Abstract
【解決手段】核酸を有機ポリカチオンに接触させ濃縮核酸を製造し、前記濃縮核酸を両媒性カチオン性脂質を含む脂質と組み合わせて、脂質:核酸複合体を形成する。さらに、疎水性側鎖に結合させた親水性ポリマーを添加することによって、安定化させる。前記複合体は、また、親水性ポリマーに結合したFab'断片のような標的部分を取り込むむことによって、特定細胞に対して特異的とする。さらに、親水性ポリマードメインおよび微粒子を、安定結合可能な疎水性ドメインを有するリンカー分子に結合させた結合蛋白質、または親水性ドメインと微粒子との安定結合を可能とする脂質部分を含有するように工学的に処理した蛋白質類を有する脂質性微粒子類。

Description
上記で説明した通り、本発明は、脂質:核酸複合体類の貯蔵寿命(保存寿命)を延長させる方法類を提供する。好適な態様において、前記複合体類は、核酸とリポゾームの併用によって形成される。しかし、前記脂質類はリポゾームとして供される必要はないことがわかっている。また、複合体化後、前記脂質:核酸複合体はもはや真の小胞として存在せず、したがって、一般的にはリポゾームとしてみなされない。脂質:核酸複合体類の調製は、当業者に公知である(たとえば、Crystal、Science 270:404−410(1995);Blaeseら、Cancer Gene Ther.,2:291−297(1995);Behrら、Bioconjugate Chem.5:382−389(1994);Remyら、Bioconjugate Chem.5:647−654(1994);およびGaoら、Gene Therapy 2:710−722(1995)を参照)。本発明の安定化された脂質:核酸複合体類の種々の成分類および構造を下記で詳細に説明する。
上記でも示唆したように、本発明の方法類は、カチオン性脂質を核酸と複合体にすることを含む。用語"カチオン性脂質"とは、生理的pHで正味の正の荷電を有する脂質種でありその数はいずれであってもよい。このような脂質類として、DODAC、DOTMA、DDAB、DOTAP、DC−CholおよびDMRIEなどが含まれるが、それらに限定されない。さらに、いくつかの市販されているカチオン性脂質類調製物類も利用でき、本発明で使用できる。これらには、たとえば、LIPOFECTIN 登録商標(GIBCO/BRL、Grand Island、New York、USAから市販されているDOTMAおよびDOPEを含むカチオン性リポゾーム類)、LIPOFECTAMINE 登録商標(GIBCO/BRLから市販されているDOSPAおよびDOPEを含むカチオン性リポゾーム類)、およびTRANSFECTAM 登録商標(Promega Corp.,Madison、Wisconsin、USAから市販されているDOGSを含むカチオン性リポゾーム類)が挙げられる。
前記脂質:核酸複合体類は、典型的には組換え技術を用いて構築された発現カセットである核酸を含有する。組換え核酸は、最初に問題の核酸を単離することで調製される。この単離された核酸を、次に、この遺伝子の発現に適したカセットまたはベクターに連結する。組換え核酸調製方法類は、当業者に公知である(Sambrookら、Molecular Cloning:A Laboratory Manual(第2版、1989)参照)。

小型のポリカチオン性分子類は、静電的荷電−荷電相互作用によって核酸類を濃縮することが公知となっている(Plumら、Biopolymers 30:631−643(1990))。核酸のポリアミン類による前処理は、したがって、カチオン性リポゾーム類を複合体化するための荷電部位の数を減らすことができる。しかし、脂質複合体形成前に核酸を濃縮し、トランスフェクション効率で測定したところ、脂質:核酸複合体類の保存寿命延長として驚くべき結果がもたらされた。このような前処理によって形成された脂質:核酸複合体類は凝集することなく、DNAに対する脂質の比率が低い場合でも安定であった。ポリアミン類、ポリアンモニウム分子類および塩基性ポリアミノ酸類のような有機ポリカチオン類、およびそれらの誘導体類も、脂質複合体形成前に核酸を濃縮するために使用される。好適な態様では、スペルミジンおよびスペルミンのようなポリアミン類を使用し、核酸を濃縮させる(たとえば、実施例1参照)。
