Abstract
【解決手段】OX40(CD134)と特異的に結合する、OX40抗体、抗OX40または抗OX40抗体と呼ばれる抗体を提供し、OX40と特異的に結合する抗体には、哺乳類抗OX40抗体、ヒト化抗OX40抗体およびキメラ抗OX40抗体が含まれ、OX40と特異的に結合する抗体および抗体部分配列(フラグメント)には、精製および単離された抗体が含まれる。さらに、それらの抗体等を含有する医薬製剤、製剤による治療法、ならびに、その他、スクリーニング方法および検出方法を含む様々な方法に有用である。

Description
本願は、2005年11月25日出願の出願第60/739,659号の優先権を主張するものであって、これは明示的に参照により本明細書に組み入れられる。
免疫系は、感染症を守るために働く多数の細胞種で構成されている。この免疫系は外来抗原の認識と、自己と非自己を区別する能力に依存している。一部の場合では、自己と非自己との境界がなくなり、自分自身の免疫系により組織の破壊がもたらされる。このような自己免疫応答は、糖尿病、多発性硬化症、および炎症性腸疾患などの衰弱性疾患を引き起こす可能性がある。このような自己免疫応答の主要メディエーターの1つがTリンパ球またはT細胞である。通常、T細胞は自己抗原に対して寛容であるが、いくつかの自己免疫障害ではこの寛容性が失われており、T細胞が健常組織に対する免疫応答を開始する。自己免疫T細胞を除去することまたはそれらの活性化もしくは生存を遮断することで病気の症状は改善する。
本発明は、OX40細胞外ドメインのアミノ酸配列中のエピトープ(例えば、配列番号50)と特異的に結合する単離または精製された抗体を提供する。抗体には、哺乳類(例えば、霊長類またはヒト)OX40配列(例えば、配列番号49)と結合するヒト抗体、ヒト化抗体およびキメラ抗体が含まれる。また、抗体には、OX40アンタゴニスト活性を有するものも含まれる。抗体には、OX40アゴニスト活性を有するものがさらに含まれる。

Claims (14)
- OX40との結合について112V8(ATCC番号PTA-7219)、112Y131(ATCC番号PTA-7218)、または112Y55(ATCC番号PTA-7220)として示されるハイブリドーマ細胞系によって産生される抗体と競合する抗体、またはそのOX40結合性抗体断片。
- OX40との結合について112V8(ATCC番号PTA-7219)、112Y131(ATCC番号PTA-7218)、または112Y55(ATCC番号PTA-7220)として示されるハイブリドーマ細胞系によって産生される抗体と同じエピトープを認識する抗体、またはそのOX40結合性抗体断片。
- 配列番号9で示される成熟重鎖可変領域配列および配列番号10で示される成熟軽鎖可変領域配列を含む抗体と同じエピトープを認識する抗体、またはそのOX40結合性抗体断片。
- 配列番号44で示される成熟重鎖可変領域配列および配列番号45で示される成熟軽鎖可変領域配列を含む抗体と同じエピトープを認識する抗体、またはそのOX40結合性抗体断片。
- 配列番号46で示される成熟重鎖可変領域配列および配列番号47で示される成熟軽鎖可変領域配列を含む抗体と同じエピトープを認識する抗体、またはそのOX40結合性抗体断片。
- 配列番号9の20番目のアミノ酸であるQから配列番号9の141番目のアミノ酸であるGで示されるアミノ酸配列を含む重鎖可変領域及び配列番号10の21番目のアミノ酸であるEから配列番号10の129番目のアミノ酸であるTで示されるアミノ酸配列を含む軽鎖可変領域を含む抗体と同じエピトープを認識する抗体、またはそのOX40結合性抗体断片。
- 請求項1〜6のいずれか一項に記載の抗体またはOX40結合性抗体断片と、製薬上許容される担体または賦形剤とを含む、医薬組成物。
- 112V8(ATCC番号PTA-7219)、112Y131(ATCC番号PTA-7218)、または112Y55(ATCC番号PTA-7220)として示されるハイブリドーマ細胞系によって産生される抗体の成熟重鎖可変領域をコードする核酸。
- 112V8(ATCC番号PTA-7219)、112Y131(ATCC番号PTA-7218)、または112Y55(ATCC番号PTA-7220)として示されるハイブリドーマ細胞系によって産生される抗体の成熟軽鎖可変領域をコードする核酸。
- 配列番号5、38、又は40を含む抗体の重鎖可変領域をコードする核酸。
- 配列番号6、39、 又は41を含む抗体の軽鎖可変領域をコードする核酸。
- 請求項8〜11のいずれか1項に記載の核酸、または前記核酸のいずれかの、重鎖可変領域をコードする核酸及び軽鎖可変領域をコードする核酸の両方、を含むベクター。
- 請求項12に記載のベクターを含む細胞。
- 請求項13に記載の細胞を使用して抗体を産生し、前記抗体を回収することを含む、OX40結合性抗体を作製する方法。
