Abstract
Description
本出願は、その全内容が参照として本明細書に組み入れられる、2002年10月11日に出願された米国特許出願第10/269,473号の一部継続出願である。
本発明は全般的に、イムノアッセイの分野、特にMHC対立遺伝子に対するペプチドの結合を検出および測定するイムノアッセイを用いることに関する。
クラスI組織適合性三元複合体は、非共有結合によって会合した三つの部分からなる。MHC重鎖と呼ばれる膜貫通タンパク質は、ほとんどが細胞表面に露出している。MHC重鎖の細胞表面ドメインはαヘリックスの二つのセグメントを含み、それらは二つの隆起を形成してそれらのあいだに溝が形成される。短いペプチドは、二つのαヘリックス間のこの溝に非共有結合によって結合し(「適合し」)、β-2ミクログロブリンの分子がMHC単量体の外側の二つのドメインに沿って非共有結合によって結合して、三元複合体を形成する。一つのMHCサブタイプ重鎖に非共有結合するペプチドは通常、もう一つのサブタイプに結合しないと考えられる。しかし、全てが同じタイプのβ-2ミクログロブリンに結合する。MHC分子は、体の細胞のほとんどによって合成され、示される。
本発明は、MHCクラスIおよびクラスII単量体を固体に固定した場合に、なおも溶液からMHC結合ペプチドを取り込んで、MHC複合体を形成することができるという発見に基づくものである。
本発明は全般的に、推定のMHC結合ペプチドに対するMHCクラスIおよびクラスII単量体、特に表面上に固定されたMHCクラスIおよびクラスII単量体の結合親和性の検出および測定に向けられるイムノアッセイに関する。固体表面に固定されたMHC単量体および改変MHC単量体はなおも、溶液からの必要な結合親和性を有するMHC結合ペプチドに結合することによってMHC複合体を形成するように再生することができる。その上、そのような結合は、そのような再生または再構成されたMHC単量体を含むMHC複合体には特異的に結合するが、MHC複合体の解離成分には結合しない立体配座依存的モノクローナル抗体を利用するような、イムノアッセイフォーマットにおいて検出することができることは本発明の発見である。
ヒトの第6染色体に存在するクラスI MHCは、三つの座、すなわちHLA-A、HLA-B、およびHLA-Cを有する。最初の二つの座は、アロ抗原をコードする多数の対立遺伝子を有する。これらは、44 kD重鎖サブユニットと、12 kDβ2ミクログロブリンサブユニットからなり、これらは全ての抗原特異性にとって共通であることが判明している。例えば、可溶性HLA-A2は、Turner, M.J.ら、J. Biol. Chem.(1977)252:7555〜7567によって記述されるように、ホモ接合ヒトリンパ芽球性細胞株J-Yから細胞質膜のパパイン消化後に精製することができる。パパインは、膜貫通領域の近位で44 kD重鎖を切断し、α1、α2、およびα3ドメインおよびβ2-ミクログロブリンを含む分子を生じる。
抗原提示細胞(APCs)の表面上のMHC糖タンパク質による抗原の提示は、抗原性タンパク質のより小さいペプチド単位へのタンパク質分解後に起こると考えられる。抗原性タンパク質内のこれらのより小さいセグメントの位置は、経験的に決定することができる。クラスI MHC結合ペプチドは、長さが約8〜約10残基、おそらく約8〜約11残基、または約8〜約12残基であると考えられる。クラスII MHC結合ペプチドは、アミノ酸の長さが約10個〜約30個、例えば約12個〜約24個であると考えられる。これらのペプチドは、アグレトープ(MHC分子によって認識される)およびエピトープ(T細胞上のT細胞受容体によって認識される)の双方を含む。エピトープは、抗原特異的T細胞受容体によって認識されるアミノ酸約5個〜6個の連続または非連続配列である。アグレトープは、ペプチドとMHCクラスI糖タンパク質との結合に関与する隣接または非隣接配列である。本発明は、そのような断片がMHC単量体または改変MHC単量体との三元複合体に組み入れられうるか否かを決定するために推定のMHC結合ペプチドを評価するためのシステム、キット、およびアッセイを提供する。
