Abstract
Description
1981年以降、マウスからは胚性幹細胞(ESC)が樹立されているが、それらに対応する物を、ラットを含む他のさまざまな哺乳動物から得ようとする試みは、成功していない。最近、多能性幹細胞がマウス及びラットの着床後卵筒期エピブラストから誘導された(Brons et al., Nature 448, 191-195 (2007);Tesar et al., Nature 448, 196-199 (2007))。これらの新規幹細胞はエピブラスト幹細胞(EpiSC)と名付けられた。EpiSCは、コロニーの形態および多様性を維持するための培養/シグナリング要件が、ヒト胚性幹細胞(hESC)によく一致するようであるが、マウスES細胞(mESC)とは、表現型およびシグナリング応答に一連の著しい相違を示す。

「多能性の」または「多能性」という用語は、3つの胚葉(内胚葉、中胚葉、および外胚葉)の全てに由来する細胞系譜に関連する特徴を集団として示す細胞タイプへの分化を適当な条件下で起こすことができる子孫を生じる能力を有する細胞を指す。多能性幹細胞は、出生前、出生後または成体動物の多くのまたは全ての組織に寄与することができる。ある細胞集団の多能性を確証するには、当技術分野で受け入れられている標準的試験(例えば8〜12週齢のSCIDマウスにおいて奇形腫を形成する能力)を使用することができるが、多能性細胞の検出には、さまざまな多能性幹細胞特徴の同定も使用することができる。細胞の多能性は、胚本来のあらゆる細胞を形成することができる完全多能性細胞、例えば胚性幹細胞およびiPSCから、3つの胚葉の全ての細胞を形成することができるが完全多能性細胞の特徴(例えば生殖細胞系伝達(germline transmission)または生物体全体を生成する能力など)の全てを示すわけではないことがある不十分または不完全多能性細胞にまでまたがる、連続したつながりである。特定の態様では、細胞の多能性が、不十分にまたは不完全に多能性である細胞から、より多能性である細胞へと、または一定の態様では、完全多能性細胞へと向上する。多能性は、例えば奇形腫形成、生殖細胞系伝達、および四倍体胚補完によって評価することができる。いくつかの態様では、本明細書において項を改めて議論する多能性遺伝子または多能性マーカーの発現を使って細胞の多能性を評価することができる。
I.序論
本発明は、ALK5阻害因子が細胞における多能性の維持と、任意で誘導とを、著しく改善するという驚くべき知見を根拠としている。ALK5の阻害因子とMAPK阻害因子(例えばMEK阻害因子、Erk阻害因子またはp38阻害因子)との組合せ、またはALK5阻害因子とFGF経路阻害因子(例えばFGF受容体阻害因子)との組合せは、以下を可能にする:
以前(例えば米国特許第5,843号;同第6,200,806号;および同第7,029,913号に)記載された従来のヒト胚性幹細胞または誘導多能性幹細胞とは著しく異なり、mESC特徴により類似している、以下に定義づける新しい機能的性質を持つ細胞の、多能性の維持;および
例えば多能性を維持するための既知の方法(例えばGSK3およびMEK阻害因子が関わるもの-WO2008/015418参照)と比較して、著しく改善された効率および細胞の安定性。
実際、ALK5の阻害因子が多能性の維持を改善するのに有効であるというのは、驚くべきことである。なぜなら、一つには、今まで当技術分野では、多能性を刺激するためにTGFβ経路をアンタゴナイズするのではなく、アゴナイズすることに、重点が置かれてきたからである。例えばWO2008/056173を参照されたい。
哺乳動物細胞の多能性を誘導または維持するために、ALK5阻害因子(または他のTGFβ/アクチビン経路阻害因子)を、任意でMAPK(例えばMEKまたはErkまたはp38阻害因子)またはFGFシグナリング経路阻害因子(例えばFGF受容体阻害因子)と共に含む、細胞培養物が提供される。いくつかの態様では、そのような阻害因子と一緒に培養された細胞が、実施例に記載する特徴、例えば限定するわけではないが、培養下でドーム状コロニーを形成すること、ESCマーカー(例えばOct4、Sox2、およびNanog)の発現、Oct4プロモーターがほぼ完全に脱メチル化されていること、Rex-1およびALP(例えば着床後段階のエピブラストおよびEpiSCには存在しないESCおよび初期エピブラストのマーカー)を発現すること、インビトロで内胚葉、神経外胚葉、および中胚葉に分化する能力、ならびにインビボ多能性特徴、例えば(例えばSCIDマウスにおいて)奇形腫を形成する能力、および非ヒト細胞の場合は、胚盤胞に注入し、受容子宮内に移植した時に、キメラ子孫を形成する能力などを持つ。その上、いくつかの態様では、培養下の細胞が、そのような特徴を、複数の細胞継代、例えば少なくとも1、2、3、4、5、7、10、20、30継代、またはそれ以上にわたり、同じ培養条件下で持ち続ける。