最近、リポゾーム中へのPEG−PEの取り込みが立体的安定性をもたらし、血液中での循環時間が長くなるという結果が得られた(Allenら、Biochim.Biophys.Acta.1066:29−36(1991);Papahadjopoulosら、Proc.Natl.Acad.Sci.USA 88:11460−11464 (1991))。本発明において、形成されたばかりの脂質:核酸複合体類中にPEG−PE(例 総脂質の1%)を挿入すると、保存寿命中前記複合体類の凝集を防止する。しかし、PEG−PEの取り込みがインビボにおけるトランスフェクション活性を阻害しなかったことおよびインビトロトランスフェクション活性は阻害されたがこれはPEG−PE末端に結合されたFab'断片を取り込むことで再獲得されたことは、驚くべき発見であった。脂質:核酸複合体中における親水性ポリマー類の存在は、保存後トランスフェクション効率で測定したところ、保存寿命延長を生じる。したがって、前記リポゾームに対してポリエチレングリコール(PEG)改変脂質類のような親水性ポリマーまたはガングリオシドGM1を添加することが望ましい。また、PEGは、脂肪酸類、スフィンゴリピド類、グリコリピド類およびコレステロールのような他の両媒性分子類によって誘導体とすることができる。上記成分類を添加すると、リポゾームへの標的部分の結合時にリポゾーム凝集を防止する。これらの成分類は、また、脂質:核酸複合体類の循環寿命を増大させる手段を提供する。
好適な態様において、本発明の脂質:核酸複合体類は抗体のFab'断片に結合され、それは、Fab'抗体断片の特異的な標的分子(例 特徴的マーカー)を有する標的細胞を前記脂質:核酸複合体が特異的に結合できるようにする標的部分として作用する。前記の高変異領域または特異的細胞表面リガンドと相互作用する別のペプチド由来の小型ペプチド類は、同様に、前記複合体類に結合することができる。一般的用語において、抗体のFab'断片とは、前記の可変領域と抗体の1腕のCH1領域を含む単量体を意味する。このような一つの好適な態様は、実施例2において記載されている。
種々の方法がリポゾーム類調製のために利用可能であり、たとえば、Szokaら、Ann.Rev.Biophys.Bioeng.9:467(1980);米国特許第4,186,183;4,217,344;4,235,871;4,261,975;4,485,054;4,501,728;4,774,085;4,837,028;4,946,787;PCT公報第WO91/17424;Szoka&Papahadjopoulos、Proc.Natl.Acad.Sci.USA 75:4194−4198(1978);Deamer&Bangham、Biochim.Biophys.Acta 443:629−634(1976);Fraleyら、Proc.Natl.Acad.Sci.USA 76:3348−3352(1979);Hopeら、Biochim.Biophys.Acta 812:55−65(1985);Mayerら、Biochim.Biophys.Acta 858:161−168(1986);Williamsら、Proc.Natl.Acad.Sci.USA 85:242−246(1988)、Liposomes、Ch.1、(Ostro編著、1983);およびHopeら、Chem.Phys.Lip.40:89(1986)に記載されている。適切な方法として、たとえば、音波処理、押し出し、高圧/均質化、微細粉末化、界面活性剤透析、小リポゾーム小胞のカルシウム誘発融合、およびエーテル注入方法などが挙げられ、全て当該分野で周知である。ひとつの方法では、不均一な大きさのマルチラメラ小胞を生成する。この方法において、小胞形成脂質類は、適切な有機溶媒中または溶媒系に溶解させ、真空中または不活性気体中で乾燥させ、薄い脂質膜を形成させる。もし所望であれば、この膜を再度三級ブタノールのような適切な溶媒に溶解させ、その後凍結乾燥させ、より均質な脂質混合物を形成させ、これは、より容易に水和された粉末様形態となる。この膜を水性緩衝液で被覆させ、典型的には15−60分にわたり攪拌下に水和させる。生成したマルチラメラ小胞の径分布は、前記脂質類をより強く攪拌することで水和させてまたはデオキシコーレートのような可溶性界面活性剤を添加することで、より小さい径にシフトさせることができる。
