Abstract
【解決手段】

Description
本出願は、発明の名称が「新規の治療タンパク質ベースの分子クラス(Novel Class of Therapeutic Protein Based Molecules)」である、2004年9月10日出願の米国特許出願番号10/939,262号(その全体が本明細書中で参考として援用される)の優先権の利益を主張する。
本発明は、ヒト及び動物被験体の病原体による感染を予防及び治療するために使用することができる治療組成物、具体的には、ウイルス感染及び細菌感染の防止及び治療のために使用することができるタンパク質ベースの治療組成物に関する。本発明はまた、アレルギー反応及び炎症反応を防止又は改善するために使用することができるタンパク質ベースの組成物に関する。本発明はまた、遺伝子治療のために使用される組換えウイルス等の組換えウイルスの形質導入効率を増加させるためのタンパク質ベースの組成物に関する。
本発明は、病原体による感染の防止及び治療のための現在の治療法が適宜得た様式で提供することがしばしば困難であり、望ましくない副作用を引き起こし、薬物耐性病原体株が生じ得ることを認識する。本発明はまた、現在のアレルギー及び炎症を治療するためのアプローチの有効性が制限されており、副作用に関連することを認識する。更に、本発明はまた、現在の組換えウイルスを投与するアプローチの形質導入効率が低く、遺伝子治療の有効性が不満足なものであることを認識する。
定義
他で定義しない限り、本明細書中で使用した全ての技術用語及び科学用語は、本発明に属する当業者に一般に理解されている意味を有する。一般に、本明細書中で使用した命名法及び下記の製造手順又は実験手順は、周知であり、当該分野で一般的に使用されている。これらの手順のために従来の方法(当該分野及び種々の一般的参考文献で提供されている方法)を使用する。用語を単数形で提供する場合、我々はこの用語の複数形も意図する。用語及び定義が参考として援用される参考文献と矛盾する場合、本出願で使用した用語は、本明細書中に記載の定義に従う。開示全体で使用されるように、以下の用語は、他で示さない限り、以下の意味を有すると理解すべきである。

本明細書中で使用される、「細胞外アンカードメイン」又は「アンカードメイン」は、標的細胞の外面又は標的細胞の外面に極めて接近して存在する物質に安定に結合することができる任意の部分である。アンカードメインは、本発明の化合物を標的細胞の外面又はその付近に保持するのに役立つ。
本発明の化合物は、病原体による細胞の感染を防止又は妨害することができるか、被験体の免役応答を調整することができるか、組換えウイルスの形質導入効率を改善することができる細胞外活性を有する少なくとも1つの治療ドメインを含む。治療活性は、非限定的な例として、結合活性、触媒活性、又は阻害活性であり得る。本発明のいくつかの実施形態では、治療活性は、病原体による細胞感染に寄与する病原体の機能を修飾又は阻害するように作用する。他の実施形態では、治療ドメインは、標的細胞又は標的生物の機能を修飾するか阻害することができる。
本発明の化合物は、選択的に、化合物のドメインを連結することができる1つ又はそれ以上のリンカーを含み得る。リンカーを使用して、化合物のドメインを最適に間隔をあけるか折り畳むことができる。リンカーによって連結された化合物のドメインは、治療ドメイン、アンカードメイン、又は化合物の安定性の増強、精製を容易にすること等の更なる機能を提供する化合物の任意の他のドメイン又は部分であり得る。本発明の化合物のドメインを連結するために使用されるリンカーは、化学的リンカー又はアミノ酸リンカー若しくはペプチドリンカーであり得る。化合物が1つを超えるリンカーを含む場合、リンカーは同一でも異なっていてもよい。化合物が1つを超えるリンカーを含む場合、リンカーの長さは同一でも異なっていてもよい。

実施例1:アプロチニン(aprotinin gene)の合成、アプロチニン融合タンパク質の精製及び試験
序論