細胞は、本発明の阻害因子と初めて接触する前に多能性であってもよいし、細胞を本発明の阻害因子の1つまたは複数と接触させた後に、(例えば適当な転写因子の導入によって、および/または多能性を誘導するための適当な小分子との接触によって)多能性が誘導されてもよい。
(例えばiPSCを作製するためまたは所望のタンパク質もしくは核酸を発現させるために)形質転換細胞が望まれる場合、本発明の阻害因子(例えばALK5阻害因子、MAPK阻害因子、FGF経路阻害因子、および任意でGSK3β阻害因子)と接触させる細胞は、接触の前に形質転換されてもよいし、かつ/または接触後に形質転換されてもよい。例えば、本発明の利点の一つは、複数の細胞継代を通した多能性細胞の維持が可能になり、したがって、細胞の多能性特徴を維持したまま、操作し、それに続いて子孫を選択することが可能になる点である。これは、トランスジェニック動物の作製(本明細書に記載するような配列特異的組換え事象によるノックアウト動物の作製を含む)には、とりわけ有用である。
一定の態様では、宿主細胞を形質転換するために、プラスミドベクターを使用することが考えられる。一般に、レプリコンと宿主細胞に適合する種に由来する制御配列とを含有するプラスミドベクターが、これらの宿主に関連して使用される。ベクターは、複製部位、および形質転換細胞の表現型選択をもたらすことができるマーキング配列を持つことができる。
受容体介在性エンドサイトーシスによって細胞に感染または侵入し、宿主細胞ゲノム中に組み込まれてウイルス遺伝子を安定に効率よく発現するという、一定のウイルスの能力は、それらを、細胞(例えば哺乳動物細胞)中に外来核酸を導入するための魅力的な候補にしてきた。本発明の核酸を送達するために使用し得るウイルスベクターの非限定的な例を以下に記載する。
核酸を送達するための、ある特定の方法では、アデノウイルス発現ベクターを使用する。アデノウイルスベクターはゲノムDNAへの組込み能力が低いことが知られているが、この性質は、これらのベクターによって得られる遺伝子導入効率の高さによって埋め合わされる。「アデノウイルス発現ベクター」は、(a)コンストラクトのパッケージングをサポートし、(b)そこにクローニングされている組織特異的または細胞特異的コンストラクトを最終的に発現させるのに十分な、アデノウイルス配列を含有するコンストラクトを包含するものとする。〜36kbの線状二本鎖DNAウイルスであるアデノウイルスの遺伝子構成の知識から、アデノウイルスDNAの大きな断片を7kbまでの外来配列で置換することが可能である(Grunhaus et al., Seminar in Virology, 200(2):535-546, 1992)。
核酸は、アデノウイルス支援トランスフェクション(adenovirus assisted transfection)を使って細胞中に導入することができる。アデノウイルス共役系(adenovirus coupled system)を用いる細胞系では、増加したトランスフェクション効率が報告されている(KelleherおよびVos, Biotechniques, 17(6):1110-7, 1994;Cotten et al., Proc Natl Acad Sci USA, 89(13):6094-6098, 1992;Curiel, Nat Immun, 13(2-3):141-64, 1994)。アデノ随伴ウイルス(AAV)は魅力的なベクター系である。というのも、これは組込み頻度が高く、非分裂細胞に感染することができるため、例えば組織培養において(Muzyczka, Curr Top Microbiol Immunol, 158:97-129, 1992)、またはインビボで、哺乳動物細胞中に遺伝子を送達するのに役立つからである。rAAVベクターの作製と使用に関する詳細は、米国特許第5,139,941号および同第4,797,368号に記載されており、これらの特許はそれぞれ参照により本明細書に組み入れられる。
レトロウイルスは、その遺伝子を宿主ゲノムに組み込む能力、大量の外来遺伝物質を運搬する能力、幅広い種および細胞タイプに感染する能力、ならびに特殊な細胞系においてパッケージングされる能力を持つことから、遺伝子送達ベクターとして有望である(Miller et al., Am. J. Clin. Oncol., 15(3):216-221, 1992)。
