Abstract
【解決手段】少なくとも2つの逆方向反復配列の間に位置する核酸配列を含んで成り、ここで当該逆方向反復配列がSBタンパク質に結合することができ、そして当該核酸フラグメントが細胞内のDNAに組込まれることができる、核酸フラグメント。このシステムはトランスポザーゼのSBファミリーの構成員(SB)又はトランスポザーゼをコードする核酸、及び隣接逆方向反復配列を有する核酸配列を含む核酸フラグメントの利用を含む。

組換DNA
遺伝子再構築−段階1:トランスポザーゼオープンリーディングフレームの再構築
アトランタ産サケ由来のTssl.1要素(GenBank受託番号L12206)を欠陥トランスポザーゼ遺伝子と並んだプライマーペアー、FTC−Start及びFTC−Stopを利用してPCR増幅し、生成物SB1を得た。次に、Tss1.2要素の欠陥トランスポザーゼ遺伝子のセグメント(L12207)をPCRプライマーFTC−3及びFTC−4を用いてPCR増幅し、次いでFTC−3及びFTC−5で更に増幅させた。PCR生成物を(プライマー配列において下線を付した)制限酵素NcoI及びBlpIで消化し、そしてクローニングしてSB1内の対応のフラグメントを置換してSB2を得た。次にニジマス由来のTsg1要素の欠陥トランスポザーゼ遺伝子の約250bpのHindIIIフラグメント(L12209)を単離し、そしてSB2内の対応の部位にクローニングしてSB3を得た。このTss1及びTsg1要素は発表されており(Radiceら、1994)、そしてS.W.Emmons氏の贈呈品である。

実施例2
逆方向反復配列を有する核酸フラグメントの調製
トランスポザーゼ遺伝子に隣接する短い54bpの非直列反復配列(IR)を有するカエノルハブジチス・エレガンス(Caenorhabditis elegans)由来の原型Tc1トランスポゾンとは対照的に、魚類のほとんどのTcEはその末端にある長い210〜250bpのIR及び各IRの先端にある直列式反復したDNA配列モチーフ(DR)を有するTcEのIR/DRサブグループに属する(Ivicsら、1996;Izsvakら、1995、共に前掲)(図1A)。しかしながら、共通IR配列は完全な反復配列ではなく(即ち、類似ではあるが、同一ではない)、ほとんどTcEとは対照的に、これらの魚類要素は本来不完全な逆方向反復配列を有することが示唆される。その一致は、IRの中央では80%未満であるが、DRでは完全であり、この類似性の非ランダム分布が、IRの中に機能的に重要な配列モチーフを維持せしめたポジティブ選別の結果でありうることを示唆している(図3)。従って、我々はDRの周辺のDNA配列が転位のためのcis作用情報及びIR内の突然変異を保有しうるが、DRの外では、おそらくはこの要素の転位する能力が損われていることはないであろうと考える。モデル基質として、我々は配列がサケ型共通配列と3.8%しか異ならないインタクトDRモチーフを有するタニクシス・アルボヌベス(Tanichthys albonubes)(以降、Tと呼ぶ)から一本のサケ型TcE基質配列を選んだ。4種類のDNA配列のDNase−保護化領域における変動は約83%から約95%で変動した。SEQ ID NO:6〜9を参照のこと。
実施例3
SBトランスポザーゼのDNA特異性
アトランタ産サケ及びゼブラダニオのゲノムの中には少なくとも2種類のTcEサブファミリー、即ちTss1/Tdr1及びTss2/Tdr2のそれぞれがある。同じサブファミリーに由来する要素は、たとえそれらが同じ種における2種類のサブファミリーの構成員ではなく、2種類の種に由来するときでさえも(例えば、Tss1及びTdr1)、約70%の核酸同一性を有するようである。例えば、Tdr1及びTdr2はそれらがコードするトランスポザーゼ及びそれらの逆方向反復配列において特徴的に異なり、そして約30%の核酸同一性しか有さない。所定のトランスポゾンサブファミリーは交差移動を避けるために互いと有意に相違し合わなくてはならない。主たる課題はトランスポザーゼの基質認識が近縁したサブファミリーのトランスポゾンの活性化を防ぐのに十分特異性であるかである。
