(実施例1:ベクター p89VP1の構築)
ウイルスストック試料は、QIAampウイルスRNAキット(Quiagen、ドイツ)による細胞培養上清からのRNA抽出の後に、最終濃度が0.01U/μlになるまで1μlのRNase阻害剤(Boehringer株式会社 Gmbh)を添加することによって、RT−PCR用に調製された。
上述した方法は、C末端のPhl p 1アレルゲン断片、すなわちペプチド 5(CVRYTTEGGTKTEAEDVIPEGWKADTAYESK;M. Focke et al. FASEB J (2001) 15:2042-4)に対して例証された。ペプチド 5のDNA配列は、鋳型としてPhl p 1のcDNA(GenBank:X78813)を用いたPCR(プライマー1Phl p 1 forward 5’−CGCGCTTAAGATGGTCCGCTACACCACCGAGGGC−3’(配列番号7):Phl p 1 reverse 5’−CGCGCTTAAGCTTGGACTCGTAGGCGGTGTCGGC−3’(配列番号8))によって増幅された。PCR産物は、p89VP1ベクターのAflII制限酵素認識部位に挿入され、かつ結果構築物は、ベクターp89P5および遺伝子産物r89P5と呼ばれる。
89VP1またはr89P5(組み換え89VP1−ペプチド 5融合タンパク質)の発現を達成するために、プラスミドは、E.coli BL21(DE3)に形質転換された。形質転換したE.coli細胞は、100mg/lのアンピシリンを含有する250mlのLB培地において、37℃において、0.4の光学濃度(600nm)になるまで培養され、かつタンパク質発現が、イソプロピル−ベータ−D−チオガラクトシダーゼ(IPTG)を1mMの最終濃度になるまで加えることによって、誘導された。E.coli細胞は、4時間後に、3500rpmにおいて、4℃における10分間の遠心分離によって回収された。精製は、Quiagen Ni−NTAおよびカラムを用いて、Qiagenの手順を用いて実施された。細胞ペレットは、5mlの6Mのグアジニン塩酸塩において1時間の変性条件下において再懸濁された。遠心分離(20分間、10000×g)の後に、上清は、1mlのNi−NTAを用いて追加の時間においてインキュベーションされた。それから、懸濁液は、カラムに装填され、5mlの洗浄緩衝液(8Mの尿素、pH6.3)を用いて2回洗浄され、かつそれから4mlの溶出緩衝液(8Mの尿素、pH3.5)を用いて溶出された。復元は、尿素のモル濃度を減少させることを伴う透析の後に、達成された。
5μgの14VP1(ヒトライノウイルス14株のVP1タンパク質)および対照(Bet v 1、Phl p 5、BSA)は、ニトロセルロース膜条辺の上にドット化された。これらの条辺は、ウサギ抗14VP1抗血清(レーン1−6)および免疫前の血清(レーン8−12)の希釈物にさらされた。結合したウサギIgG抗体は、1:1000の125I標識ロバ抗ウサギIgGを用いて検出され、かつオートラジオグラフィーによって可視化された(図3)。
89VP1の免疫原性、およびペプチド 5に対する担体として働く能力を決定するために、6週齢のメスのblab/cマウス(Charles River)の群は、以下の抗原:KLH、KLHマレイミド結合ペプチド 5(KLHP5)およびKLH EDC結合ペプチド 5(KLHP5edc)を用いて免疫された。化学結合は、イムジェクト(Imject)マレイミドmcKLHキットおよびイムジェクト免疫原EDCキット(Pierce)を用いて作製された。マレイミド基は、SH基と反応し、かつEDCは、カルボキシル基およびアミノ基と反応して、安定な結合を形成する。4匹のマウスの群は、89VP1、r89P5および89P5edcを用いて免疫され、かつ2匹のマウスは、ペプチド 5のみを用いて免疫された(それぞれ5μg、および水酸化アルミニウムを用いて1:2に混合された)。マウスは、3週間の間隔を置いて皮下に免疫され、かつ尾の血管から採血された。89VP1特異的IgG1抗体レベルは、ELISAによって決定された。
r89P5を用いた免疫化が、完全なPhl p 1と反応するIgG抗体を誘導するか否かを評価するために、同じ方法および同じマウス血清が実施例5に記載されるように使用された。ELISAプレートは、5μg/mlのrPhl p 1を用いて被われ、かつIgG1抗体の力価が決定された(図5)。
