樹状細胞(DC)は、哺乳動物免疫系内の免疫細胞であり、造血骨髄前駆細胞に由来する。より具体的には、樹状細胞は、リンパ(または形質細胞)または骨髄前駆細胞からそれぞれ生じる、リンパ系(または形質細胞様)樹状細胞(pDC)および骨髄系樹状細胞(mDC)の小分類に分類されうる。前駆体細胞のタイプによらず、前駆体細胞から未成熟な樹状細胞が産まれる。未成熟樹状細胞は、高いエンドサイトーシス活性と低いT細胞活性化能とを特徴とする。このように、未成熟樹状細胞は構成的に、そのすぐ隣にある病原体の周囲の環境を採取する。例示的な病原体には、ウイルスまたは細菌が含まれるがこれらに限定されるわけではない。採取は、toll様受容体(TLR)などのパターン認識受容体(PRR)によって達成される。樹状細胞は、toll様受容体などのパターン認識受容体によって病原体を認識すると、活性化して成熟する。
TLRは、病原体関連分子パターン(PAMP)と呼ばれる構造的に保存された分子を認識する1回膜貫通ドメインの非触媒性受容体のクラスである。PAMPは、微生物上に存在して、宿主分子とは識別される。TLRは全ての脊椎動物に存在する。TLR 13個(連続的にTLR 1〜13と呼ばれる)がヒトおよびマウスにおいて同定されている。ヒトはTLR 1〜10を含む。

D、L-ラクチドおよびグリコリドの85:15、120 kDコポリマー(PLG)(Alkermes, Cambridge, MA)を気体発泡プロセスにおいて利用して足場を形成した(Cohen S., Yoshioka T., Lucarelli, M., Hwang L.H., and Langer R. Pharm. Res. 8,713-720 (1991);参照により本明細書に組み入れられる)。GM-CSFを封入するPLGミクロスフェアを標準的なダブルエマルションを用いて作出した(Harris, L.D., Kim, B.S., and Mooney, D.J. J. Biomed.Mater. Res. 42,396-402 (1998);参照により本明細書に組み入れられる)。PLGミクロスフェア16 mgを孔形成物質であるNaClまたはショ糖(粒子サイズが250μm〜425μmとなるようにふるいにかける)150 mgと混合して、圧縮成形した。得られたディスクを高圧CO2環境において平衡にして、圧を急速に低減させて、ポリマー粒子を膨張させて相互接続構造へと融合させる。水の中に浸すことによってNaClを足場から浸出させると、90%多孔性である足場が得られた。腫瘍溶解物をPLG足場に組み入れるために、C57BL/6Jマウス(Jackson Laboratory, Bar Harbor Maine)の背部の皮下で成長させたB16-F10腫瘍の生検をコラゲナーゼ(250 U/ml)(Worthington, Lakewood, NJ)において消化して、40μmセルストレーナーを通して濾過した後、107個/mlと同等の濃度に浮遊させた。腫瘍細胞浮遊液を液体窒素中での急速凍結と融解(37℃)の4サイクルに供した後、400 rpmで10分間遠心した。腫瘍溶解物を含有する上清(1 ml)を収集してPLGミクロスフェアと共に凍結乾燥し、得られた混合物を用いてPLG足場に基づく癌ワクチンを作出した。CpG-ODNをPLG足場に組み入れるために、PEI-CpG-ODN縮合液を50%(重量/体積)ショ糖溶液60μlと共にボルテックスミキサーによって撹拌して、凍結乾燥して乾燥ショ糖と共に混合して最終重量を150 mgとした。PEI-CpG-ODN縮合体を含有するショ糖をブランク、GM-CSF、および/または腫瘍溶解物負荷PLGミクロスフェアと混合して、PLG癌ワクチンを作出した。
CpG-ODN組み入れの組み入れ効率を決定するために、CpG-ODN 50μgと共にPLG足場を調製して、クロロホルム(Sigma Aldrich)1 mlにおいて消化して、水性緩衝液2 mlによって洗浄した。水相を単離して、組み入れられたCpG-ODNの量を、Nanodrop機器ND1000(Nanodrop technologies, Wilmington, DE)を用いて吸光度の読み(光路0.2 mmで計算した260/280、および260/230比)によって決定した。同様に、CpG-ODN放出動態を決定するためにCpG-ODN負荷足場を、インキュベータ(37℃)内のリン酸緩衝生理食塩液(PBS)1 ml中に入れた。様々な時点で、PBS放出培地を収集して新鮮な培地と交換した。PLG足場に組み入れられてPBSの中に放出されたCpG-ODNの全量を経時的に分析して記録した。