送達すべき核酸は、特異的結合リガンドを発現するように操作された感染性ウイルス内に収容することができる。したがってウイルス粒子はターゲット細胞のコグネイト受容体に特異的に結合し、内容物を細胞に送達すると考えられる。レトロウイルスベクターの特異的ターゲティングを可能にするために設計された新規アプローチが、ウイルスエンベロープへのラクトース残基の化学的付加によるレトロウイルスの化学修飾に基づいて開発された。この修飾は、シアロ糖タンパク質受容体を介した肝細胞の特異的感染を可能にすることができる。
本発明と共に使用される細胞、組織または生物を形質転換するための核酸送達の適切な方法には、本明細書に記載するように、または当業者には公知であるだろうように、核酸(例えばDNA)を細胞、組織または生物中に導入することができる事実上任意の方法が包含されると考えられる。そのような方法には、DNAの直接送達、例えば、任意でFugene6(Roche)またはLipofectamine(Invitrogen)を用いる、エクスビボトランスフェクションによるもの(Wilson et al., Science, 244:1344-1346, 1989、NabelおよびBaltimore, Nature 326:711-713, 1987)、マイクロインジェクション(HarlandおよびWeintraub, J. 多能性細胞、例えば胚性幹細胞、および多能性を持つように誘導された細胞、または多能性を持つように誘導されるべき細胞は、当技術分野において公知である任意の方法に従って培養することができる。本明細書において述べるように、いくつかの態様では、多能性細胞が、ALK5阻害因子およびMEKまたはErk阻害因子の1つと共に、任意でGSK3β阻害因子および/またはLIFと共に、培養される。培養培地は、当技術分野において公知である培養培地の他の任意の成分を含むことができる。いくつかの態様では、培養培地が、細胞生存のための基礎培地成分(例えばビタミン類およびミネラル類、任意で、等張条件下にあるもの)を含む。例示的な基礎培地は、DMEMまたはその変形、例えばDMEM Knockoutである。さらに培養物にはKnock-out血清代替添加物(KSP)を補足することができる。本発明の培養物は、1つまたは複数の炭素源を含むことができる。いくつかの態様では、培養物が、L-アナリル(analyl)-L-グルタミンを含む。培養物は血清を含むこともできるし、無血清であることもできる。
本発明は、幹細胞技術(限定するわけではないが、予防的用途または治療的用途を含む)のさらなる研究および開発を可能にする。例えばいくつかの態様では、本発明の細胞(本発明の阻害因子下で培養された多能性細胞、またはそのような細胞に由来し、所望の細胞運命に沿って分化するように誘導された細胞)が、それを必要とする個体(限定するわけではないが、臓器、組織、または細胞タイプの再生を必要とする個体を含む)に導入される。いくつかの態様では、細胞が、最初は、ある個体からの生検材料中に得られ、本明細書に記載するように多能性が誘導され、任意で(例えば特定の所望する前駆細胞に)分化するように誘導され、次に、その個体中に移植し戻される。いくつかの態様では、個体中に細胞を導入する前に、その細胞が遺伝子改変される。
本発明者らは、LIFとALK5阻害因子、GSK3阻害因子、およびMEK阻害因子のカクテルとによって均質に維持することができる新規ラットiPSC(riPSC)の樹立に成功したことを、ここに報告する。riPSCは、マウスESCと共通する特徴を有し、最も重要なことには、キメラに大きく寄与をすることができる。本発明者らは、「マウスESC様」の特徴を持ち、驚いたことにMEK阻害因子およびALK5阻害因子の存在下で培養下に維持することができる、新規ヒトiPSC(hiPSC)も作製した。本発明者らの実験は、オーセンティックな胚性幹細胞をまだ入手することができないラットまたは他の種から、再プログラミングによって多能性幹細胞を作製するための枠組みを提供することになると、本発明者らは考える。
この実施例ではラット多能性細胞の作製と維持を実証する。
