ことによって調製され得る。成形錠剤は、不活性液体結合剤で湿らせた粉末状化合物を、
適切な機械で成形することによって作製され得る。非経口投与に適する本発明の製剤は、
好都合には活性化合物の滅菌水性製剤を含有し、前記製剤は、好ましくは意図される受容
者の血液と等張である。これらの製剤は、皮下、静脈内、筋肉内または皮内注射によって
投与され得る。そのような製剤は、好都合には、化合物を水またはグリシン緩衝液と混合
すること、および生じた溶液を滅菌し、血液と等張にすることによって調製され得る。

gである。

る、新規高分子組成物担体である。ELPは、逆温度相転移を受ける独特のクラスの生体
高分子に属する。それらは、転移温度(Tt)以下の温度では可溶性であるが、それらの
Ttを上回る温度で不溶性になり、凝集する[1−3]。
(i)ELPは、罹患部位で凝集するように罹患部位の局所温度以下であるTtを有す
るように設計され得る。
(ii)あるいはELPは、可溶性形態のままであるように罹患部位の局所温度を上回
るTtを有するように設計され得る。
(iii)ELPは、組成物または造影剤または標的部分の共有結合または酵素結合の
ための部位を含有し得る。
(iv)ELPはまた、ELPを罹患部位または器官に特異的に標的するペプチドまた
はタンパク質などの遺伝的にコード可能な標的部分(1以上)を含むように設計され得る
。
(2.定義)
(A)組成物:何らかの疾患に対して治療価値を有する何らかの分子。
(B)造影剤:罹患部位または器官の視覚化を提供する何らかの分子。
組成物または造影剤の例は、排他的ではないが、(i)低分子、(ii)放射性核種、
(iii)ペプチド、(iv)ペプチドミメティック、(v)タンパク質、(vi)アン
チセンスオリゴヌクレオチド、(vii)ペプチド核酸、(viii)siRNA、(i
x)金属キレート、(x)炭水化物を含む。
(3.組成物または造影剤の連結または会合)
組成物は、安定なまたは不安定な結合方式を通してELPに共有結合され得る。組成物
は、ELPと疎水的に会合し得る。組成物は、キレート化法を通してELPに連結され得
る。組成物は、二次結合を通した分子認識によりELPと会合し得る。組成物はまた、酵
素の作用を通してELPに連結され得る。組換えによって生産され得るペプチドタンパク
質などの分子の場合は、ELPと組成物は、合成またはクローン化遺伝子から適切な宿主
(大腸菌、ピキア・パストリス(pichia pastoris)、哺乳動物細胞また
はバキュロウイルス)において単一実体として生産され得る。「ELP組成物/造影剤複
合体」は、複合体の間の結合が単一実体を治療薬または造影剤として送達するために安定
であるように合成され得るか、またはELPから組成物を遊離させるためにpHまたは光
の作用、あるいは酵素の作用下で不安定であるように設計され得る。
(4.投与)
ELP組成物複合体または融合タンパク質は、(i)被験体に全身的に注入されるか(
iv、ia、ipまたはim)、(ii)罹患部位または器官に局所的に注入されるか、
(iii)経口的に送達されるか、あるいは(iv)非経口的に送達される。

ELPの合成と特性決定。ELPは、典型的には大腸菌における組換え合成によって調
製される。しかし、他の宿主も組換え合成のために使用し得る。ELPはまた、化学合成
によっても調製され得る。組換え合成の典型的な例では、重合工程は、ELPのための反
復配列(典型的にはVPGXGの約10個のペンタペプチドをコードする)についての合
成遺伝子を頭−尾方向に反復して連結する、反復方向指示型連結(recursive
directional ligation)(RDL)と呼ばれる方法によって遺伝子
レベルで実施される。プラスミドへのn回の連結後、これは、全てが同じペプチド配列を
コードするが、組成物の倍数である分子量を有する、n+1個のELP遺伝子のライブラ
リーを提供する。
イクリング(ITC)によって精製して、4つの異なるpH感受性、マレイミド活性化、
ヒドラゾンリンカーを通してドキソルビシン分子に複合する。リンカーの構造または長さ
は、ELPドキソルビシン複合体のTtにほとんど影響を及ぼさない。全ての複合体のT
tは天然ELPと同様であるためである(データは示していない)。しかし、より長いリ
ンカーを有するELPドキソルビシン複合体は、短いリンカーを有するELPドキソルビ
シン複合体よりも緩やかな転移速度を示す。pH4で、最も短いリンカーを有するELP
ドキソルビシン複合体からのドキソルビシンの放出は、72時間でほぼ80%に達した。
FaDu細胞でのインビトロ細胞培養アッセイにおいて細胞毒性に関して試験する。対照
複合体である非複合ELPは固有の細胞毒性を示さず、従って実質的なインターナリゼー
ションにもかかわらずELPが非毒性であることを示唆する(図4)。これに対し、EL
Pドキソルビシン複合体は24時間または72時間の間に実質的な細胞毒性を示し、毒性
のレベルは等しいドキソルビシン濃度のものと同様である。固形腫瘍におけるELPの蓄
積。FaDu固形癌を担持するヌードマウスに14C標識ELPを全身的に注入すること
により、生体分布試験を実施する。移植した腫瘍におけるELPの蓄積は、注入用量の1
0〜20%/グラム(%ID/g)の範囲内である。約40℃のTtを有するELPをマ
ウスに全身的に注入し、移植腫瘍を42℃に加熱したとき、蓄積は約20%ID/gであ
る。これに対し、腫瘍を加熱せずに同じELPを注入したとき、蓄積は約10%ID/g
であった。このデータは、腫瘍を加熱しないときでも、有意の濃度(%ID/g)の放射
性標識ELPが腫瘍内に局在したことを示す。これに対し、非複合形態の小さな放射性標
識分子(分子量<500Da)の注入は、腫瘍内の有意に低い蓄積を生じさせる。この実
施例は、ELPが罹患部位における有意の局在化を生じさせ得ることを明らかにする。

Claims (14)
- 被験者へ送達用の薬剤を製造するためのエラスチン様ペプチドに結合した活性物質の使
用であって、
前記活性物質が前記結合により複合体として前記被験者に投与されたとき、前記活性物
質が前記複合体としてでなはく、同量が前記被験者に投与された場合と比較して前記活性
物質のインビボでの効果が増強される、使用。 - 前記活性物質が前記結合により複合体として前記被験体に投与されたとき、前記活性物
質が前記複合体としてでなはく、同じ方法により同じ量が前記被験者に投与された場合と
比較して、(i)前記活性物質のバイオアベイラビリティーがより大きいか、(ii)前
記活性物質の半減期がより長いか、又は(iii)前記被験体において前記活性物質の全
身毒性がより少ないかのうち少なくとも1つの効果を有する請求項1に記載の使用。 - 前記活性物質が診断薬である請求項1に記載の使用。
- 前記活性物質が治療薬である請求項1に記載の使用。
- 前記活性物質が化学療法剤である請求項1に記載の使用。
- 前記化学療法剤が、(i)ドキソルビシン、(ii)パクリタキセル、(iii)シス
プラチン、および(iv)コンブレスタチンからなる群より選択される請求項5に記載の
使用。 - 前記活性物質が造影剤である請求項1に記載の使用。
- 前記造影剤がガドリニウムキレート(Gd)である請求項7に記載の使用。
- 前記活性物質が、(i)低分子、(ii)放射性核種、(iii)ペプチド、(iv)