安定化脂質:核酸複合体類(例 濃縮核酸および/または親水性ポリマーを有する)が可視可能な大きな凝集物を形成しない傾向があることおよびトランスフェクション効率上昇および保存寿命延長を示すことは、本発明の発見であった。可視可能な大きな凝集物を形成しない脂質:核酸複合体類調製のための核酸/リポゾーム比率は、当業者が決定できる。典型的には、この比率は、固定された量のたとえばプラスミドのような核酸を種々の量のリポゾーム類と混合することによって、決定される(実施例1参照)。一般的に、脂質:核酸複合体類は、核酸(例 プラスミドDNA)を同量のリポゾーム懸濁液中にピペット操作で注入し急激に混合することによって、形成される。通常、5−15nmoleのDDABまたはDOPE(上記に記載)のような脂質を含有するリポゾーム類は、1μgのプラスミドと複合体を形成するが、可視可能な大きい凝集物を形成しない。可視可能な大きな凝集物の検査は、典型的には、顕微鏡の助けをなくして実施される。脂質および核酸量の滴定の終点は、また、インビトロまたはインビボのいずれかで、非安定化対照(下記に説明)と比較してトランスフェクション効率増大をアッセイすることによって、得られる。
本発明の脂質:核酸複合体類を含む薬剤組成物類は、標準的技術にしたがって調製され、かつ、さらに薬剤学的に許容できる担体を含む。一般的に、通常の生理食塩水を、薬剤学的に許容できる担体として使用するであろう。他の適切な単体として、たとえば、水、緩衝水、等張液(例 デキストロース)、0.4%生理食塩水、0.3%グリシンなどが挙げられるが、アルブミン、リポプロテイン、グロブリン等のような安定性増強のための糖蛋白質類も含まれる。これらの組成物類は、従来周知の滅菌手法によって滅菌される。生成した水性溶液類を袋に入れ使用することもできるしまたは無菌条件下でろ過し凍結乾燥することもでき、この凍結乾燥調製物を無菌水性溶液と組み合わせて投与できる。前記組成物類は、酢酸ナトリウム、乳酸ナトリウム、塩化ナトリウム、塩化カリウム、塩化カルシウム等のようなpH調製および緩衝性の物質類、調整剤等のような、生理的条件に近似させるために必要な薬剤学的に許容できる補助剤を含むことができる。さらに、脂質:核酸複合体懸濁液は、保存時のフリーラジカルおよび脂質過酸化性傷害に対して脂質を保護する脂質保護性物質類を含むことができる。アルファトコフェロールおよびフェリオキサミンのような水溶性鉄特異的キレート剤のような脂質親和性フリーラジカルクエンチャー類が適切である。
標的組織中における脂質:核酸複合体を局在化させるためのためのひとつの手段として、投与後の血流中における脂質:核酸複合体寿命の延長がある。血流中脂質:核酸複合体寿命のひとつの尺度は、複合体投与後の選択された時点に求めた血液/網状系比である。典型的には、複合体の内部にまたは前記複合体を含む脂質に結合させてのいずれかで標識(例 蛍光マーカー、電子密度反応剤または放射性マーカー)を含有する脂質:核酸複合体類を試験生物中に注入する。その後の一定の時点において、生物を屠殺し血液中で検出した(例 発光測定によってまたはシンチレーション測定によって)標識の量を、特定組織(例 肝または脾臓)中に局在したそれと比較する。
本発明の脂質:核酸複合体類による標的細胞のトランスフェクションは、同様に、それ自体検出可能であるかまたは検出可能な生成物をコードする核酸を含有する脂質:核酸複合体類を投与することによって、求めることができる。血液試料類(例 組織生検または液体試料)を次に採取し、トランスフェクションされた核酸それ自体を検出するかまたは前記核酸の発現生成物の存在を検出することによって、トランスフェクションをアッセイする。