二倍体ラット肝臓前駆細胞株であるWB-F344(Grisham et al., Proc Soc Exp Biol Med 204, 270-279 (1993))に、レトロウイルスによってOct4、Sox2およびKlf4を形質導入した後、従来のmESC培地中のMEFフィーダー細胞上に分割した。コンパクトなアルカリホスファターゼ(ALP)陽性ESC様コロニーが形質導入の10日後に観察された(効率は約0.4%である)(図1A)。しかし、そのESC様コロニーをピックアップして同じ培地で二次培養したところ、それらは迅速に分化して、ESCの形態を失ったことから(図1B)、従来のmESC培地条件はriPSCの多能性を維持するには十分でないことが示唆された。小分子は重要な分化誘導経路を阻害することができるという考えの下、本発明者らおよび他の研究者らはこれまでに、よりロバストな方法でmESC自己複製をサポートするための小分子を同定し、使用してきた(Schugar et al., Gene Ther 15, 126-135 (2008);Xu et al., Nat Biotechnol 20, 1261-1264 (2002))。そのような化学的戦略およびmESCとEpiSC/hESCの自己複製を維持するためのシグナリングの相違に基づいて、本発明者らは、MEK阻害因子PD0325901、ALK5(TGFβシグナリングの主要I型受容体)阻害因子A-83-01、GSK3β阻害因子CHIR99021、およびFGFR阻害因子PD173074を選択し、異なる小分子の組合せがriPSCの多能性の維持に及ぼす効果を調べた。0.5μM PD0325901と3μM CHIR99021の組合せを使用すると、riPSCは培養下で短期間維持することができるが、甚だしい自発的分化を示す(図1C)。この条件下で成長させた細胞は、連続継代後は、ゆっくり増殖するようになり、分化した細胞の増殖ゆえに、培養物が劣化した。最近の研究により、CHIR99021およびFGFR阻害因子PD173074と組み合わされたPD0325901は、mESCの多能性をLIF非依存的に維持できることが実証された(Ying et al., Cell 115, 281-292 (2003))。しかし、PD0325901とCHIR99021の組合せと同様に、培地にPD173074(0.1μM)を含めることが、さらなる利益を示すことはなかった。アクチビンA/NodalシグナリングはhESCおよびEpiSCの未分化状態を維持するために重要であるが、mESC自己複製にとっては必須でないので、次に、本発明者らは、PD0325901、CHIR99021およびTGF-β阻害因子A-83-01の組合せが、riPSCの分化を抑制し、自己複製を促進することができるかどうかを調べた。興味深いことに、0.5μM PD0325901、3μM CHIR99021および0.5μM A-83-01の組合せでは、riPSCが、より均質な集団として成長し、自発的分化は実質上阻害された(図1D)。そのような条件下では、PD0325901およびCHIR99021の組合せと比較して、クローン増殖効率も有意に増加した(図5)。さらにまた、LIF自体はriPSC自己複製を持続させるのに十分でなかったものの、LIFの非存在下では極めて小さなALP陽性コロニーしか観察されず、長期間維持することもできなかった(図5)。その上、riPSCの長期自己複製にはユニークな化学的阻害因子カクテルが必要だった。riPSCは、LIF、PD0325901、A-83-01、およびCHIR99021の存在下で、30継代を超える期間、明白な分化または増殖の減少を伴わずに培養されたが、化学的阻害因子を培地から除去した後は1継代以内にESCの形態を失って分化する。この条件下においてriPSCは、培養下で典型的なドーム状コロニーを形成する点で、従来のmESCに類似していた(図1E)。免疫細胞化学により、riPSCは、Oct4(図1F)、Sox2(図1G)、SSEA-1(図1H、緑色)、Nanog(図1H、赤色)などの典型的なmESCマーカーを発現するが、SSEA3、SSEA4およびTRA-1-81などのhESCマーカーについては陰性であることが明らかになった。ラット遺伝子プライマーを使った4つのクローンriPSC株のRT-PCR分析により、内在性ラットOct4、Sox2、Nanog、Klf4、Rex-1、TDGF2、FGF4およびErasの発現が確認された(図1I)。トランスジーンに対する特異的プライマーを使用したところ、形質導入されたマウスOct4、Sox2およびKlf4遺伝子はおおむね発現停止されていることが、RT-PCR分析によって明らかになった(図1I)。ラットOct4プロモーターのメチル化状態の分析により、riPSCとWB-F344細胞の間で示差的メチル化が示された。riPSCクローンはほとんど完全な脱メチル化パターンを示し、親WB-F344細胞のそれとは異なっている(図1J)。注目すべきことに、riPSCはmESCと共通する分子特徴を持ち、特にRex-1およびALP(着床後段階のエピブラストおよびEpiSCには存在しないESCおよび初期エピブラストのマーカー)を発現する。
