Abstract

13. 表現されるべき上記自殺遺伝子配列がバリセラゾスターウィルスチミジンキナーゼであり、プロドラッグが6−メトキシプリンアラビノシド、またはその誘導体であることを特徴とする項目1の方法。
14. 該腫瘍が肝臓癌であり、該プロモーターがアルブミンプロモーター、アルファフェトプロテインブロモーター、α−アンチトリプシンプロモーター、およびホス ホエノルピルベートカルボキシナーゼプロモーターで構成されるグループから選択されることを特徴とする項目1の方法。
15. 該腫瘍が結腸癌であり、該プロモーターがカルボニックアンヒドラーゼIプロモーター、または悪性腫発生抗原プロモーターであることを特徴とする項目1の方法。
16. 該腫瘍が子宮癌であり、該プロモーターがエストロゲンプロモーター、アロマターゼサイトクロームP450プロモーター、コレステロール側鎖切断P450、および17アルファ−ヒドロキシラーゼP450プロモーターで構成されるグループから選択されることを特徴とする項目1の方法。

26. 動物またはヒトの固型腫瘍、乳頭腫、および疣贅を措置する方法において、DNA伝達システムによって個体に、自殺遺伝子と1つまたは複数の細胞毒性遺伝子を投与し、該DNA伝達システムがウィルス性ベクター、エピゾーム性ベタター、リポゾーム、陽イオン性脂質およびDNAまたはRNAなど細胞表面または抗原を識 別し、それに結合する分子構成、DNA結合分子構成、およびそれら細胞表面から細胞質へのそれら複合体を輸送することができる溶解性分子構成で構成される非ウィルス性伝達システムで構成されるグループから選択されることを特徴とするステップおよび該動物またはヒトに対してプロドラッグを投与するステップと で構成され、該プロドラッグがインビボで該自殺遺伝子によって毒性化合物に転化されることを特徴とする方法。
27. 自殺遺伝子配列がヘルペス単一体ウィルスのチミジンキナーゼ、バリセラゾスターのチミジンキナーゼ、およびバクテリア性シトシンデアミナーゼで構成されるグループから選択されることを特徴とする項目26の方法。
28. 該1つ、または複数のサイトカイン遺伝子配列がインターロイキン−1、インターロイキン−2、インターロイキン−4、インターロイキン−6、インターロイ キン−7、インターロイキン−10、インターロイキン−12、インターフェロン−α、インターフェロン−β、インターフェロン−δ、腫瘍壊死因子−α、腫瘍壊死因子−β、顆粒マクロファージコロニー刺激因子(GM−CSF)および顆粒細胞コロニー刺激因子(G−CSF)で構成されるグループから選択される ことを特徴とする項目26の方法。

37. 上記表現されるべき自殺遺伝子がバクテリア性サイトシンデアミナーゼであり、上記プロドラッグが5−フルオロシトシンまたはその誘導体であることを特徴とする、項目26の方法。
38. 上記表現されるべき自殺遺伝子配列がバリセラゾスターウィルスチミジンキナーゼであり、上記プロドラッグが6−メトキシプリンアラビノシドまたはその誘導体であることを特徴とする項目26の方法。
39. 動物またはヒトの固型腫瘍、乳頭腫、または疣贅を措置する方法において、第1のアデノウィルス性ベクターと、機能的表現のために適当な距離で順次結合 されたプロモーター、5′mRNAリーダー配列、開始部位、表現されるべき自殺遺伝子で、プロドラッグを毒性化合物に転化させることができる蛋白質をコード表現する自殺遺伝子を含んでいる核酸カセット、3′非翻訳領域およびポリアデニル化信号で構成される追加的アデノウィルス性ベクターを直接該腫瘍、乳頭 腫、または疣贅に導入するステップで、さらに該追加アデノウィルス性ベクターが機能的な表現のために適切な距離で結合されたプロモーター、5′mRNAリーダー配列、開始部位、表現されるべき サイトカイン遺伝子を含んだ核酸、3′非翻訳領域、およびポリアデニル化信号で構成されているステップとおよび該動物またはヒトにプロドラッグを投与するステップとで構成され、該プロドラッグがインビボで該自殺遺伝子によって毒性物質に転化されることを特徴とする方法。
40. 該プロモーターがラウスサルコーマウィルス−ロングターミナルリピート、サイトメガロウィルス性プロモーター、マウス白血病ウィルス−長末端反復、霊長類 ウイルス40初期および後期プロモーター、およびヘルペス単一体ウィルス−チミジンキナーゼプロモーターで構成されるグループから選択されることを特徴とする、項目39の方法。
41. 上記自殺遺伝子配列が、ヘルペス単一体ウィルスのチミジンキナーゼ、バリセラゾスターウィルス、およびバクテリア性サイトシンデアミナーゼで構成されるグループから選択されることを特徴とする項目39の方法。
42. 上記アデノウィルス性ベクターがインターロイキン−1、インターロイキン−2、インターロイキン−4、インターロイキン−6、インターロイキン−7、イン ターロイキン−10、インターロイキン−12、インターフェロン−α、インターフェロン−β、インターフェロン−δ、腫瘍壊死因子−α、腫瘍壊死因子−β、顆粒細胞−マクロファージコロニー刺激因子(GM−CSF)および顆粒細胞コロニー刺激因子(G−CSF)で構成されるグループから選択される蛋白質をコード表現する1つ、または複数のサイトカイン遺伝子配列を含んでいることを特徴とする項目39の方法。
43. 該プロドラッグがガンシクロビル、アシクロビル、1−5−ヨードウラシルFIAU、 5−フルオロシトシン、6−メトキシプリンアラビノシド、およびそれらの誘導体で構成されるグループから選択されることを特徴とする項目39の方法。
44. 該腫瘍が結腸、前立腺、乳房、肺、皮膚、肝臓、骨、膵臓、子宮、睾丸、膀胱、腎臓、頭部、または頚部癌で構成されるグループから選択されることを特徴とする項目39の方法。
45. 乳頭腫が鱗状細胞乳頭腫、脈絡膜叢乳頭腫、および喉頭乳頭腫で構成されるグループから選択されることを特徴とする項目39の方法。
46. 該疣贅が生殖器疣贅、足裏疣贅、疣贅状表皮異形成および悪性疣贅で構成されるグループから選択されることを特徴とする項目39の方法。
47. 該ガンシクロビルが約1mg/日/kg〜約20mg/日/kg体重の範囲の投与量で投与されることを特徴とする項目43の方法。

49. 該FTIUが約1mg/日/kg〜約50mg/日/kg体重の範囲の投与量で投与されることを特徴とする項目43の方法。
50. 上記表現されるべき自殺遺伝子配列がヘルペス単一体ウィルスのチミジンキナーゼであり、上記プロドラッグがガンシクロビル、アシクロビル、FIAU、またはその誘導体であることを特徴とする項目39の方法。
51. 上記表現されるべき自殺遺伝子がバクテリア性サイトシンデアミナーゼであり、上記プロドラッグが5−フルオロシトシンまたはその誘導体であることを特徴とする項目39の方法。
52. 上記表現されるべき自殺遺伝子配列がバリセラゾスターウィルスチミジンキナーセであり、上記プロドラッグが6−メトキシプリンアラビノシドまたはその誘導体であることを特徴とする項目39の方法。
53. 該腫瘍が肝臓癌であり、該プロモーターがアルブミンプロモーター、アルファフェトプロテインプロモーター、α−アンチトリプシンプロモーターおよびホスホエノルピルベートカルボキシナーゼプロモーターで構成されるグループから選択されることを特徴とする項目39の方法。
