際に考慮しなかった。LD50溶液を形成する全ての溶液は、我々が「スーパーLC」またはスーパーリキッド濃縮物と呼ぶものから、RpHPLCすなわち逆相高圧液体クロマトグラフィーを使用して製造した。
安定性pH試験。これは、安定性とpHの試験の両方であった。これは、「変換」前すなわち形態1と、「変換」後すなわち形態2のペプチドとを比較する。この試験は配列番号119のペプチドを使用した。この試験は、熱に加えて、pHを下げると、すなわち酸またはpHを7以下にするための任意の手段によってペプチド溶液のpHを下げると、形態1から形態2へのペプチドの「変換」が増加する結果になることを示す。
安定性pH試験の結果は図8〜10に示されている。
5μLの注入体積を用いた。結果は下に記載されている。
「変換」なしのアイソフォーム。我々は、配列番号119が高温で分子量中の18ダルトンを失ってアイソフォームを形成し得ることを示した。実施例2では、我々は粉末としての配列番号119の元々の形態、形態1を、形態2に「変換」するためにオートクレーブ処理し、これをジュウイチホシウリハムシの幼虫群の飼料に添加することによって試験した場合に、殺虫効果が約5倍に増加することを示した。しかし、ハイブリッドペプチドである配列番号119におけるこの変形は、天然型ペプチドである配列番号121のようなペプチドでは注目されていなかった。配列番号119とは対照的に、配列番号121にはN末端Glnが存在し、これはそれ自体がNH3を失って、すなわち分子量中の17ダルトンを失ってN−Pyrへと環化する場合がある。これら2つの化学的変性、H2Oを失うこととNH3を失うことは、これらの2つのプロセスで失う分子量が非常に近いことから区別することは困難である。我々は、これらの化学的変性の両方が配列番号121のような配列(我々はこれを天然型ハイブリッドペプチドとも呼ぶ)に生じ得るかどうかを評価するために、分析用HPLCと高感度TOF LC/MS法を使用した。以下のデータは、このプロセスについて我々が本明細書で記載しているような適切な条件に置いた場合に天然型ハイブリッドペプチドに「変換」が生じ得るかを示している。
5μLの試料を、流速5μm/分でWaters BEH130 C−18 Symmetryカラム(0.3mmID×15cm)に注入した。0.1%の移動相B(0.1%のギ酸が入った水)から40%の移動相B(0.1%のギ酸が入った100%アセトニトリル)まで25分かけて直線的な濃度勾配をかけ、25.5分で85%のB、27.5分で85%のB、そして28分で0.1%のBとすることで逆相分離を行った。
10〜30μLμLの試料を、流速1mL/分でWaters C−18 X−Bridgeカラム(4.6mmID×50mm)に注入した。99%の移動相A(0.1%のギ酸が入った水)から95%の移動相B(0.1%のギ酸が入った100%アセトニトリル)まで6分かけて直線的な濃度勾配をかけ、11分で95%のB、11.2分で1%のBで、15分かけて逆相分離を行い、全体の実行時間は18分であった。
LC−MSシステムは、エレクトロスプレーイオン化源を備えたWaters/MicromassZQスペクトロメーターから構成された。試料は、流速1mL/分でZorbax SB−C18カラム(2.1×30mm)に注入した。逆相分離は、ダイオードアレイ検出器(210〜300nm)を使用して、96%の移動相A(0.1%のギ酸が入った水)から98%の移動相B(0.07%のギ酸が入った100%アセトニトリル)まで直線的な濃度勾配をかけることにより3.1分かけて行った。

I)ペプチドヒドラジド
ペプチドヒドラジドは、ペプチドラクトン(パート1参照)とヒドラジンから作られ、ペプチドヒドラジドを形成する。ペプチドヒドラジドは、本質的に3段階の手順で作られる。ペプチドラクトンをヒドラジン一水和物と混合する。混合物を溶液になるまで撹拌してペプチドヒドラジドを形成する、そしてペプチドヒドラジドを精製する。
ペプチドヒドラゾンとペプチドヒドラジドは重要な中間体である。ペプチドに望まれる官能基が何であるかに応じて、様々な種類のペプチドヒドラゾンを製造することができる。ここでは我々は異なるペプチドヒドラゾンの様々な実施例を示す。ヒドラゾンの例は実施例8〜11に示されている。当業者はこれらが実施例を限定しない代表的で例示的なものに過ぎず、他の試薬や条件も使用できることを理解するであろう。
そのネイティブな酸形態のペプチド、パート1で述べられたペプチドラクトン、及びパート2のペプチドヒドラジドを表す典型的な式が示される。他のヒドラジド及びヒドラゾンは実施例8〜11で述べる。
この実施例では、ペプチドヒドラジド(I)を合成するための2つの方法を示す。最初の方法の実施例6(a)では、ペプチドラクトンの出発溶液は比較的純度の高い、HPLC分取したものである。2つ目の方法の実施例6(b)では、ペプチドラクトンの出発溶液は純度が低く、形態1と形態2の両方を含んでいる。すなわち、ペプチドラクトンと混ざったペプチドが存在する。両方の手順でも、ペプチドヒドラジドの同じマススペクトルが得られる。
実施例7では我々はその典型的な酸の形態、形態1すなわちペプチドの形態の毒性ペプチドと、ここで示される式でラベリングされるようなペプチドヒドラジド、すなわちペプチドヒドラジド(I)へと変換された後の同じ毒性ペプチドとを比較する。注射のために次の試料が準備される:
1.100ng/μLのヒドラジド(I)水溶液。この溶液を水で希釈して50ng/μLと5ng/μLの溶液を作製する。
2.100ng/μLのハイブリッド+2水溶液。この溶液を水で希釈して50ng/μLと5ng/μLの溶液を作製する。
適切な標識をした、注射されるハエの容器を用意し、18ゲージの針で容器の蓋に空気穴を開ける。ハエを選択する。完全に孵化して1日目及び2日目のハエを使用する。1日目とは完全に孵化した日である。CO2ガスラインを開き、針でCO2ガスを入れることにより箱の中でハエを動かなくする。ハエが動かなくなった後、ハエをCO2プレートに移し、これらを眠らせたままにする。ハエの重さを量り、12〜18mgの質量を有するものを注射の生物学的分析に使用する。

