Description

組換え<Dig>抗体、抗体断片、および融合タンパク質を設計、作製、最適化、および特徴付けするのに必要な条件は、タンパク質配列および(DNA)配列の情報が入手可能なことである。従って、ハイブリドーマクローン19-11に由来する「最初の」マウス<Dig>抗体のVHドメインおよびVLドメインについて、この情報が得られなければならない。この後に行う必要のある実験工程は、(i)<Dig>産生19-11ハイブリドーマ細胞からRNAを単離すること、(ii)このRNAをcDNAに変換し、次いで、cDNAをVHを有するPCR断片およびVLを有するPCR断片に変換すること、ならびに(iii)大腸菌において増やすために、これらのPCR断片をプラスミドベクターに組込み、これらのDNA(および推定タンパク質)配列を決定することである。
Rneasy-Kit(Qiagen)を適用することによって、RNAを5x10e6個の抗体発現ハイブリドーマ細胞(クローン19-11)から調製した。簡単に述べると、沈殿させた細胞をPBSで1回洗浄し、沈殿させ、その後に、溶解のために500μl RLT-Puffer(+β-ME)に再懸濁した。細胞をQiashredder(Qiagen)に通すことによって完全に溶解し、次いで、製造業者のマニュアルに記載のようにマトリックスを介した精製手順(ETOH, RNeasyカラム)に供した。最後の洗浄工程の後に、RNAを50ul RNaseフリー水に溶解してカラムから回収した。回収したRNAの濃度は、1:20に希釈した試料のA260およびA280を定量することによって求めた。単離したRNA試料の完全性(質、分解の程度)は、ホルムアミド-アガロースゲル上での変性RNAゲル電気泳動によって分析した(Maniatis Manualを参照されたい)。これらのRNAゲル電気泳動の例を図1に示した。個々のバンドはインタクトな18sおよび28sリボソームRNAに相当する。これらのバンドが損なわれていない(強度比が約2:1)ことからRNA調製物の質が良好であることが分かる。19-11ハイブリドーマから単離したRNAを凍結し、分注して-80Cで保管した。
標準的な5'-RLM RACEプロトコールについて述べられた反応を用いて、FirstChoice Kit(Ambion)試薬キットを適用することによって、後の(RACE-)PCR反応のためのcDNAを19-11 RNA調製物から調製した。PCR反応にはPwo DNAポリメラーゼを使用した。このために、10ugの19-11 RNAまたは対照RNA(マウス胸腺由来)を適用し、5'RACEアダプターを組込むために述べられたように処理した。本発明者らは「アウターPCR」反応を適用する必要がなく、「インナーPCR」に直接取りかかった。すなわち、これは、5'RACE Inner Primer(キット由来)およびC-κ特異的プライマーまたはCH1特異的プライマーのいずれかからなるプライマーペアを組み合わせることを伴った。VL領域を増幅するためのcκプライマー配列は、
であった。VH領域を増幅するためのCH1プライマー配列は、
であった。これらのプライマーの組み合わせの場合、PCRを行うために60℃のアニーリング温度が適しており、55〜65C/(グラジエントPCR)の温度が適用されてきた(94C 0.5分、55〜65C 1分〜72C 1分、35サイクル、72Cで10分の伸長により完了)。抗体VH領域を含有するDNA断片またはVL領域を含有するDNA断片の特異的増幅の成功は、600bp〜800bpのDNA断片が別個に出現することによって反映される。図2は、これらの規定されたDNA断片が19-11 RNAから得られたことを示している。これらのDNA断片は、<Dig>ハイブリドーマ19-11のVHコード配列およびVLコード配列を含有する。
標準的な分子生物学的技法(Maniatis Manual)によるアガロースゲル抽出および後の精製によって、VHコードPCR断片およびVLコードPCR断片を単離した。正確に製造業者の説明書に従ってpCR bluntII topo Kit(Invitrogen)を適用することによって、Pwoによって作製された精製PCR断片をベクターpCR bluntII topoに挿入した。Topo連結反応物を形質転換によって大腸菌Topo10-one-shotコンピテント細胞に導入した。その後に、VLを含有するインサートまたはVHを含有するインサートを有するベクターを含有する大腸菌クローンを、LB-カナマイシン寒天プレート上にあるコロニーとして特定した。その後に、これらのコロニーからプラスミドを調製し、EcoRIを用いた制限消化によって、ベクターに望ましいインサートが存在することを確認した。ベクターバックボーンは、インサートの両側に隣接するEcoRI制限認識部位を含有するので、EcoRiによって遊離可能な、約800bp(VLの場合)または600bp(VHの場合)のインサートによって、インサートを有するプラスミドが判明した。VHおよびVLの複数のクローンに対する自動DNA配列決定によって、19-11 VLおよびVHのDNA配列および推定タンパク質配列を決定した。<Dig>クローン19-11のVL配列をSeq.ID NO.1に示し、<Dig>クローン19-11のVH配列をSeq.ID NO.2に示した。
抗体配列をヒト化する目的は、マウス由来の最初の抗体の全機能を保持しているが、ヒト免疫系が「異物」と認識する配列もしくは構造が全く無い(またはごくわずかしかない、または無関係な配列もしくは構造を有する)分子を作製することである。この難題に対処することができる様々な手順が利用可能であり、公開されている(Almagro JC, Fransson J Humanization of antibodies. Frontiers in bioscience : a journal and virtual library; 2008 Jan 1;13:1619-33, Hwang WY, Foote J Immunogenicity of engineered antibodies. Methods (San Diego, Calif.); 2005 May;36(1):3-10)。
mu<Dig>19-11 Fv領域の本発明者らのインシリコ構造モデルの基礎は、実験により決定されたVH mRNA配列およびVL mRNA配列から推定されたタンパク質配列である(実施例1に記載、Seq.ID NO.1およびSeq.ID NO.2)。これらの配列によってコードされるタンパク質の構造モデルを、マウス抗体のFvドメインのホモロジーモデリングとエネルギー最小化によってインシリコで作製した。このために、ホモロジーモデリングのために適用するCDRおよびフレームワーク配列を別々に、PDB(Protein DataBank)上でホモロジー検索した。それぞれのCDRおよびフレームワークについて、さらに多くのホモログ構造が重ね合わされた。その後に、軽鎖および重鎖の異なる部分からモデルを構築した後に、複合体の(エネルギー)最小化を行った。本発明者らのホモロジーモデリング手順に起因するmu<Dig>19-11 Fv領域の構造モデルを図3に示した。予想された構造のかなり特殊な特徴の1つは、VH-VL境界面に深く入り込んでいるように見えるよく目につく空洞である。この狭い空洞が形成する主な決定要因は、VHの長いCDR3ループである。空洞の内部は、1個のメチオニン(深部にある残基)、2個のセリン、2個のプロリン、数個のチロシン(壁に隣接する)によって裏打ちされている。この抗体が認識する抗原ジゴキシゲニンは、深い空洞に入り込んでハプテンのように結合すると考えるのが理にかなっている。
抗ジゴキシゲニン抗体のヒト化したVH配列およびVL配列をさらに最適化するために、本発明者らは実験により親(マウス)抗体の構造を決定した。このために、周知の最新法(パパイン消化)を適用して、精製IgGのプロテアーゼ消化によってFab断片を作製した。(Fcを除去する)プロテインAクロマトグラフィーによって、残りのFc断片からFab断片を分離し、その後に、タンパク質断片を除去するためにサイズ排除クロマトグラフィー(Superdex200 HiLoad 120ml 16/60ゲル濾過カラム, GE Healthcare, Sewden)に供した。
望ましいヒト化VHドメインおよびヒト化VLドメインに相当するアミノ酸配列が、CDRグラフティングならびに結合特異性および親和性を調節するさらなる変異の導入に基づく手順を適用することによって規定された。この手順の基礎となる基本原理は、ヒト生殖系列の中での、マウス配列と異なる各アミノ酸配列の「スコア値」の帰属(attribution)である。このスコアは、抗原認識能力または複合体安定性に及ぼすアミノ酸電荷の推定上の影響によって規定される。低いスコアおよび相対的な高い使用頻度に基づいて、ヒト生殖系列が選択される。結果として生じるヒト化分子にt細胞エピトープがごくわずかしか無いように、または全く無いようにするために、このヒト化手順にはTEPITOPE分析(T細胞エピトープを予測する)が含まれる。最初に、この手順によって規定された「ヒト」配列は、スコアが高すぎる場合(負の干渉の高い可能性を示す)、(最初の)マウス配列と交換する必要がある場合がある。これは、CDRまたはCDR配列の周囲領域におけるアミノ酸変化に最も頻繁に必要とされる。場合によっては、安定性および機能性を保持するために、マウス残基への「逆突然変異」がCDRだけでなくフレームワークの中でも必要とされる。