マウスの免疫化およびELISA分析は、項5に記載されているように実施された。完全なオオアワガエリ花粉抽出物は、ELISAプレート上にまぶされ、かつIgG1抗体の力価が決定された(図6)。
アレルギー性患者のIgE抗体のrPhl p 1に対する結合を阻害するペプチド由来ウサギIgの能力を決定するために、ELISAプレートは、1μg/mlのrPhl p 1を用いて被われ、洗浄され、かつブロッキングされた。プレートは、1:100に希釈したウサギ抗ペプチド(89P5、KLHP5)、ウサギ抗rPhl p 1とともに、かつ対照を目的として対応する免疫前の血清とともに、前インキュベーションされた。洗浄の後に、プレートは、Phl p 1アレルギー性患者からのヒト血清(1:3に希釈)とともにインキュベーションされ、かつ結合したIgEは、マウス抗ヒトIgE(Pharmingen、1:1000)を用いて、かつそれからPOX結合ヒツジ抗マウスIgG(Amaersham Bioscience)を用いて検出された。抗ペプチド血清をともなう前インキュベーションによって達成されたIgE結合の阻害%は、以下のように(100ODi/ODp×100)算出された。
3匹のマウスの群は、KLH、KLHP5およびKLHPedcを用いて免疫された。4匹のマウスの群は、89VP1、r89P5および89P5edcを用いて4回免疫され、かつ2匹のマウスは、ペプチド 5のみを用いて免疫された(それぞれ5μg)。脾臓細胞は、最後の免疫の後における10日目に採られ、かつ単一の培養細胞は、ペプチド 5(P5)、89VP1、KLH、Con Aならびに陽性対照および陰性対照として培地をそれぞれ用いて、96ウェルプレートにおいて3つ1組にして、刺激された。4日後に、0.5μCiの放射性の[3H]チミジンが、各ウェルに加えられた。それから、細胞は、15時間後に単一フィルタプレート上において、パッカードセルハーベスター(Packard Cell Harvester)を用いて回収された。細胞関連放射性は、ベータ計測器を用いて測定された。刺激は、予想された場合において誘導され、かつy軸に示されている。刺激に使用された抗原は、x軸に示されている。各箱は、マウスの免疫に使用された抗原のデータを表している(図8)。2以上のすべての値は、陽性として数える。KLHおよびKLHP5の免疫マウスは、KLHを用いて刺激された場合にのみ陽性であり、かつペプチド 5マウスは、完全に陰性である。また、KLHP5dec群は、ELISAの結果と対応して陰性である。r89P5、89P5decおよび89VP1の免疫マウスから得られる細胞は、89VP1を用いた刺激の後にのみ増殖した。未処置の対照マウスは、すべての場合において増殖を示さない。これらの結果は、T細胞エピトープが、担体89VP1によって提供され、かつペプチド 5によって提供されないことを示している。
無作為に選ばれた個人の5つのヒト血清は、秋に採られ、かつ5つは、冬に採られた。14VP1、89VP1およびrPhl p 1に対するIgG1、IgG2、IgG4およびIgAの抗体レベルは、実施例5に記載されたようにELISAによって決定された。ヒトIgG1、IgG2、IgG4およびIgA4は、1:2000のPOX結合ヒツジ抗マウスIgG(Amersham Bioscience)を用いて、(BD Pharmingen)1:1000を検出された。秋および冬に採られた血清の抗14VP1および89VP1のIgG1の高い力価を検出することができた(図9)。抗rPhl p 1のIgG1抗体の力価は、非常に低かった。IgG2、IgG4およびIgA抗体を、すべての場合において、非常に低いレベルにおいて検出することができた。異なるHRV株のVP1タンパク質は、近縁関係にあり、かつその交差反応性が他の研究において示されている。
5人のPhl p 1アレルギー性患者の血清が採られ、かつELISA実験が、実施例5に記載のように実施された。ELISAプレートは、rPhl p 1および89VP1を用いて被われ、かつ特異的なIgM、IgG1、IgG2、IgG3、IgG4、IgAおよびIgE抗体の力価が、決定された(図10)。抗rPhl p 1のIgG1抗体よりも抗89VP1 IgG1の方が、検出され得た。