正確に折り畳まれたHLA重鎖単量体の検出
本実験は、固体支持体に結合させたMHC単量体が、三元複合体を形成するように再構成できること、および立体配座依存的抗MHCモノクローナル抗体によって認識され、特異的に結合されうることを証明する。言い換えれば、固体支持体に結合したMHC単量体は正確に折り畳まれて、MHC結合ペプチドに結合するであろう。下記の表1は、ペプチド結合の際に正確に折り畳まれたHLA単量体を検出する主要な段階を要約している(図1も参照のこと)。
抗HLA-FITC抗体の較正
本実施例において、BSA-ビオチン-アビジンコーティング96ウェルマイクロタイタープレートを蛍光測定アッセイに用いるために調製した。結合ペプチドMart-1 26-35Lとの三元複合体におけるHLA-A2m単量体を、様々な濃度の抗HLA-ABC-FITCまたは濃度0、0.25、0.5、1、2、および4 μg/mlの抗HLA-FITCモノクローナル抗体と共にインキュベートした。特に、それぞれの抗体濃度に関して、HLA単量体を濃度0、0.0078、0.0156、0.3125、0.0625、0.125、0.25、および0.5 μg/mlで加えた。
抗HLA-FITCモノクローナル抗体の特異性
A.立体配座特異性
抗HLA-クラスI-FITC抗体から産生されたシグナルが、強調されたHLA単量体の解離の程度の関数として異なるか否かを決定するために実験を計画した。実験のために用いられる特定のHLA単量体は、三元複合体において結合ペプチドMart1 27-35を含むHLA重鎖単量体HLA-A*0201であった。
表2に示すように、ヒト対立遺伝子A2、A3、A11、B7、B8に関する異なるHLA重鎖単量体と共に一つのマウス対立遺伝子Kdを抗HLA-クラスI-FITC抗体と共にインキュベートした。
濃度5 μg/mlのMart1 27-35ペプチドとの三元複合体におけるビオチン結合MHC単量体を、アビジンコーティングプレートに結合させた。プレートを糖含有緩衝液によって4℃〜8℃で一晩飽和した後、プレートを30℃および湿度19%で一晩乾燥させた。プレートをこれらの条件で乾燥させた後、HLA単量体は、三元複合体から解離したことが判明した。したがって、結合アッセイにおいてプレートを用いるための調製の際に低いpHで単量体からMHC結合ペプチドを剥離させる必要がなかった。

A.試薬
特に明記していなければ、精製化学薬品をメルク(Merck、ダームスタット、ドイツ)およびカルロエルバ(Carlo Erba、ロデノ、イタリア)から得た。ビオチン化BSAと共にアビジンは、イムノテック(Immunotech、マルセイユ、フランス)から得た。LUMITRAC-600白色96ウェルマイクロタイタープレートは、グライナー(Greiner、PN 655074 LUMITRAC 600(フリッケンハウゼン、ドイツ))から得た。SA-PEと共にHLA-A*0301/EBV HLA重鎖は、イムノミクス(Immunomics、サンディエゴ、カリフォルニア州)から得た。FITCを結合させた抗HLA-クラスI-モノクローナル抗体(クローン:B9.12.1)は、イムノテック抗体製造サービス(Antibody Manufacturing Services of Immunotech)から得た。パート番号:IM1838。この抗体は、マウスIgG2aモノクローナル抗体である。
白色96ウェルマイクロタイタープレートの各ウェルを5 μg/mlビオチン化BSAのPBS溶液200μlによってコーティングして、プレートを4℃で16時間インキュベートした。プレートを洗浄して、5 μg/mlアビジン溶液200μl/ウェルを加えた。次に、プレートを4℃でさらに16時間インキュベートした。その後、プレートを洗浄して、ブロッキング乾燥溶液を加えた。プレートを再度さらに16時間インキュベートした。その後、溶液を捨てて、プレートをペーパータオルの上でたたいて水分を除去した。次に、プレートを特殊な乾燥室に24時間入れた。その後、プレートを使用するまで個々に自動ロックバッグに入れた。
