本発明は、天然のヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン;ならびにヒトプラスミノーゲン中の対応するドメインに相同なC末端ドメイン活性化部位およびセリンプロテアーゼドメインを有するポリペプチドをコードするヌクレオチド配列を含んでなるポリヌクレオチドを提供し;該ポリペプチドは固定されたリシンに結合する。該N末端クリングルドメインは、天然のヒトプラスミノーゲンのクリングル1若しくはクリングル4に相同であり得る。
ィブリンに対する結合親和性より高い、部分的に切断されたフィブリンに対する結合親和性を表し得る。
完全長プラスミンのフィブリンおよびアンチプラスミン結合特性を有する単純なグリコシル化されていない分子を提供するため、本発明はプラスミノーゲンの欠失変異体を提供する。本明細書でδ−プラスミノーゲンと称される本変異体中で、クリングル1に相同なドメインと活性化部位の間の天然のアミノ酸配列の少なくとも一部分が欠失されている。一局面において、ヒトプラスミノーゲンの天然のクリングル1ドメインに相同なドメインを、実質的に、該ドメイン間に残存するプラスミノーゲン活性化部位を含有する介在する天然の配列のみを用い、プラスミノーゲンのセリンプロテアーゼ部分、若しくはその相同な機能的アナログに直接結合し得る。
ポリペプチドの「ドメイン」および「領域」という用語は、別の方法で逆に示されない限り、本明細書で使用されるとおり一般に同義である。「クリングル」若しくは「セリンプロテアーゼ」などのような十分に認識された構造若しくは機能の呼称と一緒に列挙される場合、こうした用語は、こうした呼称に対応するポリペプチド構造に関連することが普遍的に認識かつ理解される、少なくとも何らかの特徴(1種若しくは複数)に関するポリペプチドの特徴を紹介することができる。

本発明はまた、本発明の単離された核酸分子を包含するベクター、該組換えベクターで
遺伝子的に工作されている培養宿主細胞、および組換え技術によるδ−プラスミ(ノーゲ)ンポリペプチドの製造にも関する。

本発明のポリヌクレオチドは、本発明のポリペプチドの変形物および特定の例をコードするものを包含する。例えば、表現型上サイレントのアミノ酸置換の作成方法に関する指針はBowie,J.U.ら、“Deciphering the Message in Protein Sequences:Tolerance to Amino Acid Substitutions,”Science 247:1306−1310(1990)に提供され、ここで著者はタンパク質が驚くべきことにアミノ酸置換に寛容であることを示している。本発明の範囲内のいかなる数の置換もこうした一般的原則の応用により得ることができるとは言え、置換に関する特定の指針について、クリングル1ドメインの構造および機能に関する本明細書で引用される参考文献が当業者により調べられ得る。
δ−プラスミ(ノーゲ)ンは、米国第2003/0012778 A1号明細書;およびNovokhatny,V.ら、J.Thromb.Haemost.1(5):1034−41(2003)(双方は引用することにより本明細書に組み込まれる)に記述される方法および組成物に従って、治療的使用のため処方し得る。例えば、低pH(約2.5から約4まで)、低緩衝能力の緩衝液をδ−プラスミンの製剤に使用し得る。加えて、プラスミン、ミニプラスミンおよび/若しくはマイクロプラスミンで実施されるところの当業者に既知の他の方法および処方を使用して、本発明のδ−プラスミンを治療的投与のため処方し得る。
δ−プラスミノーゲンのアミノ酸配列は配列番号2に示す。δ−プラスミノーゲンをコードする推定の配列を、大腸菌(E.coli)発現およびmRNAの安定性のためコドンを最適化して、配列番号1に示されるところのDNA配列を生じた。

δ−プラスミノーゲン配列をコードするDNAを多様な細胞に形質転換し、そして、1mM IPTG(イソプロピルチオ−β−D−チオガラクトピラノシド)による誘導後のタンパク質の過剰発現をSDS−PAGEにより分析した。Arg、IleおよびLeuをコードする希な大腸菌(E.coli)tRNAを発現するよう工作した細胞型BL21(DE3)RIL(Stratagene、カリフォルニア州ラホヤ)細胞を、δ−プラスミノーゲンの産生に使用した。
δ−プラスミノーゲンベクターを含有するBL21(DE3)RIL細胞の単一コロニーを使用して、5mlのLB/アンピシリン(100μg/ml)/クロラムフェニコール(50μg/ml)に接種し、そして振とう機上37℃で8時間インキュベートした。その後、50μlのアリコートを、新鮮培地中でのさらなる増殖のため、培養細菌懸濁液から採取した。該処置を、6mlの細菌培養物および250mlの培地を用いて16時間後に反復した。培養物を振とうしながら37℃で約1.0のOD600nmまで増殖させ、そしてIPTGを1mMの最終濃度まで添加した。培養物を追加の5時間増殖させた。細胞を5,000×gでの遠心分離により収集し、そして細胞ペレットを20mM EDTAを含有する20mMトリス pH8.0に溶解しかつ−80℃で凍結させた。