14VP1を注入したウサギの血清の、完全なウイルスに対する抗体結合は、完全なHRV14ウイルス(レーン2および5)および対照として5μgの精製14VP1(レーン3および6)を用いて確認された。ウイルス抽出物は、12%のSDS−PAGEによって分離され、かつニトロセルロース膜上にブロットされた。1:500のウサギ抗14VP1抗血清(レーン1−3)および1:500の免疫前の血清(レーン4−6)がHRV14および14VP1に対する結合に関して試験された。結合したIgGは、125I標識されたロバ抗ウサギ抗体を用いて検出され、かつオートラジオグラフィーによって可視化された(図11)。
HRV14の組織培養投与量50(TDC50)が決定された。従って、1%FSCおよび40mMのMgCl2のMEM−Eagleにおける、1:102−1:108のウイルス希釈が実施され、かつHeLa Ohio細胞とともに、24ウェルプレート、34℃、加湿した5%CO2雰囲気において、3日間培養された。また、ウイルスなしの対照が播かれた。
ペプチドは、アプライドバイオシステム(Applied Biosystem)ペプチド合成機モデル433Aにおいて、HBTU活性化(0.1mmolの小規模サイクル)を伴うFmocストラテジーを用いて、Focke et al. Faseb J (2001) 15:2042に記載のように合成された。Phl p 1アレルゲンの詳細な分析の後に、溶媒にさらされるアミノ酸が豊富な、長さが31アミノ酸(P1:3026ダルトン)から38アミノ酸(P5:3853ダルトン)の範囲にある6つのPhl p 5由来ペプチドが、調製された(表3)。
15.1. IgE反応性の欠如
6つのPhl p 5由来ペプチドのIgE反応性を分析するために、単離したPhl p 5由来ペプチドおよびKLH結合Phl p 5由来ペプチドは、29人の草本花粉アレルギー性患者からの血清を用いたELISAによって、IgE結合能に関して完全なrPhl p 5と比較された(表4)。
CD203cの上方制御は、アレルゲン誘導好塩基球の活性および脱顆粒に対する代用マーカーとして説明されている(Hauswirth et al., J Allergy Clin Immunol 2002; 110:102)。従って、草本花粉アレルギー性患者の好塩基球に対するCD203cを測定することによる、完全なrPhl p 5アレルゲンおよびペプチドの等モル濃度の混合物のアレルギー誘発性活性は、比較された。
草本花粉アレルギー性患者からの好塩基球におけるCD203cの決定は、アレルギー誘発活性がPhl p 5由来ペプチドを用いて観察され得ないことを示している。
16.1. 組み換えアレルゲンおよびアレルゲン抽出物
精製組み換えPhl p 5は、Vrtala et al. J of Immunol (1993) 151:4773-4781に記載のように、E.coliにおいて発現された。
HPLC精製ペプチドは、製造者の助言に従ってKLHに対して結合され、かつコンジュゲーションキット(Conjucation Kit)(Sigma、アメリカ)を用いて精製された。
KLH結合ペプチドを用いて免疫化した後に誘導される抗体が、rPhl p 5、天然のPhl p 5および他の草本花粉種からのPhl p 5関連草本花粉アレルゲンを認識したか否かを調べるために、ELISA試験が実施された。ELISA検出に関して、プレート(Nunc Maxisorp、デンマーク)は、花粉アレルゲン抽出物(100μg/ml:フレウム プラテンセ、ロリウム ペレンネ、ポア プラテンシス、ダクティリス グロメラタ、セカーレ ケレアーレ、トゥリティクム アエスティブム、アベナ サティバ、ホルデウム ブルガレ、アントクサントゥム オドラトゥム)または精製組み換えアレルゲン(5μg/ml:rPhl p 5)を用いて被われた。ELISAプレートは、洗浄され、かつブロッキングされ、続いて1:2500に希釈したウサギ抗血清および免疫前の血清とともにインキュベーションされた。結合したウサギIgGは、HRP結合ヤギ抗ウサギIg抗血清(Jackson Immunresearch、ペンシルバニア)を用いて検出された。結果は、<5%の誤差を有する、2つ1組の決定の平均値を表している(図13、表5)。
ペプチドのそれぞれを用いた免疫化は、最初の免疫から4週間目において検出可能になり、かつ2回目の免疫の後に増強される、強いPhl p 5特異的IgG応答を誘導した。ペプチド 2を用いた免疫化は、最も高いPhl p 5特異的IgG応答を誘導し、ペプチド6、4、5および最も低い応答を誘導した1と続く。ロリウム ペレンネ、ポア プラテンシス、ダクティリス グロメラタ、セカーレ ケレアーレおよびホルデウム ブルガレにおけるアレルゲンの群5に対する反応性を欠如した抗ペプチド 1抗体を除いて、他のペプチド抗血清が、試験された草本花粉抽出物のそれぞれと交差反応した。