アッセイ技法は以下の通りであった。HLA単量体を三元複合体において0.25 μg/mlで含み、トリス10 mM、NaCl 150 mM、EDTA 0.5 mM、NaN3 0.1%、BSA 0.2%において希釈した各試料200μl/ウェルを、アビジンコーティングプレートのウェルにローディングして、軌道を描くシェーカーにおいて室温の暗所で1時間インキュベートした。次に、ウェルを、0.05%TWEEN 80(登録商標)を含む9 g/l NaCl溶液300μlを用いて自動洗浄機(SLT、ザルツブルグ、オーストリア)によって3回すすいだ。次に、2 μg/ml FITC-結合抗HLA-クラスI抗体200 μl/ウェルを加えた。プレートを軌道を描くシェーカーにおいて室温の暗所で45分間インキュベートして、3回洗浄し、トリス10 mM、NaCl 150 mM、EDTA 0.5 mM、NaN3 0.1%、BSA 0.2%を200 μl/ウェル加えた。FITC蛍光は、これらのパラメータに従って、パーキンエルマーLS-50B蛍光光度計によって測定した:

様々な濃度のHLA-A*0201/Mart1再構成単量体を、抗HLA-クラスI-FITC mAbの様々な濃度と共にインキュベートした。図6に示すように、全てのHLA重鎖単量体濃度に関して1〜2 μg/mlの抗HLA-クラスI mAb濃度で平衡に達した。
三元複合体におけるHLA単量体に関する抗HLA-クラスI-FITC抗体の特異性を異なるヒト対立遺伝子に対して調べた。以下のHLA単量体/ペプチド三元複合体のそれぞれに関して上記のように用量反応曲線を調製した。
− HLA-A*0201/Mart1 2635L
− HLA-A*0301/EBV
− HLA-B*0702/HIV
− HLA-B*0801/HIV
− HLA-A*1101/EBV
− H-2Db/HA1
ペプチド-MHC オフ率の測定
有効なCD8+ T細胞反応を得るためには、クラスI MHC分子は、長期間にわたって十分な量で多様な配列の多くのペプチドに結合しなければならない。多くの腫瘍細胞は、抗原性ペプチドがそれらを提示するクラスI MHC分子に対して十分に結合しないために、免疫応答を逃れるように思われる。ペプチドがMHC分子に対して効率よく結合しない場合、循環中のT細胞は、MHC三元複合体を認識せず、それらを提示する細胞は消失しないであろう。
Claims (67)
- 表面に一つまたはそれ以上のMHC単量体または改変MHC単量体が結合し、該単量体が、溶液から適したMHC結合ペプチドを組み入れる、固体表面を含むシステム。
- 単量体がMHCクラスIであって、から単量体が再構成条件でMHC結合ペプチドを組み入れる、請求項1記載のシステム。
- 固体表面がビーズである、請求項1記載のシステム。
- 固体表面がマイクロタイタープレートに存在する、請求項1記載のシステム。
- 固体表面がハイスループットシステムでのスクリーニングに適している、請求項1記載のシステム。
- 固体表面に対する単量体の結合が可逆的である、請求項1記載のシステム。
- 固体表面に対する単量体の結合が切断可能である、請求項1記載のシステム。
- 固体表面が第一の結合リガンドによってコーティングされ、単量体のC末端に第二の結合リガンドが提供され、第一のリガンドが第二のリガンドに特異的に結合する、請求項1記載のシステム。
- 第一の結合リガンドが、アビジン、ストレプトアビジン、ニュートラアビジン(neutravidin)、ストレプタクチン(StrepTactin)および単量体アビジンから選択され、第二の結合リガンドがビオチンである、請求項7記載のシステム。
- 第二の結合リガンドがC末端を通して単量体に結合している、請求項8記載のシステム。
- 単量体がMHCクラスI単量体であって、かつ単量体が変性条件で変性し、再生条件でMHC結合ペプチドを組み入れるように再構成される、請求項1記載のシステム。
- 変性条件がpH約2〜約4を含む、請求項11記載のシステム。
