Abstract
Description
[発明の概要]
本発明は、LbLコーティングを医療用デバイス、好ましくは眼用デバイス、より好ましくはコンタクトレンズに適用する方法であって、
(a)医療用デバイスを第一のポリイオン性材料を含有する第一のコーティング溶液と接触させて、第一のポリイオン性材料の最内層を医療用デバイス上に非共有結合的に適用する工程であって、第一のコーティング溶液中の第一のポリイオン性材料の濃度がLbLコーティングの親水性を増大させるのに十分に高く、また医療用デバイスが表面改質されていない工程、
(b)第一のポリイオン性材料の最内層を有する医療用デバイスを、第一のポリイオン性材料の電荷と反対の電荷を有する第二のポリイオン性材料を含有する第二のコーティング溶液と接触させて、第一のポリイオン性材料の最内層および最内層の上部に第二のポリイオン性材料の一層からなる第一の多価電解質二重層を形成する工程、
(c)第一の多価電解質二重層を有する医療用デバイスを第一のポリイオン性材料または第二のポリイオン性材料の電荷と反対の電荷を有する第三のポリイオン性材料を含有する第三のコーティング溶液と接触させて、第一の多価電解質二重層の上部に、第一のポリイオン性材料または第三のポリイオン性材料の層を形成する工程、
(d)第一の多価電解質二重層および第一の多価電解質二重層の上部に第一のポリイオン性材料または第三のポリイオン性材料の一層を有する医療用デバイスを、第二のポリイオン性材料または第一もしくは第三のポリイオン性材料の電荷と反対の電荷を有する第四のポリイオン性材料を含有する第四のコーティング溶液と接触させて、第一のポリイオン性材料または第三のポリイオン性材料の層および第二のポリイオン性材料の一層からなる第二の多価電解質二重層を形成する工程、ならびに、場合により、
(e)1以上のさらなる多価電解質二重層を構築するために、工程(c)〜(d)を1以上の回数繰り返す工程、
を含む方法を提供する。
次に、本発明の実施態様を詳細に参照し、その一つ以上の例を以下に述べる。各例は、本発明の説明として提供するものであり、本発明を限定するものではない。実際、本発明の範囲または精神を逸することなく、種々の変形および変更を本発明に加えることができることが当業者には明らかであろう。たとえば、一つの実施態様の一部として例示または記載される特徴を、他の実施態様で使用してさらなる実施態様を得ることもできる。したがって、本発明は、添付の特許請求の範囲およびその均等物の範囲に入るような変形および変更を包含することを意図する。本発明の他の目的、特徴および態様は、以下の詳細な説明に開示されるか、あるいは以下の詳細な説明から自明である。本説明が例示的な実施態様の記載にすぎず、本発明のより広い態様を限定するものとして意図するものではないということが、当業者に理解されよう。
(i)場合により変性剤単位を含む、ポリアリルアミン(PAH)ホモ−またはコポリマー;
(ii)ポリエチレンイミン(PEI);
(iii)場合により変性剤単位を含む、ポリビニルアミンホモ−またはコポリマー;
(iv)ポリ(ビニルベンジル−トリ−C1〜C4−アルキルアンモニウム塩)、例えば、ポリ(ビニルベンジル−トリ−メチルアンモニウムクロリド);
(v)脂肪族もしくは芳香脂肪族二ハロゲン化物および脂肪族N,N,N’,N’−テトラ−C1〜C4−アルキル−アルキレンジアミンのポリマー、例えば、(a)1,3−二塩化もしくは二臭化プロピレンまたは二塩化もしくは二臭化p−キシリレンと(b)N,N,N’,N’−テトラメチル−1,4−テトラメチレンジアミンのポリマー;
(vi)ポリ(ビニルピリジン)またはポリ(ビニルピリジニウム塩)のホモ−またはコポリマー;
(vii)式:
の単位を含むポリ(ハロゲン化N,N−ジアリル−N,N−ジ−C1〜C4−アルキル−アンモニウム);