EDTA、0.15M NaCl、0.15Mアルギニン−HClを含有する8倍容量の0.1Mトリス−HCl pH8.0に対し、緩衝溶液の頻繁な交換を伴い8〜10時間にわたり透析した。
を伴うAMICONフィルターを使用しておよそ10〜20mlに濃縮し、そして10mM EDTA、0.15M NaClを含有する100倍容量の0.1Mトリス pH8.0に対し一夜透析した。この物質を遠心分離して微粒子を除去し、その後リシンアフィニティー樹脂(リシン−セファロース4B;Amersham Biosciences、ニュージャージー州ピスカタウェイ)を通過させた。δ−プラスミノーゲンは、0.2Mのε−アミノカプロン酸(εACA)を含有するトリス緩衝生理的食塩水、pH8.0を使用して樹脂から溶出した。
精製したδ−プラスミノーゲンは、還元(ジチオスレイトール処理)および非還元タンパク質のSDS−PAGE分析により、35〜40kDaの領域の単一バンドとして出現した(図5を参照されたい)。MALDI質量分析により測定されたその正確な分子量は、37,198Daという期待された値に非常に近い37,089Daであった。
フィブリンと相互作用する能力を保持するかどうかを決定するため、δ−プラスミノーゲンのフィブリンへの結合をtPAによるそのその後の活性化および生じる血餅溶解により評価したマイクロタイタープレートアッセイで、そのフィブリン結合特性を試験した。この目的上、100μlの5mg/mlのフィブリノーゲンを、マイクロタイタープレートの各ウェル中でトロンビンと重合させた。多様な濃度のδ−プラスミノーゲンをフィブリン塊の上面に添加しかつ37℃で1時間インキュベートした。フィブリン塊が未だ無傷でありかつウェルに付着していた間に、プレートをPBSで徹底的に洗浄した。洗浄後、tPAの0.1〜mg/ml溶液を各ウェルに添加し、そしてプレートを37℃で2時間インキュベートした。結果として塊のいくつかが完全に溶解され、そしていくつかは部分的に溶解された一方、非常に少量のδ−プラスミノーゲンを含むウェルおよび対照ウェルは事実上無傷のままであった。線維素溶解の程度を、1M NaOH中で再構成した最初の塊の残部の280nmの吸光度を測定することによりモニターした。吸光度の値を、δ−プラスミノーゲン濃度の関数としてプロットした。
10mM EDTAおよび0.15M NaClを含有する0.1Mトリス緩衝液、pH8.0に対し透析したδ−プラスミノーゲンを、本質的にプラスミンについて以前に記述された(Marder,V.J.ら、Thromb Haemost.、86(3):739−45(2001)(引用することにより組み込まれる))とおり、セファロース4Bに固定したウロキナーゼを使用して、δ−プラスミンに活性化した。活性化は室温で起こり、そしてS−2251の活性の増大によりリアルタイムでモニターした。バッチごとに変動した(典型的には1〜2mg/ml)δ−プラスミノーゲンの量に依存して、インキュベーション時間は30〜60分であった。S−2251の活性がプラトーに達した活性化の完了に際して、ウロキナーゼ−セファロースを濾過し、そして活性のδ−プラスミンをベンズアミジン−セファロース(Pharmacia)に捕捉した。δ−プラスミンは、低pH緩衝液(0.2Mグリシン、pH3.0、0.3M NaCl、0.2M εACA)を使用して樹脂から溶出した。

δ−プラスミンのアミド分解活性を、プラスミン基質D−Val−Leu−Lys−p−ニトロアニリド(S−2251)(DiaPharma、オハイオ州ウエストチェスター)を使用して検査した。PBS緩衝液中pH7.4、25℃で、S−2251のMichaelis−Menten定数(Km)は138μMであることが見出された(表2)。該製剤のkcatは510min−1であることが見出された。4−グアニジノ安息香酸4−ニトロフェニル塩酸塩(pNPGB)滴定(Chase,T.とE.Shaw、Methods Enzymol.197:20−27(1970))を使用して、機能的活性部位のパーセントは67%であることが見出された。活性部位のパーセントについてkcatを補正して、755±45min−1のkcatを決定した。この値は、完全長のプラスミンについて(760±23min−1)およびマイクロプラスミン(全5個のクリングルを欠く)について(795±24min−1)同一アッセイで決定された値に非常に近かった(図9を参照されたい)。これらのデータは、クリングルの存在若しくは非存在が、セリンプロテアーゼドメインの触媒活性に影響を及ぼさないことを示す。