17.1. ペプチド特異的IgGによる、rPhl p 5aに対するアレルギー性患者のIgE結合の阻害
遮断抗体が免疫療法において重要な役割を果たすことを示されているので、ペプチドの遮断抗体を誘導する能力に関する情報は、重要である。
18.1. リンパ球増殖アッセイ
最も低いと見込まれるT細胞反応性を有するペプチドを同定して、治療関連副作用を最小化するために、T細胞反応性が、リンパ球増殖アッセイによって試験された。末梢血単核細胞(PBMC)は、2人の草本花粉アレルギー性患者から、Ficoll(Amersham Pharmacia Biotech、イギリス)密度勾配遠心分離によって単離された。PBMC(2×105)は、96ウェルプレート(Nunclone;Nalge Nunc International、デンマーク)の中の、2mMのL−グルタミン(SIGMA、アメリカ)、50μMのベータメルカプトエタノール(SIGMA)および1mlごとに0.1mgのゲンタマイシン(SIGMA)を補った200μlの無血清のUltra Culture培地(BioWhittaker、ロックランド、ME)において、加湿した雰囲気において37℃および5%のCO2において、3つ1組にして培養された。細胞は、異なる濃度の合成ペプチド(ウェル毎に1.25、0.6および0.3μg)および比較を目的として、ウェル毎に4Uのインターロイキン−2(Boehringer Mannheim、ドイツ)および培地単独を用いて刺激された。培養から6日後に、ウェル毎に0.5μCiの[3H]チミジンが加えられ、その16時間後において、取り込まれた放射性がマイクロベータシンチレーションカウンター(Wallac ADL、ドイツ)を用いて液体シンチレーションカウントによって測定された。cpmの平均値は、3つ1組から算出され、かつ刺激指標(SI)は、抗原刺激またはインターロイキン−2刺激および未刺激対照によって得られたcpmの商として算出された。
ネコアレルギーのワクチン接種に好適なペプチドを得るために、長さ30から36アミノ酸であり、かつ分子の全体の範囲に渡る5つのペプチドが、Fel d 1の公知のアミノ酸配列に従って設計された。
ワクチン接種に好適化された低アレルゲン誘導性ペプチドを同定するために、Fel d 1由来合成ペプチドに対する血清IgE反応性が調べられた。
5つのFel d 1由来合成ペプチドのIgE結合能は、14人のネコアレルギー性患者からの血清を用いて、完全なrFel d 1アレルゲンのIgE結合能と比較された。ELISAプレート(Nunc Maxisorb、デンマーク)は、Fel d 1由来合成ペプチドまたは対照としてのrFel d 1を用いて被われ(0.5μg/ウェル)、洗浄され、かつブロッキングされた。それから、プレートは、ネコアレルギー性患者および非アトピー性の個人からの、1:5希釈した血清を用いて、4℃において夜通しインキュベーションされた。結合したIgE抗体は、1:2500に希釈したホースラディッシュペルオキシダーゼ標識した抗ヒトIgE抗体(KPL、アメリカ)を用いて検出された。
IgE結合は、効果細胞結合特異的なIgEの交差結合をも要求するタイプIのアレルギー性反応の誘導に対して、十分条件ではないが、必要条件であるので、Fel d 1由来合成ペプチドの実際のアレルギー誘導性活性は、好塩基球活性化アッセイにおいて試験される。これらのアッセイは、好塩基球活性化に対するマーカーとしていずれもが認識されている表面マーカーCD203c、およびCD63のアレルゲン特異的な上方制御を検出する(Hauswirth et al. J Allergy Clin Immunol. (2002) 110:102-109)。
Fel d 1由来合成ペプチドは、IgE結合に関して欠陥があることを示された。アレルゲン特異的IgG抗体の誘導を目的とするワクチン接種の候補分子として、ペプチドは、特異的なアレルゲン構造を保持している必要があり、かつ完全なアレルゲンに対する特異的なIgG免疫応答を依然として誘導することができる必要がある。Fel d 1由来合成ペプチドがこの要求を満たすか否かを見出すために、ウサギにおける免疫実験が実施された。
改良されたSITに関して所望の候補分子は、低下したIgE関連副作用という利点を与えるだけでなく、低下したT細胞媒介副作用という利点も与える。Fel d 1由来合成ペプチドT細胞活性化特性を調べるために、リンパ球増殖アッセイが実施された。
