Abstract
Description
A.プロタミノバクター生体触媒の調製
砂糖工場製濃厚ジュース(乾燥物質含有量65%)8kg、トウモロコシ浸出液2kg、(NH4)2HPO40.1kg及び蒸留水89.9kgからできている、必要ならpH7.2に調整された滅菌栄養基質の10mlで株プロタミノバクター・ルブルム(CBS574.77)の継代培養から細胞を濯ぎ出す。この懸濁液を上記組成の栄養溶液200mlが入っている1リットルフラスコ中でのシェーカー前培養のための接種材として用いる。
この生体触媒を調製するために、砂糖工場製濃厚ジュース(乾燥物質含有量=65%)8kg、トウモロコシ浸出液2kg、(NH4)2HPO40.1kg及び蒸留水89.9kgからできている、必要ならpH7.2に調整された滅菌栄養基質の10mlで株シュードモナス・メソアシドフィラMX−45(FERM BP3619)の継代培養から細胞を濯ぎ出した。この懸濁液を前記栄養溶液200mlが入っている1リットルフラスコ中でのシェーカー前培養のための接種材として用いた。
A)のようにして得られた固定化細胞をB)のようにして得られた固定化細胞と1:1重量比で混合し、温度自動調節であり得る及び25〜30℃に温度自動調節されているカラム反応器に一緒に充填し、そしてDM(乾燥物質)含有量が35〜45%のスクロース溶液を連続的に流し通す。この場合における流量は、用いたスクロースの少なくとも97%が所望のトレハルロース含有組成物(これはビタロースを意味する)に変換されるように調整する。

残存スクロースを、場合により、このようにして得られたトレハルロース含有組成物(これはビタロースを意味する)から、以下のようにしてそれをH+イオンが負荷された強酸性カチオン交換材中で、又は、カラム反応器中の適する触媒で処理することによって除去した。
100cm3の強酸性カチオン交換体(例えばLewatit(登録商標)OC1052)を60℃に温度自動調節された適するガラスカラムに充填し、公知の方法によりHClで再生することによってH+イオンを負荷した。

この実施例では、生成物組成に対するシュードモナス・メソアシドフィラMX−45(FERM BP3619)とプロタミノバクター・ルブルムCBS574.77のいくつかの比の影響を解析した。
次に、甘味剤としてスクロースとグルコースの混合物を用いたゼリー用の4つのレシピ、及び、本発明のトレハルロース含有組成物(これはビタロースを意味する)を用いた4つのレシピを調製する。調製されたゼリーを貯蔵及びTPA分析で比較試験した。
上記の、実施例3Aで調製したサンプルを比較試験で詳細に分析した、すなわち、いくつかの増粘剤(ペクチン、ゼラチン、スターチ及び/又はアラビアガム)を含有しているスクロース/グルコース組成物をトレハルロース含有組成物(これはビタロースを意味する)に対して比較した。
ii)密閉PE(ポリエチレン)バッグに25℃で8週間貯蔵
TPA試験及び貯蔵挙動についての分析の時間間隔は、O(貯蔵の開始)、1、2、4、8週間後に行われた。
4.1)シリアルバー
相対的な甘味の強さの関係を明らかにするために、以下の溶液群を、各場合、15人の試験者による3点比較試験で互いを比較した。
b)2つの7%濃度スクロース溶液対実施例1Dによる甘味剤の17.5%濃度溶液。
c)2つの7%濃度スクロース溶液対実施例1Dによる甘味剤の18.5%濃度溶液。
実施例1Dの甘味剤でアイスクリームを製造するために、22.1kgの生クリーム(乾燥質量中40%脂)、58.1kgの全乳(DM中3.7%脂)及び4.5kgのスキムミルクを15kgの実施例1Dの甘味剤及び0.3kgの安定剤と混合し、ホモジナイズし、殺菌した。
低カロリーストローベリージャムを製造するために、1kgの刻みイチゴを1kgの実施例1Cの甘味剤並びに8gの中度エステル化ペクチン及び7gの酒石酸と3分間一緒にボイルし、準備したビンにビン詰めにした。
血糖応答(血糖インデックス、GI)及びインスリン応答(血中インスリンの指標、II)に対するトレハルロースの影響をイソマルツロースとの対比で調査した。
以下、本発明の実施形態を示す。
(1)(a)スクロース含有組成物を適切な条件下でシュードモナス属及びプロタミノバクター属微生物の細胞又は細胞抽出物と接触させ、(b)トレハルロース含有組成物を生成させる、トレハルロース含有組成物の調製方法。
(2)スクロース含有組成物が、水性媒体に溶解又は懸濁されている、(1)に記載の方法。
(3)細胞又は細胞抽出物が、少なくとも1つの担体に固定化されている、(1)又は(2)に記載の方法。
(4)両方の属の微生物の細胞又は細胞抽出物が、同一の担体に共固定化されている、(1)〜(3)のいずれか1に記載の方法。
(5)(1)〜(4)のいずれか1に記載の方法に従って調製されたビタロース(トレハルロース含有組成物)。
(6)20〜95重量%(乾燥質量基準)のトレハルロース含量を含んでいる、(5)に記載のビタロース(トレハルロース含有組成物)。
(7)8〜50重量%(乾燥質量基準)のイソマルツロース含量を含んでいる、(5)又は(6)に記載のビタロース(トレハルロース含有組成物)。
(8)(5)〜(7)のいずれか1に記載のビタロース(トレハルロース含有組成物)及び少なくとも1種の添加剤を含んでいる、ヒト又は動物の消費用製品。

- グルコースを持続して放出し且つ同時に低インスリン応答を引き起こすことができるように設計された、ヒト又は動物の消費用製品を調製するための請求項5〜17のいずれかに記載のビタロース(トレハルロース含有組成物)もしくは製品又はトレハルロースの使用。
- 製品が、医薬製品である、請求項18に記載の使用。
- 請求項5〜17のいずれかに記載の組成物又は製品を、それを必要としている対象に適切な量で適用し、それによって低インスリン応答及び持続したグルコースの放出を引き起こさせる、グルコース又はインスリン代謝関連病態又は疾患の治療方法。
- インスリン関連疾患が、糖尿病、代謝症候群、耐糖能障害、インスリン抵抗、過脂肪、過体重、肥満、高脂血、ガン、肝疾患、インスリン感受性及び関節硬化からなる群から選択される、請求項20に記載の方法。
- 請求項5〜17のいずれかに記載の組成物又は製品を、それを必要としている対象に適切な量で適用する、対象における歯の疾患の予防及び治療方法。
- 請求項5〜17のいずれかに記載の組成物又は製品を、それを必要としている対象に適切な量で適用する、消費者の口中におけるプラーク形成を低減させるための、好ましくは請求項22に記載の方法。
- 請求項5〜17のいずれかに記載の組成物又は製品を、それを必要としている対象に適切な量で適用する、対象の精神的及び/又は身体的能力を改善させるための方法。
