Abstract
Description
(a)リン脂質含有開始材料を、撹拌下、C1〜C3アルコール及びその混合から選択された脂肪族アルコールを含む抽出剤と、前記リン脂質の少なくとも一画分の前記抽出剤への移転を生じさせるのに十分な時間、接触させることと、
(b)遠心力を適用することを含む工程によって、得られた混合物を、残余ラフィネートから、リン脂質濃縮抽出物に分離することと、を含む、リン脂質含有供給材料を、1つ又は2つ以上のリン脂質が濃縮された2つ又はそれ以上の画分へ画分化するための、複数の混合及び分離ステージを含む抽出工程に関し、各分離ステージからの前記リン脂質含有抽出物が、少なくとも一部分、先行ステージに戻るか、さらに先行の混合ステージに戻り、最終リン脂質濃縮抽出物が、第一残余ラフィネートから分離される。
図1は、本工程の好ましい実施形態を描写している。ここで、リン脂質供給タンク(1)と、熱交換機(図示せず)を含む抽出剤タンク(2)が、最終抽出剤出口(4)と最終ラフィネート出口(4)を有する、多ステージ遠心液体−液体交換器(3)に流体連結する。抽出剤供給が、リン脂質供給に対して向流に、抽出機(3)内入り、最終抽出剤が、抽出剤容器(5)及びラフィネート容器(6)中に回収される。タンク(1)及び(2)両方には、実際の実験に必要な流れ、従って抽出比率を調節及び制御するために、フローメータを備える。温度制御設備が、熱交換機内、及び遠心抽出器入口及び出口の両方に設置される。
抽出器:Rousselet Robatel(France)から得た多ステージ遠心液体−液体抽出器を本実験に使用した。抽出器は、中心ロータに接続した6回転ボウルを含み、最大回転速度2,900rpm、最大流速(2相)25〜30l/hであった。ボウルは、0.39lの有用容量を有した。
未精製大豆レシチンを、それぞれ2.5%wt.及び4.55% wt.水を含むエタノールで抽出した。表1は、種々の実行の結果に適用した条件を描写している。
Claims (16)
- リン脂質含有供給材料を、1つ又は2つ以上のリン脂質が濃縮された2つ又はそれ以上の画分に画分化するための、複数の混合及び分離ステージを含む、向流抽出工程であって、以下の段階、すなわち、
(a)リン脂質含有開始材料を、撹拌下、C1〜C3アルコール及びその混合から選択された脂肪族アルコールを含む抽出剤と、前記リン脂質の少なくとも一画分とを、前記抽出剤内へ移転させるのに十分な時間、接触させる段階と、
(b)遠心力を適用することを含む工程によって、得られた混合物を、残余ラフィネートから、リン脂質濃縮抽出物に分離する段階と、を含み、
各分離ステージからの前記リン脂質濃縮抽出物が、少なくとも一部、先行ステージに戻るか、さらに上流の混合ステージに戻り、最終リン脂質濃縮抽出物が、第一残余ラフィネートから分離される、向流抽出工程。 - (a)において、前記リン脂質含有開始材料が、並流又は向流混合操作において前記抽出剤と接触する、請求項1に記載の工程。
- 前記リン脂質抽出物が、1つ又は2つ以上のホスファチジルコリン(PC)、ホスファチジルエタノールアミン(PE)、ホスファチジルイノシトール(PI)及び/又はホスファチジン酸(PA)を含む、請求項1又は請求項2に記載の工程。
- 前記最終ラフィネート相が、12重量%未満のホスファチジルコリン(PC)を含む、請求項1〜3のいずれか1項に記載の工程。
- 前記最終抽出物が、少なくとも25重量%のホスファチジルコリン(PC)と、50重量%超のアセトン不溶物(AI)とを含む、請求項1〜4のいずれか1項に記載の工程。
- 前記脂肪族アルコールがエタノールである、請求項1〜5のいずれか1項に記載の工程。
- 前記脂肪族アルコール中の水濃度が、0〜10重量%の範囲内である、請求項1〜6のいずれか1項に記載の工程。
- 段階(a)及び(b)が、少なくとも部分的に、多ステージ抽出装置中で実行され、多ステージ抽出装置は各ステージに対して、
i)ロータ
ii)2つの液体流が混合される、前記ロータに連結した混合チャンバであって、混合チャンバが、
iia)前記混合チャンバ中に配置された定常攪拌器と、
iib)その中で液体流が、前記混合チャンバによって発生する遠心力によって分離される沈降チャンバと、を含む混合チャンバと、が含まれる、工程。 - 前記定常攪拌器が、定常ディスクを含み、前記混合することが、前記定常ディスクと回転している混合チャンバ間の速度差を通して達成される、請求項8に記載の工程。
- 少なくとも2つの抽出及び分離ステージを含み、対応する抽出剤が、向流又は並流様式において、1つ又は2つ以上のステージにて、対応するラフィネートに加えられる、請求項1〜9のいずれか1項に記載の工程。
- 抽出比率が、1〜5の範囲内であり、前記抽出剤相に、1〜10重量%の量で水が含まれ、前記抽出剤相が、25℃〜70℃の温度を有し、2〜10の抽出ステージが、分離段階に対して、2〜20.000gを用いて使用される、請求項1〜10のいずれか1項に記載の工程。
- 前記リン脂質含有供給にレシチンが含まれる、請求項1〜11のいずれか1項に記載の工程。
- レシチンが、大豆レシチン、トウモロコシレシチン、菜種レシチン、米油レシチン、ヒマワリレシチン、綿種レシチン、パーム油レシチン、海産油レシチン、バイオマスレシチン、ピーナッツレシチン、卵黄レシチン、ミルクレシチン及び/又は脳レシチン、より好ましくは大豆レシチン又はヒマワリレシチンから選択される、請求項12に記載の工程。
- 少なくとも一部のリン脂質を、ラフィネート及び/又は抽出物相から単離することをさらに含む、請求項1〜13のいずれか1項に記載の工程。
- 請求項1〜14のいずれか1項に記載の工程に従って入手可能な、リン脂質濃縮抽出物又はリン脂質減損ラフィネート。
- 食品、好ましくはパン製品、栄養素、菓子、インスタント食品、マーガリン、スプレッド、動物餌製品及び/又は医薬品組成物のため、又は放出剤又は工業用乳化剤としての、リン脂質濃縮抽出物又はリン脂質減損ラフィネートの利用。