インフルエンザウイルスタンパク質血球凝集素(HA)は、主なインフルエンザエンベロープタンパク質であるHAはウイルス感染において重要な役割を果たす。HAの重要性は、HAが宿主免疫応答によって産生された防御中和抗体の主な標的であるという事実によって証明されている(Hayden,FG.(1996)In Antiviral drug resistance(ed.D.D.Richman),pp.59−77.Chichester,UK:John Wiley & Sons Ltd.)。HAはウイルス感染において2つの異なる機能を有することが現在明らかである。第1に、HAは、シアル酸細胞受容体へのウイルスの付着を担う。第2に、HAは、ウイルスエンベロープの細胞膜との融合の誘発による標的細胞へのウイルスの侵入を媒介する。
アプロチニンは、58個のアミノ酸残基及び3つの鎖内ジスルフィド結合を有する一本鎖ポリペプチドである(配列番号1)。アプロチニンのアミノ酸配列を、図1に示す。アプロチニン及びアプロチニン融合タンパク質をコードする遺伝子を、テンプレートとして大腸菌発現のために至適化したコドンとの重複オリゴヌクレオチドを使用したPCRによって合成する。PCR産物を、pCR2.1−TOPOベクター(Invitorgen)にクローン化する。配列決定後、遺伝子を、発現ベクターpQE(Qiagen)にサブクローン化する。ベクターは、組換えタンパク質を容易に精製するための精製タグ(Hisx6)を保有する。コンストラクトを使用して、大腸菌を形質転換する。LB−アンピシリン培地中で中対数期まで成長させた形質転換細胞を、標準的プロトコールに従ってIPTGによって誘導する。細胞をペレット化し、超音波処理によってリン酸緩衝化生理食塩水(PBS)に溶解する。His6精製タグを有する酵素を、ニッケルカラム(Qiagen)を使用して精製する。

アプロチニン及びアプロチニン融合タンパク質のトリプシン阻害活性を、以前に詳述されている測光アッセイによって測定する(Fritz H and Wunderer G.(1983)Arzneim−Forsch 33:479−494)。簡単に述べれば、このアッセイでは、アプロチニンは、Na−ベンゾイル−L−アルギニン−p−ニトロアニリド(BzArgpNA又はL−BAPA)(Sigma)のトリプシン触媒加水分解を阻害し、これを、405nmで測光的に追跡する。他のトリプシン単位(UBAPA)は、1分間あたりの1μmolの基質の加水分解に対応する。1阻害単位(IUBAPA)は、2つのトリプシン単位の活性を50%減少させ、これは、1UBAPAのトリプシンの阻害に数学的に対応する。アプロチニンの比活性を、IUBAPA/mgポリペプチドで示す。
標的としてヒトプラスミンを使用した表面プラズモン共鳴アッセイ(すなわち、BIAcore分析(BIAcore,Piscataway,NJ))を使用して、種々のリンカーに対する二量体及び三量体アプロチニンの親和性を、単量体アプロチニンと比較する。同様に、標的としてヘパリンを使用したBIAcoreアッセイを使用して、GAG結合アプロチニン融合タンパク質とヘパリンとの間の親和性を分析する。
インフルエンザウイルスのストック
インフルエンザウイルス株を、ATCC及びSt.Jude Children’s Research Hospitalの貯蔵所から入手する。インフルエンザウイルスに関与する全ての実験を、生物安全レベルIIで行う。
ウイルスストックの感染力及び力価を、2種のプラークアッセイ(従来のアッセイ及び修正したアッセイ)によって決定する(Tobita,K,Sugiura,A,Enomoto,C and Furuyama,M.(1975)Med Microbiol Immnuol 162:9−14;Zhimov OP,Ovcharenko AV and Bukrinskaya AG.(1982)Arch Virol 71:177−183)。従来のプラークアッセイは、ウイルス滴定法として日常的に使用されている。これは、MDCK単層へのウイルス感染直後に外因性のトリプシンを添加した寒天を重層する必要がある(Tobita,K,Sugiura,A,Enomoto,C and Furuyama,M.(1975)Med Microbiol Immnuol 162:9−14)。この方法は、非切断HAを有する全てのウイルス粒子の活性化によって試験されるウイルスストックの感染力を人為的に増加させる。