上記にも述べたように、用語"保存寿命"とは、前記脂質:核酸複合体がその生物活性を失うまで(4℃の緩衝液中のような規定された条件下における)保存可能な期間を称するものとして本文で使用している。本発明で保存寿命決定のためにアッセイする前記の生物活性とは、静脈注射後哺乳類細胞にトランスフェクションする脂質:核酸複合体の能力である。
本発明の脂質:核酸複合体類とともに特異的標的部分を使用して、特異的細胞または組織を標的とすることができる。一の態様において、抗体または抗体部分のような標的部分を親水性ポリマーに付着させ、複合体形成後において脂質:核酸複合体と組み合わせる。したがって、一般的エフェクター脂質:核酸複合体と併用して標的部分を使用すると、前記複合体を特異的細胞類および組織類への運搬のために都合よく調整できる能力が付与される。
本発明は、また、上述の脂質:核酸複合体類を調製するキット類を提供する。このようなキット類は、上記のような容易に入手可能な材料類および試薬類から調製できる。たとえば、下記の材料類のいずれかひとつまたはそれ以上を含むことができる:リポゾーム類、核酸(濃縮または非濃縮)、親水性ポリマー類、Fab'断片類のような標的化部分で誘導体とした親水性ポリマー類、および指示書類。実にさまざまなキット類および成分類を本発明にしたがって調製でき、キットを使用するユーザーおよびユーザーの特定の必要性に応じて調製できる。たとえば、このキットは、上記のように前記複合体が特異的細胞種を標的とするようにいくつかの標的化部分のいずれかひとつを含むことができる。
A.全般
本発明は、また、表面結合蛋白質類を有する脂質性微粒子類を調製することを提供する。上記で背景の項でも述べたように、従来技術は、抗体類のような可溶性蛋白質類は極めて大きいため、それらの可溶化傾向が、脂質性微粒子に安定に結合するために蛋白質に結合したリンカー分子の疎水性領域の傾向を圧倒することを示唆する。この教示は、ゆえに、およそ同一サイズかまたは大きいリンカー分子に結合した小型のペプチド類はリンカー分子の疎水性領域が脂質性微粒子に安定に結合するのを許す一方、蛋白質よりもはるかに小さいリンカー分子に結合した蛋白質類はそうすることができないであろうということであった。
本発明の用途において、蛋白質は最初に(a)疎水性ドメイン、(b)末端で前記の疎水性ドメインに結合した親水性ポリマー鎖、および(c)蛋白質分子上の1個以上の官能基と反応性で前記疎水性ドメインの反対側の末端あるいはその近くで親水性ポリマー鎖に結合した化学基、を含むリンカー分子に結合できる。このようなリンカー分子類は、当該技術で公知である(Allen&Martin、US5,527,528;Shahinian&Sylvius、Biochim.Biophys.Acta.,1239:157−167(1995);Zalipskyら、J.Controlled Release 39:153-161、1996;Kirpotinら、Biochemistry、36:66−75(1997))。
A.材料類および方法類
1.脂質類&他の試薬類
小型のカチオン性リポゾーム類は、下記のようにして5%(w/v)デキストロース溶液中で調製した。クロロホルム中DDABまたは他のカチオン性脂質類は、DOPEまたは/およびコレステロールと所望のモル比で混合し、溶媒をゆっくりとロータリエバポレータで50℃で減圧下で除去した。50℃に前加温した5%デキストロース溶液で乾燥した脂質膜を水和させ、容器をアルゴン中で密封した。水和脂質懸濁液をバスソニケータ(Lab Supplies,Hickville、N.Y.)中で5−10分間、50℃で音波処理した。リポゾーム類の最終濃度は5mMカチオン性脂質であり、リポゾーム類の大きさは、ダイナミック光分散によって測定し、195±65nmであった。音波処理したリポゾーム類を使用するまで、4℃でアルゴン中に保存した。