riPSCの発生能を調べるために、インビトロ分化アッセイを行った。免疫染色により、riPSCは、標準的な胚様体分化法の下で、内胚葉(アルブミンおよびPdx1)(図2A、2B)、神経外胚葉(βIII-チューブリン、Tuj1)(図2C)および中胚葉(brachyury)(図2D)誘導体に分化し得ることが示された。次に、本発明者らはriPSCのインビボ発生能を調べた。重症複合免疫不全(SCID)マウスへの移植後に、riPSCは、神経上皮様構造(外胚葉)、気道上皮(内胚葉)、軟骨様構造(中胚葉)および平滑筋(中胚葉)を含む3つの胚葉の全てからなる奇形腫を形成した(図2E〜2H)。最も注目すべきことに、Brown-Norwayラット(黒毛)胚盤胞(n=18)への注入後に、3匹のラットが生まれ、その全てが甚だしい毛色キメリズムを示した(図2I)。しかし生殖細胞系伝達はまだ検出されていない。総合すると、上記の結果から、ラットEpiSCとは異なる本発明者らのriPSCは、多能性mESC様ラット幹細胞株と定義された。
この実施例では、マウス胎性幹細胞と類似する状態にあるヒト多能性細胞を誘導し、それを維持するための培養培地を実証する。
1981年以降、マウスからは胚性幹細胞が樹立されているが(Martin, G.R., Proc Natl Acad Sci USA 78, 7634-7638 (1981))、それらに対応するものをラットなどの他の動物から得ようとする試みは、完全には成功していない(Demers et al., Cloning Stem Cells 9, 512-522 (2007);Ruhnke et al., Stem Cells 21, 428-436 (2003), 2003;Schulze et al., Methods Mol Biol 329, 45-58 (2006);Ueda et al.「Establishment of rat embryonic stem cells and making of chimera rats」PLoS ONE 3, e2800 (2008))。遺伝子再プログラミングと化学的アプローチとを併用することにより、本発明者らは、コロニー形態および培養要件/シグナリング応答に関して従来のmESCの重要な特徴を有する、新規mESC様ラットおよびヒト多能性幹細胞を作製することができた。小分子のユニークなカクテルの下で、本発明者らの安定なriPSCは、インビボでキメリズムに大きく寄与する能力を有していた。ラットは、生理学的研究および行動学的研究に、より適しており、多重遺伝子性ヒト疾患の優れたモデルである。しかし、インビボで多能性を有するラット幹細胞を入手することができないため、この貴重なモデルの活用は妨げられていた。本発明者らによるラット多能性細胞の樹立および適当な化合物カクテルを使った戦略は、生物医学研究者にとって、標的遺伝子破壊(gene-targeted)ラットを作製する道を開くと考えられる。本発明者らのhiPSCは、よりロバストに成長し、従来のmESCに類似しかつ従来のhESCとは異なる新規な多能性状態を表すようである。
細胞培養およびウイルス形質導入:University of North CarolinaのWilliam B. Coleman教授のご厚意によって分譲された二倍体ラットWB-F344細胞(Grisham et al., Proc Soc Exp Biol Med 204, 270-279 (1993))を、継代第7代において、マウスOct4、Klf4およびSox2用のpMXsベースのレトロウイルス(Addgene)により、記載のとおり(Takahashi, K.およびYamanaka, S., Cell 126, 663-676 (2006))、形質導入した。24時間後に、1×105個の形質導入WB-F344細胞を、100mmディッシュ中のX線不活化CF1 MEF上に播種し、mESC成長培地:Knockout(商標)DMEM、20%Knockout血清代替添加物、1%Glutamax、1%非必須アミノ酸、1%ペニシリン/ストレプトマイシン、0.1mM β-メルカプトエタノールおよび103U/ml mLIF(Millipore)と共に培養した。10日後に、mESC成長培地中のMEFフィーダー細胞上で増殖させるために、riPSCコロニーをピックアップし、MEK阻害因子PD0325901(Stemgent、0.5μM)、ALK5阻害因子A-83-01(Tocris Bioscience、0.5μM)、およびGSK3β阻害因子CHIR99021(Stemgent、3μM)で処理した。