実施例4
SBトランスポザーゼによるDNAの転位
以下の実験は当該合成サケ型SBトランスポザーゼが転位の複雑な工程を全て実行することを実証する。即ち、DNA分子を認識し、基質DNAを切り出し、そしてそれを細胞のDNA、例えば細胞染色体に挿入する。これはSBトランスポザーゼを含まず、それ故組込みも非相同性認識を介して達成するコントロールサンプルとは対照的である。
核の中に入り込み、そして逆方向反復配列内のその作用部位に特異的に結合する能力に加えて、完全に活性なトランスポザーゼはトランスポゾンを切り出し、そしてそれを組込むものと期待される。SBトランスポザーゼのC末端半分において、3つのタンパク質モチーフがDD(34)E触媒ドメインを構築する;2個の不変アスパラギン酸残基D(153)及びD(244)、並びにグルタミン酸残基E(279)。最後の2つは34個のアミノ酸で離れている(図2)。インタクトDD(34)E枠がTc1及びTc3トランスポザーゼの触媒機能のために必須である(van Luenenら、1994 Cell 79,293-301;Vos and Plasterk,1994、前掲)。
細胞を10%の胎児牛血清の添加したDMEMの中で培養し、トランスフェクションの1日前に6cmプレーン上に播種し、そしてBRL由来のリポフェクチンを用いて5μgのElutip(Schleicher and Schuell)−精製プラスミドDNAでトランスフェクションした。DNA脂質複合体との5時間のインキュベーション後、細胞をリン酸緩衝食塩水(PBS)中の15%のグリセロールで30sec「グリセロールショック」にかけ、PBSで1回洗い、次いで血清含有培地を再供給した。トランスフェクションの2日後、トランスフェクション化細胞をトリプシン処理し、2mlの血清含有DMEMの中に再懸濁し、そしてこの細胞懸濁物のアリコート1ml又は0.1mlのいずれかを600μg/mlのG418(BRL)を含む培地の数枚の10cmプレートの上に播種した。2週間の選別後、細胞クローンを拾い、そして個々の培養物の中で増殖させるか又はPBS中の10%のホルムアルデヒドで固定し、PBS中のメチレンブルーで30min染色し、脱イオン水でよく洗い、風乾し、そして写真撮影した。
実施例5
様々な種に由来する細胞内でのDNAの転位
トランスポザーゼ活性の宿主要件を、マウス由来のLMTK及びヒト由来のHela、並びにゼブラダニオ由来の胚細胞の3種類の脊椎動物細胞を用いて評価した。
実施例6
SBトランスポゾン由来の安定な遺伝子発現
トランスポゾンシステムは、例えば遺伝子治療、及びバイオリアクターシステムのための動物染色体への遺伝子導入の目的のために、その導入する遺伝子が確実に発現される限りにおいて、遺伝子導入のために機能的であろう。ねむれる美女(sleeping beauty)トランスポザーゼ媒介導入後の遺伝子発現の精度を決定するため、我々はSV40プロモーターの指令下にあるグリーンフルオレセントタンパク質(GFP)遺伝子を含むトランスポゾンとねむれる美女トランスポザーゼをコードするin vitro合成したmRNAとを第1ゼブラダニオ胚の中にマイクロインジェクションした。胚形成の際にGFPの多少の発現を示す注射した胚のうちの34個を成熟に至るまで増殖させ、そして野生型ゼブラダニオと交配させた。このような交配から、我々は34匹の魚のうち4匹がGFP遺伝子をその子孫に伝達できることを見い出した(表1)。A,B,C及びDと表示するこれらの4匹のFo魚の子孫におけるGFPの発現を評価し、そしてその魚を成長させた。この4匹のオリジナル始祖から、GFP遺伝子の伝達率は約2%〜12%(表1)に範囲し、平均約7%であった。これらの魚におけるGFPの発現は同じ組織タイプ中の全ての個体においてほぼ同一であり、GFP遺伝子の発現が卵及び精子を介する伝達を経て復活することを示唆する。これらのデーターは生殖系がGFP遺伝子の発現に関してモザイクであり、そして遺伝子の発現が安定であることを示唆する。魚DのF1子孫を互いと交配させた。この場合、我々は約75%の伝達を予測し、そして実際に69/70(77%)のF2魚が同じ組織において同等レベルでGFPタンパク質を発現することを見い出した。これは、SBトランスポゾンシステムが少なくとも2世代の動物を通じて確実に発現できる遺伝子を導入する能力の更なる証拠である。