DC株、JAWSII(ATCC, Manassas, VA)をインビトロ実験のために用いて、20%FBS(Invitrogen, Carlsbad, CA)および5 ng/ml GM-CSFを添加したα-MEM(Invitrogen, Carlsbad, CA)において維持した。DC活性化に及ぼすCpGに富むオリゴヌクレオチド(CpG-ODN)のインビトロ効果を決定するために、JAWSII細胞を5μg/ml CpG-ODN 1826、5'-tcc atg acg ttc ctg acg tt-3'(SEQ ID NO:10)(Invivogen, San Diego, CA)と共に24時間培養して、0、50、または500 ng/ml GM-CSFの存在下で12時間培養した。DC活性化に及ぼす縮合CpG-ODNの効果を査定するために、混合物をボルテックスミキサーによって撹拌しながらODN-1826溶液をPEI溶液中に滴下することによってCpG-ODNをPEI分子と縮合した(Huang YC, Riddle F, Rice KG, and Mooney DJ. 黒色腫腫瘍溶解物および様々な用量のGM-CSFおよび/またはPEI-CpG-ODN縮合体10μgを有するPLG足場をC57BL/6Jマウスの下左脇腹皮下に埋め込んだ。動物にB16-F10黒色腫細胞(ATCC, Manassas, NJ)105個を、背部頚部への皮下注射によって14日後に投与した。動物を腫瘍成長の発生(およそ1 mm3)に関してモニターして、腫瘍が20〜25 mm(最も長い直径)まで成長した場合には人道的理由から屠殺した。組織学検査に関して、腫瘍を注入後20〜25日目に生検を行ってZ固定液(Anatech, Battle Creek, MI)において固定して、ヘマトキシリンおよびエオジンに関して染色した。T細胞浸潤に関して腫瘍組織を調べるために、アビジン-ビオチン-ペルオキシダーゼVectastain Elite ABCキット(Vector Laboratories)を用いて免疫ペルオキシダーゼ染色を行った。用いた一次抗体はGK 1.5(CD4)および53-6.72(CD8)であり、染色を、DAB+基質色素原(DAKO, Carpinteria, CA)を用いて顕色させた。腫瘍試料(n=3または4)からの切片をNikon光学顕微鏡(Indianapolis, IN)によって40倍および100倍で可視化して、陽性染色T細胞を手動で計数した。PLG癌ワクチンをまた、既に記述されているように(Dranoff G., et al. Proc. Natl. Acad. Sci. USA. 90, 3539-3543(1993);参照により本明細書に組み入れられる)、GM-CSFを発現するように遺伝子改変されてその後放射線照射(3500 rad)されたB16-F10黒色腫細胞を用いた一般的な細胞に基づくワクチンと比較した。放射線照射腫瘍細胞(5×105個)をC57BL/6Jマウスの皮下に注射して、マウスにB16-F10黒色腫細胞105個を14日後に投与した。
本試験における値は全て、平均値±S.D.として表記した。群のあいだの有意差をStudentのt検定によって分析し、0.05未満のP値は有意であると見なした。

実施例1:GM-CSFを負荷したPLG装置
GM-CSF 3μgを負荷したPLGマトリクスをC57BL/6Jマウスの皮下嚢に埋め込んだ。マクロ孔性PLGマトリクスは、GM-CSF、危険シグナル、および癌抗原を既定の空間的時間的様式でインビボで提示し、かつ動員されたDCがプログラムされる際のDCの居所として役立つ。これらのマトリクスは、常在DCを有効に動員するために、最初の5日以内にその生物活性GM-CSF負荷のおよそ60%を放出し、その後次の10日間にわたって生物活性GM-CSFをゆっくりと持続的に放出した(図11A)。
合成CpG-ODN分子をPEIと縮合させて、それによって、細胞膜との会合を促進して、膜貫通輸送を増強することによって細胞インターナリゼーションを促進する陽性荷電の低分子PEI-CpG-ODN縮合体が得られた(図2)。PEIのアミン基とODNのリン酸基のあいだの電荷比7および15でのODN縮合によって、最適な粒子サイズおよび陽電荷(図2BおよびC)が得られたが、高用量でのPEI毒性が原因で、電荷比7を実験において利用した。