インビトロでのiPSCの分化:riPSCまたはhiPSCのインビトロ分化は、標準的胚様体分化法によって行った。riPSCまたはhiPSCを0.05%トリプシン-EDTAで解離し、超低接着100mmディッシュ中、10%FBSを補足したDMEM培地で培養して、胚様体(EB)を形成させた。培地を1日おきに交換した。1週間後にEBを収集し、10%FBSを含むDMEM培地に入れて、マトリゲル被覆6穴プレートに移した。3〜7日後に、免疫細胞化学分析のために細胞を固定した。細胞培養製品は、特に言及した場合を除いて全て、Invitrogen/Gibco BRLから入手した。
胚盤胞からラットES細胞を誘導するために、透明帯除去ラット胚盤胞(E4.5)を、20%Knock-out血清代替添加物(KSR)、1%非必須アミノ酸、1000U/mlマウスLIF、1%Glutmax、3μM CHIR99021、0.5μM PD0325901、0.25μM A-83-01(または2μM SB431542)を補足したKnock-out DMEM培地と共に、X線不活化CF1フィーダー細胞上に播種した。3〜5日後に、ICM由来細胞塊を、アキュターゼ(Accutase)によって解離し、新しいフィーダー上に移した。典型的ES細胞形態を持つコロニーをピックアップし、アキュターゼで解離してから、新しいフィーダー上に播種した。樹立されたラットES細胞を上記の培地で培養し、約3日ごとに継代した(1:6)。ラットES細胞とriPSCはどちらも、長期自己複製には、LIFおよび阻害因子カクテル(CHIR99021、PD0325901、A-83-01)の存在を要求する。ラットES細胞およびriPSCは、例えばOct4、Sox2、NanogおよびSSEA-1などのマウスES細胞の多能性マーカーを発現するが、従来通りに培養されたヒトES細胞によって発現されたSSEA-3、SSEA-4、TRA-1-61およびTRA-1-81などのマーカーは発現しない。ラットES細胞とriPSCはどちらも、マウスEpiSCでは発現されないICMマーカーRex-1およびALPを発現する。
ヒトおよびサルES細胞をマウスES細胞様(初期ICM)多能性状態に転換するために、20% Knock-out血清代替添加物(KSR)、1%非必須アミノ酸、1% Glutmax、10ng/ml bFGFを補足したDMEM/F-12培地で、ヒトES細胞(Hues9)およびサルES細胞(R366.4)を培養した。細胞が50%コンフルエントに達した時に、培地を、2μMリジン特異的デメチラーゼ1阻害因子(パルネート)を補足したAdvance DMEM/F-12、1×N2、1×B27、1% Glutmax、50μg/ml BSA培地に切り換えた。3日後に、10μg/mlヒトLiF、3μM CHIR99021、0.5μM PD0325901、および2μM SB431542を含有するがパルネートは含有しない同じ培地で、細胞を培養した。甚だしい分化にもかかわらず、コンパクトなマウスES細胞様コロニーが、約1週間の処置後に、目に見えるようになった。転換されたヒト/サルES細胞を上記の培地で培養し、約4〜5日ごとに継代(1:6)した。細胞は、Oct4、Sox2、Nanog、SSEA-3、SSEA-4、TRA-1-61、TRA-1-81などの多能性マーカーを発現し、ICMマーカーRex-1およびALPも発現する。
従来のマウス胚性幹細胞(ESC)は、着床前胚盤胞の内部細胞塊(ICM)の多能性細胞に由来し、それに相当する。それらは、無限に自己複製することができ、インビトロで全ての細胞タイプを生じる能力を持ち、最も重要なことには、胚盤胞に戻した時に、インビボで、生殖細胞系を含む動物全体に寄与する能力を持つ。最近になって、着床後段階のエピブラストから、エピブラスト幹細胞(EpiSC)と呼ばれる異なるタイプの多能性細胞が得られた(Brons et al., Nature 448, 191-195, 2007;Tesar et al., Nature 448, 196-199, 2007)。