実施例7
挿入突然変異誘発及び遺伝子発現のためのSBトランスポゾン
ゲノム内及びゲノム間で一の染色体位置から別の位置へと移動するその固有の能力に基づき、転位可能な要素を以下の生物を含む所定の生物の遺伝子操作を変革した:細菌(Gonzalesら、1996 Vet.Microbiol.48,283-291;Lee and Henk,1996.Vet.Microbiol.50,143-148)、ショウジョウバエ(Ballinger and Benzer,1989 Proc.Natl.Acad.Sci.USA 86,9402-9406;Bellenら、1989 Genes Dev.3,1288-1300;Spradlingら、1995 Proc.Natl.Acad.Sci.USA 92,10824-10830)、C.エレガンス(Plasterk,1995.Meth.Cell.Biol.,Academic Press,Inc.pp.59-80)及び様々な植物種(Osborne and Baker,Curr.Opin.Cell Biol,7,406-413(1995))。トランスポゾンはトランスポゾンタギング、エンハンサートラッピング及び遺伝子導入のための有用なベクターとしての仕事をする。しかしながら、全てではないにしても大半の経済的に重要な動物はかかる道具を包いている。その簡潔性及び様々な生物において機能するその明白な能力のため、SBはDNAトランスポゾン技術が現在適用できない種のための効率的なベクターとしての有用性を発揮するであろう。
実施例8
遺伝子マッピングのためのマーカーとしてのSBトランスポゾン
導入遺伝子及びその他の遺伝子座をマッピングするための反復要素も同定した。DANAは、先祖SINE要素への短い配列の挿入により集成されたと思われる異なるカセットの異常なサブ構造を有するレトロポゾンである。DANAはダニオ(Danio)系の中で約4×105コピー/ゲノムで増幅した。DANAとほぼ同じくらいに豊富なAngel要素は魚類遺伝子付近に見い出せる逆方向反復配列である。DANA及びAngel要素は共にゲノムの中でランダムに分布され、そしてメデリアン(Medelian)状で分れているようである。DANA及びAngel要素に対して特異的なプライマーを利用するPCR増幅は魚類ストック間での多形態のスクリーニング及び遺伝子導入配列の移行のための遺伝子マーカーとして利用できる。相互的散在反復配列−PCR(IRS−PCR)は多形態DNAを検出するのに利用できる。IRS−PCRはメデリアン状で遺伝される多形態フラグメントを供する反復配列特異的プライマーを利用し、反復要素の隣接するゲノムDNAを増幅する(図10A)。多形態DNAフラグメントはIRS−PCRにおいてDANA又はAngel特異的プライマーにより作成でき、そして検出可能な多形態バンドの数は、SB様トランスポゾン等のゼブラダニオゲノム内の反復配列に対する様々なプライマーの組合せにより有意に増大しうる。

核酸フラグメント及びトランスポザーゼを多能性又は全能性細胞に導入し、ここで当該核酸フラグメントは少なくとも2つの逆方向反復配列の間に位置する核酸配列を含んで成り、ここで当該逆方向反復配列はSBタンパク質に結合し、そしてここで当該核酸フラグメントは細胞内のDNAの中に組込まれることができ、そしてここで当該トランスポザーゼはSEQ ID NO:1と少なくとも80%のアミノ酸配列同一性を有するSBタンパク質である;そして
前記細胞を動物へと成長させる;
工程を含んで成る方法。 - 前記多能性又は全能性細胞が卵母細胞、胚細胞、卵及び幹細胞から成る群より選ばれる、請求項63記載の方法。
- 前記導入工程が:
マイクロインジェクション;
エレクトロポレーション;
前記核酸フラグメントと陽イオン脂質小胞体又はDNA凝縮試薬との組合せ;及び
当該核酸フラグメントをウィルスベクターに組込み、次いでそのウィルスベクターを前記細胞と接触させること;