抗ジゴキシゲニン抗体のヒト化VH配列およびヒト化VL配列のさらなる最適化を適用して、ジゴキシゲニンに対する親和性がさらに高いモジュールを作製した。実験により決定された予測構造、ならびにインシリコで計算された予測構造(前記を参照されたい、実験による構造決定のない構造モデリングに基づく)に基づいて、本発明者らは、変化すると親和性に影響し得る3つの位置を特定した。これらは、VHドメインの(Kabat位置)Ser49、Ile57、およびAla60に位置した(図46)。アミノ酸VHSer49とAlaを交換、VHIle57とAlaを交換、およびVHAla60とProを交換することによって、SEQ ID NO 36およびSEQ ID NO 37として列挙した配列を有する抗体誘導体が作製された。この配列からなる結合実体を発現させ、標準的なプロテインAおよびサイズ排除技術を用いて精製することができた(実施例3の「組換えヒト化<Dig>抗体、断片、および二重特異性融合タンパク質の組成、発現、および精製」を参照されたい)。結果として生じた分子は完全に機能し、ヒト化親分子と比較してジゴキシゲニンに対する改善した親和性を示した。これは表面プラズモン共鳴(BiaCore)実験によって証明された(詳細については、実施例4の「組換え<Dig>抗体、断片、および二重特異性融合タンパク質とジゴキシゲニン化抗原との結合」を参照されたい)。これらの実験の結果を図46に示し、VH49、VH57、およびVH60変異を導入することによって、ジゴキシゲニンに対する親和性が約10倍改善したと判明した。この後に、dig結合可変領域の実験により決定された構造を検査することによって、これらの位置の関係が確かめられた。親和性が改善したジゴキシゲニン結合モジュールは、様々な二重特異性抗体誘導体用および多重特異性抗体誘導体用のハプテン結合実体として適用することができる。
さらに詳細な特徴付けのために、ヒト化IgGを形成するために、または他の抗体配列との二重特異性融合タンパク質を作製するために、マウス<Dig>モジュールおよびヒト化<Dig>モジュールをヒト抗体の定常領域と組み合わせた。ヒト化<Dig>IgG、ならびにDigおよび他の標的(例えば、受容体型チロシンキナーゼHer2またはIGF1R)に結合する二重特異性誘導体の作製には、(i)このような分子のアミノ配列およびヌクレオチド配列を設計および規定すること、(ii)これらの分子を、トランスフェクトされた哺乳動物培養細胞において発現させること、ならびに(iii) これらの分子を、トランスフェクトされた細胞の上清から精製することが必要であった。
(最初の)マウスmu<Dig>19-11 Fv領域の結合特異性を有するヒト化IgGを作製するために、本発明者らは、前記で規定されたヒト化VH配列とIgG1のCH1-CH2-CH3のN末端をインフレームで融合した。同様に、本発明者らは、前記で規定されたヒト化VL配列とCκのN末端をインフレームで融合した。結果として生じたhu<her2><Dig>IgG H鎖およびL鎖のアミノ酸配列を、Seq.ID NO.7、 Seq.ID NO.8、およびSeq.ID NO.9に示した。hu<Dig>の結合特異性ならびに受容体型チロシンキナーゼHer2に対する特異性を含む二重特異性抗体誘導体を作製するために、本発明者らは、ヒト化したVH配列およびVL配列によって規定される<Dig>単鎖Fvモジュールと、以前に述べられた<Her2>抗体(例えば、米国特許第5,772,997号)のH鎖のC末端をインフレームで融合した。以前に述べられているVH44-VL100ジスルフィド結合を導入することによって、この<Dig>scFvモジュールをさらに安定化した(例えば、Reiter Y, Brinkmann U, Lee B, Pastan I Engineering antibody Fv fragments, for cancer detection and therapy: disulfide-stabilized Fv fragments. Nature biotechnology; 1996 Oct; 14(10):1239-45)。Her2ならびにジゴキシゲニンに結合する、結果として生じた二重特異性抗体誘導体のアミノ酸配列を、Seq.ID NO.7、 Seq.ID NO.8、およびSeq.ID NO.9に示した。
FreeStyle(商標)293発現系を用いて製造業者の説明書(Invitrogen, USA)に従って、ヒト胚腎臓293-F細胞の一過的トランスフェクションによって、<Dig>IgGおよび二重特異性抗体誘導体を発現させた。このために、対応する二重特異性抗体の軽鎖および重鎖を、原核生物選択マーカーおよび真核生物選択マーカーを有する発現ベクターの中に構築した。これらのプラスミドを大腸菌において増幅し、精製し、その後に、一過的トランスフェクションに適用した。細胞を取り扱うために、Current Protocols in Cell Biology (2000), Bonifacino, J.S., Dasso, M., Harford, J.B., Lippincott-Schwartz, J. and Yamada, K.M.(eds.), John Wiley & Sons, Incに記載のように、標準的な細胞培養法を使用した。懸濁FreeStyle(商標)293-F細胞をFreeStyle(商標)293 Expression培地に入れて37℃/8%CO2で培養し、トランスフェクションの日に、細胞を1〜2x106生細胞/mlの密度で新鮮培地に播種した。250ml最終トランスフェクション体積のために333μlの293fectin(商標)(Invitrogen, Germany)ならびに250μgの1:1モル比の重鎖プラスミドDNAおよび軽鎖プラスミドDNAを用いて、DNA-293fectin(商標)複合体をOpti-MEMI培地(Invitrogen, USA)に溶解して調製した。トランスフェクションの7日後に、IgGまたは二重特異性抗体を含有する細胞培養上清を、14000gで30分の遠心分離および滅菌フィルター(0.22μm)に通す濾過によって清澄化した。精製まで上清を-20℃で保管した。
発現プラスミドトランスフェクションの7日後に、HEK293細胞上清を収集した。この中に含まれる組換え抗体(誘導体)を上清から、プロテインA-Sepharose(商標)(GE Healthcare, Sweden)を用いたアフィニティクロマトグラフィーおよびSuperdex200サイズ排除クロマトグラフィーによって二段階で精製した。簡単に述べると、二重特異性抗体および三重特異性抗体を含有する清澄化培養上清を、PBS緩衝液(10mM Na2HPO4、1mM KH2PO4、137mM NaCl、および2.7mM KCl、pH7.4)で平衡化したHiTrap ProteinA HP(5ml)カラムにアプライした。結合しなかったタンパク質を平衡化緩衝液によって洗い流した。二重特異性抗体を0.1Mクエン酸緩衝液、pH2.8によって溶出させ、タンパク質含有画分を0.1ml 1M Tris、pH8.5によって中和した。次いで、溶出したタンパク質画分をプールし、Amicon Ultra遠心フィルター装置(MWCO:30K, Millipore)によって3mlの体積まで濃縮し、20mM ヒスチジン(Histidin)、140mM NaCl、pH6.0で平衡化したSuperdex200 HiLoad 120 ml 16/60ゲル濾過カラム(GE Healthcare, Sweden)にロードした。精製された抗体および誘導体のタンパク質濃度は、Pace et. al., Protein Science, 1995, 4, 2411-1423に従ってアミノ酸配列に基づいて計算されたモル吸光係数を用いて、280nmでの光学密度(OD)とバックグラウンド補正として320nmでのODを測定することによって求めた。単量体抗体画分をプールし、瞬間冷凍し、-80℃で保管した。後のタンパク質分析および特徴付けのために、試料の一部を用意した。
以下で述べる分析は、ヒト化手順によって完全な結合活性を保持している<Dig>誘導体が得られたかどうか評価するために行った。このために、Biacore T100またはBiacore 3000機器(GE Healthcare Bio-Sciences AB, Uppsala)を用いて表面プラズモン共鳴(SPR)技術によって、組換え<Dig>誘導体の結合特性を分析した。このシステムは分子相互作用研究用に十分に確立している。これは、リガンド/分析物結合を連続リアルタイムモニタリングし、従って、様々なアッセイ設定において会合速度定数(ka)、解離速度定数(kd)、および平衡定数(KD)を求めることができる。SPR技術は、金でコーティングされたバイオセンサーチップ表面近くの屈折率を測定することに基づく。屈折率の変化から、固定化されたリガンドと、溶解状態で注入された分析物の相互作用によって引き起こされる表面の質量変化が分かる。分子が、表面にある固定化リガンドに結合すれば質量が増大し、解離すれば質量が減少する。結合研究を行うために、捕捉用の抗ヒトIgG抗体を、アミンカップリング化学を用いてCM5バイオセンサーチップ表面に固定化した。フローセルを、0.1M N-ヒドロキシスクシンイミドおよび0.1M 3-(N,N-ジメチルアミノ)プロピル-N-エチルカルボジイミドの1:1混合物によって5μl/minの流速で活性化した。特に断りのない限り、10μg/mlの抗ヒトIgG抗体を酢酸ナトリウム、pH5.0に溶解して注入した。これによって、約12000RUの表面密度が得られた。参照対照フローセルを同様に処理したが、捕捉用抗体の代わりにビヒクル緩衝液のみで処理した。1Mエタノールアミン/HCl pH8.5を注入することによって表面をブロックした。ヒト化タンパク質変種の結合と、最初のハイブリドーマ19-11に由来するマウス<Dig>IgGの結合を比較するために、抗ヒトIgG抗体について前記で述べられたのと同様に、捕捉用抗マウスIgG抗体をCM5バイオセンサーチップ表面に固定化した。組換え<Dig>誘導体の機能を評価するために、(i)ヒト化IgG、(ii)hu<Dig>scFvを有する融合タンパク質、または(iii)ジスルフィド安定化scFvを含む組換えhu<Dig>モジュールの結合を、様々なジゴキシゲニン化抗原を用いてアッセイした。結果として生じた結合親和性を、組換えヒト化モジュールが得られたマウス「野生型」DIG-IgGの結合と比較した。