プラスミドpCMV/IVS−luc+は、下記のようにして構築した。CMVプロモータおよび合成IgEイントロンを、含有する断片をpBGt2.CATからSpeIおよびHindIIIを用いて切りとり、pBSIIKS+中にクローニングした。SV40後期ポリ(A)シグナルを含む改変蛍ルシフェラーゼ(luc+)をコードするcDNAを、pGL3−Basic Vector(Promega)からHindIIIおよびSalIによって切断し、前記切断部の下流のpBS−CMV−IVSクローンに入れた。Qiagen社(Chatsworth、CA)によって採択され考案されたアルカリ融解操作法を用いて、プラスミド類を精製した。プラスミド純度は、260nm対280nmの吸光度比によって測定し、1−2mg/mlの濃度で10mMTris−Clおよび1mM EDTA含有pH8.0の緩衝液中に保存した。
トランスフェクション実験前に、一定量のプラスミドを種々の量のリポゾーム類に対して混合することによって、大きい凝集物ではない複合体類を形成するための最適DNA/リポゾーム比を求めた。一般に、トランスフェクション複合体類は、プラスミドを等量のリポゾーム懸濁液中にピペット操作でいれその後急激に混合することによって、形成させた。通常、8−12nmのDDABを含有する、リポゾーム類は、可視可能な大凝集物を形成することなく、1μgのプラスミドと複合体を形成するであろう。このような複合体類は、過剰の正の荷電を有しているが、しかし、まだ、4℃における保存中に時間とともに凝集し4日後にはトランスフェクション活性を消失する傾向がある。インビトロ実験のためには非常に希釈された複合体類を必要とするが、1μgDNA当たり5nmoleのDDABを含むカチオン性脂質:プラスミドDNA複合体類("CLDC")を使用した。脂質:プラスミドDNA複合体類が大凝集物を形成しトランスフェクション活性を時間とともに喪失しないようにするため、2つの手法を用いた。(1)PEG−PEのような少量の親水性ポリマー(約1%モル比)を調製後数分以内の脂質:プラスミドDNA複合体類に取り込ませること;および/または(2)リポゾーム類と混合する前に、プラスミドをポリアミン(例 DNA1μg当たり約0.05乃至5.0nmoleのスペルミジン)で濃縮すること。前記ポリアミン類の最適量は、大凝集物形成前にDNAに対してポリアミン類を滴定することによって、決定した。これらの複合体類の大きさは、ダイナミック光分散によって範囲410±150nmであると推定された。
精製されたルシフェラーゼは、ルミノメータを校正し相対的ルシフェラーゼ比活性の対照スタンダードを構築するための標準として、Boehringer Mannheimから購入した。組織抽出物中におけるレポータ遺伝子発現は、ルミノメータで測定した相対的光単位を標準曲線に従って重量単位に変換することによって、ナノグラム量で示した。細胞または組織中で発現したルシフェラーゼは、化学的細胞溶解によって抽出した。効果的な溶解緩衝液は、pH7.8の0.1Mりん酸カリウム緩衝液、1%トリトンX−100、1mMのDTTおよび2mMのEDTAで構成されていた。
1."ヘルパー"脂質の最適化
カチオン性リポゾーム類のインビトロ遺伝子運搬のための使用は、Felgnerらがかれらの研究を公表して以降、広く普及した(Felgnerら、Proc.Natl.Acad.Sci.USA 84:7413−17(1987))。後日、DOPEがインビトロ遺伝子トランスフェクションのためのこれまでで最も効率的な"ヘルパー"脂質であることが確立され(Felgnerら、J.Biol.Chem.269:2550−2561(1994))、この結果は、数箇所の研究室で確認された(Farhoodら、Gene Therapy for Neoplastic Diseases、pp.23−55(Huber&Lazo編著、1994);Zhouら、Biochim.Biophys.Acta 1189:195−203(1994))。インビトロ研究に基づいて、いったん正に荷電された脂質:プラスミドDNA複合体類が細胞膜に結合すると、DOPEは、膜融合によって細胞質運搬を促進することができると示唆されている(Zhouら、Biochim.BioPhys.Acta.1189:195−203(1994))。FriendらはDOTMA/DOPE脂質:プラスミドDNA複合体類が直接形質膜に融合することの形態的証拠を全く得ているというものではないものの、彼らは、融合事象の可能性を排除していない(Friendら、Biochim.BioPhys.Acta 1278:41−50(1996))。彼らは、前記複合体類がエンドサイトーシスの作用を受けカチオン性脂質類がエンドソーム性/ライソゾーム性の膜を破壊し、その後DNA複合体類が細胞質に逃げ込み最終的に核内に入りこむのを促進すると示唆している。