1.初代ヒト上皮細胞の短期間培養。従来のin vitroインフルエンザウイルス感染を、主に培養培地に外因性トリプシンを添加したMDCK細胞で行う。トリプシンがin vivoでインフルエンザウイルスを活性化するプロテアーゼではないので、これは生理学的にかけ離れており、本明細書中で提案した研究には不適切である。これまで報告されている外因性プロテアーゼを使用しないでインフルエンザウイルスの成長を支持することができるin vitro組織培養モデルは非常に限られており、腎臓起源の霊長類細胞を含む初代培養物、孵化卵の尿膜腔及び羊膜腔を裏打ちする細胞、胎児気管臓器培養、及び初代ヒトアデノイド上皮細胞である(Endo Y,Carroll KN,Ikizler MR and Wright PF.(1996)J Virol 70:2055−2058)。これらのうち、初代ヒトアデノイド上皮細胞を使用した最も最近の研究がヒト条件に最も近い。この場合、Endo et.al.(Endo Y,Carroll KN,Ikizler MR and Wright PF.(1996)J Virol 70:2055−2058)は、ヒトアデノイドの手術サンプルから上皮細胞を単離し、上皮細胞をコラーゲンマトリクス(Vitrogen 100,Celtrix Laboratories,Palo Alto,California)を含むトランスウェルインサート(Transwell insert)(Costar,Cambridge,Mass)上で培養した。細胞を、成長因子及び微量元素を補足した50%HamF12及び50%得イーグル最初基本培地中で維持した。細胞は、10〜14日目でコンフルエンシーに達し、大部分が単層として維持されたが、繊毛細胞の不連続なパッチも存在し、通常の繊毛活動をコンフルエンシー到達後1〜3週間維持された。この系では、インフルエンザA型ウイルスは106PFU/mlの力価且つ0.001の感染多重度で成長した(Endo Y,Carroll KN,Ikizler MR and Wright PF.(1996)J Virol 70:2055−2058)。感染中、進行性の細胞変性効果も認められた。この系の最も大きな欠点は、新鮮なヒトアデノイド組織が必要なことである。
アプロチニン融合タンパク質の抗ウイルス効果
1.感染前の処置。アプロチニン融合タンパク質を、種々の濃度で初代ヒト細胞培養物に添加し、細胞を1時間インキュベートする。細胞を新鮮な培地で洗浄し、直ちに0.01〜1のMOIのインフルエンザウイルスと共にインキュベートする。1時間後に細胞を再度洗浄し、3〜5日間培養する。種々の時点の上清中のウイルスの力価及び感染力を、2つのプラークアッセイによって測定する。ウイルス感染に起因する細胞変性効果を、生きた細胞のクリスタルバイオレットによる染色及び実験終了時の570nmでの吸光度の測定による定量によって評価する。アプロチニン融合タンパク質による細胞防御率を、100×{(アプロチニン処置サンプル−非処置感染サンプル)/(非感染コントロール−非処置感染サンプル)}によって計算する。細胞防御についての薬物有効性を、50%の細胞保護を達成するその有効濃度(EC50)によって記載する。HA活性化は、新規の放出されたウイルス粒子に対してのみ起こるので、最初のウイルス感染ラウンドは正常に起こり、ウイルス力価は感染後24時間で上昇する。しかし、第2ラウンドの開始から、ウイルスの感染力は低下し、ウイルス力価はアプロチニン処置の結果として漸減する。この実験の結果により、1回の予防的処置でのその有効性によって種々の異なるアプロチニン融合タンパク質型が識別される。
アプロチニン融合タンパク質がインフルエンザHAタンパク質の切断の阻害によってインフルエンザウイルス感染を阻害することを証明するために、ヒト初代上皮細胞培養物を、MOI 1にてインフルエンザウイルスに感染させる。アプロチニン融合タンパク質を、ウイルス接種直前又はウイルス感染直後のいずれで培養物に添加する。感染6.5時間後、培養物を、冷メチオニンを欠き、且つ100μCi/ml(パルス)の濃度の35S標識メチオニン(Amersham)を含むMEM中で1時間インキュベートする。その後、細胞を、10倍濃度の冷メチオニンを含むMEMで2回洗浄し、MEM中で更に3時間インキュベートする(追跡)。標識後、細胞を放射免疫沈降アッセイ(RIPA)緩衝液に溶解し、HAを、感染のために使用したウイルスの特定の株に対する抗血清(抗インフルエンザ血清は、ATCC及びCenter of Disease Control and Preventionから入手することができる)によって沈殿させ、免疫複合体を、プロテインG−Sepharose(Amersham)によって精製する。サンプルを、SDS−PAGEによって分画し、オートラジオグラフィを行う。アプロチニン融合タンパク質で処置されていないサンプルでは、HA1及びHA2が主なHA種であると予想される一方で、アプロチニン処理サンプルでは、HA0が存在する主なHA型であると予想される。
