Abstract
【解決手段】ムクロジ科植物の抽出物を含有することを特徴とするウレアーゼ活性阻害剤を提供する。本発明のウレアーゼ活性阻害剤の原料である“レイシ”は、ムクロジ科の常緑高木の果樹で、学名をLitchi chinensis、和名をレイシ(茘枝)あるいはライチといい、中国南部原産で熱帯・亜熱帯地方において栽培されている。また、“ランブータン”は、レイシと同じムクロジ科で、学名をNephelium lappaceumといい、東南アジア原産で中型から大型の熱帯の果樹である。本発明に用いるレイシ抽出物およびランブータン抽出物は市販品を利用することができる。本発明に係るウレアーゼ活性阻害剤の形態は特に問わず、錠菓、キャンディー、顆粒剤、飲料などの通常の医薬品、医薬部外品、飲食品の形態を採用できる。

1.レイシ種子の50%エタノール抽出物 1.0
2.砂糖 79.8
3.グルコース 19.0
4.ショ糖脂肪酸エステル 0.2
<製法>
成分1〜3に70%(v/v)エタノールを適量加えて練和し、押出し造粒した後に乾燥して顆粒を得る。成分4を加えて打錠成形し、1gの錠菓を得る。当該錠菓を1日3錠摂取することで、レイシ種子の50%エタノール抽出物を30mg/日摂取できる。
配合量(g)
1.ランブータン果皮の熱水抽出物 3.0
2.砂糖 77.8
3.グルコース 19.0
4.ショ糖脂肪酸エステル 0.2
<製法>
成分1〜3に70%(v/v)エタノールを適量加えて練和し、押出し造粒した後に乾燥して顆粒を得る。成分4を加えて打錠成形し、1gの錠菓を得る。当該錠菓を1日3錠摂取することで、ランブータン果皮の熱水抽出物を90mg/日摂取できる。

1.レイシ種子の100%エタノール抽出物 2.0
2.マルチトール 70.0
3.デンプン糖化物 28.0
<製法>
成分1〜3を120〜170℃で加熱溶解し、金型にて固化させ、3gの飴を得る。当該飴を1日3個摂取することで、レイシ種子の100%エタノール抽出物を180mg/日摂取できる。
配合量(g)
1.ランブータン果皮の50%エタノール抽出物 1.0
2.マルチトール 70.0
3.デンプン糖化物 29.0
<製法>
成分1〜3を120〜170℃で加熱溶解し、金型にて固化させ、3gの飴を得る。当該飴を1日3個摂取することで、ランブータン果皮の50%エタノール抽出物を90mg/日摂取できる。

1.レイシ種子の熱水抽出物 3.0
2.乳糖 82.0
3.セルロース 15.0
<製法>
成分1〜3に70%(v/v)エタノールを適量加えて練和して押出し造粒し、乾燥して顆粒剤を得る。当該顆粒剤は、1回1gずつ1日3回摂取することで、レイシ種子の熱水抽出物を90mg/日摂取できる。

1.ランブータン果皮の100%エタノール抽出物 1.0

3.セルロース 15.0
<製法>
成分1〜3に70%(v/v)エタノールを適量加えて練和して押出し造粒し、乾燥して顆粒剤を得る。当該顆粒剤は、1回1gずつ1日3回摂取することで、レイシ種子の熱水抽出物を30mg/日摂取できる。

3.クエン酸 7.0
4.香料 適量

<製法>
成分5に成分1〜4を加え、撹拌溶解して濾過し、加熱殺菌して1000mLとし、これを50mLガラス瓶に充填する。当該飲料は、1日1本摂取することでレイシ種子の50%エタノール抽出物を50mg/日摂取できる。
配合量(g)
1.ランブータン果皮の50%エタノール抽出物 1.0
2.ショ糖 60.0
3.クエン酸 7.0
4.香料 適量
5.精製水で全量を1000mLとする。
<製法>
成分5に成分1〜4を加え、撹拌溶解して濾過し、加熱殺菌して1000mLとし、これを50mLガラス瓶に充填する。当該飲料は、1日1本摂取することでランブータン果皮の50%エタノール抽出物を50mg/日摂取できる。
実施例1〜6で得られたレイシ種子抽出物およびランブータン果皮抽出物のウレアーゼ活性阻害効果を、ナタ豆由来ウレアーゼを用いて試験した。即ち、エッペンチューブに100mMのリン酸緩衝液に溶解した諸種の濃度の試料溶液20μL、100mMのリン酸緩衝液に溶解したナタ豆由来ウレアーゼ(シグマ製)溶液(0.8U/mL)20μLを添加し、37℃で15分間静置反応させた。次いで、リン酸緩衝液に溶解した60mMの尿素溶液40μLを添加し、37℃で15分間静置反応させた後に、1N硫酸を20μL添加することにより反応を停止させた。得られた反応液に、溶液A(フェノール5.0g及びニトロプルシドナトリウム25mgを蒸留水500mLに溶解させたもの)500μLと溶液B(300mLの蒸留水にリン酸水素二ナトリウム2.2g及び水酸化ナトリウム2.5gを溶解し、有効塩素10%以上の次亜塩素酸ナトリウム3.0mLを添加し、蒸留水で500mLとしたもの)500μLを添加し、65℃で20分間反応させた。反応後、630nmにおける吸光度を測定した。また、対照として、試料非添加コントロールについても同様に測定した。なお、阻害活性(%)は以下の式にて算出した。
阻害活性(%)=(対照群の吸光度−試料添加群の吸光度)/対照群の吸光度×100
表1に示すように、レイシ種子抽出物およびランブータン果皮抽出物はウレアーゼ活性阻害効果を示した。
Claims (5)
- ムクロジ科植物の抽出物を含有することを特徴とするウレアーゼ活性阻害剤。
- レイシ抽出物および/またはランブータン抽出物を含有することを特徴とするウレアーゼ活性阻害剤。
- 請求項1または請求項2記載のウレアーゼ活性阻害剤を含有することを特徴とする医薬品。
- 胃炎の予防剤または治療剤であることを特徴とする請求項3記載の医薬品。
- 胃潰瘍または十二指腸潰瘍の予防剤または治療剤であることを特徴とする請求項3記載の医薬品。