脂質:プラスミドDNA複合体類の構造的安定性およびトランスフェクション活性の関係は、これまで公表された報告ではあまり詳細に説明されていない。脂質に対するDNAの比率を変更することによって正味で負の荷電から正の荷電にすることで、脂質:プラスミドDNA複合体類の大きい凝集物を避けるため、スクリーニング操作を確立した。種々のDNA/脂質比率にある各粒子カチオン性脂質の脂質:プラスミドDNA複合体類を調製し、結果として生成した安定処方類および準安定処方類をインビボトランスフェクションのために使用した。DNA1μg当たりカチオン性脂質8−12nmoleを含む複合体類は、最大のインビボトランスフェクション活性を有することがわかった。しかし、これらの複合体類のトランスフェクション活性は、時間とともに低下した。脂質:プラスミドDNA複合体類形成操作を改変することなく、数日以内に可視可能な凝集物があり、4℃において1ヶ月間保存した後、トランスフェクション活性は1000倍を超える程低下し、ほとんどバックグランドレベルまで低下した(図4)。したがって、保存中高いインビボトランスフェクション活性を保持できる、安定化脂質:プラスミドDNA複合体類の処方が保証される。
新鮮形成脂質:プラスミドDNA複合体類中にPEG−PE(総脂質の1%)を挿入することは、保存時にこの複合体類が凝集することを防止できるだけではなく、このPEG−PE含有複合体類は、インビボにおいて妥当な高いトランスフェクション活性を示し、PEG−PEを含まない複合体類に比較してわずかに低い活性であった(図4)。前記複合体類中へPEG−PEを取り込むことは、PEG−PEの百分率増をもたらし、トランスフェクション活性の阻害が用量に関連して生じることが解る(結果は示していない)。予測外ではあるが、PEG−PE含有複合体類を4℃において保存すると、ゆっくりではあるが当初の活性を回復することは、図4に示した通りである。PEG−PEによるトランスフェクションに及ぼす阻害効果の機序面についておよび低温保存後の活性回復については、現時点では公知でない。
PEG−PEの脂質:核酸複合体類の保存寿命延長における役割に加えて、ポリアミンで濃縮された核酸は、また、前記複合体類の保存寿命に対して同様の予測外の増大をもたらした。濃縮DNAで形成された脂質:プラスミドDNA複合体類は凝集することなく、DNAに対する低比率の脂質において安定であった。図4は、このような調製物のインビボトランスフェクション活性レベルとその保存時の動向を示している。再び、ポリアミンで前処理せず複合体形成後すぐに使用した試料の活性と比較した際、トランスフェクション活性の予測外の増大は、熟成させたポリアミン処理脂質:プラスミドDNA複合体類で見られた。脂質−界面活性剤ミセル中においてプラスミドを脂質と複合体化させることによって、安定なカチオン性脂質/DNA複合体類を得るための異なる手法は、最近公表された(Hoflandら、Proc.Natl.Acad.Sci.USA 93:7305−7309(1996))。しかし、トランスフェクション効率の30%しか、インビトロ15%血清中でこのような複合体類によって保持されず、インビボ結果については、全く報告されなかった。
