Description
本願は、引用することにより本明細書に組み込まれる、2007年11月29日に出願された米国仮出願60/991,148に対する35 USC §119の下で優先権を請求する。
1つの態様において、本発明は
a)天然ヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン、ここで、クリングルドメイン内の最後の4個のアミノ酸残基はV、P、QおよびCである;および
b)ヒトプラスミノーゲンにおける対応するドメインに相同なC末端ドメイン活性化部位およびセリンプロテアーゼドメイン
を有するポリペプチド(ここで、該ポリペプチドは固定化リシンに結合する)をコードするヌクレオチド配列を含んでなるポリヌクレオチドを提供する。
a)天然ヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン、ここで、クリングルドメイン内の最後の4個のアミノ酸残基はV、P、QおよびCである;および
b)ヒトプラスミノーゲンにおける対応するドメインに相同なC末端ドメイン活性化部位およびセリンプロテアーゼドメイン

b)段階a)において提供されるポリペプチドを1つもしくはそれ以上のペプチド結合を切断するために十分な条件下でプロテアーゼと接触させ、それにより1つもしくはそれ以上の組換えプラスミンポリペプチドを生成せしめること
を含んでなる。1つの態様において、提供することは適当な宿主において配列番号:1に示されるような配列に対応する配列もしくはその縮重バリアントを有するオープンリーディングフレームを発現させることを含んでなる。別の態様において、ポリペプチドは配列番号:2に示されるようなアミノ酸配列を有する。
本発明者等は、その構造からの4個のクリングルの欠失にもかかわらず天然プラスミノーゲン様特性を有する(TAL6003)−プラスミノーゲンと本明細書において呼ばれる新規組換えプラスミノーゲンポリペプチドもしくはそのバリアントを見出した。これらの(TAL6003)−プラスミノーゲンもしくはそのバリアントは、チモーゲンのクリングルドメインとセリンプロテアーゼドメインとの間に位置するArg−Valペプチド結合のタンパク質分解切断を少なくとも伴う活性化事象の後に機能性プラスミン酵素(本明細書において(TAL6003)−プラスミンと呼ばれる)に活性化されるようになることができるチモーゲンである。
i)(TAL6003)−プラスミノーゲンの活性化後に生成される(TAL6003)−プラスミンのより低い分子量(例えば、1つの態様において約36,911〜約37,039Da)は、増加した比活性(タンパク質のmg当たり)をもたらす;
ii)比較的低い分子量と合わせて、天然タンパク質に存在する少なくとも2個のグリコシル化部位(図3参照、すなわち、N289およびT346)の欠如は、比較的安価な細菌および酵母発現系を用いたこのタンパク質の組換え生産を容易にする;
iii)(TAL6003)−プラスミノーゲンは、プラスミノーゲンアクチベーターtPA、ウロキナーゼおよびストレプトキナーゼにより活性化することができる;
iv)天然ヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリング
ルドメインの存在は、血栓溶解効能に重要であるプラスミンのフィブリン結合特性を維持する;
v)天然ヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン上のα2−抗プラスミン結合部位の存在は、(TAL6003)−プラスミンがプラスミンのこの生理的阻害剤により迅速に阻害されることを可能にする(出血を防ぐことができる特性);
vi)(TAL6003)−プラスミンのより小さいサイズは、α2−マクログロブリンによるそれらの阻害を容易にし、さらに天然プラスミンと比較して出血性合併症の機会を減らすことができる。特定の態様において、完全な消化されていないフィブリン(オーゲン)(fibrin(ogen))の主要結合部位を保持するクリングル5の欠如は、循環するフィブリノーゲンの減少した枯渇を伴って(TAL6003)−プラスミンの使用を可能にすることができる;
vii)天然ヒトプラスミノーゲンのクリングルドメインに相同な単一のN末端クリングルドメイン(ここで、クリングルドメイン内の最後の4個のアミノ酸残基はV、P、QおよびCである)の存在は、セリンプロテアーゼドメインへの天然様連結(すなわち、ヒトプラスミノーゲンのクリングル5ドメインとセリンプロテアーゼドメインとの間の天然に存在するドメイン連結点と同様の連結)を提供する;そして
viii)組換え(TAL6003)−プラスミノーゲンの発現後に、そのN末端を切断し戻して(cleaved back)(例えば、活性化中に切断し戻して)天然様N末端を提供し得る。
他にそれに反して示されない限り、ポリペプチドの「ドメイン」および「領域」という用語は、本明細書において用いる場合に一般に同義語である。「クリングル」もしくは「セリンプロテアーゼ」などのようなよく認識された構造もしくは機能表示と一緒に列挙される場合、そのような用語はそのような表示に対応するポリペプチド構造と関連すると一般に認識されそして理解される少なくともいくつかの特徴(1つもしくは複数)に関連するポリペプチド特性を導入する。