- 再構成単量体には存在するが変性単量体には存在しない立体配座エピトープに対して特異的に結合する抗MHC抗体をさらに含む、請求項1記載のシステム。
- 再構成条件がpH約7〜約8.5を含む、請求項1記載のシステム。
- β2-ミクログロブリンをさらに含む、請求項1記載のシステム。
- 単量体がHLAクラスIである、請求項14記載のシステム。
- 再構成された単量体には特異的に結合するが、変性単量体には結合しない、抗MHCクラスIモノクローナル抗体をさらに含む、請求項15記載のシステム。
- システムがβ2-ミクログロブリンおよび約8個〜約12個のアミノ酸の適したHLA結合ペプチドをさらに含み、再構成された単量体が再構成条件でβ2-ミクログロブリンおよび適したペプチドに結合する、請求項17記載のシステム。
- モノクローナル抗体がハイブリドーマB9.12.1によって産生される、請求項17記載のシステム。
- 単量体がHLAクラスIIである、請求項1記載のシステム。
- MHC結合ペプチドに結合した単量体と、MHC結合ペプチドを欠損する単量体とを区別するモノクローナル抗体をさらに含む、請求項20記載のシステム。
- 単量体がHLAクラスIIであって、システムが約10〜約30個のアミノ酸の適したHLA結合ペプチドを含み、再構成された単量体が再構成条件で適したペプチドに結合する、請求項21記載のシステム。
- 単量体によってコーティングされた固体表面が乾燥型である、請求項1、17、または22記載のシステム。
- 請求項1、17、または22記載のシステムを含むキット。
- 説明書をさらに含む、請求項23記載のキット。
- MHC単量体が再構成型で結合する対照ペプチドをさらに含む、請求項24記載のキット。
- 以下の段階を含む、MHCクラスI単量体または改変MHCクラスI単量体と、その推定のMHC結合ペプチドとの結合を決定する方法:
複数のMHC単量体または改変MHC単量体が結合した固体表面を推定のMHC結合ペプチドの存在下および非存在下で再構成条件でインキュベートする段階であって、該単量体が変性され、再構成条件で適したMHC結合ペプチドを含む複合体を形成するように再構成される段階、ならびに
MHC複合体に結合するが、MHC複合体の解離した成分には結合しないモノクローナル抗体に接触させた後のMHC単量体に対する結合を決定して、該抗体の結合により、単量体と推定のMHC結合ペプチドとが結合していることが示される段階。 - 変性条件がpH約2〜約4の範囲を含む、請求項27記載の方法。
- モノクローナル抗体の存在下で再構成条件で単量体に関する標準的なMHC結合ペプチドと共に単量体を別々にインキュベートすることをさらに含み、測定段階が、標準的なペプチドによって引き起こされた抗体の結合を、推定のMHC結合ペプチドによって引き起こされた抗体の結合と比較することをさらに含む、請求項27記載の方法。
- 単量体がHLAクラスIであって、モノクローナル抗体が抗MHC-クラスI抗体であって、かつ再構成条件が、単量体の再構成のために十分なβ2-ミクログロブリンが存在することを含む、請求項29記載の方法。
- 単量体がHLAサブクラスA、B、またはCである、請求項30記載の方法。
- 以下の段階を含む、MHCクラスII単量体または改変MHCクラスII単量体と、その推定のMHC結合ペプチドとの結合を決定する方法:
推定のMHC結合ペプチドの存在下および非存在下で複数のMHC単量体または改変MHC単量体が結合した固体表面を、適したペプチドローディング条件でインキュベートして、適したMHC結合ペプチドを含む複合体を形成する段階、ならびに
MHC単量体と推定のMHC結合ペプチドとの結合を決定する段階。 - 単量体がHLAクラスIIであって、決定段階が、MHC単量体と推定のMHC結合ペプチドとの結合を決定するモノクローナル抗体に複合体を接触させることを含む、請求項32記載の方法。
- 抗体が、MHC結合ペプチドに結合した単量体と、MHC結合ペプチドに結合していない単量体とを区別する、請求項32記載の方法。
- 適したペプチドローディング条件がpH約4〜約8の範囲を含む、請求項32記載の方法。