最後に、凍結乾燥による脂質:プラスミドDNA複合体類の安定化のため、条件を確立した。5%(w/v)デキストラン水溶液中における音波処理で懸濁させたDDAB/Cholで構成したリポゾーム類を、方法類で記載したようにDNAと1:10比で(DDAB 1nmoleに対するDNAのμg)で混合すると、活性を喪失することなく凍結乾燥できた。脂質:プラスミドDNA複合体類を形成したデキストランの最終濃度は、8%(w/v)であった。凍結乾燥した調製物は、精製水を添加することによって再構成し、静脈注射後のマウス肺中におけるそれらのトランスフェクション活性をルシフェラーゼレポータ遺伝子発現によって測定した。再構成した調製物の凍結及び融解は、前記活性に影響を及ぼさなかった(通常、組織蛋白質1mg当たり1−2ngのルシフェラーゼ蛋白質)。
A.Fab'断片の調製
濃縮DNAは、3種の異なる脂質組成物類と実施例1に記載の方法を用いて複合体としたが、下記の改変を行った。第1の複合体は、DDAB/DOPE(1/1)により作成され、上記のようにDNAのみで複合体となったカチオン性リポゾーム類を生成した。第2の複合体は、PEGの最終位置でマレイミドで誘導体とした1%PEG−PEを有するDDAB/DOPE(1/1)により作成され、DNAとの複合体化後添加された立体安定化成分を有するCLDCを生成した。第3の複合体は、PEGの最終位置にマレイミド残基に対する遊離のチオール基を介して付着したヒト化抗HER−2抗体のFab'断片で誘導体とした1%PEG−PEを有するDDAB/DOPE(1/1)により作成された。これによって、DNAと複合体形成後添加された立体安定化成分に付着した標的リガンドを有するCLDCを産生した。
上記実施例1で記載したように細胞をトランスフェクションしたが、脂質:プラスミドDNA複合体の保存はしなかった。2種の細胞系統をこの実施例で用いた。第1の細胞系統はMCF−7であり、この細胞系統の細胞は、HER−2レセプターを過剰発現しない。これらの細胞を、10%ウシ胎児血清を有するDME H−21および5%CO2中で培養した。第2の細胞系統はSK−BR3細胞類であり、それらの細胞は、HER−2レセプターを過剰発現し、5%CO2中のウシ胎児血清を添加したMcCoy's5A培地で培養した。両者の場合、細胞(約5×104細胞/ウェル)は、上記のように脂質と複合体としたプラスミドDNA12μgと37℃で4時間、トランスフェクションさせかつインキュベーションした。その後上清をアスピレーションし、新鮮培地を添加し、細胞を37℃で24時間インキュベーションした。細胞をその後PBS(Ca/MG非含有)で洗浄し採取し、上記のようなルシフェラーゼアッセイのために溶解緩衝液中に懸濁させた。

前記脂質性微粒子と前記誘導体化した親水性ポリマーとが、一緒にインキュベーションしたときに安定に結合する、前記キット。 - 前記標的部位が、抗体、抗体の断片、抗体のFab'断片、一本鎖Fv抗体断片、レセプター蛋白質、リンホカイン、サイトカイン、酵素、ホルモン、成長因子又は核酸結合蛋白質である、請求項30に記載のキット。
- 前記標的部位が細胞表面マーカーと特異的に結合する、請求項30に記載のキット。
- 前記親水性ポリマーがPEG、PEG−PE、PEG−DSPE、PEG連結界面活性剤又は合成ポリマーである、請求項30に記載のキット。
- 蛋白質に結合した脂質微粒子を調製するキットであって、前記キットは、
(1)前記脂質性微粒子を含む容器、
(2)前記蛋白質で誘導体化した親水性ポリマーを含む容器、を含み、
前記脂質性微粒子と前記蛋白質で誘導体化した親水性ポリマーとが、一緒にインキュベーションしたときに安定に結合する、前記キット。 - 前記親水性ポリマーがPEG、PEG−PE、PEG−DSPE、PEG連結界面活性剤又は合成ポリマーである、請求項34に記載のキット。
- 蛋白質に結合した脂質微粒子を調製するキットであって、前記キットは、
(1)前記脂質性微粒子を含む容器、
(2)親水性ポリマー鎖及び疎水性アンカーを有する蛋白質を含む容器、を含み、
前記脂質性微粒子と、親水性ポリマー鎖及び疎水性アンカーを有する前記蛋白質とが、一緒にインキュベーションしたときに安定に結合する、前記キット。 - 前記蛋白質が抗体、抗体の断片、抗体のFab'断片、一本鎖Fv抗体断片、レセプター蛋白質、リンホカイン、サイトカイン、酵素、ホルモン、成長因子又は核酸結合蛋白質である、請求項36に記載のキット。