Claims (23)
- 次の工程を含み、任意選択的にはこれらの工程を文字順で含む、毒性ペプチドを含むペプチドの活性または毒性の増加方法。
a)水と前記ペプチドを混合して、液体中にある前記ペプチドまたは半液体中にある前記ペプチドの形態の、水溶液または水性エマルションを形成する工程であって、前記水溶液または水性エマルションが少なくとも10%の水からなる工程;
b)前記ペプチドの前記水溶液または水性エマルションのpHを測定する工程;
c)前記溶液またはエマルションのpHをpH7.0未満に調整する工程 - 前記水溶液または水性エマルションが、約1.0〜約6.5、約2.0〜約6.0、約2.5〜約5.5、約3.0〜約5.0、約3.0〜約4.0のpHに調整される、または約3.2、3.4、3.5、3.6、または3.8のpHに調整される、請求項1に記載の方法。

を含む、前記製品がペプチドヒドラゾン(II)である請求項16に記載の方法及び製品。 - 複合グリコールを強酸または弱酸で酸性化することと、ヒドラジドを添加してよく混ぜてペプチドグリコールヒドラゾンを製造することとを含む、昆虫捕食者のペプチドをペプチドヒドラジドからペプチドヒドラゾンへと変換するプロセスによって作られる、ペプチドヒドラジド製品の製造方法及び前記ペプチドヒドラジド製品。
- a)O−[2−(6−オキソカプロイルアミノ)エチル]−O’−メチルポリエチレングリコール(IV)と呼ばれる化合物のエタノール混合物のストック溶液に酢酸を1滴添加すること、
b)工程aからの、酢酸で処理されたO−[2−(6−オキソカプロイルアミノ)エチル]−O’−メチルポリエチレングリコール(IV)(分子量約2’000)のストック溶液を使用し、ヒドラジド(I)の水溶液にこれを添加すること、
c)混合し、室温で放置すること、
d)O−[2−(6−オキソカプロイルアミノ)エチル]−O’−メチルポリエチレングリコール(IV)(分子量約2’000)のストック溶液の残部を分割して添加し、混合後終夜放置しておくことでペプチドヒドラゾン(III)を生成すること、
を含む、前記製品がペプチドヒドラゾン(III)である請求項18に記載の方法及び製品。 - アクリルケトンをヒドラジドに添加してヒドラゾンを形成することを含む、昆虫捕食者ペプチドをペプチドヒドラジドからペプチドヒドラゾンへと変換するプロセスによって製造される、ペプチドヒドラジドケトン製品の製造方法及び前記ペプチドヒドラジドケトン製品。
- エタノールに入ったアクリルケトン(V)をヒドラジド(I)水溶液に添加して混合することを含む、前記製品がペプチドヒドラゾン(VI)である請求項20に記載の方法及び製品。
- PEG4ケトン(VIII)をヒドラジド(I)の水溶液に添加して混合することでヒドラゾン(IX)を形成することを含む、前記製品がペプチドヒドラゾン(IX)である請求項20に記載の方法及び製品。
- 共有結合している2H+OまたはH2Oまたは分子のうちのいずれか1つ以上をペプチドから除去することとして記述される方法であって、本明細書に記載されているまたは明細書もしくは請求項中に見出されるような単独または組み合わせでの、いずれかの条件、温度、圧力、及びpHまたは酸性条件の下で、共有結合している2H+Oまたは分子のうちのいずれか1つ以上が取り除かれた任意の毒性ペプチドを含み、本明細書及び請求項に記載の手順のいずれかによって製造されたいずれかの前記ペプチド、ヒドラジド、またはヒドラゾンを含む方法、により製造されるペプチドの調製方法及びまたはペプチド、及びまたは前記方法によって製造される殺虫性組成物、または、本明細書及び請求項に記載の方法のいずれかによって製造され、その後昆虫の生息場所への適用に好適な製剤中で使用される、前記ペプチドの殺虫性組成物としてのこれらのペプチドのいずれかの使用。