δ−プラスミンの血栓溶解効率を、以下の実験プロトコルを使用して、カテーテル補助(catheter−assisted)血栓溶解のin vitroモデル(Novokhatny,V.ら、J Thromb Haemost.、1(5):1034−41(2003)(引用することにより組み込まれる))で試験した。
Claims (21)
- 天然のヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン;ならびにヒトプラスミノーゲン中の対応するドメインに相同なC末端ドメイン活性化部位およびセリンプロテアーゼドメインを有するポリペプチドであって、固定されたリシンに結合するポリペプチドをコードするヌクレオチド配列を含んでなるポリヌクレオチド。
- N末端クリングルドメインが、天然のヒトプラスミノーゲンのクリングル1若しくはクリングル4に相同である、請求項1に記載のポリヌクレオチド。
- コードされるポリペプチドが、配列番号2に示される配列に最低90%若しくは95%、または98%同一である、請求項1に記載のポリヌクレオチド。
- コードされるポリペプチドが、配列番号2に示される配列に最低98%同一である、請求項1に記載のポリヌクレオチド。
- コードされるポリペプチドが配列番号2に示される配列である、請求項1に記載のポリヌクレオチド。
- ポリヌクレオチドのヌクレオチド配列が、配列番号1に示される配列、若しくはその縮重バリアントである、請求項1に記載のポリヌクレオチド。
- ヌクレオチド配列が、天然のヒトプラスミノーゲンのクリングル1若しくはクリングル4ドメインに相同な1個のN末端クリングルドメイン;ならびにヒトプラスミノーゲン中の対応するドメインに相同なC末端ドメイン活性化部位およびセリンプロテアーゼドメインを有するポリペプチドをコードする、請求項1に記載のポリヌクレオチド。
- ヌクレオチド配列が、天然のヒトプラスミノーゲンのクリングル1若しくはクリングル4ドメインに最低90%同一な単一のN末端クリングルドメイン;ならびにヒトプラスミノーゲンの活性化部位およびセリンプロテアーゼドメインに最低90%同一なC末端ドメインを有するポリペプチドをコードする、請求項1に記載のポリヌクレオチド。
- 天然のヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン;ならびにヒトプラスミノーゲン中の対応するドメインに相同なC末端ドメイン活性化部位およびセリンプロテアーゼドメインを含んでなるポリペプチドであって、該ポリペプチドが固定されたリシンに結合する、上記ポリペプチド。
- N末端クリングルドメインが、天然のヒトプラスミノーゲンのクリングル1若しくはクリングル4に相同である、請求項9に記載のポリペプチド。
- ポリペプチドが、ミニプラスミンの線維素溶解活性のα2−アンチプラスミンによる阻害速度より最低約5倍より速い阻害速度の、α2−アンチプラスミンにより阻害される線維素溶解活性を表す、請求項9に記載のポリペプチド。
- 阻害速度が、ミニプラスミンの阻害速度より最低約10倍、20倍、30倍若しくは40倍より速い、請求項11に記載のポリペプチド。
- 固定されたリシンが、リシン−アガロース、リシン−ヒドロゲル、リシン−架橋アガロースよりなる群から選択される固体支持体マトリックスに結合されたリシンである、請求項1に記載のポリペプチド。
- 固定されたリシンがリシン−架橋アガロースである、請求項13に記載のポリペプチド。
- ポリペプチドが、ミニプラスミンのフィブリノーゲンに対する結合親和性よりも低いフィブリノーゲンに対する結合親和性を表す、請求項1に記載のポリペプチド。
- ポリペプチドが、ミニプラスミンの部分的に切断されたフィブリンに対する結合親和性よりも高い、部分的に切断されたフィブリンに対する結合親和性を表す、請求項1に記載のポリペプチド。
- プラスミノーゲン活性化部位およびプラスミノーゲンセリンプロテアーゼドメインに対しN末端に位置する単一のクリングルドメインを含んでなるポリペプチドであって、該クリングルドメインは、天然のヒトプラスミノーゲンのクリングル5とよりも天然のヒトプラスミノーゲンのクリングル1若しくはクリングル4との最低1残基より大きいアミノ酸配列の同一性を有し、かつ、ヒトプラスミノーゲンのクリングル1および4の天然の配列に関しての該単一のクリングル領域の保存的置換が、クリングル5との同一性比較の目的上天然の配列と異なると考えられない、上記ポリペプチド。
- ポリペプチドが、配列番号2に示されるところのアミノ酸配列、およびその保存的置換を有する、請求項17に記載のポリペプチド。
- ポリペプチドが、配列番号2に示されるアミノ酸配列の位置76の相対位置に類似の相対位置に1アルギニン残基を有する、請求項17に記載のポリペプチド。
- 請求項1に記載のポリヌクレオチドを含んでなる発現ベクター。
- 請求項20の発現ベクターを含んでなる培養細胞。