様々な形式でジゴキシゲニン結合モジュールを細胞標的化実体に接続することができる。例えば、このために、「古典的な」抗体断片およびFabまたはFvなどの抗体由来モジュールを適用できるだけでなく、文献に以前に記載されたことのある単一ドメイン抗体様実体も適用することができる。(図6に例示され、多くの分析の例として用いられる)H鎖とのC末端融合に加えて、さらなる形式が本発明者らの研究室において作製されており、機能が評価された。図47aは、マスター分子として完全長IgGをベースとする選ばれた分子を示す。これらは、抗原発現標的細胞への、ハプテンを介したペイロード送達を成し遂げるために作製された。これらは、例として、(ジスルフィド安定化)scFvとIgGのL鎖のC末端との融合、単鎖FabとCH3またはC-κいずれかのC末端との融合、六価分子を作製するようなscFv(および/またはscFab)とL鎖ならびにH鎖との融合を含む。さらに、単鎖リンカーのないジスルフィド安定化Fvを含有する三価実体を含めて三価実体が首尾良く作製され、ハプテンを介したペイロード送達のために適用することができる。融合蛍光タンパク質を含有する二重特異性実体も作製した。様々な形式を作製するために適用されたアミノ酸配列をSEQ ID NO 38〜SEQ ID NO 45として列挙した。全ての分子を哺乳動物細胞において発現させ、標準的なプロテインAおよびサイズ排除技術を用いて良好な収率で精製することができた(実施例3の「組換えヒト化<Dig>抗体、断片、および二重特異性融合タンパク質の組成、発現、および精製」を参照されたい)。
ジゴキシゲニン化ペプチドと、組換えDig結合モジュールを含有する二重特異性抗体誘導体との複合体は、ペプチドに温和な生物物理学的挙動および改善したPKパラメータを付与し得る。さらに、このような複合体は、ペプチドを、二重特異性抗体変種によって認識される抗原を示す細胞に標的化することができる。これらの複合体は、2つの高親和性Dig結合部位において、2つのジゴキシゲニン化ペプチドが結合する(部位ごとに1つのジゴキシゲニン化ペプチドが結合する)1本のヒト化<標的>-<Dig>IgGからなる。このペプチドは、ジゴキシゲニン化されているのにもかかわらず、ならびに抗体と複合体化している間に良好な生物学的活性を保持していることが望ましい。また、二重特異性抗体誘導体の細胞表面標的結合部位は、複合体を形成したジゴキシゲニン化ペプチドの存在下で、その結合特異性および親和性を保持していることが望ましい。
確立したプロトコール(FastMoc 0.25mmol)に従って、Fmoc化学を用いる自動化Applied Biosystems ABI 433Aペプチド合成機において、ペプチド合成を行った。反復サイクルにおいて、対応するFmocアミノ酸を連続してカップリングすることによって、ペプチド配列を組み立てた。カップリング工程ごとに、樹脂(3x2.5min)を、N-メチルピロリドンに溶解した20%ピペリジンで処理することによって、N末端Fmoc基を除去した。カップリングは、DMFに溶解したHBTU/HOBt(各1mmol)およびDIPEA(2mmol)によって活性化されたFmoc保護アミノ酸(1mmol)を用いて行った(45〜60分間ボルテックスする)。カップリング工程ごとに、NMPに溶解したAc2O(0.5M)、DIPEA(0.125M)、およびHOBt(0.015M)の混合物で処理する(10分間ボルテックスする)ことによって、未反応アミノ基をキャップした。各工程の間に、樹脂をN-メチルピロリドンおよびDMFによって大規模に洗浄した。立体障害アミノ酸を自動化二重カップリングに組込んだ。このために、カップリングサイクルの間にキャッピング工程なく、樹脂を1mmolの活性化基本要素によって2回処理した。標的配列が完了したら、標準的なアミノ酸カップリング条件を用いて、Fmoc-12-アミノ-4,7,10-トリオキサドデカン酸(TEG-スペーサー)をFAM5BおよびINF7ペプチドとカップリングした。その後に、全てのペプチド配列(スペーサーを有するFAM5BおよびINF7、スペーサーを有さないメリチン、FALLv1、およびFALLv2)のアミノ末端にFmoc-Cys(Trt)-OHを取り付けた。最後のFmoc脱保護の後に、ペプチド樹脂をフィルターフリットに入れ、トリフルオロ酢酸、水、およびトリイソプロピルシラン(19mL:0.5mL:0.5mL)の混合物によって2.5時間処理した。切断溶液を濾過し、冷(0℃)ジイソプロピルエーテル(300mL)を添加してペプチドを沈殿させて、無色の固体を得た。無色の固体をジイソプロピルエーテルによってくりかえし洗浄した。粗生成物を酢酸/水混合物に再溶解し、凍結乾燥し、その後に、0.1%TFAを含有するアセトニトリル/水勾配を用いた調製用逆相HPLC(Merck Cromolith prep RP-18e カラム, 100x25mm)によって精製した。
対応するシステイン改変ペプチド(6〜20mg)を0.1M KPO4緩衝液(1mL)に溶解した溶液に、100μLのDMFに溶解した等モル量のジゴキシゲニン-3-カルボキシ-メチル-エチルアミドマレイミドを添加した。反応混合物を周囲温度で2〜20時間、穏やかに混合し、濾過し、標的化合物を、0.1%TFAを含有するアセトニトリル/水勾配を用いた調製用逆相HPLC(Merck Cromolith prep RP-18eカラム, 100x25mm)によって単離した。凍結乾燥の後に、ジゴキシゲニン-ペプチド結合体を無色の固体として得た。
組換え<IGF1R>-<Dig>二重特異性抗体および<Her2>-<Dig>二重特異性抗体をカップリング反応のタンパク質成分として使用した。これらの分子の組成および精製は実施例1に記載されている。
生理活性化合物の特異的標的化を目的としたどの技術でも対処する必要のある非常に重要なトピックの1つは、化合物の生物学的活性を保持しなければならないことである。さらに、標的化モジュールの特異性および活性はペイロードの取り付けによる影響を受けてはならない。本発明者らが説明する二重特異性抗体技術は、生理活性ペプチドに対して2つの調整工程を有する。このうち1つは標的化モジュールの改変である。第1の工程において、本発明者らは、ジゴキシゲニンと生理活性ペプチドを共有結合によりカップリングする。第2の工程において、このジゴキシゲニン化ペプチドは、大きなタンパク質である二重特異性抗体誘導体と複合体を形成する。ペプチド活性を保持するために、両工程について改変ペプチドの活性を保証することが重要である。すなわち、活性アッセイによって、(i)ジゴキシゲニン化(digoxygenation)の後にペプチドの機能が保持されていること、(ii)ジゴキシゲニン化ペプチドとマウス<Dig>またはヒト化<Dig>との複合体形成の後に機能が保持されていることが示される必要がある。最後に、(iii)最終複合体において標的化モジュールの結合特異性および親和性がなお保持されていることが示される必要がある。
ジゴキシゲニンによるペプチドメリチン、FALLv1、およびFALLv2の付加または変化によって生物学的活性が変化するかどうか評価するために、本発明者らはインビトロアッセイを行った。これらのペプチドは細胞傷害性であるので、これらの生物学的活性は死細胞数をモニタリングすることによって容易に分析することができる。この数を測定するために、CytoTox-Gloアッセイ(Promega Madison, WI)を使用した。
ハプテンとの共有結合だけでなく、ペプチドと大きな二重特異性抗体分子との複合体形成もこれらの生物学的活性が影響を及ぼすことがある。IgGに由来する二重特異性分子は大きなタンパク質であるので(ペプチドサイズの10〜40倍)、このような分子がペプチドの到達を立体的に妨げ、従って、生物学的活性を妨害し得ることをアプリオリに排除することができない。逆もまた同じであり、ペプチド複合体形成が、二重特異性抗体誘導体の中にある標的化モジュールの特異的な抗原結合機能を妨害し得る可能性がある。これらのトピックに対処するために、本発明者らは、抗体-ペプチド複合体の特異的な抗原結合機能ならびに抗原発現腫瘍細胞株に対するペプチド-抗体複合体のインビトロ活性を分析した。
これらのBiacore実験のために、組換えにより産生されたRTK IGF1R可溶性細胞外ドメインを適用して、<IGF1R>-<Dig>-Dig-ペプチド複合体を特徴付けた。ジゴキシゲニン化ペプチドとの相加的結合を示すために、ジゴキシゲニン化INF7(SEQ. ID. NO.21)およびFALLv1(SEQ. ID. NO.18)を使用した。抗体を、0.1%BSAを含むHBS-P(10mM HEPES、pH7.4、150mM NaCl、0.005%Surfactant P20)で希釈し、10μl/minの流速で注入した。接触時間(会合相)は、50nMの濃度の抗体の場合、1分であった。DIG-INF7の場合、50nMの濃度のジゴキシゲニン化INF7を使用し、90nMの濃度のジゴキシゲニン化FALLv1を使用した。50nMの濃度の可溶性IGF1R断片を使用した。ジゴキシゲニン化ペプチドおよびsIGF1RをHBS-P(10mM HEPES、pH7.4、150mM NaCl、0.005%Surfactant P20)で希釈し、10μl/minの流速で相加的に注入した。接触時間(会合相)は3分であり、解離時間(ランニング緩衝液による洗浄)は各分子について3分であった。0.85%H3PO4の再生溶液を10μl/minの流速で120秒間注入した後に、各結合サイクルの後に、共有結合しなかったタンパク質を除去するために、10mM NaOHを10μl/minの流速で120秒間注入した。シグナルは1シグナル/秒の割合で検出した。全ての相互作用を25℃で行った。これらのアッセイの結果(図16)から、結合の順番に関係なく、ジゴキシゲニン化ペプチドとのIgG複合体は細胞表面抗原に対する結合特異性および親和性を保持していることが分かる。