- 適した条件で、単量体と、単量体に関する標準的なMHC結合ペプチドとを別々にインキュベートする段階をさらに含み、決定段階が、標準的なペプチドの結合を、推定のMHC結合ペプチドによって引き起こされた結合と比較することを含む、請求項32記載の方法。
- 単量体がHLAサブクラスD、DR、DP、またはDQである、請求項32記載の方法。

固体表面に結合した少なくとも一つの変性MHC単量体または改変MHC単量体を、推定のMHC結合ペプチド、および対応する再構成MHC単量体を含む第一の複合体における立体配座エピトープに特異的に結合するが、MHC複合体の解離成分には結合しないモノクローナル抗体と共にインキュベートする段階であって、インキュベーションが再構成条件で行われる段階;ならびに
推定のMHC結合ペプチドを含むMHC複合体に対するモノクローナル抗体の結合を、単量体と既知のMHC結合ペプチドとを含む対応する複合体に対するモノクローナル抗体の結合と比較する段階。 - 再構成条件が温度約4℃〜約37℃の範囲を含む、請求項47記載の方法。
- 再構成条件が温度約4℃〜約8℃の範囲を含む、請求項47記載の方法。
- 再構成条件がpH約7〜約8.5の範囲を含む、請求項47記載の方法。
- 再構成条件が適した再構成緩衝液が存在することを含む、請求項47記載の方法。
- 単量体が、再構成条件でβ2-ミクログロブリンにさらに結合し、かつモノクローナル抗体が抗MHCクラスIモノクローナル抗体である、請求項47記載の方法。
- 単量体が、HLA-A、HLA-B、およびHLA-Cから選択される、請求項51記載の方法。
- モノクローナル抗体がハイブリドーマB9.12.1によって産生される、請求項53記載の方法。
- 以下の段階を含む、推定のMHC結合ペプチドに関するMHCクラスII単量体または改変MHCクラスII単量体の結合親和性の程度を決定する方法:
固体表面に結合させた少なくとも一つの変性MHCクラスII単量体または改変MHCクラスII単量体を、推定のMHC結合ペプチド、および単量体を含む第一の複合体におけるエピトープに特異的に結合するが、MHC複合体の解離成分には結合しないモノクローナル抗体と共に、適した結合条件でインキュベートする段階;ならびに
推定のMHC結合ペプチドを含むMHC複合体に対するモノクローナル抗体の結合を、単量体と既知のMHC結合ペプチドとを含む対応する複合体に対するモノクローナル抗体の結合と比較して、結合の差によって、推定のMHC結合ペプチドに関する単量体の結合親和性の相対度が示される段階。 - 結合条件がpH約4〜約8の範囲を含む、請求項55記載の方法。
- インキュベート段階が約12時間〜約48時間の間である、請求項47または55に記載の方法。
- 抗体が検出可能な標識と共に提供され、かつ結合の比較が、抗体の結合に起因する検出可能な標識によって産生されるそれぞれのシグナルの差を検出することを含む、請求項47または55に記載の方法。
- 抗体が蛍光標識によって標識され、かつ結合の比較が、抗体の結合に起因するそれぞれの蛍光の差を検出することを含む、請求項58記載の方法。
- 蛍光標識がフルオレセインイソチオシアネート(FITC)である、請求項59記載の方法。
- 固体表面がマイクロタイタープレートのウェルまたはビーズであり、かつ検出段階が、蛍光光度計によって蛍光を読み取ることを含む、請求項47または55に記載の方法。
- 検出段階が、ハイスループットスキャニングを用いて蛍光を検出することをさらに含む、請求項61記載の方法。
- 単量体がHLA-D、DR、DP、またはDQサブクラスから選択される、請求項55記載の方法。
- 単量体がキメラである、請求項47または55に記載の方法。
- 推定のMHC結合ペプチドを含むMHC複合体と既知のペプチドとを、推定のMHC結合ペプチドの相対的解離速度を示すために十分な期間、解離-試験温度を含む条件でインキュベートした後に、結合の差を比較する、請求項47または55に記載の方法。
- 解離-試験温度が約4℃〜約37℃の範囲である、請求項65記載の方法。
