Claims (21)
- a)光に応答して第1のイオンが細胞膜を通過することを可能にするように適合された光応答性タンパク質と;
b)第2のイオンが細胞膜を通過することを可能にすることによって第1のイオンの細胞膜の通過に応答する応答タンパク質と
を含む、融合ポリペプチド。 - a)光応答性タンパク質が、配列番号:1、4、5、および6の1つと少なくとも90%のアミノ酸配列同一性を有するアミノ酸配列を含む光活性化イオンポンプであるか;または
b)光応答性タンパク質が、配列番号:8〜15および29の1つに記載のアミノ酸配列と少なくとも90%のアミノ酸配列同一性を有するアミノ酸配列を含む光活性化イオンチャンネルである、
請求項1に記載の融合ポリペプチド。 - 前記応答タンパク質が、配列番号:19中で記載されるASIC2aポリペプチドアミノ酸配列と、少なくとも80%、少なくとも85%、少なくとも90%、少なくとも95%、少なくとも98%、少なくとも99%または100%のアミノ酸配列同一性を有するアミノ酸配列を含む、請求項1に記載の融合ポリペプチド。
- 小胞体(ER)搬出シグナルをさらに含み、任意で、該ER搬出シグナルがアミノ酸配列FCYENEV(配列番号:47)を含む、請求項1〜4のいずれか一項に記載の融合ポリペプチド。
- 請求項1〜5のいずれか一項に記載の融合ポリペプチドをコードするヌクレオチド配列を含む核酸。
- 請求項6に記載の核酸を含む、組換え発現ベクター。
- 請求項7に記載の組換え発現ベクターにより遺伝学的に改変された細胞。
- 細胞の膜電位の調節のためのシステムであって、
a)光に応答して第1のイオンが細胞膜を通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸と;
第2のイオンが細胞膜を通過することを可能にすることによって第1のイオンの細胞膜の通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸;及び
b)光により標的場所を照射するように構成された装置、または
a)請求項6に記載の核酸もしくは請求項7に記載の組換え発現ベクター;及び
b)光により標的場所を照射するように構成された装置
を含む、該システム。 - 前記光活性化タンパク質がイオンポンプもしくはイオンチャンネルであるか、または前記応答タンパク質がイオンポンプ、イオンチャンネル、イオン交換タンパク質、もしくはイオンコトランスポーターである、請求項9に記載のシステム。
- 前記装置が、約350〜約750nmまたは約530nm〜約560nmの範囲の波長を有する光により標的場所を照射するように構成される、請求項9に記載のシステム。
- 前記装置が以下の1つまたは複数を行うように構成される、請求項9に記載のシステム:
a)光により標的場所を一定して照射する;
b)光のパルスにより標的場所を照射する;
c)光の波長及び/または強度を調節する;
d)光のパルスの周波数及び/または継続期間を調節する;ならびに
e)使用者インプットに応答して標的場所を照射する。 - 前記使用者インプットが、光の波長、光の強度、光パルスの継続期間、光パルスの周波数、及び/または標的場所を含む、請求項12に記載のシステム。
- 前記装置が対象中に埋込まれるように適合される、請求項9に記載のシステム。
- 光に応答して第1のイオンが細胞膜を通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸と;
第2のイオンが細胞膜を通過することを可能にすることによって第1のイオンの細胞膜の通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸と
を含む、医薬組成物。 - 光に応答して第1のイオンが細胞膜を通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸と;
第2のイオンが細胞膜を通過することを可能にすることによって第1のイオンの細胞膜の通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸と
を含む、細胞。 - 光に応答して細胞の膜電位を調節するためのインビトロの方法であって、活性化用の波長の光に細胞を曝露する段階を含み、細胞が、
a)光に応答して第1のイオンが細胞膜を通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸と;
b)第2のイオンが細胞膜を通過することを可能にすることによって第1のイオンの細胞膜の通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸と
により遺伝学的に改変される、該方法。 - a)第1の核酸が、光に応答して複数の水素イオンが細胞膜を外向きの方向で通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含み;かつ
b)第2の核酸が、複数のナトリウムイオンが細胞膜を内向きの方向で通過することを可能にし、細胞膜の持続的脱分極を引き起こし、それによって電圧ゲート型ナトリウムイオンチャンネルを不活性化し、その活性を阻害することによって、細胞膜の外部表面上またはその付近での水素イオンの存在に応答する応答タンパク質をコードするヌクレオチド配列を含む、
請求項17に記載の方法。 - a)光に応答して第1のイオンが標的細胞の膜を通過することを可能にするように適合された光活性化タンパク質をコードするヌクレオチド配列を含む、第1の核酸と;
b)第2のイオンが該膜を通過することを可能にすることによって第1のイオンの標的細胞の膜の通過に応答する応答タンパク質をコードするヌクレオチド配列を含む、第2の核酸と
を含む、対象における病態を治療する方法における使用のための製品であって、
該方法が該製品により対象の標的細胞を遺伝学的に改変する段階と、病態について対象を治療するために、活性化用の波長の光に標的細胞を曝露する段階と
を含み、該第2のイオンの膜の通過により該病態が治療される、製品。 - 前記標的細胞が神経細胞であり、
前記光活性化タンパク質が、光に応答して複数の水素イオンが神経細胞の膜を外向きの方向で通過することを可能にするように適合され;かつ
前記応答タンパク質が、複数のナトリウムイオンが膜を内向きの方向で通過し、ここで膜を通る複数のナトリウムイオンの通過が膜を脱分極させ、神経細胞の膜中の電圧ゲート型ナトリウムチャンネルを不活性化し、それによって神経学的病態を治療することを可能にする、請求項19に記載の製品。 - 前記標的細胞が神経細胞であり、
前記光活性化タンパク質が、光に応答して複数の水素イオンが神経細胞の膜を通過することを可能にするように適合され;かつ
前記応答タンパク質が、複数のナトリウムイオンが膜を通過し、ここで膜を通るナトリウムイオンの通過が膜を脱分極させ、逆行性で伝導される活動電位を引き起こす神経細胞の膜中の複数の電圧ゲート型ナトリウムチャンネルを不活性化し、それによって神経細胞またはその一部内の逆行性で伝導される活動電位を選択的に阻害することを可能にする、
請求項19に記載の製品。