完全なrFel d 1に対するアレルギー性患者のIgE抗体の結合を阻害する、ペプチド誘導ウサギIgGの能力は、ELISA競合アッセイにおいて試験された。ELISAプレート(Nunc Maxisorb、デンマーク)は、rFel d 1(0.05μg/ウェル)を用いて被われ、洗浄され、かつブロッキングされた。それから、プレート結合rFel d 1は、1:100に希釈したウサギ抗ペプチド抗血清(単一の抗ペプチド抗血清および抗ペプチド抗血清の混合物が使用された)、ウサギ抗rFel d 1抗血清、および対照を目的としてまた、それぞれのウサギ免疫前の血清とともに、前インキュベーションされた。プレートが洗浄された後に、それらは、ネコアレルギー性患者からの、1:5に希釈したヒト血清とともにインキュベーションされた。結合したIgE抗体は、1:2500に希釈したホースラディッシュペルオキシダーゼ標識抗ヒトIgE抗体(KPL、アメリカ)を用いて検出された。抗ペプチド抗血清との前インキュベーションによって達成されたIgE結合の阻害%は、以下のように(IgE結合の阻害%=100−O.D.I/O.D.P×100(O.D.Iは、ウサギ免疫血清との前インキュベーションの後に測定された光学密度であり、かつO.D.Pは、ウサギ免疫前の血清との前インキュベーションの後に測定された光学密度である))算出された。
Bet v 1に由来する、表面露出B細胞エピトープは、治療的ワクチン接種および予防的ワクチン接種のモデルマウスにおける防御性のBet v 1特異的IgG応答を誘導することを、すでに示されている(Focke M et al. Clin Exp Allergy (2004) 34:1525-1533)。Focke M et al. (2004) において、6つのBet v 1由来ペプチドがマウスの免疫化の前に担体分子KLHに対して結合された。本実施例において、これらのペプチド(表1)を用いて誘導されたBet v 1特異的IgG応答が、Bet v 1アレルゲンではなく、担体に由来するT細胞エピトープの助けによって促進されることが示される。LFPKVAPQAISSVENIEGNGGPGTIKKISF(配列番号20)、GPGTIKKISFPEGFPFKYVKDRVDEVDHTN(配列番号21)およびVDHTNFKYNYSVIEGGPIGDTLEKISNEIK(配列番号22)の配列を有するBet v 1由来ペプチドが、関連するT細胞応答を誘導しないことが、驚くことに見出され、かつT細胞応答の大部分が、担体分子であるKLHに対して対象化されていることが証明され得た(図18および19)。この知見は、治療的なワクチン接種の間における副作用の低下にとって、かつ予防的ワクチン接種の間における潜在的な感作の危険を低下させることにとって、非常に重要なものである。あらゆるIgE反応性を欠如するが、アレルゲン由来T細胞エピトープを含むアレルゲン由来ペプチドが、T細胞活性化に起因する副作用を誘導したことは、過去に証明されている。Bet v 1について例証されているような、IgEおよびT細胞反応性を欠如するアレルゲン由来ペプチドは、治療的ワクチン接種の間において、IgE媒介副作用もT細胞媒介服作用も誘導しない。予防的ワクチン接種に使用される場合に、ペプチドは、Bet v 1特異的T細胞の準備刺激なしに、Bet v 1特異的防御性IgG応答を誘導する。これは、続くアレルギー性感作のための道を敷き得るワクチンを介した、アレルギー性免疫応答を前準備刺激することの危険を最小化すべきである。
Der p 2由来ペプチドのIgE結合能は、表13に従ったペプチドを採用し、かつイエダニアレルギーにかかっている個体の血清を用いる、実施例15.1および20.1に記載のように決定された。
26.1. ペプチドの設計
IgE結合活性能に対する、ペプチド長における変動の影響を研究するために、Phl p 5由来ペプチドのT細胞反応性および免疫原性の変異体が、少数のアミノ酸だけ、ペプチド 1(P1)の長さを伸ばすこと、およびペプチド 2(P2)の長さを減らすことによって設計された(表15)。