EpiSCは、長期間、自己複製することができ、インビトロで多能性であると共に、奇形腫アッセイにおいてインビボでも多能性であると思われるが、mESCとは対照的に、ICMに組み入れられてキメリズムに寄与する能力を持たないことから、EpiSCはICM由来のESCよりも進んだ/後期の発生段階の多能性に由来し、それに相当することが確認され、それらを「再プログラム」してICM段階の多能性細胞に戻すことは、インビボ環境下でさえ、できないことが示唆される。従来のヒトESCは、胚盤胞を使って得られたものではあるが、いくつかの遺伝子発現、コロニー形態(すなわち扁平なコロニー)ならびに自己複製および分化におけるシグナリング応答を含む数多くの特徴に関して、EpiSCに極めて密接に対応すると思われる。EpiSC/hESCは、(よりコンパクトでドーム状のコロニー形態を持つ)mESCとは、機能的および機構的に、他にも多くの点で異なる。例えば、mESCは、白血病抑制因子(LIF)および骨形成タンパク質(BMP)の下(Ying et al., Cell 115, 281-292, 2003)またはMEKおよび/もしくはFGFRの阻害下(Ying et al., Nature 453, 519-523, 2008)で自己複製するが、EpiSC/hESCは、自己複製に関してMAPK、FGF、およびTGFβ/アクチビン/Nodal経路活性に依存するようであり、MEK、FGFRおよび/またはALK4/5/7阻害因子で処理すると迅速に分化する(Brons et al., Nature 448, 191-195, 2007;Li et al., Differentiation 75, 299-307, 2007;Peerani et al., EMBO J 26, 4744-4755, 2007;Tesar et al., Nature 448, 196-199, 2007)。また、所定の分化条件下で、mESCはBMP処理に応答して中胚葉系譜に分化するが、EpiSC/hESCは栄養芽層または原始内胚葉細胞を生じる(Brons et al., Nature 448, 191-195, 2007;D'Amour et al., Nat Biotechnol 23, 1534-1541, 2005;Xu et al., Nat Biotechnol 20, 1261-1264, 2002)。これらの観察は、EpiSCとhESCとが本質的に類似しているという概念を強く裏付けており、mESCとEpiSC/hESCは2つの別個の多様性状態、すなわち着床前胚盤胞のICMを表すmESC様状態と、着床後のエピブラストを表すEpiSC様状態とを表すので、エピブラスト状態(従来のhESCを含む)を、ICM状態に転換し戻すことができるかどうかという、魅力的な仮説を提起する。mESCまたはmEpiSCからのキメリズムに寄与する能力には明確な相違があるので(これは、EpiSCからmESCへの機能的変換の決定的確認になるだろう)、マウス系は、そのような興味をそそる過程を研究するための理想的なプラットフォームになり、おそらくは新しいタイプのICM/mESC様ヒト多能性細胞を従来のhESCから作製するための基礎を提供する。
細胞培養:マウスEpiSC株はPaul Tesar博士(Case Western Reserve University)から分譲されたものである。EpiSC(EpiSC-5株、雄)は、以前記述されたように10ng/ml bFGFを補足したヒトESC培地中の照射CF1 MEF上で維持した(Tesar et al., Nature 448, 196-199, 2007)。EpiSCを1mg/ml IV型コラゲナーゼ(Invitrogen)で3〜4日ごとに継代した。R1 mESCは、20%KSR(Invitrogen)、0.1mM 2-ME(Sigma-Aldrich)、2mM L-グルタミン(Invitrogen)、0.1mM NEAA(Invitrogen)、および103単位/mlの組換えマウス白血病抑制因子(LIF)(ESGRO、Millipore)を補足したKnockout DMEM(Invitrogen)からなる従来のmESC成長培地を使って照射CF1 MEF上で培養した。mESCおよび転換細胞は、0.05%トリプシン/EDTAを使って、単細胞懸濁液として、3日ごとに継代し、通常の培養については、1cm2あたり1.0×104細胞の密度で播種した。フィーダーフリー培養の場合は、1×N2(Invitrogen)、1×B27(Invitrogen)、0.1mM 2-ME、2mM L-グルタミン、0.1mM NEAA、50μg/ml BSAフラクションV(GIBCO)、103単位/ml LIFおよび10ng/ml BMP4(R&D)を補足したKnockout DMEMからなる既知組成培地中、ゼラチン被覆組織培養皿上で、細胞を成長させる。