例えば上述のような発明対象のシステム及び装置またはその構成要素、ならびに標的組織中で活動電位を光遺伝学的に調節するために発明対象のシステム及び/または装置をどのように使用するかについてのインストラクションを少なくとも含むキットも提供される。いくつかの実施形態において、キットは、発明対象のポリヌクレオチド、ベクターまたは医薬組成物のうちの1つまたは複数を含むことができる。本開示の実施形態に従うキットは、1つまたは複数の装置(1つもしくは複数の送達装置、1つもしくは複数の光生成装置及び/または1つもしくは複数の制御装置等)も含むことができる。
神経細胞に、光活性化プロトンポンプタンパク質(eArch3.0)及び酸感受性ナトリウムイオンチャンネル応答タンパク質(ASIC2a)をコードするポリヌクレオチドをトランスフェクションした。タンパク質は神経細胞中で発現し、神経細胞の原形質膜中に存在した。560nmの光のパルスはeArch3.0を活性化し、速い外向きプロトン電流を引き起こし、原形質膜の早期過分極をもたらした。次いで、細胞外プロトンは、ASIC2aによって運搬された内向きカチオン電流を活性化し、持続的膜脱分極及び後続する生来の電圧ゲート型ナトリウムチャンネルの不活性化をもたらし、脱分極ブロック及び誘発されたスパイキングの抑制を引き起こした。結果を図1中で示す。
海馬の培養ニューロンに、酸感受性ナトリウムイオンチャンネル応答タンパク質(ASIC2a)をコードするポリヌクレオチドをトランスフェクションした。タンパク質は神経細胞中で発現し、神経細胞の原形質膜中に存在した。1000pA電流を10Hzで細胞の中へ注入し、ホールセルパッチクランプ記録を収集した。1000pA電流パルス注入に応答して、外向き電流成分はスパイキングを阻害した。しかしながら、2000pA電流パルス注入の間に、ASIC構成要素によって引き起こされた脱分極ブロックのみがスパイキングを阻害するのに十分であった。結果を図2中で示す。挿入図は、1秒の緑色光パルスに応答した各々の細胞の電圧クランプトレースを示す。
神経細胞に、光活性化プロトンポンプタンパク質(eArch3.0)及び酸感受性ナトリウムイオンチャンネル応答タンパク質(ASIC2a)をコードするポリヌクレオチドをトランスフェクションした。タンパク質は神経細胞中で発現し、神経細胞の原形質膜中に存在した。スパイクは、10Hzで閾上(高信頼性スパイキング)電流パルス注入を使用して、ベースラインで誘発された。光が適用された場合、eArch3.0媒介性過分極はスパイキングを阻害するのに不十分であったが、ASIC2a媒介性脱分極は、脱分極ブロックに起因して、光パルスの残りを通してスパイキングを強く抑制した。右側の挿入図は、提供された外向き電流及び内向き電流の振幅により、560nmの光の1秒のパルスに対する同じニューロンの電圧クランプ応答を示す。結果を図3中で示す。
神経細胞に、光活性化プロトンポンプタンパク質(eArch3.0)及び酸感受性ナトリウムイオンチャンネル応答タンパク質(ASIC2a)をコードするポリヌクレオチドをトランスフェクションした。タンパク質は神経細胞中で発現し、神経細胞の原形質膜中に存在した。この実施例において、内向き電流:外向き電流の比は小さく(約1)、したがって、ASIC構成要素によって引き起こされた脱分極は、脱分極ブロックを引き起こすこと及び誘発されたスパイキングに打ち勝つことには不十分であった。結果を図4中で示す。

バイスタンダー実験
すべての実験は、Stanford Administrative Panel on Laboratory Animal Careによって承認されたプロトコル下で行われた。
バイスタンダー効果の電気生理学的特性
神経活性における、「バイスタンダーニューロン」(本明細書において、間接的に曝露されているが、直接は発現してはいないニューロンとして定義される)による光遺伝学媒介性変化の存在が試験された。2つの光遺伝学的標的化戦略を用いた。第一に、CHR2(H134R)、eArch3.0、eNpHR3.0またはカルモジュリンキナーゼIIαプロモーターによって駆動されるYFP対照(AAV5−CamKII−(オプシン)−eYFP)のうちの1つを含有する光遺伝学的コンストラクトを、海馬のCA1領域中の片側に注入した。これらのニューロンの対側への軸索投射に起因して、次いで、対側のCA1中のオプシン発現軸索によって囲まれた、非発現ニューロン(バイスタンダーニューロン)から記録することができた(図15A及び15C)。脱分極オプシンCHR2(H134R)(今後ChR2と称される)、2つの過分極オプシン(膜標的化発現について促進された)(プロトンポンプのアーキロドプシン(eArch3.0)及びクロライドポンプのハロロドプシン(eNpHR3.0))、及びYFP対照へのバイスタンダー応答を、一致した実験条件下で海馬の調製物において比較した。第2のカテゴリーのバイスタンダーニューロンは遺伝子導入マウス系統Thy1−ChR2(18系統)を使用して同定し、その場合、ChR2は層V皮質ニューロン中で発現し、バイスタンダーニューロンは表面的な皮質中に位置し、それらはChR2を発現する膜突起によって囲まれるが、ChR2を自らは発現していない(図15B及び15D)。

により対象の標的細胞を遺伝学的に改変する段階と;
病態について対象を治療するために、活性化用の波長の光に標的細胞を曝露する段階と
を含む、該方法。 - 前記光活性化タンパク質がイオンポンプである、請求項67に記載の方法。
- 前記イオンポンプが、水素イオンポンプ、ナトリウムイオンポンプ、カリウムイオンポンプ、クロライドイオンポンプまたはカルシウムイオンポンプである、請求項68に記載の方法。
- 前記光活性化タンパク質がイオンチャンネルである、請求項67に記載の方法。

a)光に応答して複数の水素イオンが神経細胞の膜を外向きの方向で通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸;及び
b)複数のナトリウムイオンが膜を内向きの方向で通過し、ここで膜を通る複数のナトリウムイオンの通過が膜を脱分極させ、神経細胞の膜中の電圧ゲート型ナトリウムチャンネルを不活性化し、それによって神経学的病態を治療することを可能にすることによって、神経細胞の膜を通る複数の水素イオンの通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸
により対象の神経細胞を遺伝学的に改変する段階と;
病態について対象を治療するために、活性化用の波長の光に神経細胞を曝露する段階と
を含む、該方法。 - 前記光活性化タンパク質が水素イオンポンプである、請求項78に記載の方法。
- 前記光活性化タンパク質が水素イオンチャンネルである、請求項78に記載の方法。
- 前記応答タンパク質がナトリウムイオンポンプである、請求項78に記載の方法。

- 神経細胞またはその部分内で逆行性で伝導される活動電位を選択的に阻害する方法であって、
a)光に応答して複数の水素イオンが神経細胞の膜を通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸;及び
b)複数のナトリウムイオンが膜を通過し、ここで膜を通るナトリウムイオンの通過が膜を脱分極させ、逆行性で伝導される活動電位を引き起こす神経細胞の膜中の複数の電圧ゲート型ナトリウムチャンネルを不活性化することを可能にすることによって、水素イオンの神経細胞の膜の通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸
により神経細胞を遺伝学的に改変する段階と;
その中の逆行性で伝導される活動電位を阻害するために、活性化用の波長の光に神経細胞またはその部分を曝露する段階と
を含む、該方法。 - 前記光活性化タンパク質が水素イオンポンプである、請求項85に記載の方法。