本発明のポリヌクレオチドには、1個もしくはそれ以上のヌクレオチドを伴い得る置換、欠失および/もしくは付加を有するバリアントが包含される。バリアントは、コーディング領域、非コーディング領域もしくは両方において改変することができる。コーディング領域における改変は、保存的もしくは非保存的アミノ酸置換、欠失もしくは付加をもたらすことができる。これらの中で特に好ましいのは、(TAL6003)−プラスミン(オーゲン)タンパク質もしくはその一部の特性および活性を改変しない、サイレント置換、付加および欠失である。またこれに関して特に好ましいのは保存的置換である(以下を参照)。

プロモーターには、CMV前初期プロモーター、HSVチミジンキナーゼプロモーター、初期および後期SV40プロモーター、ラウス肉腫ウイルス(RSV)のもののようなレトロウイルスLTRのプロモーター、ならびにマウスメタロチオネイン−Iプロモーターのようなメタロチオネインプロモーターが包含される。
本発明のポリヌクレオチドには、本発明のポリペプチドのバリエーションおよび特定の例をコードするものが包含される。例えば、表現型の上でサイレントのアミノ酸置換を行う方法に関するガイダンスは、Bowie,J.U.et al.,“Deciphering the Message in Protein Sequences:Tolerance to Amino Acid Substitutions,”Science 247:1306−1310(1990)に提供され、ここで、著者等はタンパク質がアミノ酸置換に驚くほど寛容であることを示す。本発明の範囲内の置換のいくつでもそのような一般的原理の適用により得ることができるが、置換に関する特定のガイダンスには、クリングル1ドメインの構造および機能に関して本明細書に引用される参考文献を当業者は調べることができる。

結合型炭水化物鎖、N末端もしくはC末端終端のプロセシング、アミノ酸バックボーンへの化学部分の連結、N結合型もしくはO結合型炭水化物鎖の化学修飾、ならびに原核生物培養宿主細胞における(TAL6003)−プラスミノーゲンポリペプチドの発現に適応させたベクターおよび構築物の結果としてのN末端メチオニン残基の付加が包含される。ポリペプチドはまた、タンパク質の検出および単離を可能にするために酵素、蛍光、同位体もしくは親和性標識のような検出可能な標識で修飾することもできる。
(TAL6003)−プラスミン(オーゲン)は、両方とも引用することにより本明細書に組み込まれる、米国特許出願第10/143,112号;およびNovokhatny,V.,et al.,J.Thromb.Haemost.1(5):1034−41(2003)に記述される方法および組成物に従って治療用途に調合することができる。例えば、低pH(約2.5〜約4)、低緩衝能バッファーを(TAL6003)−プラスミンの調合に用いることができる。さらに、プラスミン、ミニ−プラスミンおよび/もしくはミクロ−プラスミンで実施されるような、当業者に既知である他の方法および調合を治療的投与用の本発明の(TAL6003)−プラスミンを調合するために用いることができる。
(TAL6003)−プラスミノーゲンのアミノ酸配列を配列番号:2に示す。(TAL6003)−プラスミノーゲンをコードするヌクレオチド配列を有するポリヌクレオチドをエシェリキア・コリ発現およびmRNA安定性のためにコドンを最適化して配列番号:1に示されるようなDNA配列を提供した。このポリヌクレオチドをエシェリキア・コリ発現ベクターpET24b(+)(Novagen;Madison,WI)のNdeIおよびBamH1部位にクローン化して細胞質タンパク質を生成せしめた。
(TAL6003)−プラスミノーゲンをコードするDNAを含んでなる発現ベクターをBL21(DE3)RIL(Stratagene,La Jolla,CA)、BL21(DE3)(遺伝子型:F−ompT hsdSB(rB −mB −)gal dcm(DE3))(EMB Biosciences,Inc.,San Diego,CA)およびBLR(DE3)(遺伝子型:F−ompT hsdSB(rB −mB −)gal dcm(DE3)Δ(srl−recA)306::Tn10(TetR))を包含する様々な細胞に形質転換し、そして1mMのIPTG(イソプロピル−ベータ−D−チオガラクトピラノシド)による誘導後のタンパク質過剰発現をSDS−PAGEにより分析した。発現概算は、振盪フラスコにおける1Lの細胞培養物当たり少なくとも約250mgであった。