Phl p 5由来ペプチド1、2およびこれらの変異体1a、2bのIgE反応性を分析するために、0.2μgのペプチド/ドットを適用し、かつ7人の草本花粉アレルギー性患者からの血清(p1−p7)および非アトピー性患者からの血清(NHS)を用いて、ドットブロットアッセイが実施された。結合したIgEは、125I標識抗ヒトIgE(Phadia、Uppsala、スウェーデン)を用いて検出された。rPhl p 5は、陽性対照として使用され、かつHSAは、陰性対照として使用された。患者は、rPhl p 5に対して反応するが、ペプチドおよびペプチドの変異体に対して反応しない(図21)。
2人のアレルギー性患者からのPBMCは、異なる濃度の、Phl p 5由来ペプチド 1、2、それらの変異体 1a、2bおよび対照を目的としたrPhl p 5を用いて、刺激された。ペプチドを用いて得られた刺激指標は、rPhl p 5を用いて得られた刺激指標よりも有意に低かった(図22)。
ウサギは、KLH結合Phl p 5由来ペプチドおよび変異体を用いて免疫された。ELISA実験が用いられて、ペプチドおよびそれらの変異体に対する、得られたウサギ抗血清のIgG反応性を測定した(表16)。ペプチドおよびそれらの変異体を用いた免疫化は、ペプチドおよび対応する変異体を認識する交差反応性IgG抗体を誘導した。
a.抗ペプチド 1血清(抗P1)は、ペプチド 1変異体(1a)と交差反応し、かつ抗ペプチド 1a血清(抗P1a)は、ペプチド 1と交差反応する。
b.抗ペプチド 2血清(抗P2)は、ペプチド 2変異体(2b)と交差反応し、かつ抗ペプチド 2b血清(抗P2b)は、ペプチド 1と交差反応する。
ヒトIgEがrPhl p 5に対して結合することを阻害する、ウサギ抗ペプチド2および2b IgGの能力は、競合ELISAにおいて研究された。rPhl p 5を結合させたELISAプレートは、抗P2、抗P2b、および対照を目的とした抗Phl p 5抗血清を用いて前インキュベーションされた。それから、プレートは、12人の草本花粉アレルギー性患者からの血清にさらされた。rPhl p 5に対するIgE結合の阻害%は、表17に示される。抗ペプチド 2抗血清および抗ペプチド 2b抗血清は、患者のrPhl p 5に対するIgE結合を同程度に阻害する。
ヒトライノウイルスは、100を越えて異なる株からなる。この中和試験において、1つの株のライノウイルス感染がまた、他の株のVP1特異的抗体よって阻害され得ることは、示される。HeLa細胞は、等しい密度において、ウェルに播種された。100TCD50のHRV14は、抗14VP1−および抗89VP1−抗体の希釈物(未希釈;1:2−1:32を1−6のウェル)を用いて前インキュベーションされ、かつレーンAおよびDのそれぞれにおけるウェルに加えられた。レーンBおよびCにおいてTCD50のHRV89は、抗14VP1−および抗89VP1抗体の希釈物のそれぞれを用いて前インキュベーションされ、かつ細胞に対して加えられた。3日後に、生細胞が、紫に染色された。抗89VP1抗体および抗14VP1抗体は、類似の様式において、HRV14の感染を阻止する。また、14VP1および89VP1に対して産生された抗体は、同じ濃度までのHRV89の感染を阻害する。
Claims (57)
- アレルゲンから得られる少なくとも1つの低アレルギー誘発性分子から構成される低アレルギー誘発性タンパク質であって、当該少なくとも1つの低アレルギー誘発性分子が少なくとも1つの第2の非アレルギー誘発性タンパク質またはこれらの断片と融合されているかまたは接合されている、低アレルギー誘発性タンパク質。
- 上記少なくとも1つの低アレルギー誘発性分子が、上記少なくとも1つの第2のタンパク質またはこれらの断片のN末端および/またはC末端に対して融合されていることを特徴とする請求項1に記載の低アレルギー誘発性タンパク質。
- 上記少なくとも1つの第2のタンパク質が、ウイルスタンパク質、特にRNAもしくはDNAウイルスタンパク質、細菌タンパク質、真菌タンパク質または原生生物タンパク質であることを特徴とする請求項1または2に記載の低アレルギー誘発性タンパク質。
- 上記ウイルスタンパク質がカプシドタンパク質であることを特徴とする請求項3に記載の低アレルギー誘発性タンパク質。
- 上記ウイルスタンパク質の少なくとも1つが、ヒト病原性ウイルス、好ましくはピコルナウイルス科のウイルスから得られることを特徴とする請求項3または4に記載の低アレルギー誘発性タンパク質。
- ピコルナウイルス科の上記ウイルスが、ライノウイルス属、好ましくはヒトライノウイルス、特にヒトライノウイルス89または14の種に属することを特徴とする請求項5に記載の低アレルギー誘発性タンパク質。