(viii)四級化アクリル酸またはメタクリル酸ジ−C1〜C4−アルキル−アミノエチルのホモ−またはコポリマー、例えば、ポリ(塩化2−ヒドロキシ−3−メタクリロイルプロピルトリメチルアンモニウム)などのポリ(2−ヒドロキシ−3−メタクリロイルプロピルトリ−C1〜C2−アルキルアンモニウム塩)ホモポリマー、または四級化ポリ(メタクリル酸2−ジメチルアミノエチル)もしくは四級化ポリ(ビニルピロリドン−co−メタクリル酸2−ジメチルアミノエチル);
(ix)EP−A−456,467に開示されているようなPOLYQUAD(登録商標);または
(x)ポリアミノアミド(PAMAM)、例えば、直鎖PAMAM、またはアミノ基を末端に有するStarbust(商標)PAMAMデンドリマー(アルドリッチ)などのPAMAMデンドリマーである。

コーティングされたコンタクトレンズのCoFの測定
CoFは、コンタクトレンズの眼の上での動きの容易さを測定するパラメーターの一つである。高いCoFは、眼上皮を機械的に損傷する可能性を増大させる。コンタクトレンズのCoFは、以下のように、スレッドオンブロック(sled-on-block)型摩擦テスターにより測定することができる。ある負荷(例えば、約2.0g)の下に、コンタクトレンズを、生物学的に適切な基材に対して、所定の速度で、前後に滑らせ、垂直力(N)および切線分力(FT)を測定する。コンタクトレンズのCoFは、式μ=FT/Nに基づいて計算される。
コーティングされたコンタクトレンズの接触角の測定
コンタクトレンズの平均接触角(付着液滴)は、マサチューセッツ州、ボストンにあるAST, Inc.からのVCA2500XE接触角測定装置を用いて測定した。この装置は、前進もしくは後退接触角または付着(静的)接触角を測定することが可能である。測定は、好ましくは、完全に水和された材料上で行われる。
ポリアクリル酸(PAA)溶液:PolyScience社からの、分子量約90,000を有するポリアクリル酸の二つの溶液を、適量の材料を水に溶解して調製し、所望の濃度のPAA溶液とした。PAA濃度は、PAA中の繰返し単位に基づいて計算した。溶解した後、ポリアニオン性のPAA溶液のpHは、pHが約2.5になるまで、1N塩酸を加えて調整した。
PAAおよびPAH溶液:PAAおよびPAH溶液は、実施例3に記載したように調製した。
コーティングC:PAA/PAHの4.5層の二重層を有するコーティングを、フルオロシロキサンヒドロゲル材料、ロトラフィルコンA(CIBA vision)製のソフトコンタクトレンズ上に形成した。コンタクトレンズは次のようにしてコーティングした:(a)手で、PAA溶液(0.0001M,pH2.5)に30分間浸漬して、レンズ上にコーティングの最内層を形成し;(b)水すすぎをせずに、手で、PAH溶液(0.0001M,pH2.5)に5分間浸漬し;(c)水すすぎをせずに、手で、PAA溶液(0.0001M,pH2.5)に5分間浸漬し;(d)工程(b)〜(c)を3回繰り返して、レンズ上に4.5層(すなわち、PAA/PAH/PAA/PAH/PAA/PAH/PAA/PAH/PAA)を構築した。コーティングされたレンズの各々を、リン酸緩衝塩溶液(PBS)(pH約7.2)で満たしたガラス瓶に入れて密封し、オートクレーブ処理した。オートクレーブ処理後、コーティングされたレンズを含む瓶は、レンズの特性評価まで開けなかった。
Claims (14)
- 生体適合性LbLコーティングを医療用デバイスに適用する方法であって、
(a)前記医療用デバイスを第一のポリイオン性材料を含有する第一のコーティング溶液と接触させて、第一のポリイオン性材料の最内層を前記医療用デバイス上に非共有結合的に適用する工程であって、第一のコーティング溶液中の第一のポリイオン性材料の濃度がLbLコーティングの親水性を増大させるのに十分に高く、また第一のコーティング溶液との接触の前に、前記医療用デバイスが表面改質されていない工程、