(TAL6003)−プラスミノーゲンベクターを含有するエシェリキア・コリ細胞(例えば、BL21(DE3)RIL、BL21(DE3)もしくはBLR(DE3))の単一コロニーを用いて5mlのLB/カナマイシン(30μg/ml)に接種し、そして振盪器上で37℃で8時間インキュベーションした。その後で、新しい培地におけるさらなる培養用に50μlのアリコートを培養細菌懸濁液から取った。6mlの細菌培養物および250mlの培地で16時間後に該手順を繰り返した。培養物を振盪しながら37℃で〜1.0のOD600nmまで培養し、そしてIPTGを1mMの最終濃度になるように加えた。培養物をさらに5時間培養した。細胞を5,000xgで遠心分離により採取し、そして細胞ペレットを20mMのEDTAを含有する20mMのTris pH8.0に溶解し、そして−80℃で凍結させた。

精製された(TAL6003)−プラスミノーゲンは、還元(ジチオスレイトールで処理する)および非還元タンパク質のSDS−PAGE分析により35〜40kDa領域におけるバンドとして現れた。MALDI質量分析により決定されるその分子量は約38,140Daであり、それは予想される値に近い。
るその後の活性化および結果として起こる血栓溶解により評価するマイクロタイタープレートアッセイにおいてそのフィブリン結合特性を試験した。この目的のために、100μlの5mg/mlフィブリノーゲンをマイクロタイタープレートの各ウェルにおいてトロンビンで重合させた。様々な濃度の(TAL6003)−プラスミノーゲンをフィブリン血栓の上部に加え、そして37℃で1時間インキュベーションした。プレートをPBSで十分に洗浄し、その間にフィブリン血栓は依然として完全なままでありそしてウェルに結合している。洗浄した後に、tPAの0.1mg/ml溶液を各ウェルに加え、そしてプレートを37℃で2時間インキュベーションした。結果として、血栓のいくらかは完全に溶解され、そしていくらかは部分的に溶解され、一方、非常に少量の(TAL6003)−プラスミノーゲンを有するウェルおよびコントロールウェルは実質的に元のままであった。線維素溶解の程度は、1MのNaOHにおいて再構成した最初の血栓の残りの280nmの吸光度を測定することによりモニターした。吸光度値を(TAL6003)−プラスミノーゲン濃度の関数としてプロットした。
精製された(TAL6003)−プラスミノーゲン溶液へのSKの添加は、(TAL6003)−プラスミンへの(TAL6003)−プラスミノーゲンの転化をもたらす。タンパク質を2mg/mlに濃縮し、そして50%グリセロールで1:1に希釈して25%グリセロール中1mg/mlの溶液を生成せしめた。溶液を室温にし、そしてストレプトキナーゼを1:100のモル比のSK:(TAL6003)−プラスミノーゲンで加えた。反応物を撹拌せずに室温で4.5hrインキュベーションした。次に、0.5Mの最終濃度になるようにNaClを加えて反応を遅くした。SDS−PAGEによる活性化の分析により、90%の収率の活性化タンパク質が示された。