から成る群より選ばれる方法を含んで成る、請求項63記載の方法。 - 前記ウィルスベクターがレトロウィルスベクター、アデノウィルスベクターもしくはアデノ関連ウィルスベクター、又はヘルペスウィルスから成る群より選ばれる、請求項65記載の方法。
- 前記動物がマウス、魚類、有蹄類、鳥類又はヒツジである、請求項63記載の方法。
- 核酸を細胞内のDNAの中に導入するための方法であって:
少なくとも2つの逆方向反復配列の間に位置する核酸配列を含んで成る核酸フラグメントを細胞に導入することを含んで成り、ここで当該逆方向反復配列はSBタンパク質に結合することができ、そして当該核酸フラグメントはSBタンパク質の存在下で細胞内のDNAの中に組込まれることができる、方法。 - 前記方法がSBタンパク質を細胞に導入することを更に含んで成る、請求項68記載の方法。
- 前記SBタンパク質がSEQ ID NO:1と少なくとも80%の同一性を含んで成るアミノ酸配列を有する、請求項68記載の方法。

約10%のSDS−ポリアクリルアミドゲル上での約35kD〜約40kDの分子量幅;並びに

を含んで成るタンパク質であって、非相同性組換により得られるレベルと比べ、細胞の核酸に核酸フラグメントを導入する速度において少なくとも約5倍の向上を有する、タンパク質。 - 細胞内の核酸配列を移動させるための方法であって:
請求項84又は85記載のタンパク質を請求項1記載の核酸フラグメントを含むDNAを有する細胞の中に導入する工程を含んで成り、ここで当該タンパク質は細胞のDNA内の第一位置から細胞のDNA内の第二位置へと当該核酸を移動させる、方法。 - 前記細胞のDNAがゲノムDNAである、請求項87記載の方法。
- 前記細胞DNA内の第一位置が染色体外DNAである、請求項87記載の方法。
- 前記細胞DNA内の第二位置が染色体外DNAである、請求項87記載の方法。
- 前記タンパク質を前記細胞の中に核酸として導入する、請求項87記載の方法。
- 細胞のゲノム内の遺伝子を同定するための方法であって:
核酸フラグメント及びSBタンパク質を細胞に導入する、ここで当該核酸フラグメントは少なくとも2つの逆方向反復配列の間に位置する核酸配列を含んで成り、ここで当該逆方向反復配列は前記SBタンパク質に結合でき、そしてここで当該核酸フラグメントは当該SBタンパク質の存在下で細胞内のDNAの中に組込まれることができる;
当該細胞のDNAを前記核酸配列を切断することのできる制限エンドヌクレアーゼで消化する;
前記逆方向反復配列を同定する;
当該逆方向反復配列に近い核酸を配列決定し、オープンリーディングフレームからDNA配列を得る;そして
このDNA配列をコンピューターデーターベース内の配列情報と比較する;
工程を含んで成る方法。 - 前記制限エンドヌクレアーゼが6塩基認識配列を認識する、請求項92記載の方法。
- 前記消化工程が、消化したフラグメントのクローニング又は消化したフラグメントのPCR増幅を更に含んで成る、請求項93記載の方法。
- プロモーターに作用可能式に連結された遺伝子を含んで成る安定な遺伝子導入脊椎動物であって、当該遺伝子及びプロモーターに逆方向反復配列が隣接し、そしてこの逆方向反復配列がSBタンパク質に結合する、脊椎動物。
- 前記SBタンパク質がSEQ ID NO:1又はSEQ ID NO:1に対して少なくとも80%の相同性を有するアミノ酸配列を含んで成る、請求項95記載の安定な遺伝子導入脊椎動物。
- 前記脊椎動物が魚類である、請求項96記載の安定な遺伝子導入脊椎動物。
- 前記脊椎動物がゼブラダニオである、請求項97記載の安定な遺伝子導入脊椎動物。
- 前記脊椎動物がマウスである、請求項96記載の安定な遺伝子導入脊椎動物。
- 1又は複数の下記の配列と結合できるトランスポザーゼ活性を有するタンパク質:
SEQ ID NO:4,SEQ ID NO:5,SEQ ID NO:6,SEQ ID NO:7,SEQ ID NO:8,SEQ ID NO:9又はSEQ ID NO:10。 - SBタンパク質に特異的に結合できる抗体。