序論
インフルエンザウイルスは、RNAウイルスのオルトミクソウイルス科ファミリーに属する。A型及びB型は共に、宿主細胞に由来する脂質エンベロープ中に封入された8つのセグメント化マイナス鎖RNAゲノムを有する。ウイルスエンベロープは、以下の3つのタンパク質から構成されるスパイクで覆われている:宿主細胞受容体にウイルスを付着させてウイルスと細胞膜との融合を媒介する血球凝集素(HA);宿主細胞からの新規のウイルスの放出を促進するノイラミニダーゼ(NA);及びイオンチャネルとしての機能を果たす少数のM2タンパク質。インフルエンザA型ウイルスについて、HA及びNAは共に抗原連続変異及び抗原不連続変異を受け、ウイルスサブタイプを、そのHAタンパク質とNAタンパク質との間の血清学的相違によって識別する。全部で15のHA型(H1〜H15)及び9つのNA型(N1〜N9)が存在し、これまでに3つのHA(H1〜H3)及び2つのNA(N1及びN2)のみが、ヒトインフルエンザA型ウイルスで見出されている(Granoff,A.& Webster,R.G.,ed.Encyclopedia of Virology,2nd Edition,Vol 2)。インフルエンザA型ウイルスと対照的に、インフルエンザB型ウイルスでは異なる抗原サブタイプは認められない。
NEU2、NEU4、及びM.ビリジファシエンス(M.viridifaciens)の酵素をPBS及び50%グリセロール中にて−20℃で保存する。ウエルシュ菌(C.perfringens)及びA.ビスコサス(A.viscosus)の酵素を、10mM酢酸緩衝液(pH5)中にて4℃で保存する。タンパク質調製物(prep)を、HPLC及びSDS−PAGE電気泳動によって特徴づける。酵素の比活性及び安定性を、シアリダーゼアッセイによってモニタリングする。
1.インフルエンザウイルスのストック
インフルエンザウイルス株を、ATCC及びSt.Jude Children’s Research Hospitalの貯蔵所から入手する。ウイルスストックを、0.3%ウシ血清アルブミン及び0.5マイクログラムトリプシン/mlを補足した最少基本培地(MEM)中にてMadin−Darbyイヌ腎臓(MDCK)細胞上で成長させる。48〜72時間のインキュベーション後、培養培地を低速遠心分離によって明澄化する。ウイルス粒子を、25%スクロースクッションによる超遠心によってペレット化する。精製ウイルスを、50%グリセロール−0.1M Tris緩衝液(pH 7.3)に懸濁し、−20℃で保存する。ウイルス力価を、プラークアッセイ(Tobita,K,Sugiura,A,Enomoto,C and Furuyama,M.(1975)Med Microbiol Immnuol 162:9−14)又は50%のMDCK細胞が感染する必要があるウイルスの用量であるTCID50によって決定する。

2.Neu5Acアルファ(2,3)−Galを有する株には、具体的には、動物単離物A/duckUkraine/1/63、A/duckMemphis/928/74、A/duckhokk/5/77、A/Eq/Miami/1/63、A/Eq/Ur/1/63、A/Eq/Tokyo/71、A/Eq/Prague/71等が含まれる(Connor,RJ,Kawaoka,Y,Webster,RG and Paulson JC.(1994)Virology 205:17−23)。
このアッセイを使用して、各酵素が受容体Neu5Acアルファ(2,6)−Gal及びNeu5Acアルファ(2,3)−Galを破壊する効率を迅速に決定する。