ジゴキシゲニン化低分子化合物と、組換えDig結合モジュールを含有する二重特異性抗体誘導体との複合体は、温和な生物物理学的挙動および改善したPKパラメータを低分子化合物に付与し得る。さらに、このような複合体は、化合物を、二重特異性抗体変種によって認識される抗原を示す細胞に標的化することができる。これらの複合体は、2つの高親和性Dig結合部位において、2つの(部位ごとに1つの)ジゴキシゲニン化ペプチドが結合する1本のヒト化<標的>-<Dig>IgGからなる。化合物は、ジゴキシゲニン化されているのにもかかわらず、ならびに抗体と複合体化している間に生物学的活性を保持していることが望ましい。また、二重特異性抗体誘導体の細胞表面標的結合部位は、複合体を形成したDig化合物の存在下で、その結合特異性および親和性を保持していることが望ましい。本技術を評価するために本発明者らが例として使用した低分子化合物はドキソルビシンである。ドキソルビシンおよびジゴキシゲニン化ドキソルビシン(=Dig-Dox)の生物学的活性は、インビトロでヒト腫瘍細胞株に対する細胞傷害作用を確かめることによって評価することができる。
ドキソルビシンをSigma-Aldrichから入手した。ジゴキシゲニンとドキソルビシンをカップリングするために、本発明者らは以下の手順を行った。すなわち、ジゴキシゲニン-3-O-メチルカルボニル-ε-アミノカプロン酸-N-ヒドロキシスクシンイミドエステル(20mg, 30.4μmol)をDMF(500μL)に溶解した溶液に、トリエチルアミン(8.4μL, 60.8μmol)を添加し、結果として生じた混合物をすぐに、ドキソルビシン-HCl(16.6mg, 30.4μmol)をDMF(500μL)に溶解した溶液に移した。反応混合物を周囲温度で2時間混合し、濾過し、標的化合物を、0.1%TFAを含有するアセトニトリル/水勾配を用いた調製用逆相HPLC(Merck Cromolith prep RP-18eカラム, 100x25mm)によって単離した。凍結乾燥の後に、ドキソルビシン-ジゴキシゲニン結合体を無色の固体(25.2mg, 76%)として得た。分析用HPLC:tR=13.9分(Merck Chromolith Performance RP-18e、100x4.6mm、水+0.1%TFAアセトニトリル/水+0.1%TFA 80:20, 25分);ESI-MS(陽イオンモード):m/z:C58H75N2O18の計算値:1087.5;測定値:1087.6[M+H]+。ドキソルビシンの分子量は579.98Daである。結果として生じたドキソルビシン-Dig結合体の分子量は1087.24Daである。抗体との複合体形成の時点まで、本発明者らは、DMSOに溶解した結合体を分注して-20℃で保管した。図18はドキソルビシン-ジゴキシゲニン結合体の構造を示す。

ドキソルビシンのジゴキシゲニン化によって生物学的活性が変化するかどうか評価するために、本発明者らは、ドキソルビシンまたはジゴキシゲニン化ドキソルビシンの存在下で増殖させた様々な細胞株を用いてバイオアッセイを行った。その後に、細胞生存を分析した。図20、21、および48aは、ドキソルビシンおよびジゴキシゲニン化ドキソルビシンの生物学的活性を評価するために行ったこれらの細胞生存アッセイの結果を示す。これらのアッセイのために、本発明者らはKPL-4細胞、H322M細胞、またはMDA-MB-468細胞を播種し、プレートに一晩付着させた。翌日、細胞を、示された濃度のドキソルビシンまたはジゴキシゲニン化ドキソルビシンで48時間処理した。次いで、細胞を溶解し、CellTiter Glo Luminescent Cell Viability Assay(Promega, Madison, WI)を適用することによって、細胞生存を評価した。これらのアッセイの結果(図20、21、および48a)から、H322M細胞上でインキュベートした時に、ジゴキシゲニン化ドキソルビシンは生物学的活性の一部を保持していることが分かる。KPL-4細胞は、使用した濃度のジゴキシゲニン化ドキソルビシンとの曝露後に影響を全く示さない。細胞生存アッセイのIC50値は、H322M細胞とインキュベートした時に、非改変ドキソルビシンについては25μMであり、ジゴキシゲニン化ドキソルビシンについては>1000μMであった。KPL-4細胞の場合、細胞生存アッセイのIC50値は、非改変ドキソルビシンについては2.6μMであり、ジゴキシゲニン化ドキソルビシンについては>1000μMであった。細胞生存アッセイのIC50値は、MDA-MB-468細胞とインキュベートした時に、非改変ドキソルビシンについては17μMであり、ジゴキシゲニン化ドキソルビシンについては>1000μMであった。このことから、ジゴキシゲニンによって改変されるとドキソルビシンの活性が大幅に失われることが分かる。この活性低下の理由は、ジゴキシゲニン化ドキソルビシンの機能は失われないが、細胞膜浸透が限定されるからである。この分子は、元々のドキソルビシンより非常に大きく、従って、生体膜に容易に浸透することができない。この限定を実験的に図22に示した。すなわち、免疫蛍光から、ドキソルビシンは膜に浸透し、核にある作用部位に蓄積することが分かる。対照的に、ジゴキシゲニン化ドキソルビシンの大半は細胞に曝露されるとエンドソームの中に蓄積し、従って、細胞内の作用部位に到達しない。それにもかかわらず、ジゴキシゲニン化ドキソルビシンの分子機能は依然として保持されている。これは、エンドソームエスケープ試薬を同時適用することによって証明することができる。エンドソームエスケープ試薬はジゴキシゲニン化ドキソルビシンを細胞質および核に入らせ、結果として、細胞傷害性を大幅に高める(以下を参照されたい)。
ペイロード活性を腫瘍細胞に特異的に送達するために抗体-複合体の特異的な抗原結合機能を利用できるかどうか分析するために、本発明者らは、免疫蛍光研究の後に、標的化ドキソルビシンの生物学的活性を確かめるインビトロ細胞傷害アッセイを行った。Dig-Doxは、514nmレーザーを用いた励起による免疫蛍光によって視覚化できるのに対して、発光は520〜560nmの波長の間で検出される。このために、本発明者らは、細胞上での、<IGF1R>-<DIG>と複合体を形成したdig-doxの標的化および蓄積を光学的に視覚化することができた。図22はIGF1R発現MCF7細胞のIF分析を示し、IGF1R-DIGおよびDig-Doxの複合体は細胞表面に蓄積した。これらの研究の結果から、dig-doxは二重特異性抗体の標的化部分によって標的細胞に特異的に送達されることが分かる。
標的抗原を有する組織または細胞を特異的に画像化するために、ジゴキシゲニン化蛍光基質と、組換えDig結合モジュールを含有する二重特異性抗体誘導体との複合体を適用することができる。これらの複合体は、2つの高親和性Dig結合部位において、画像化技術によって視覚化可能な2つのジゴキシゲニン化基質が結合する(部位ごとに1つのジゴキシゲニン化基質が結合する)1本のヒト化<標的>-<Dig>IgGからなる。画像化化合物は、ジゴキシゲニン化されているのにもかかわらず、ならびに抗体と複合体化している間にその特性(蛍光)を保持していることが必要である。また、二重特異性抗体誘導体の細胞表面標的結合部位は、複合体を形成したDig化合物の存在下で、その結合特異性および親和性を保持していることが望ましい。
ジゴキシゲニン化Cy5を作製するために、モノボックエチレンジアミンを用いて、DIG-カルボキシメチル-NHSエステル(DE3836656)を変換した。その後に、bocを除去し、遊離したアミンをCy5-NHSエステル(GE Healthcare, PA15106)と反応させた。DIG-Cy5を精製するために、RP18カラムを用いたHPLCを行った。溶出剤Aは、0.1%TFAを含有するH2Oであり、溶出剤Bは、0.1%TFAを含有するアセトニトリルであった。60分にわたって行った溶出の間に、溶出剤Bの濃度を0%から100%に上げた。

Cy5のジゴキシゲニン化によってCy5の蛍光特徴が変化するかどうか評価するために、本発明者らはCy5の励起スペクトルおよび発光スペクトルを比較し、これと、新しく作製したDig-Cy5のスペクトルおよび二重特異性抗体との複合体の中にあるDig-Cy5のスペクトルを比較した。表4は、これらの分析の結果をまとめている。すなわち、Cy5とジゴキシゲニンとの結合体化ならびにDig-Cy5と抗体との複合体形成はCy5の蛍光特徴を妨害しない。本発明者らは、抗体を介した標的化およびインビボ画像化のために、Dig-Cy5およびDig-Cy5-複合体を適用できると結論付けた。
ジゴキシゲニン化Cy5の複合体形成が二重特異性標的化モジュールの結合特徴を変えるかどうか評価するために、本発明者らは表面共鳴分析を適用した。抗体親和性を確かめるために、Her2(ならびにIGF1R)の細胞外ドメインを抗原として使用した。これらの分析は実施例4に詳述されている。表5はこれらの分析の結果をまとめている。すなわち、Dig-Cy5と抗体との複合体形成は標的化モジュールの結合親和性を妨害しない。
画像化試薬を腫瘍細胞に特異的に送達するために抗体-複合体の特異的な抗原結合機能を利用できるかどうか分析するために、本発明者らはインビボで近赤外蛍光画像化研究(NIRF)を行った。これらの研究のために、50μgの<Her2>-<Dig>-Dig-Cy5または<IGF1R>-<Dig>Dig-Cy5複合体を、皮下腫瘍異種移植片を有する免疫不全マウスに静脈内注射した。これらの異種移植片は、抗原Her2(KPL4細胞)または抗原IGF1R(H322M細胞)を発現していた。その後に、CRIのMaestroシステムを用いてNIRF画像化を行った。動物を測定用チャンバーに入れ、色素に応じて規定されたスペクトル波長アレイにわたって蛍光を測定した。様々な既定のスペクトル、例えば、自己蛍光および色素シグナルの分離を可能にする特殊なソフトウェアによって、得られた画像の各ピクセルのスペクトル情報を解析した。様々な試料を比較するために、これらの分離した特異的なシグナルをMaestroソフトウェアによって定量した。