成長曲線実験のために、ゼラチン被覆12穴プレート中、フィーダーフィリー条件下で、細胞を培養した。二つ一組にした細胞試料を、各ウェルに1×105細胞の密度でプレーティングした。各時点(24時間間隔)について、二つ一組のウェルの細胞をトリプシン処理し、血球計を使って計数した。これらのカウント数を平均し、プロットした。ALK阻害因子A-83-01、SB431542、MEK阻害因子PD0325901、GSK3阻害因子CHIR99021、およびFGF受容体阻害因子PD173074はStemgent Inc.から購入した。パルネートはSigmaから購入した(P8511)。
新しく作製されたhiPSCを特徴づけるために、リアルタイムPCRを利用して、hiPSCの遺伝子発現を解析した。ヒトES細胞株Hues9(hES施設、Harvard University、http://mcb.harvard.edu/melton/hues/)を対照として使用した。リアルタイムPCR分析により、Hues9細胞と比較して、本発明のhiPSCは、Gbx2(5倍)、Dppa3(2倍)およびKlf4(2.5倍)などといった一定の遺伝子を、より高いレベルで発現することがわかる。これらのマーカーは、マウスES細胞でも高度に発現するが、マウスEpiSCでは高度に発現しないことがわかる。リアルタイムPCRアッセイに使用したプライマーは、

Claims (57)
- 少なくとも1回の細胞分裂にわたって多能性細胞を培養する方法であって、
細胞の多能性を維持しつつ、少なくとも1回の細胞分裂を可能にするように、十分な量の
a. ALK5阻害因子、および
b. MEK阻害因子、Erk阻害因子、p38阻害因子、およびFGF受容体阻害因子のうちの1つまたは複数から選択される第2の化合物;および
c. 十分な時間にわたる十分な栄養素
の存在下で、多能性動物細胞を培養する工程
を含む、前記方法。 - 培養工程が、一定量のGSK3β阻害因子の存在下で細胞を培養することをさらに含む、請求項1記載の方法。
- GSK3β阻害因子がCHIR99021である、請求項2記載の方法。
- 培養工程が、一定量の白血病抑制因子(LIF)の存在下で細胞を培養することをさらに含む、請求項1記載の方法。

複製し、かつ多能性を維持している、単離された多能性動物細胞であって、
ただし、マウス胚性幹細胞(mESC)ではない、前記単離された多能性動物細胞。 - ヒト細胞である、請求項41記載の単離された細胞。
- ヒト胚性幹細胞である、請求項42記載の単離された細胞。
- ラット細胞である、請求項41記載の単離された細胞。

- ALK5およびMEKの阻害下で多能性を維持する、請求項41記載の単離された細胞。
- 従来通りに培養されたhESC、エピブラスト幹細胞、およびヒト誘導多能性細胞と比較して、より高レベルのE-カドヘリンを発現する、請求項41記載の単離された多能性動物細胞。
- 従来通りに培養されたhESC、EpiSC、およびヒト誘導多能性細胞と比較して、2倍高いレベルのE-カドヘリンを発現する、請求項47記載の単離された多能性動物細胞。
- 従来通りに培養されたhESC、エピブラスト幹細胞、およびヒト誘導多能性細胞と比較して、Gbx2、Dppa3、Klf4、およびRex1を含むマーカーを、より高レベルに発現する、請求項41記載の単離された多能性動物細胞。

(a) 不完全多能性細胞を、ヒストンデアセチラーゼ阻害因子、ヒストンH3K4脱メチル化の阻害因子またはH3K4メチル化の活性化因子から選択されるエピジェネティック修飾因子と接触させる工程;
(b) 工程(a)の後に、細胞を、(i)ALK5阻害因子、(ii)MEK阻害因子、Erk阻害因子、またはp38阻害因子、および(iii)FGF受容体阻害因子のうちの2つまたはそれ以上と共に、エピジェネティック修飾因子の非存在下で培養する工程であり、それにより、前記不完全多能性哺乳動物細胞と比較して、より完全に多能性である細胞を作製する、工程
を含む、前記方法。 - (c) 工程(b)の後の不完全多能性哺乳動物細胞を、(i)ALK5阻害因子、(ii)MEK阻害因子、Erk阻害因子、またはp38阻害因子、および(iii)FGF受容体阻害因子、および(iv)GSK3阻害因子と共に培養する工程
をさらに含む、請求項51記載の方法。 - 培養工程(a)および/または(b)および/または(c)が、不完全多能性哺乳動物細胞を白血病抑制因子(LIF)の存在下で培養することをさらに含む、請求項51または52記載の方法。