MDCK細胞のコンフルエントな単層を、種々の濃度のシアリダーゼで1時間処置し、緩衝液で2回洗浄し、種々のインフルエンザウイルス株を感染させる。1時間のインキュベーション後、細胞を再度洗浄して非結合ウイルスを除去する。細胞表面上のウイルス結合部位の減少を確立するために、細胞に寒天を重層し、37℃でインキュベートする。シアリダーゼ処置細胞中のプラーク数を、コントロール細胞と比較する。或いは、細胞を通常の培地中で37℃で培養し、培養培地中のウイルス力価を、TCID50として種々の培養期間で測定する。
初代ヒト気管支上皮細胞を購入し(Clonetics)、製造者の説明書に従って、添加最少培地で培養する。種々の濃度のシアリダーゼを培養培地に添加する。7〜10日間にわたって細胞成長を測定する。また、細胞を、微視的細胞変性効果について定期的に観察する。
1.アンカードメインとしてのGAG結合配列の選択
全ての性質(抗ウイルス活性、毒性、安定性、産生の容易さ等が含まれる)が最良である1つのシアリダーゼを選択する。次いで、我々は、これをGAG結合配列を遺伝学的に連結し、pQEベクターに融合遺伝子をサブクローン化し、大腸菌由来の融合タンパク質を発現及び精製する。

正常な細胞成長及び形態に対する融合タンパク質の効果を、種々の濃度の融合タンパク質との初代ヒト気管支上皮細胞との培養、細胞の成長曲線の追跡、及び任意の微視的細胞変性効果の観察によってモニタリングする。
機能ドメイン及びアンカードメインから構成される融合タンパク質を、多数のより異なる適用のためにデザインする。多数の他の感染性微生物(パラミクソウイルス(Wassilewa,L.(1977)Arch Virol 54:299−305)、コロナウイルス(Vlasak,R.,Luytjes,W.,Spaan,W.and Palese,P.(1988)Proc Natl Acad Sci USA 85:4526−4529)、ロタウイルス(Fukudome,K.,Yoshie,O.and Konno,T.(1989)Virology 172:196−205)、緑膿菌(Ramphal,R.and PyIe,M.(1983)Infect Immun 41:339−44)等)も細胞受容体としてシアル酸を使用することが公知であるので、例えば、本明細書中で提案したシアリダーゼ融合タンパク質を、インフルエンザに加えて他のウイルス及び細菌による感染に対する治療薬/予防薬としても使用することができる。例えば、ヘパリン結合ドメインに融合したアプロチニンは、インフルエンザに加えて、パラインフルエンザウイルス等の活性化に宿主セリンプロテアーゼを必要とする他のウイルスの感染を防止/治療するために使用される融合タンパク質を作製することができる。
巨大細菌シアリダーゼに関する公開された論文によれば、51kDaのアルスロバクター・ウレアファシエンス(Arthrobacter ureafaciens)シアリダーゼ、ウエルシュ菌(C.perfringens)由来の71kDaのシアリダーゼ、及びアクチノミセス・ビスコサス(Actinomyces viscosus)由来の113kDaのシアリダーゼは、種々のシアル酸抱合体に対する比活性及び広範な基質特異性が類似しているようである(Biology of the Sialic Acids(1995),270−273;Corfield et al.,Biochem.J.,(1981)197(2),293−299;Roggentin et al.,Biol.Chem.Hoppe Seyler,(1995)376(9),569−575;Teufel et al.,Biol.Chem.Hoppe Seyler,(1989)370(5),435−443)。第3のシアリダーゼ(ミクロモノスポラ・ビリジファシエンス(Micromonospora viridifaciens)由来の68kDaの酵素)は、インフルエンザウイルス受容体を破壊することも公知である(Air and Laver,Virology,(1995)211(1),278−284;(1995),270−273)。
シアリダーゼ融合タンパク質を産生するために、発現コンストラクトを、大腸菌BL21に形質転換した。1つのコロニーを、2.5mlのLBブロスに接種し、震盪しながら37℃で一晩成長させた。午前中に、2mlの一晩培養物を500mlのTB培地を含む2リットルの震盪フラスコに接種し、培養物を、OD600=4.0まで(2〜4時間)、震盪しながら37℃で成長させた。最終濃度が1mMのIPTGの添加によってタンパク質発現を誘導し、撹拌しながら3時間継続した。5,000×gで10分間の遠心分離によって細胞を採取した。細胞を1回洗浄し(PBS中に再懸濁し、再度遠心分離する)、15mlの溶解緩衝液に再懸濁した。