抗原発現細胞に核酸を特異的に標的化するために、siRNAなどのジゴキシゲニン化核酸と、組換えDig結合モジュールを含有する二重特異性抗体誘導体との複合体を適用することができる。このような複合体は、ペプチドを、二重特異性抗体変種によって認識される抗原を示す細胞に標的化することができる。これらの複合体は、2つの高親和性Dig結合部位において、2つの(部位ごとに1つの)ジゴキシゲニン化核酸が結合する1本のヒト化<標的>-<Dig>IgGからなる。核酸は、ジゴキシゲニン化されているのにもかかわらず、ならびに抗体と複合体化している間に機能を保持していることが望ましい。また、二重特異性抗体誘導体の細胞表面標的結合部位は、複合体を形成したジゴキシゲニン化核酸の存在下で、その結合特異性および親和性を保持していることが望ましい。
1.オリゴリボヌクレオチドの合成および精製:
ABI394合成機(Applied Biosystems)を用いた固相上でのホスホルアミダイト技術に従って、10μmolスケールでオリゴリボヌクレオチドを合成した。合成は、多孔性ガラスで作られた固体支持体(CPG, 520Å, 75μmol/gのローディング、Prime Synthesis, Aston, PA, USAから入手、または3'-PT-アミノ-Mod. C6 CPG、37μmol/gのローディング、Glen Research, Sterling, Virgina, USAから入手)上で行った。通常のRNAホスホルアミダイト、2'-O-メチルホスホルアミダイト、および2'-Fホスホルアミダイト、ならびに補助試薬はProligo(Hamburg, Germany)から購入した。合成サイクルを変更することなく、対応するホスホルアミダイト(GE Healthcare, Munich Germanyから入手)を用いて、Cy5蛍光色素を5'末端に取り付けた。固相合成が完了した後に、支持体に結合しているオリゴマーの切断および脱保護を行った。次いで、AKTA Explorerシステム(GE Healthcare)においてSource 15Q, SPSC-150, 150x8mmカラム(Bischoff, Leonberg, Germany)を用いた陰イオン交換(AEX)HPLCによって、粗オリゴマーを精製した。
ジゴキシゲニン-3-O-メチルカルボニル-ε-アミノカプロン酸-N-ヒドロキシ-スクシンイミド(Roche, Basel, Switzerland)をジメチルスルホキシド(DMSO)(Fluka, Buchs, Switzerland)に溶解した。この溶液を、緩衝液(0.1Mホウ酸Naを含む0.1M KCl、pH8.5)に溶解した精製アミノ改変RNAに添加した。反応はAEX HPLCによって管理した。定量反応の場合には、3M NaOAc、pH=5.2およびエタノール(1:32)を用いた沈殿によって、結合体化RNAを単離した。反応が定量反応でなければ、精製工程を行った。この場合、AKTA Explorerシステム(GE Healthcare)においてDNAPac PA-100 22x250mm(Dionex, Idstein, Germany)を用いたAEX HPLCによって、オリゴマーを精製した。緩衝液Aは、6M尿素、10mM NaClO4、20mM Tris、1mM EDTA;pH7.4、20%ACNであり、緩衝液Bは、6M尿素、500mM NaClO4、20mM Tris、1mM EDTA;pH7.4、20%ACNであった。4.5mL/min(60℃)の流速を使用した。260nm、280nmでUVをトレースし、Cy5の場合は643nmを記録した。55分以内に25%B〜55%Bの勾配を使用した。適切な画分をプールし、3M NaOAc、pH=5.2およびエタノール(1:32)によって沈殿させた。遠心分離後に、ペレットを水に溶解した。
等モルのRNA溶液を組み合わせることによって相補鎖をアニーリングした。混合物を凍結乾燥し、望ましい濃度となるように適量のアニーリング緩衝液(100mM NaCl、20mMリン酸ナトリウム、pH6.8)を用いて再構成した。この溶液を95℃の水浴に入れ、3時間以内に室温まで冷却した。
組換え<IGF1R>-<Dig>二重特異性抗体および<Her2>-<Dig>二重特異性抗体をカップリング反応のタンパク質成分として使用した。これらの分子の組成および精製は実施例1に記載されている。
ネイティブ質量分析を適用して、タンパク質複合体の分子量を求めることができる。本技術は、変性質量分析とは対照的に中性pHの水性揮発性溶媒を用いて行われ、質量分析中の非共有結合タンパク質複合体の解離または破壊を最小限にするように最適化されている(Sharon M, Robinson CV (2007), The role of mass spectrometry in structure elucidation of dynamic protein complexes, Annu Rev Biochem. 2007 ;76: 167-93.; Heck AJ (2008), Native mass spectrometry: a bridge between interactomics and structural biology, Nat Methods. 5(11):927-33.)。それにもかかわらず、試料調製中および質量スペクトル取得中の複合体解離を阻止するためには、分析されるタンパク質複合体はある程度安定していることが依然としてネイティブ質量分析に必要な条件である。
生理活性化合物の特異的標的化を目的としたどの技術でも対処する必要のある非常に重要なトピックの1つは、化合物の生物学的活性を保持しなければならないことである。さらに、標的化モジュールの特異性および活性はペイロードの取り付けによる影響を受けてはならない。本発明者らが説明する二重特異性抗体技術は、生理活性核酸に対して2つの調整工程を有する。このうち1つは標的化モジュールの改変である。第1の工程において、本発明者らは、ジゴキシゲニンと生理活性核酸、例えば、siRNAを共有結合によりカップリングする。第2の工程において、このジゴキシゲニン化核酸(siRNA)は、大きなタンパク質である二重特異性抗体誘導体と複合体を形成する。核酸の活性を保持するために、両工程について改変核酸の活性を保証することが重要である。すなわち、活性アッセイによって、(i)ジゴキシゲニン化の後に核酸の機能が保持されていること、(ii)ジゴキシゲニン化核酸と、標的化モジュールを含有する<Dig>との複合体形成後に機能が保持されていることが示される必要がある。最後に、(iii)最終複合体において標的化モジュールの結合特異性および親和性がなお保持されていることが示される必要がある。
ジゴキシゲニンによるsiRNAの付加または変化によって生物学的活性が変化するかどうか評価するために、本発明者らはインビトロアッセイを細胞培養において行った。このために、本発明者らは、Eg5 mRNAを不活性化するsiRNAの活性を評価することにした。Eg5 siRNA活性の直接的な作用は、そのコグネイトmRNAのダウンレギュレーションである。これは、bDNA(分枝DNA)アッセイによって定量することができる。bDNA(分枝DNA)アッセイは、細胞内の特異的なmRNAの量を検出する(Burns et al., 1999, Molecular Endocrinology)。これらのアッセイを行うために、本発明者らは、規定された数のそれぞれの細胞タイプを96ウェルプレートに播種し、一晩付着させた。翌日、細胞を望ましい量のある特定のsiRNAでトランスフェクトするか、mRNAレベルに対する分析しなければならない作用のある薬剤で処理した。24時間後に、mRNAレベルを定量するために、製造業者(Affymetrix)の説明書に従って、QuantiGeneキットプロトコールを行った。簡単に述べると、細胞溶解産物を遺伝子特異的プローブセットの存在下で捕捉プレートに移し、次いで、53℃で一晩インキュベートした。ウェルを洗浄した。次いで、ウェルをAmplifierおよびアルカリホスファターゼ連結標識プローブと連続して53℃でインキュベートし、インキュベーションの間に洗浄を行った。最後の洗浄の後に、発光アルカリホスファターゼ基質ジオキシタン(dioxitane)を添加し、53℃で30分間インキュベートした。InfiniteF200ルミネッセンスリーダー(Tecan Austria, Groding)を用いて、発光を検出した。Eg5を標的とするsiRNAの生物学的活性はbDNA分析によって確かめることができるだけでなく、成長細胞におけるEg5 mRNA枯渇によって引き起こされる表現型によっても確かめることができる。Eg5は、キネシン様タンパク質ファミリーに属するモータータンパク質である。Eg5はKSP(キネシン紡錘体タンパク質)とも知られ、二極性紡錘体の形成に必須であり、紡錘極の適切な分離に必要とされる。Eg5が枯渇すると特徴的な単極紡錘体が形成し、紡錘体チェックポイントが活性化する。これにより有糸分裂停止が引き起こされ、最終的にアポトーシスへとつながる(Tao et.al., Molecular and cellular biology. 2007 Jan; 27(2):689-98)( Tao W. et.al., Cancer Cell. 2005;8:49-59)。このために、 siRNAによってEg5が不活性化すると、多くの場合、培養細胞への細胞傷害性表現型が媒介される。従って、Eg5 siRNAの生物学的活性は、生細胞の数をモニタリングすることによって分析することができる。
