Description
本出願は、2012年3月20日に出願の米国仮特許出願第61/613231号の優先権を主張し、この出願は、その全体が参照により本明細書に援用される。
大うつ病性障害は、低調な気分、自殺思考、意欲の減少および喜びを経験することができないことによって特徴づけられる。この重度の精神医学的疾患の有病率にもかかわらず、最も一般的に処方される治療的介入の、選択的セロトニン再取り込み阻害薬は、たいてい効果がなく、および重篤な有害な副作用を有する。
本明細書に使用される、「異種」という用語は、核酸に対する言及において、その天然環境にない(すなわち、人の手によって変化された)遺伝子産物(ポリペプチドまたは核酸)をコードする核酸をいう。たとえば、異種核酸は、1つの種から別の種に導入された核酸を含む。また、異種核酸は、いくつかの方法で変化された(たとえば、変異され、複数コピーに付加されて、非天然プロモーターまたはエンハンサー配列、その他に連結された)生物にとって天然の遺伝子を含む。異種核酸は、核酸のcDNA形成を含むヌクレオチド配列を含んでもよく、cDNA配列は、センス(mRNAを産生するように)またはアンチセンス配向(mRNA転写物に相補的であるアンチセンスRNA転写物を産生するように)のいずれで発現してもよい。異種核酸は、いくつかの態様において、異種核酸配列が異種遺伝子によって、または染色体における遺伝子配列と共にコードされるタンパク質のための遺伝子に関連した天然に見いだされないか、または天然において見いだされない染色体の部分に関連する(たとえば、遺伝子が通常発現されない座位において発現される遺伝子)プロモーターなどの調節エレメントを含むヌクレオチド配列に典型的には連結される点で、内因性核酸から区別することができる。
本開示は、うつ病の非ヒト光遺伝学的動物モデルを提供する。動物モデルは、うつ病を治療するための薬剤を同定するために、およびうつ病の治療のための治療ストラテジーの標的を同定するために有用である。
本開示は、動物のニューロンにおいて光応答性オプシン(たとえば、光応答性イオンチャネル、光応答性イオンポンプ、その他)を発現する非ヒト動物を提供する。光に対する光活性化されたオプシンの曝露による光応答性オプシンの活性化が動物の挙動を調節する。詳細な態様において、光応答性オプシンの光活性化は、動物においてうつ病を誘導する。その他の態様において、光応答性オプシンの光活性化は、うつ病を軽減する。
上記の如く、いくつかの場合において、導入遺伝子(たとえば、光応答性オプシンをコードするヌクレオチド配列を含む核酸)は、哺乳動物の細胞のサブセットにのみ存在する。たとえば、いくつかの場合において、導入遺伝子は、脳細胞にのみ存在する。これらの態様の一部において、導入遺伝子は、細胞のサブセットのゲノム(ランダムな組み込み部位にて、またはターゲットされた組み込み部位にてのいずれか)に組み込まれる。その他の場合において、導入遺伝子は、エピソームのままである。

光遺伝学は、生組織のターゲットされた細胞において、さらに自由に移動する哺乳類およびその他の動物内でさえ、機能する無処置の生物学的システムでペースを保持するために必要な時間精度(ミリ秒−時間尺度)で、特異的なイベントを制御するために使用される遺伝学的および光学的な方法の組み合わせをいう。光遺伝学は、ニューロン膜電位の時間的に正確な操作を可能にし、一方で特異的ターゲティング機構を介して細胞−タイプ分解能を維持する、ターゲットニューロン細胞の原形質膜に対する迅速な光応答性チャネルまたはポンプタンパク質の導入を必要とする。光に対する応答における神経細胞膜過分極または脱分極を促進するために使用することができる任意の微生物オプシンを使用してもよい。たとえば、光応答性クロライドポンプのハロロドプシンファミリー(たとえば、NpHR、NpHR2.0、NpHR3.0、NpHR3.1)およびGtR3プロトンポンプを、光に対する応答における神経細胞膜過分極を促進するために使用することができる。加えて、光応答性カチオンチャネルタンパク質(たとえば、ChR2、SFOs、SSFOs、C1V1s)のチャネルロドプシンファミリーのメンバーは、光刺激に対する応答における神経細胞膜脱分極または脱分極誘導されたシナプスの枯渇を促進するために使用することができる。
いくつかの場合において、非ヒト動物モデルの神経細胞において発現される光応答性オプシンは、光応答性イオンポンプ(たとえば、光応答性クロライドポンプ)である。たとえば、光応答性クロライドポンプのハロロドプシンファミリーの1つまたは複数のメンバーは、神経細胞の原形質膜上に発現される。いくつかの態様において、1つまたは複数の光応答性クロライドポンプは、VTAにおけるニューロンの原形質膜上に発現される。その他の態様において、1つまたは複数の光応答性クロライドポンプは、mPFCにおけるニューロンの原形質膜上で発現される。

本非ヒト動物モデルを使用して、うつ病の治療における治療上の介入のためにさらなる標的を同定することができる。したがって、本開示は、うつ病を促進するタンパク質を同定する方法を提供し、このようなタンパク質は、うつ病を治療するための潜在的な治療上の標的(たとえば、標的の活性を調節し、およびこれによりうつ病を治療する薬物を同定するために使用することができる標的)であると考える。
本非ヒト動物モデルを使用して、うつ病以外の障害の治療のために、うつ病を誘導する効果のために、開発されている薬物を試験することができる。本非ヒト動物モデルにおいてうつ病の症候を誘導する薬物は、うつ病以外の障害を治療することにおける適合性を再評価されることを必要としてもよいし、本非ヒト動物モデルにおけるうつ病の症候をもはや誘導しないが、うつ病以外の障害を治療することにおける有効性を保持するように、化学修飾されることを必要としてもよいし、または警告ラベルにおいて薬物がおそらくうつ病の症候を誘導するかもしれないという可能性を含むことを必要としてもよい。
中脳腹側被蓋野(VTA)ドーパミンニューロンの活動が、意欲および快活症の正常なレベルの維持に必要であったかどうかを調査した。VTA DAニューロンの活動を選択的に阻害することがうつ病様挙動を敏感に誘導し得るかどうかを試験するため、光遺伝的技術(21〜23)によって可能になる細胞タイプ特異的ターゲティングおよび正確な時間操作を利用した。VTA DAを阻害するために、琥珀色光(eNpHR3.0)(23)の照明においてニューロン膜を過分極化する増強されたハロロドプシンを発現したものは、複数のうつ病アッセイにおいてこれらの動物を試験する前に、TH::Creマウス(24)のVTAに二重にloxPが導入されたcre依存的ウイルスの構築物を注射することによって、チロシン水酸化酵素(TH)陽性ニューロンにおいて選択的に発現した(図1A)。

- 前記膜輸送シグナルが、配列番号:17に示したアミノ酸配列を含む、請求項5に記載の方法。
- 前記うつ病アッセイは、強制水泳試験、尾懸垂試験または条件付き場所嗜好試験である、請求項1から7のいずれか一項に記載の方法。
- 前記決定することは、過分極化光応答性オプシンポリペプチドを活性化する波長の光に前記VTAを曝露することの後に、または並行して実施される、請求項1に記載の方法。