コンストラクト番号2によってコードされたAR−AvCDタンパク質のシアリダーゼ活性をアッセイし、ウエルシュ菌(C.perfringens)(Sigma,St.Louis,MO)及びA.ウレアファシエンス(A.ureafaciens)(Prozyme,San Leandro,CA)から精製した天然のシアリダーゼの活性と比較した。更に、アンフィレグリンGAG配列(配列番号7)をNeu2ヒトシアリダーゼ(配列番号8)と融合したコンストラクトから産生された融合タンパク質もシアリダーゼ活性についてアッセイした。
AR−AvCD融合タンパク質のN末端は、一定の条件下で部分的に切断され、程度は小さいが精製AR−AvCD調製物中のタンパク質が不均一になる。この問題を解決するために、我々は、シアリダーゼ融合タンパク質のN末端を至適化するためのアプローチをデザインした。N末端に無作為なアミノ酸を有するAR−AvCDを含むライブラリーを、以下のように構築した。AR−AvCDを、遺伝子の5’末端上にアニーリングするプライマーがアミノ酸2及び3に対応する位置に無作為化配列を含むプライマー対を使用したPCRによって増幅した。プライマーのヌクレオチド配列及びコードされるアミノ酸配列を以下に示す。

ARコンストラクトが実際にシアリダーゼ融合タンパク質の細胞表面活性を改善するかどうかを評価するために、我々は、コンストラクト番号2(図7に示す配列番号24)又はコンストラクト番号1(His6−AvCD、図5に示す配列番号17)で形質転換した大腸菌から精製したタンパク質を初代ヒト気管支上皮細胞とインキュベートし、十分な洗浄後に細胞結合シアリダーゼ活性を測定した。コンストラクト番号2タンパク質(配列番号25)とインキュベートした細胞について、10%までのシアリダーゼが細胞に結合していることが見出され、細胞結合シアリダーゼ活性は、コンストラクト番号2タンパク質の投入濃度によって用量依存的様式で増加した。しかし、コンストラクト番号1タンパク質(配列番号18)とインキュベートした細胞は、バックグラウンドレベルのシアリダーゼ活性しか示さなかった。更に、我々は、コンストラクト番号2タンパク質又はコンストラクト番号1タンパク質のいずれかでMDCK細胞を処理し、細胞表面上の残存アルファ(2,6)結合シアル酸レベルを測定した(図8)。100mU/ウェル未満の酵素活性と同一レベルで、コンストラクト番号2タンパク質は、コンストラクト番号1タンパク質より顕著に高い有効性を示した。これらの結果は、ARドメインが実際にシアリダーゼ機能を増強させることを示す。
インフルエンザウイルスのストック
インフルエンザウイルス株を、ATCC及びSt.Jude Children’s Research Hospitalの貯蔵所から入手する。インフルエンザウイルスに関与する全ての実験を、生物安全レベルIIで行う。
コンストラクト番号2AR−AvCDタンパク質がインフルエンザウイルスから細胞を防御する能力を評価するために、我々は、最初にMDCK細胞をコンストラクト番号2から作製したAR−AvCD又はA.ウレアファシエンス(A.ureafaciens)から単離した広範な細菌シアリダーゼで処置し、広範囲に選択したヒトインフルエンザウイルス(IFV)(ヒトIFV AのH1、H2、及びH3サブタイプ、ヒトIFV Bが含まれる)及びトリIFV株で細胞を攻撃した。図9に示すように、コンストラクト番号2から作製した融合タンパク質は、80〜100%の細胞防御を示し、これは、A.ウレアファシエンス(A.ureafaciens)シアリダーゼの効果に匹敵していた。
我々は、細胞ベースのELISA法を使用して、AR−AvCDタンパク質(コンストラクト番号2を使用して作製)及びAR−G4S−AvCDタンパク質(コンストラクト番号3を使用して作製)によってIFV増幅阻害を評価した(Belshe et al.,J Virol.,(1988)62(5),1508−1512)。