- 上記アレルゲンが、主要なカンバ花粉アレルゲン、特にBet v 1およびBet v 4;主要なオオアワガエリ花粉アレルゲン、特にPhl p 1、Phl p 2、Phl p 5、Phl p 6およびPhl p 7;主要なイエダニアレルゲン、特にDer p 1およびDer p 2;主要なネコアレルゲン、特にFel d 1およびFel d 2;主要なミツバチアレルゲン;主要なスズメバチアレルゲン;プロフィリン、特にPhl p 12;オリーブアレルゲン、特にOle e 1;パリエタリア ユダイカアレルゲン、特にPar j 2;ブタクサアレルゲン、特にAmb a 1;ヨモギ花粉アレルゲン、特にArt v 1;ならびに貯蔵庫ダニアレルゲン、特にLep d 2からなる群から選択されることを特徴とする請求項1〜6のいずれか1項に記載の低アレルギー誘発性タンパク質。
- 低アレルギー誘発性分子が、低下したIgE結合能を示すことを特徴とする請求項1〜7のいずれか1項に記載の低アレルギー誘発性タンパク質。
- 低アレルギー誘発性分子が、低下したT細胞反応性を示すことを特徴とする請求項1〜8のいずれか1項に記載の低アレルギー誘発性タンパク質。

- 上記ベクターが発現ベクターであることを特徴とする請求項12に記載のベクター。
- 上記ベクターが、細菌ベクター、真菌ベクター、昆虫ベクター、ウイルスベクターまたは哺乳類ベクターであることを特徴とする請求項12または13に記載のベクター。
- 請求項11に記載の核酸分子または請求項12〜14のいずれか1項に記載のベクターを包含する宿主。
- 請求項1〜10のいずれか1項に記載の低アレルギー誘発性タンパク質に対して対象化された抗体。
- 上記抗体がモノクロナル抗体またはポリクロナル抗体であることを特徴とする請求項16に記載の抗体。
- 請求項1〜10のいずれか1項に記載の低アレルギー誘発性複合体または請求項16もしくは17に記載の抗体を包含するワクチンの調合物。
- 上記調合物が、少なくとも1つの補佐剤、薬学的に受容可能な賦形剤および/または防腐剤をさらに包含することを特徴とする請求項18に記載の調合物。
- 上記調合物が、10ng〜1g、好ましくは100ng〜10mg、特に0.5μg〜200μgの上記低アレルギー誘発性タンパク質または抗体を包含することを特徴とする請求項18または19に記載の調合物。
- ヒトもしくは動物におけるウイルス感染および/またはアレルギーを処置するかまたは予防する薬剤の製造に対する、請求項1〜10のいずれか1項に記載の低アレルギー誘発性タンパク質または請求項16もしくは17に記載の抗体の利用。
- 上記薬剤が、少なくとも1つの補佐剤、薬学的に受容可能な賦形剤および/または保存剤をさらに包含することを特徴とする請求項21に記載の利用。
- 上記薬剤が、能動免疫または受動免疫に使用されることを特徴とする請求項21または22に記載の利用。
- 上記薬剤が、10ng〜1g、好ましくは100ng〜10mg、特に0.5μg〜200μgの上記低アレルギー誘発性タンパク質または抗体を包含することを特徴とする請求項21〜23のいずれか1項に記載の利用。
- 上記薬剤が、0.01mg/kg体重〜5mg/kg体重、好ましくは0.1mg/kg体重〜2mg/kg体重の量において個体に投与されることを特徴とする請求項21〜24のいずれか1項に記載の利用。
- 個体におけるアレルギーおよび/またはウイルス感染の診断に対する、請求項1〜10のいずれか1項に記載の低アレルギー誘発性複合体または請求項16もしくは17に記載の抗体の利用。
- 薬剤もしくはワクチンにおける担体としての、またはウイルス感染、特に風邪の診断に対する、ピコルナウイルス属のウイルスから得られるウイルスカプシドタンパク質の利用。
- 上記ウイルスが、ヒトライノウイルス、特にヒトライノウイルス89または14の種に属することを特徴とする請求項27に記載の利用。
- C末端および/またはN末端の切り詰め(truncation)を有し、かつ野生型のPhl p 5と比較して低下したIgE結合能を示すPhl p 5から得られる低アレルギー誘発性分子。
- 切り詰めた(truncated)上記分子が、低下したT細胞反応性を示すことを特徴とする請求項29に記載の分子。
- 切り詰めた上記Phl p 5が、Phl p 5の93〜128、98〜128、26〜53、26〜58、252〜283番目のアミノ酸またはこれらの配列変異によって構成されることを特徴とする請求項30に記載の分子。