(b)第一のポリイオン性材料の最内層を有する前記医療用デバイスを、第一のポリイオン性材料の電荷と反対の電荷を有する第二のポリイオン性材料を含有する第二のコーティング溶液と接触させて、第一のポリイオン性材料の最内層および最内層の上部に第二のポリイオン性材料の一層からなる第一の多価電解質二重層を形成する工程、
(c)第一の多価電解質二重層を有する前記医療用デバイスを、第一のポリイオン性材料または第二のポリイオン性材料の電荷と反対の電荷を有する第三のポリイオン性材料を含有する第三のコーティング溶液と接触させて、第一の多価電解質二重層の上部に第一のポリイオン性材料または第三のポリイオン性材料の層を形成する工程、
(d)第一の多価電解質二重層および第一の多価電解質二重層の上部に第一のポリイオン性材料または第三のポリイオン性材料の一層を有する前記医療用デバイスを、第二のポリイオン性材料または第一もしくは第三のポリイオン性材料の電荷と反対の電荷を有する第四のポリイオン性材料を含有する第四のコーティング溶液と接触させて、第一のポリイオン性材料または第三のポリイオン性材料の層および第二のポリイオン性材料の一層からなる第二の多価電解質二重層を形成する工程、ならびに、場合により、
(e)さらなる多価電解質二重層を構築するために、工程(c)および(d)を何回か繰り返す工程、
を含む方法。 - 工程(a)と(b)の間、工程(b)と(c)の間、工程(c)の間、工程(d)と(c)の間に少なくとも一つのすすぎ工程を含み、すすぎ工程が前記医療用デバイスをすすぎ溶液と接触させることにより行われる、請求項1の方法。
- 工程(a)と(b)の間、工程(b)と(c)の間、工程(c)の間、工程(d)と(c)の間にすすぎ工程を含まない、請求項1の方法。
- 前記接触の少なくとも一つが、医療用デバイスに溶液を吹き付けることにより生じる、請求項1の方法。
- 前記接触の少なくとも一つが、医療用デバイスをコーティング溶液に浸漬させることにより生じる、請求項1の方法。
- 前記方法が、(a)〜(d)の工程を3〜20回繰り返すことを含む、請求項1の方法。
- 第一のコーティング溶液中の第一のポリイオン性材料の濃度が、第二、第三または第四のコーティング溶液中の第一、第二、第三または第四のポリイオン性材料の濃度の少なくとも3倍である、請求項1の方法。
- 第一のコーティング溶液中の第一のポリイオン性材料の濃度が、第二、第三または第四のコーティング溶液中の第一、第二、第三または第四のポリイオン性材料の濃度の10〜25倍である、請求項7の方法。
- 第三のコーティング溶液が第一のポリイオン性材料を含有する、請求項7の方法。
- 第三のコーティング溶液が第三のポリイオン性材料を含有する、請求項7の方法。
- 第四のコーティング溶液が第二のポリイオン性材料を含有する、請求項7の方法。
- 第四のコーティング溶液が第四のポリイオン性材料を含有する、請求項7の方法。
- 前記医療用デバイス上への生体適合性LbLコーティングの形成において、少なくとも三種の異なるポリイオン性材料を使用する、請求項7の方法。
- 生体適合性LbLコーティングが潤滑性のあるコーティング材料またはポリクォートの少なくとも一層を含み、潤滑性のあるコーティング材料が、PAMAMデンドリマー、PAAm−co−PAA、PVP−co−PAA、グリコサミノグリカン、フコイダン、ポリアスパラギン酸、ポリグルタミン酸、カルボキシメチルセルロース、カルボキシメチルデキストラン、アルギネート、ペクチン、ゲラン、カルボキシアルキルキチン、カルボキシメチルキトサン、硫酸化多糖、糖タンパク質およびアミノアルキル化多糖からなる群より選択される、請求項7の方法。