AR−AvCD又はAR−G4S−AvCDタンパク質(コンストラクト番号2及び3から作製)の細胞傷害性を評価するために、MDCK細胞を、低密度で96ウェルプレートに播種し、10%FBS、及び、ウェルあたり20UまでのAR−AvCDタンパク質、又はAR−G4S−AvCDタンパク質を含むDMEM中で5日間培養した(両シアリダーゼは、全実験中に完全な活性を保持していた)。AR−AvCD又はAR−G4S−AvCD処置ウェル又はコントロールウェル中の細胞密度を、クリスタルバイオレットでの細胞の染色及び570nMでの吸光度の測定によって毎日決定した。最も高い濃度のAR−AvCD又はAR−G4S−AvCD(100U/ml)を含む培養物でさえも、細胞成長は阻害されなかった。従って、AR−AvCD又はAR−G4S−AvCDのIC50(細胞成長が50%阻害される薬物濃度である)は、約100U/mlである。
フェレットに、ヒト非適合インフルエンザウイルスを感染させてヒトに匹敵する疾患の徴候を得て、ザナミビル(Relenza)等の抗ウイルス化合物で処置することができる(Mendel et al.,Antimicrob Agents Chemother,(1998)42(3),640−646;Smith and Sweet,Rev.Infect.Dis.,(1988)10(1),56−75;Reuman et al.,J.Virol.Methods,(1989)24(1−2),27−34)。我々の化合物のin vivo有効性を評価するために、我々は、フェレットモデルにおけるAR−AvCDタンパク質(コンストラクト番号2から作製)を試験した。具体的には、試験によって血清中の抗血球凝集素抗体の存在について陰性であった24匹の幼若メスフェレット(0.5〜0.8kg)(Marshall Farms,North Rose,NY)を、本研究に含めた。2種の動物を各ケージに入れ、試験前に3日間馴化させた。動物を、以下の3つの群に無作為に分けた:8匹を、薬物希釈緩衝液及びウイルス攻撃で処置し、12匹をAR−AvCD及びウイルス攻撃で処置し、4匹をAR−AvCDのみで処置した。500U/mlのシアリダーゼ活性及び0.7mg/mlのタンパク質濃度のリン酸緩衝化生理食塩水(PBS)に溶解したAR−AvCDの調製物を本研究で使用した。薬物処置群の動物に、合計で約1mg/kgの投薬量レベルに達する各用量の1mlのAR−AvCDを投与した。
細菌
肺炎球菌(S.pneumoniae):異なる血清型の10種の被包性株を、ATCCに寄託されている臨床分離株から選択する。細菌を凍結ストックとして維持し、3%ヒツジ血液(Difco&Micropure Medical Inc.)を含むトリプシンダイズ寒天プレート上に5%CO2下にて37℃で18時間継代した。肺炎球菌(S.pneumoniae)を放射性同位体で標識するために、1〜2日目のプレート培養物から接種物を取り、70μCiの[3H]リジン/mlを含むリジン欠損トリプシンダイズブロスに添加し、5%CO2下にて37℃でインキュベートした。各培養物の成長を、595nmでの光の吸収によってモニタリングする。対数後期で細菌を採取し、遠心分離(13,000rpm×3分)によって2回洗浄し、L−15培地(フェノールレッドを含まない)+0.1%BSA(L−15−BSA)に再懸濁した(Cundell and Tuomanen,Microb.Pathog.,(1994)17(6),361−374)(Barthelson et al.,Infect.Immun.,(1998)66(4),1439−1444)。
全ての[3H]標識細菌を、洗浄後にL−15−BSAに懸濁し、Petroff−Hausserチャンバーでの目視による計数によって細菌の濃度を決定し、シンチレーションカウンティングによって放射能を決定し、[3H]標識細胞の比活性を計算する。7cpm/1000細胞又はそれを超える細菌の調製物を使用する。細菌を、5×108細胞/mlに希釈する。BEAS−2B細胞単層を、5×107細菌を含む[3H]標識細菌懸濁液と、5%CO2下にて37℃でインキュベートした。30分後、非結合細菌を、L−15−BSAで5回の洗浄によって除去する。WD−HAE組織に付着した細菌を、シンチレーションカウンティングによって定量した。