様々な形式でジゴキシゲニン結合モジュールを様々な細胞標的化実体に接続することができる。ヒトIGF1受容体またはHer2を認識する抗体とのC末端融合に加えて、様々な細胞表面抗原を認識する様々な他の抗体を、ハプテンを介したペイロード送達のためのビヒクルに変換した。作製され、精製され、および特徴付けられた例には、ヒトCD22抗原、ヒトCD33抗原、Lewis Y癌関連炭水化物抗原、ヒトおよびマウスのVEGF受容体2または受容体CDCP1を認識する<Dig>含有送達ビヒクルが含まれる。図47に記載の形式の組み合わせを適用して、1つの表面標的分子にある2つ(またはそれより多い)別々の標的または別々のエピトープを認識する分子でさえもDig標的化実体と組み合わせることができる。図51aおよびbは、様々な表面標的分子を発現する抗原発現標的細胞への、ハプテンを介したペイロード送達を実現するために作製された選択された分子を示す。様々な細胞表面抗原を取り扱う標的ペイロード送達用のビヒクルを作製するために適用されたアミノ酸配列をSEQ ID NO 51〜SEQ ID NO 60として列挙した。CDCP1を認識する二重特異性標的化実体を作製するために用いられたアミノ酸配列は、出願EP09011046.1に記載されている。全ての分子を哺乳動物細胞において発現させ、標準的なプロテインAおよびサイズ排除技術を用いて良好な収率で精製することができた(実施例3の「組換えヒト化<Dig>抗体、断片、および二重特異性融合タンパク質の組成、発現、および精製」を参照されたい)。図51c〜gは、様々な標的抗原を認識する様々な形式のこれらの全ての分子が、ペイロード送達の必要条件である標的化特異性ならびにジゴキシゲニン結合能力および親和性を完全に保持していたことを示す。すなわち、これは、表面プラズモン共鳴(BiaCore)実験(詳細については、実施例4の「組換え<Dig>抗体、断片、および二重特異性融合タンパク質とジゴキシゲニン化抗原との結合」を参照されたい)、ならびにFACS分析(データは示さず)によって証明された。FACS分析の場合、標的-Dig二重特異性抗体を細胞とインキュベートし、その後に、蛍光標識ジゴキシゲニンと別々にインキュベートした。これらの実験の結果を図51c〜gにまとめた。データから、細胞表面標的抗原ならびにジゴキシゲニンに対する結合特異性および親和性は、親抗体またはジゴキシゲニン結合部分と比較して変化しないことが判明した。従って、ハプテンを介した標的ペイロード送達のビヒクルとして、多くの異なる標的抗原を認識する多くの異なる形式およびモジュールを適用することができる。これらの標的化ビヒクルの一部、特に、腫瘍細胞上に高密度にある、内部移行される抗原を認識する標的化ビヒクルは、標的ペイロード送達に特に適しているかもしれない。例えば、腫瘍細胞に豊富にあるLeY炭水化物抗原を認識する二重特異性分子は、核酸および他のペイロードを腫瘍細胞に送達するのに非常に効果的な担体である(下記の実施例15、Leyを介したDPC標的化を参照されたい)。
siRNAはかなり大きなサイズがあり、高度に荷電しているので、二重特異性標的化モジュールとsiRNAとの複合体形成は標的化モジュールの特異的な抗原結合機能を妨害し得る可能性がある。これらのトピックに対処するために、本発明者らは、(i)抗体-siRNA複合体の特異的な抗原結合機能ならびに(ii)これらの複合体がsiRNAを抗原発現標的細胞にインビトロで標的化する能力を分析した。最後に、本発明者らは、(iii)インビボでの腫瘍異種移植片モデルにおけるsiRNA標的化のNIRF画像化によって、抗体-siRNA複合体の機能を確かめた。
<標的>-<Dig>Dig-siRNA複合体の抗原結合機能を確かめるために、本発明者らは、<Her2>-<Dig>について実施例4に述べられたのと同じ実験設定を用いて表面プラズモン共鳴を適用した。<IGF1R>-<Dig>の場合、実施例4に記載の方法の以下の変更を適用した。すなわち、2μg/mlの抗ヒトIgG抗体を、酢酸ナトリウム、pH5.0に溶解して注入した。これによって、約600RUの表面密度が得られた。それぞれの結合サイクルの後に共有結合していないタンパク質を除去するために、0.85%H3PO4を5μl/minで60秒間注入し、次いで、5mM NaOHを5μl/minで60秒間注入することによって、再生を行った。これらのBiacore実験のために、組換えにより産生されたRTK Her2可溶性細胞外ドメインを用いて、<Her2>-<Dig>-Dig-siRNA複合体を特徴付け、組換えにより産生されたRTK IGF1R可溶性細胞外ドメインを適用して、<IGF1R>-<Dig>-Dig-siRNA複合体を適用した。これらのアッセイの結果(表8にまとめた)から、ジゴキシゲニン化siRNAとのIgG複合体は細胞表面抗原に対して結合特異性および親和性を保持していることが分かる。この表は、<Her2>-<Dig>または<IGF1R>-<Dig>二重特異性標的化モジュールに対するジゴキシゲニン化siRNAの親和性も示している。
<Her2>-<Dig>-Dig-siRNA複合体および<IGF1R>-<Dig>-Dig-siRNA複合体が抗原発現細胞へのペイロードの特異的標的化を媒介するかどうか分析するために、本発明者らは、siRNAをDigならびに蛍光標識によって同時に標識できるという事実を利用した。それによって、顕微鏡観察または他の画像化技術によってsiRNAの局在を視覚化することが可能になる。
<Her2>-<Dig>-Dig-siRNA複合体および<IGF1R>-<Dig>-Dig-siRNA複合体が、細胞培養実験だけでなく生きている動物でも抗原発現細胞へのペイロードの特異的標的化を媒介するかどうか分析するために、本発明者らは、siRNAをDigならびに蛍光標識によって同時に標識できるという事実を利用した。それによって、画像化技術によって、生きている動物におけるsiRNAの局在を視覚化することが可能になる。このタスクのために本発明者ら適用した技術はインビボでの近赤外蛍光画像化(NIRF)である。これらの研究のために、50ugの<Her2>-<Dig>-Dig-siRNA-Cy5または<IGF1R>-<Dig>Dig-siRNA-Cy5複合体を、皮下腫瘍異種移植片を有する免疫不全マウスに静脈内注射した。これらの異種移植片は、抗原Her2(KPL4細胞)または抗原IGF1R(H322M細胞)を発現していた。その後に、実施例10に記載のようにMaestroシステムを用いてNIRF画像化を行った。
mRNAの特異的破壊を媒介するために、siRNAは標的細胞の細胞質に到達しなければならない。従って、特異的siRNA活性を送達するための重要な要因の1つは、分子が細胞に送達されるだけでなく(これは実施例12および実施例13において証明されている)、十分な量のsiRNAがこれらの細胞の細胞質に導入されなければならないことである。このために、これらの分子は、少なくとも1回、生体膜に浸透しなければならない。生物製剤は膜を容易に通過しないので、このプロセスは、siRNA活性を効果的に送達するために克服しなければならないボトルネックである。このボトルネックを克服する手段は、膜浸透、膜を通るタンパク質移行、または膜破壊プロセスが関与し得るエンドソームエスケープ機構もしくは小胞エスケープ機構でもよい。siRNAを細胞質に送るプロセスの助けとなり得る要因の1つは、(i)内部移行後に二重特異性標的化部分からsiRNAペイロードを放出する能力(siRNAペイロードの内部移行は実施例13に示されている)である。これによって細胞質内への進入が容易になり得る。なぜなら、導入される必要のある実体はsiRNA標的化複合体全体より小さい(Dig-siRNAのみである)からである。標的細胞の細胞質へのsiRNAの導入を容易にし得る別の原理は、(ii)標的化複合体と、エンドソーム機能モジュレーターまたはエンドソームエスケープ/破壊モジュールとの組み合わせである。
ジゴキシゲニン化siRNAと<標的>-<Dig>二重特異性標的化モジュールとの複合体は規定されており、<Dig>モジュールの親和性が十分なために安定している。他方で、Dig-siRNAとタンパク質との結びつきは共有結合でなく、抗体-ハプテン相互作用からなる。複合体の非共有結合カップリング方式が内部移行後のペイロード放出を媒介できるか分析するために、本発明者ら二重標識siRNA複合体を用いた蛍光顕微鏡実験を行った。これらの実験のために、本発明者らは、Cy5標識Dig-siRNAを適用して、細胞内のsiRNAの位置を突き止めた。標的化複合体のタンパク質部分を視覚化するために、ウサギ抗ヒトκ軽鎖抗体を適用した後に、Alexa-fluor488標識ヤギ抗ウサギ抗体とインキュベーションした。両実体も、異なる励起チャンネルおよび発光チャンネルで顕微鏡によって視覚化することができた。両チャンネルからシグナルを視覚化したものを重ね合わせると、抗原発現細胞への適用後にタンパク質およびsiRNAの経路を同時に追跡する機会が得られる。視覚化技術および顕微鏡観察技術の実験上の詳細は実施例12に記載されている。図33および図34は、これらの共染色実験の結果を示す。Her2発現KPL4細胞を<Her2>-<Dig>Dig-siRNA-Cy5複合体に37℃で曝露し、その後に、顕微鏡によって分析した(図37)。同様に、IGF1R発現H322M細胞を<IGF1R>-<Dig>Dig-siRNA-Cy5複合体に37℃で曝露し、その後に、顕微鏡によって分析した(図40)。どちらの例も、複合体と細胞表面との結合およびその後の内部移行が観察された。初期には、タンパク質およびsiRNAシグナルは細胞表面およびエンドソーム内に共存した。このことから、完全複合体と標的細胞が結合し、完全複合体が標的細胞に内部移行したことが分かる。後期には、本発明者らは、siRNA関連シグナルからのタンパク質関連シグナルの分離を観察した。この分離は、<Her2>-<Dig>Dig-siRNA-Cy5を用いたKPL4細胞において、ならびに<IGF1R>-<Dig>Dig-siRNA-Cy5に曝露されたH322M細胞において観察された。