- 切り詰めた上記Phl p 5が、Phl p 5の132〜162、217〜246、176〜212番目のアミノ酸またはこれらの配列変異によって構成される請求項29に記載の分子。
- C末端および/またはN末端の切り詰めを有し、かつ低下したIgE結合能を示すFel d 1から得られる低アレルギー誘発性分子。
- 切り詰めた上記分子が、低下したT細胞反応性を示すことを特徴とする請求項33に記載の分子。
- 切り詰めた上記Fel d 1が、Fel d 1の鎖 1の1〜34もしくは35〜70番目のアミノ酸、Fel d 1の鎖 2の1〜34、35〜63もしくは64〜92番目のアミノ酸またはこれらの配列変異によって構成されることを特徴とする請求項33または34に記載の分子。
- C末端および/またはN末端の切り詰めを有し、かつ低下したIgE結合能を示すDer p 2から得られる低アレルギー誘発性分子。
- 切り詰めた上記分子が、低下したT細胞反応性を示すことを特徴とする請求項36に記載の分子。
- 切り詰めた上記Der p 2が、Der p 2の1〜33、21〜51、42〜73、62〜103、98〜129番目のアミノ酸またはこれらの配列変異によって構成されることを特徴とする請求項36または37に記載の分子。

- 低下したIgE結合能を示し、かつ任意に低下したT細胞反応性を示す請求項29〜39のいずれか1項に記載の分子の少なくとも2つを包含する低アレルギー誘発性融合タンパク質。
- 上記分子の少なくとも2つが同じアレルゲンから得られる場合に、上記分子が野生型のアレルゲンにおける断片の順番とは異なる順番において互いに融合されていることを特徴とする請求項40に記載の融合タンパク質。
- 請求項29〜39のいずれか1項に記載の低アレルギー誘発性分子または請求項40もしくは41に記載の融合タンパク質をコードする核酸分子。
- 請求項42に記載の核酸分子を包含するベクター。
- 上記ベクターが発現ベクターであることを特徴とする請求項43に記載のベクター。
- 上記ベクターが、細菌ベクター、真菌ベクター、昆虫ベクター、ウイルスベクターまたは哺乳類ベクターであることを特徴とする請求項43または44に記載のベクター。
- 請求項18に記載の核酸分子または請求項43〜45のいずれか1項に記載のベクターを包含する宿主。
- 請求項29〜39のいずれか1項に記載の低アレルギー誘発性分子または請求項40もしくは41に記載の融合タンパク質に対して対象化された抗体。
- 上記抗体が、モノクロナル抗体またはポリクロナル抗体であることを特徴とする請求項47に記載の抗体。
- 請求項29〜39のいずれか1項に記載の低アレルギー誘発性分子、請求項40もしくは41に記載の融合タンパク質または請求項46もしくは47に記載の抗体を包含するワクチン調合物。
- 上記調合物が、少なくとも1つの補佐剤、薬学的に受容可能な賦形剤および/または防腐剤をさらに包含することを特徴とする請求項49に記載の調合物。
- 上記調合物が、10ng〜1g、好ましくは100ng〜10mg、特に0.5μg〜200μgの上記低アレルギー誘発性分子または抗体を包含することを特徴とする請求項49または50に記載の調合物。
- 個体におけるアレルギーを処置するかまたは予防する薬剤の製造に対する、請求項29〜39のいずれか1項に記載の低アレルギー誘発性分子、請求項40もしくは41に記載の融合タンパク質、請求項42に記載の核酸分子、請求項43〜45のいずれか1項に記載のベクター、または47もしくは48に記載の抗体の利用。
- 上記薬剤が、少なくとも1つの補佐剤、薬学的に受容可能な賦形剤および/または防腐剤をさらに包含することを特徴とする請求項52に記載の利用。
- 上記薬剤が、10ng〜1g、好ましくは100ng〜10mg、特に0.5μg〜200μgの上記免疫原性分子、核酸分子、ベクター、宿主または抗体を包含することを特徴とする請求項52または53に記載の利用。
- 上記薬剤が、0.01mg/kg体重〜5mg/kg体重、好ましくは0.1mg/体重〜2mg体重の量において個体に投与されることを特徴とする請求項52〜54のいずれか1項に記載の利用。
- 上記個体が、アレルギーになる危険性にさらされていることを特徴とする請求項52〜55のいずれか1項に記載の利用。
- 個体におけるアレルギーの診断またはアレルギー療法の推移の監視に対する、請求項29〜39に記載の低アレルギー誘発性分子、請求項40に記載の融合タンパク質、または請求項47もしくは48に記載の抗体の利用。
