図5に示す形状及び構成の微小突起(360μmの微小突起長さ及び190個/cm2の微小突起密度)を有する円形シート(シート面積は2cm2)を備えたチタンの微小突起部材を、モデル・タンパク質ワクチンオバルブミンで被覆した。フルオロセインでタグを付けたオバルブミンの200mg/mLのコーティング水溶液を準備した。コーティングのためには、微小突起部材をこの溶液に短時間浸漬し、送風して乾燥させ、室温で一晩乾燥させた。その後の分析で、微小突起部材が200から250μg/cm2でオバルブミンによって被覆されたことが実証された。
ドライコーティングしたオバルブミンで第2の実験を実行し、衝撃後の投与量及び異なるチタン微小突起部材を使用し、適用時間を長くした手作業の適用を比較した。フルオロセインでタグを付けたオバルブミンの200mg/mLのコーティング用水溶液を準備した。微小突起部材(微小突起長さ214μm、保持形体なし、微小突起292個/cm2、2cm2の円盤)をコーティング溶液に短時間浸漬し、送風して乾燥して、室温で一晩乾燥させた。その後の分析で、微小突起部材が200から250μg/cm2でオバルブミンによって被覆されたことが実証された。
Claims (3)
- 薬剤を被験者の角質層を通して伝達する方法であって、
1つ又は複数の角質層に穿刺する微小突起を有する微小突起部材を提供するステップと、
前記微小突起部材を前記被験者の皮膚部位付近に配置するステップと、
衝撃力で前記微小突起部材を突き刺すステップとを含み、それによって前記微小突起部材は、前記角質層との衝突時に前記微小突起部材の1cm2当たり約0.05〜3ジュールの範囲のエネルギを10ミリ秒以内の貫入期間にわたって与え、それによって前記角質層穿刺微小突起の少なくとも1つが、前記被験者の前記角質層を通る微小スリットを形成し、さらに、
前記微小スリットを通して前記薬剤を伝達するステップを含む方法。 - 薬剤を被験者の角質層を通して伝達する方法であって、
1つ又は複数の角質層に穿刺する微小突起を有する微小突起部材を提供するステップと、
衝撃力で前記微小突起部材を突き刺すステップとを含み、それによって前記微小突起部材は、前記角質層との衝突時に前記微小突起部材の1cm2当たり約0.1〜0.3ジュールの範囲のエネルギを1から10ミリ秒の範囲の貫入期間にわたって与え、それによって前記角質層穿刺微小突起の少なくとも1つが、前記被験者の前記角質層を通る微小スリットを形成し、さらに、
前記微小スリットを通して前記薬剤を伝達するステップを含む方法。 - 薬剤を被験者の角質層を通して伝達する方法であって、
1つ又は複数の角質層を穿刺する微小突起を有する微小突起部材を提供するステップと、
第1衝撃力を提供するような構成である衝撃アプリケータを提供するステップと、
前記微小突起部材を前記被験者の皮膚部位付近に配置するステップと、
前記衝撃アプリケータを、前記微小突起部材との動作上の関係で前記皮膚部位付近に配置するステップと、
前記微小突起部材に前記第1衝撃力を与えるために前記衝撃アプリケータを起動するステップとを含み、それによって前記微小突起部材は、前記角質層との衝突時に前記微小突起部材の1cm2当たり約0.1〜0.3ジュールの範囲のエネルギを1から10ミリ秒の範囲の貫入期間にわたって与え、それによって前記角質層穿刺微小突起の少なくとも1つが、前記被験者の前記角質層を通る微小スリットを形成し、さらに、