mRNAの特異的破壊を媒介するためには、十分な量のsiRNAを、これらの細胞の細胞質内に導入しなければならない。細胞質においてRNAi機構の成分はsiRNA二重鎖を特異的に認識し、アンチセンスsiRNA鎖を、RNA誘導性サイレンシング複合体と呼ばれるタンパク質複合体に組込む。次いで、アンチセンス鎖はそれぞれのmRNAと対になるとRISCによってmRNAが切断され、それによって、特異的なmRNA破壊が媒介される。
脂質をベースとするトランスフェクション試薬が、<DIG>二重特異性抗体によって特異的に標的化されたsiRNAのエンドソームエスケープを助ける能力を調べた。従って、脂質をベースとする市販のトランスフェクション試薬であるDharmaFECT(Dharmaconにより提供)を使用した。
siRNAの特異的標的化を目的としたどの技術でも対処する必要のある非常に重要なトピックの1つは、生物学的活性のために核酸が標的細胞の細胞質に到達しなければならないことである。核酸は高度に荷電しており、それ自体では膜を容易に通過しないので、標的化された(および内部移行した)siRNAが細胞の細胞質に放出されることがsiRNA送達の大きなボトルネックである。
ジゴキシゲニン化DPCを作製するために、NHS-ジゴキシゲニンを様々な比でPBAVE-CDM-PEG反応ミックスに添加するという変更を加えて、他に記された手順を適用した(Rozema DB et. al., Dynamic PolyConjugates for targeted in vivo delivery of siRNA to hepatocytes. Proceedings of the National Academy of Sciences of the United States of America; 2007 Aug 7; 104(32): 12982-7 PMID:17652171、特に、WO2008/0022309、米国特許出願公開第US2008-0152661A1号、同第US2008-0287630号、同第US2008-0281074A1号、同第US2008-0287628A1号、同第US2008-0269450A1号、米国特許第7,098,032号、同第7,019,113号、同第6,919,091号)。簡単に述べると、ジゴキシゲニン化DPCを作製するために、Hepes緩衝液(500mM, pH8)に溶解した2.5μMのポリブチルアミノビニルエーテル(PBAVE)溶液を、(等張性グルコース緩衝液または1xリン酸緩衝食塩水PBSに溶解した)0.07M N-ヒドロキシスクシンイミドNHS-DIG溶液と混合する。混合物を室温で30分間インキュベートする。このために適用することができるPBAVE:NHS-Dig比にはモル比1:1または様々な他の比が含まれる。30分間のインキュベーション工程の後に、CDM-PEG550溶液(0.11M)をPBAVE-DIG複合体に1:35の比(Dig-PBAVE:CDM-PEG)で添加し、もう60分間室温でインキュベートする。その後に、1.3ml Sephadex G-50スピンカラムを適用することによって、カップリングされていない過剰なCDM-PEGを除去する。NHS-DigおよびCDM-PEGをPBAVEスキャフォールドに取り付けると、pH感受性のカルボキシジメチルマレイン酸無水物CDM結合が形成する。
<標的>-<Dig>Dig-DPC複合体の抗原結合機能を確かめるために、本発明者らはFACSアッセイを適用した。これらのアッセイのために、<IGF1R-Dig>-DPC複合体を、IGF1Rを発現するH322M細胞に適用した。図40に示したように、FACS分析をH322M細胞株およびRaji細胞株に対して行った。7.5nMのモル濃度の抗体または複合体および30nMの濃度の二次抗体(必要な場合、すなわち、対照)を与えた細胞をPBS+5%FCSに懸濁した。一次抗体を氷上で30分間インキュベートし、洗浄工程の後に、二次抗体を氷上で30分間添加した。BD FACS Canto IIによって試料を測定する前に、細胞を洗浄した。APC(対照)またはCy3(DPCを含有する試料)を検出するのに適した適切な波長で、シグナルを検出した。H322Mの場合、IGF1Rが発現しており、結合体とカップリングされたIGF1R-DIG二重特異性抗体のシグナルが得られる。Raji細胞株はCD22を発現し、この細胞株にはCD22-DIG+結合体の陽性シグナルがある。両細胞株とも、かなり高い二次抗体濃度によって引き起こされる中等度のバックグラウンドシグナルを示す。しかしながら、結合体とカップリングされた二重特異性抗体には二次抗体がアッセイに無いので、これらの試料において検出されるシグナルは全て特異的な抗原結合によるものである。本発明者らの実験分析から、IGF1R.Dig::Dig-DPC複合体は、IGF1Rを発現するH322M細胞に結合することが証明された。対照的に、この分子は、IGF1Rを発現しないRaji細胞またはRamos細胞に結合しない。これにより、DPCとカップリングされた二重特異性抗体について結合および特異性は保持されていることが判明した。さらなる実験から、CD22抗原を発現するRamos細胞およびRaji細胞は、CD22抗原を認識するDig二重特異性抗体と複合体を形成したDPCに結合することが証明された。このことは、DPCとカップリングされた二重特異性抗体について結合および特異性が保持されていることを裏付けている。
抗体-DPC複合体がインビボでDPCを腫瘍に標的化するのに十分な安定性があるかどうかという問題に対処するために、動物実験を行った。これらの実験のために、Cy3標識ジゴキシゲニン化DPCと複合体を形成したCy3標識Her2-Dig二重特異性抗体を、Her2発現KPL4腫瘍異種移植片を有する動物に注射した。腫瘍におけるDPCの蓄積は近赤外蛍光画像化(NIRF)によって様々な時点で検出され、(二重特異性抗体と複合体を形成していない)非標的化Cy3標識Dig-DPCの蓄積と比較された。図42は、この分析の結果を示す。すなわち、標的化されていないDPCは抗原陽性細胞において蓄積がほとんどないか、または蓄積を全く示さない。対照的に、二重特異性Dig-抗体によって標的化されたDPCは腫瘍に蓄積した明らかな証拠を示す。本発明者らは、DPCをインビボで望ましい標的部位に標的化するためにDig.二重特異性抗体を使用できると結論付けた。
LeYは、結腸、胃、卵巣、乳房、および肺の多くの粘液性癌ならびに一部の表皮癌の表面に見られるLeYファミリーの炭水化物抗原である。この抗原を標的化するために、二重特異性LeY-DIG抗体を構築した。<LeY>配列は、LeY抗原に対するマウス抗体であるモノクローナル抗体B3に由来した(Pastan, et al., 1991, Cancer research)。B3は限られた数の正常組織としか反応しないので、癌を治療するための理想的な候補である(Brinkmann et al., 1991, PNAS)。さらに、LeY抗原は、前述した癌腫に由来する細胞に非常に高密度にあるので、この抗原と結合するとLeYおよび<LeY>の複合体は高い内部移行率を示す。
SEC-MALLS(サイズ排除クロマトグラフィーマルチアングルレーザー光散乱)は、生体分子をゲル濾過カラムによって分離し、3種類の検出システム:UV/可視光、屈折率(RI)、および光散乱(LS)に送る分析法である。UV/可視光およびRIの検出器から試料濃度の情報が得られる。LSは、溶解状態にある高分子および広範囲の粒子を特徴付けるための非侵襲的な技法である。ほとんどの特徴付け法とは対照的に、LSは外部の較正標準を必要としない。この意味では、LSは絶対的な技法である。ここで使用したWyatt Technology機器は、絶対的な分子特徴付けのために、以下の2種類の光散乱測定を行う。
・古典的光散乱/静的光散乱:ここでは、散乱光の強度は角度の関数として測定され、モル質量、rms半径、および第2ビリアル係数(A2)を得ることができる。ある種の粒子の場合、古典的光散乱からサイズ、形状、および構造を得ることができる。
・準弾性光散乱(QELS)または動的光散乱(DLS):QELS測定では、高速フォトンカウンター(fast photon counter)を用いて散乱光シグナルの時間依存的な変動を測定する。QELS測定値から、高分子または粒子の流体力学半径(hydrodynamic radius)を求めることができる。
<標的>-<Dig>二重特異性抗体を用いて、DIG標識カーゴを細胞に特異的に送達することができる(実施例6)。持ち上がった問題の1つは、<標的>-<Dig>二重特異性抗体を用いてDIG標識タンパク質も細胞に標的化できるかということである。この問題に答えるために、本発明者らは、DIG部分を、前記のように高感度緑色蛍光タンパク質(eGFP)と結合体化した。eGFPは、スペクトル特徴が改善され、蛍光、光安定性が増大し、主励起が488nmにシフトしている、オワンクラゲ(Aequorea Victoria)に由来するGFPの変異体である(Heim R, Cubitt A, Tsien R (1995). Nature 373(6516):663-4.)。
<LeY>-<DIG>二重特異性抗体を用いて、DIG標識タンパク質eGFPを標的細胞に向けることができるかどうか分析するために、本発明者らはFACS分析を使用した。eGFPは蛍光を発するので(主励起488nm、主発光509nm)、このタンパク質の存在は、一般的に用いられるFITCフィルターセットによって検出することができる。本発明者らは、2つの[1:2]DIG部分または3つの[1:3]DIG部分とカップリングされた2種類のeGFP変種を作製した。
細胞表面結合受容体を標的とする抗体は、多くの場合、結合すると内部移行する。内部移行後に、標的に結合した抗体はエンドソーム区画に輸送される。これらの区画は連続的に酸性化される。eGFPなどのpH依存性フルオロフォアの助けを借りると、抗体の内部移行をモニタリングすることができる。