DCの有効なCpG刺激は、細胞内局在化を必要とすることから、PEI縮合体のDC活性化に及ぼす効果を評価した。インビトロでPEI-CpG-ODNによって刺激されたDCは、裸のCpG-ODNによって刺激されたDCと比較して増強されたレベルのCD86、MHCII、およびCCR7発現を示し、これは縮合体のDC取り込みと強く相関した(図4AおよびB)。DCは、PEI-CpG-ODNの細胞取り込み時に、細針状の樹状突起の発達と大きい膜膨張とを含む活性化された形態を示し、これによって成熟DCはT細胞を「包み込んで」強い細胞-細胞相互作用を促進することができる。PEI-CpG-ODNによって刺激されたDCの活性化状態は、TNF-αおよびLPS(図3C)によって刺激された陽性対照の状態を代表したか、またはそれを超えており、PEI-CpG-ODN縮合体は、インビトロでCCL19に対するDCの遊走を非刺激DC(図4D)に対して3倍増加させた。
DCを持続的に動員してプログラムするための感染症模倣システムは、宿主DCをPLGマトリクスに誘引するためにGM-CSFの同時放出によって作製されたが、PEI-CpG-ODN縮合体は、おそらくプラスミドDNAに関して示されているように、動員されたDCに複合体を局所に取り込ませる静電結合により、マトリクス内にほとんどが保持された(25日間で>80%)(図6)。特に、最適化された場合、このアプローチによって、インサイチューでマトリクスに常在するMHCIIおよびCCR7発現DCの数はそれぞれ、およそ2.5倍および4.5倍増加した(GM-CSFまたはCpG-ODN送達単独に対して)(図7AおよびB)。興味深いことに、高用量のPEI-CpG-ODN(>50μg)によって、比較的低いMHCII発現および増強されたCCR7発現が得られ、このことは低用量と比較してDC機能の差次的調節を示している(図7A)。最適なCpG-ODNシグナル伝達(〜10〜25μg)は、インサイチューで阻害性GM-CSFレベル(>40 ng/ml)の存在下でDC活性化を増強し、この感染症模倣システムは、エクスビボプロトコルにおいて一般的に投与される活性化DCの数(>106個)をもたらした(図7AおよびB)。
次に、免疫応答を生成するために持続的にDCを動員およびプログラムすることができるか否かを、黒色腫モデルにおいて試験した。このワクチンは、動物の50%が80日間の時間枠において腫瘍を形成しなかったことから(図9)、有意な防御を提供し、この結果は、広く研究された細胞に基づく治療によって得られた結果と驚くほど類似であった(図9)。lys+CpGを投与された動物は、処置後140日でも37.5%が腫瘍を有さず、免疫防御を達成した。
骨髄およびリンパ球系列の双方の造血前駆細胞は、侵入する病原体に対して特異的応答を伝えることができる特異的防衛機構がその各々に備わっている2つの主なカテゴリーのDC、すなわち形質細胞様DC(pDC)および通常の(conventional)DC(cDC)への分化能を有する。この柔軟性によって、おそらくDCサブセットの動員および生成を、所望の免疫応答を誘発させることに最も熟達させることが可能になる。cDCには、長い突出した樹状突起を有する古典的(classical)DC形態を示し、それによって抗原処理およびT細胞に対する抗原提示にそれらを特に適合させる、CD11c+CD11b+およびCD11c+CD8α+細胞が含まれる。pDCは、細菌またはウイルスDNAにおける非メチル化CpGジヌクレオチド配列などの「危険シグナル」に応答して大量の1型インターフェロンを産生することができる丸い非樹状細胞である。

Claims (30)
- 開き、相互接続したマクロ孔を有する足場組成物、
1つまたは複数の樹状細胞を動員して該足場組成物内に一時的に常在させる動員組成物、および
該足場組成物内の樹状細胞を活性化する感染症模倣免疫モジュレーターを含み、該感染症模倣免疫モジュレーターが縮合したシトシン-グアノシンオリゴヌクレオチド(CpG-ODN)を含む、展開組成物
を含む埋め込み可能な装置。 - 縮合したCpG-ODNが5〜50μg存在する、請求項1記載の埋め込み可能な装置。
- 縮合したCpG-ODNがPEI-CpG-ODNを含む、請求項1記載の埋め込み可能な装置。
- 腫瘍抗原をさらに含む、請求項1記載の埋め込み可能な装置。