プラスミン基質D−Val−Leu−Lys−p−ニトロアニリド(S−2251)(DiaPharma,West Chester,OH)を用いて(TAL6003)−プラスミンのアミド分解活性を調べた。
(TAL6003)−プラスミンの線維素溶解効能は、以下の実験プロトコルを用いて血栓溶解アッセイのインビトロモデルにおいて試験した。
Claims (16)
- 配列番号2の配列に少なくとも95%同一であるポリペプチドであって、
a)天然ヒトプラスミノーゲンのクリングルドメインに対応する単一のN末端クリングルドメインであって、該クリングルドメイン内の最後の4個のアミノ酸配列がVPQCである、N末端クリングルドメイン;そして
b)天然ヒトプラスミノーゲンの対応するドメインに一致するC末端ドメイン活性化部位およびセリンプロテアーゼドメイン
を含み、固定化リシンに結合するポリペプチドをコードするヌクレオチド配列を含むポリヌクレオチド。 - コードされるポリペプチドが配列番号2に示される配列である、請求項1に記載のポリヌクレオチド。
- 前記ポリヌクレオチドのヌクレオチド配列が、配列番号1に示される配列又はその縮重バリアントである、請求項1に記載のポリヌクレオチド。
- 前記ポリペプチドが、ミニ−プラスミンのフィブリノーゲンに対する結合親和性より低いフィブリノーゲンに対する結合親和性を示す、請求項1に記載のポリヌクレオチド。
- 前記ポリペプチドが、ミニ−プラスミンの部分的に切断されたフィブリンに対する結合親和性より高い部分的に切断されたフィブリンに対する結合親和性を示す、請求項1に記載のポリヌクレオチド。
- 前記ポリペプチドが、参照ポリペプチドと比較して減少した免疫原性を有し、該参照ポリペプチドが前記ポリペプチドの一次アミノ酸配列と、参照ポリペプチドの単一のN末端クリングルドメインの最後の4個のアミノ酸配列がVPQCではない点を除き、同一である一次アミノ酸配列を有する、請求項1に記載のポリヌクレオチド。
- 前記ポリペプチドが、クリングルドメインとセリンプロテアーゼドメインとの間に位置するArg−Valペプチド結合のタンパク質分解性切断に少なくとも関与する活性化事象の後に機能的プラスミンへと活性化できる、請求項1に記載のポリヌクレオチド。
- 請求項1〜7のいずれか1項に記載のポリヌクレオチドによりコードされるポリペプチド。
- 前記ポリペプチドが、α2−アンチプラスミンによるミニプラスミンの線維素溶解活性の阻害の速度よりも、少なくとも5倍速い阻害の速度で、α2−アンチプラスミンにより阻害される線維素溶解活性を示す、請求項8に記載のポリペプチド。
- 前記ポリペプチドが、配列番号2で示されるアミノ酸配列を有する請求項8に記載のポリペプチド。
- 前記ポリペプチドが、配列番号2に示されるアミノ酸配列の位置85と同等の相対的位置にアルギニンを有する、請求項8に記載のポリペプチド。
- 請求項1のポリヌクレオチドを含んでなる発現ベクター。
- 請求項12に記載の発現ベクターを含んでなる細胞。
- 異なるN末端を有する1又は複数の組換えプラスミンポリペプチドを製造する方法であって、当該方法が以下の:
a)請求項8に記載のポリペプチドを提供し;
b)工程a)で提供されたポリペプチドを、1又は複数のペプチド結合を切断するために十分な条件下でプロテアーゼと接触させて、それにより異なるN−末端を有する1又は複数の組換えプラスミンポリペプチドを形成する
を含む、前記方法。 - 前記ポリペプチドが、配列番号2に示されるアミノ酸配列を有する、請求項14に記載の方法。