ジゴキシゲニン化ペイロードを標的化ビヒクルにカップリングするためにジゴキシゲニン結合モジュールを適用することは、ハプテンを介したペイロード送達を実現する技術的可能性の1つである。しかしながら、この考えは、ペイロードを捕捉し、標的化モジュールに接続する様々なハプテンまたは他の実体に発展することができる。例えば、siRNA送達のために、核酸に結合する二重特異性抗体および抗体誘導体を適用して、siRNAを標的化ビヒクルに接続することができる。
様々なペプチド配列、いわゆる細胞浸透ペプチド(CPP)を用いて、siRNA分子をヒト癌細胞に移入できることが示されている。TATペプチドは、CPP機能を有するペプチドを用いてトランスフェクション能力を得るための公開された一例である。しかしながら、今までに、siRNAとCPP由来モジュールのトランスフェクションと、標的化特異性を容易に組み合わせることはできなかった。DIG-siRNAを様々な標的細胞に標的化する能力が示されたジゴキシゲニン結合二重特異性抗体(実施例13を参照されたい)を適用して、DIG-siRNAおよびsiRNA結合CPPの複合体を抗原発現標的細胞に標的化することができる。これを実現するために、CPPはsiRNA-抗体標的化複合体に取り付けられる必要がある。CPPは、ある場合には、核酸と複合体を自発的に形成することができる。CPP様機能を有するペプチドを核酸結合実体に接続することによって、ペプチドとsiRNAをさらに安定して結合することができる。このような実体は、負に荷電したリン酸バックボーンと正に荷電したアルギニンアミノ基とがイオン相互作用するために二本鎖siRNA分子に結合するオリゴアルギニン配列またはポリアルギニン配列でもよい。CPPをsiRNAに接続するために、さらに構造化されたペプチドドメインまたはタンパク質ドメイン、例えば、一本鎖または二本鎖の核酸および核酸誘導体に結合するドメインも適用することができる。

本発明者らは、<標的-DIG>二重特異性抗体が、DIG標識siRNA-脂質ナノ粒子の送達機構を妨害することなくDIG標識siRNA-脂質ナノ粒子を細胞に誘導できるかどうかという問題を調べた。この問題に対処するために、本発明者らは、
(i)siRNA-脂質ナノ粒子のDIGカップリング脂質成分を合成し、
(ii)これらのDIG標識siRNA-脂質ナノ粒子を用いて処方し、
(iii)DIG標識siRNA-脂質ナノ粒子と<標的-DIG>二重特異性抗体との複合体を作製し、
(iv)<標的-DIG>二重特異性抗体と組み合わせたDIG標識siRNA-脂質ナノ粒子の標的化特性およびmRNAサイレンシング特性をインビトロで評価した。
NHS活性化DIG(Roche Diagnostics GmbH, Mannheim, Germany)をアミン官能化DSPE-PEG2000(Avanti Polar Lipids, Inc., Alabaster, AB)とカップリングすることによって、DSPE-PEG2000-DIGを合成した。
DIG標識siRNA-脂質ナノ粒子は、(7.1mol%、57.1mol%、34.4mol%、および1.4mol%)の比の1,2-ジステアロイル-3-ホスファチジルコリン(DPPC)(Avanti Polar Lipids, Inc., Alabaster, AB)、2,2-ジリノレイル-4-ジメチルアミノエチル-[1,3]-ジオキソラン(社内で合成)、コレステロール(Sigma-Aldrich Chemie GmbH, Munich, Germany)、およびポリエチレングリコール(PEG) 2000-脂質からなった。総含有率(1.4mol%)のPEG2000-脂質は、Sunbright GM-020CE、1.4%、1.36%、1%、または0.4%のα[3'-(1,2-ジミリストイル-3-プロパノキシ)-カルボキサミド-プロピル]-ω-メトキシ-ポリオキシエチレン(NOF Europe, Grobbendonk, Belgium)、および0%、0.04%、0.4%、または1%のDSPE-PEG2000-DIG(前記を参照されたい)からなった。脂質およびコレステロールをエタノールに溶解した。AHA1-siRNA、DIGとカップリングされたAHA1-siRNAまたはルシフェラーゼ(LUC)-siRNAをクエン酸塩緩衝液、pH4に溶解した。適量のエタノール脂質混合物をsiRNA緩衝水溶液に素速く注入することによって、siRNA-脂質ナノ粒子を形成した。その後に、10kD MWCO Slide-A-Lyzer(登録商標)カセット(Thermo Scientific, Rockford. IL)を用いて、siRNA-脂質ナノ粒子をリン酸緩衝食塩水(PBS)に対して透析した。siRNA-脂質ナノ粒子調製物のpH値を確認し、siRNA-脂質ナノ粒子のサイズ分布およびゼータ電位を、Zetasizer Nano-ZS(Malvern Instruments LTD, Malvern, UK)における動的光散乱によって求めた。siRNA捕捉の程度を定量するために、変更を加えたRiboGreenアッセイ(Invitrogen GmbH, Darmstadt, Germany)を行った。siRNAの総含有率は、siRNA-脂質複合体をメタノール:クロロホルム(10:1、vol:vol)に溶解した後に分光測定によって求めた。siRNA-脂質ナノ粒子(RLX-107-RLX-115)の平均粒径は119nm〜128nm(zaverage値)であり、多分散指数は0.02〜0.1であった。PBS中でのゼータ電位は-3〜-4mVであった。カプセル化効率は95%〜96%であり、siRNAの最終濃度は0.4mg/ml〜0.5mg/mlであった。
1pmolのAHA1-siRNA、DIGとカップリングされたAha1-siRNA、またはルシフェラーゼ(LUC)-siRNAを含有するDIG標識siRNA-脂質ナノ粒子を、腫瘍関連LewisYオリゴ糖およびDIGに対する3.24pmolの二重特異性抗体<LeY-DIG>、もしくはシアル酸結合膜貫通タンパク質CD22およびDIGに対する3.24pmolの二重特異性抗体<CD22-DIG>、またはダルベッコリン酸緩衝食塩水(D-PBS)(Invitrogen, Carlsbad, CA)と、最終体積200μl のOpti-MEM(登録商標)(Invitrogen GmbH, Darmstadt, Germany)中で室温で30分間インキュベートした。
MCF-7細胞を80μlの培地(RPMI1640 Medium、10%ウシ胎児血清、100U/mlペニシリン/ストレプトマイシン; Biochrom AG, Berlin, Germany)に溶解して、96ウェルプレートに15,000個の細胞/ウェルの密度で播種し、細胞インキュベーター(37℃、95%H2O、5%CO2)に入れて一晩インキュベートした。DIG標識siRNA-脂質ナノ粒子を<標的-DIG>二重特異性抗体とプレインキュベーションした後に(前記を参照されたい)、20μlの混合物をMCF-7細胞に添加して、1nM siRNAを含有し、抗体を含有しない最終体積100μl(ABなし)、または1nM siRNAおよび3.24nMの<LeY-DIG>(LeY)もしくは<CD22-DIG>(CD22)抗体を含有する最終体積100μlにした。細胞を細胞インキュベーターに入れて12時間インキュベートした。対照として、細胞を、同じ抗体濃度であるが、siRNAまたはsiRNA脂質ナノ粒子なしでインキュベートもした(ABのみ)。さらなる対照として、細胞を、製造業者の説明書に従って、市販のトランスフェクション試薬Dharmafect2(Dh2)(Dharmacon Inc., Lafayette, CO)と複合体を形成した50nMのAHA1-siRNAまたはルシフェラーゼ-siRNAによってトランスフェクトした。インキュベーション後に細胞を溶解し、市販のbDNA-定量システム(Affymetrix Inc., Fremont, CA)を用いてAHA1-mRNA濃度およびGAPDH-mRNA濃度を定量した。AHA1-mRNAシグナルをGAPDH-mRNAシグナルに対して基準化し、Dh2を用いたルシフェラーゼ-siRNAトランスフェクション後(100%になるように設定した)と比較して、AHA1/GAPDH-mRNA比を報告した。
Claims (21)
- ハプテンおよび標的タンパク質に対して特異的な二重特異性抗体であって、ハプテンに結合する第1の抗原結合部位および標的タンパク質に結合する第2の抗原結合部位を含み、ハプテンは核酸と結合体化される、二重特異性抗体。
- ハプテンがビオチンおよびポリエチレングリコール(PEG)からなる群より選択される、請求項1記載の二重特異性抗体。
- ハプテンがジゴキシゲニンである、請求項2記載の二重特異性抗体。
- a)2つの完全長抗体重鎖および2つの完全長抗体軽鎖からなる単一特異性二価抗体であって、それぞれの鎖は可変ドメインを1つしか含まない、単一特異性二価抗体、
b)2つのペプチドリンカー、
c)2つの単一特異性一価単鎖抗体であって、それぞれが、抗体重鎖可変ドメイン、抗体軽鎖可変ドメイン、および該抗体重鎖可変ドメインと該抗体軽鎖可変ドメインとの間の単鎖リンカーからなる、2つの単一特異性一価単鎖抗体
を含む、請求項1〜3のいずれか一項記載の二重特異性抗体。 - DIGに特異的に結合する抗原結合部位が、SEQ ID NO.3、変異S49A、I57A、およびA60Pを有するSEQ ID NO 3、またはSEQ.ID NO.2より選択される重鎖可変ドメイン、ならびにSEQ ID NO 5またはSEQ.ID NO.1より選択される軽鎖可変ドメインを含む、請求項3または4記載の二重特異性抗体。
- DIGに特異的に結合する抗原結合部位が、重鎖Fd断片(VHおよびCH1)としてSEQ.ID NO.4またはSEQ.ID NO.36、ならびにL鎖としてSEQ.ID NO.6またはSEQ.ID NO.37を含む、請求項3〜5記載の二重特異性抗体。
- 抗体がヒト化されている、請求項1〜6のいずれか一項記載の二重特異性抗体。