BEAS−2B細胞を、1〜50mUのAR−AvCDと2時間インキュベートする。上記のように、インフルエンザ桿菌(H.influenzae)株及び肺炎球菌(S.pneumoniae)株を使用して、細胞接着アッセイを行う。正のコントロールとして偽処置細胞を使用する。AR−AvCDの有効性を、細菌付着に対して50%阻害を達成するための酵素の量であるEC50として定量する。
in vitro実験
公開された手順と類似の方法で、AR−AvCDの効果を証明する実験を行う(Bals et al.,J Virol.,(1999)73(7),6085−6088)。十分に分化した気道上皮(WDAE)細胞の単層を、transwell中で維持する(Karp et al.,Methods Mol.Biol.,(2002)188,115−137;Wang et al.,J Virol.,(1998)72(12),9818−9826)。細胞表面からシアル酸を排除するために、培養培地を、0.5〜10単位のAR−AvCDが溶解された無血清培地と置換する。細胞を、30分間〜6時間処置する。細胞単層を洗浄し、AAVで形質導入し、形質導入効率を、標準的な手順を使用して推定する。いくつかのtranswellを、コントロールの目的で用いるために培地のみ(AR−AvCDなし)で処置する(基本形質導入効率)。更なるコントロールには、脱シアルの細胞傷害性効果を評価するためのAR−AvCDのみで処置したtranswellが含まれ得る。形質導入細胞の検出を容易にするためにレポーターウイルスを使用する。レポーターAAV及びその使用の例は、文献に記載されており、AAV−CMV−eGFP、AAV2LacZ(Bals et al.,J Virol.,(1999)73(7),6085−6088;Wang et al.,Hum.Gene Ther,(2004)15(4),405−413)、及びアルカリホスファターゼ(Halbert et al.,Nat.Biotechnol.,(2002)20(7),697−701)が含まれる。効率を、固定し、適切な基質(lacZ又はAP含有ウイルスを使用する場合)で処置した細胞の光学顕微鏡法又は生きた細胞の蛍光顕微鏡法(GFPを使用する場合)によって推定する。NHBE初代上皮細胞(Cambrex,Walkersvile,MD)を使用してNexBioで行った実験に従って、transwellあたり10単位のAR−AvCDを使用した場合に1時間未満で最大量のシアル酸が除去される。使用した他の細胞株(例えば、MDCK)は、はるかに少ないAR−AvCDの投与量で脱シアルするようになる(1時間で0.1U)。従って、我々は、10UのAR−AvCDで2時間のWDAE処理で接近可能なシアル酸を除去し、AAVでのWDAE細胞の形質導入を有意に増強するのに十分であると予想する。
動物モデルにおけるAR−AvCD処置の効果を証明するために、以前に記載の実験に類似の実験を行う(Flotte et al.,Proc.Natl.Acad.Sci.U.S.A,(1993)90(22),10613−10617;Halbert et al.,Nat.Biotechnol.,(2002)20(7),697−701)。AAV投与の数時間前(1〜6時間前)に、以前に公開されたプロトコールに従って、AR−AvCDを、エアゾール又は凍結乾燥AR−AvCD粉末の鼻吸入によってマウスの肺に送達させる(Flotte et al.,Proc.Natl.Acad.Sci.U.S.A,(1993)90(22),10613−10617)。前に記載のように、レポーター遺伝子(アルカリホスファターゼ)を保有するAAVを鼻吸入によって送達し、4週間後にマウスを安楽死させ、固定した肺中の形質導入細胞を検出する(Halbert et al.,J Virol.,(1998)72(12),9795−9805)。
前に記載の実験方法を使用して(Cocchiara et al.,J Neuroimmunol.,(1997)75(1−2),9−18)、本発明の化合物による処置により、肥満細胞によるヒスタミン放出を誘導する基質P(SP)を妨害することを証明する。別の実験組を使用して(Stenton et al.,J Pharmacol.Exp.Ther.,(2002)302(2),466−474)、本発明の化合物による処置により、2つのPAR活性化ペプチドによって刺激された肥満細胞によるβ−ヘキソサミニダーゼ放出が阻害される(PARは、プロテイナーゼ活性化受容体を示す)。
