Abstract
Description
(1.動物)
野生型マウス(c57/Black 6)およびt−PA欠損マウス(t−PA−/−マウス)(c57/Black 6)(Carmelietら、1994)は、Peter Carmeliet博士,Leuven,Belgiumによって提供された。
t−PAまたはDSPAα1のいずれかの注入後の、脳組織におけるタンパク質分解活性の評価を、電気泳動分析によって実施した(Granelli−PipernoおよびReich,1974)。7日間の期間にわたる海馬内への注入後に、マウスを麻酔し、次いで、心臓を通して(transcardially)PBSで灌流し、そして脳を取り出した。海馬領域を取り出し、エッペンドルフ管に移し、そしてプロテアーゼインヒビターを含まない等体積(w/v)(約30〜50μm)の0.5%NP−40溶解緩衝液(0.5% NP−40、10mM Tris−HCl(pH7.4)、10mM NaCL、3mM mgCl2、1mm EDTA)中でインキュベートした。脳抽出物を、携帯型ガラスホモジナイザでホモジナイズし、そして氷上で30分間維持した。次いで、サンプルを遠心分離し、そして上清を除去した。存在するタンパク質の量を決定した(Bio−Rad試薬)
(3.プロテアーゼのザイモグラフィ分析)
サンプルおよび脳組織抽出物におけるタンパク質分解活性を、Granelli,PipernoおよびReich(1974)の方法に従って、電気泳動分析によって決定した。組換えタンパク質(100nMまで)または脳組織抽出物(20μg)を含むサンプルを、非還元条件下で(10%)SDS−PAGEに供した。ゲルをプレートから取り出し、1% Triton X 100中で2時間洗浄し、次いで、重合したフィブリノゲンおよびプラスミノゲンを含有するアガロースゲル上に重ねた(Granelli,PipernoおよびReich,1974)。タンパク質分解されたゾーンが現れるまで、加湿チャンバ内でゲルを37℃でインキュベートする。
カイニン酸損傷モデルは、Tsirkaら(1995)の研究に基づいた。動物に、アトロピン(4mg/kg)を腹腔内(i.p.)に注射し、次いで、ペントバルビタールナトリウム(70mg/kg)のi.p.注射で麻酔した。その後、マウスを、定位フレーム中に配置し、100μlのPBSまたは組換えヒトt−PA(0.12mg/ml,1.85μM)またはDSPAα1(1.85μM)のいずれかを含むミクロ浸透圧ポンプ(Alzetモデル 1007D、Alzet CA、USA)を、肩甲骨間に皮下移植した。これらのポンプを、滅菌チューブを介して脳カニューレに連結し、そしてバー開口部を通って挿入し、正中栓付近に液体を導入するために、座標ブレグマ−2.5mm、座標中外側0.5mmおよび座標背腹側1.6mmで、頭蓋を通した。このカニューレを、所望の位置で固定し、そしてこれらのポンプを、それぞれの溶液を時速0.5μlで合計7日間注入させた。
KA注射5日後、これらの動物を麻酔し、そして経噴門的に30mlのPBSで還流し、続いて70mlの4%パラホルムアルデヒド溶液で還流し、同じ固定液中で後固定し、続いて30%ショ糖中でさらに24時間インキュベーションした。脳の冠状切片(40μm)を、次いで、凍結ミクロトーム上で切断し、そしてチオニン(BDH、Australia)で対比染色するかまたは以下に記載されるように、免疫組織化学試験のために処理した。
海馬のCA1−CA3亜領域内でのニューロン喪失の定量を、以前(Tsirkaら,1995;Tsirkaら,1996)に記載されたとおりに行った。全ての処置群からの背側海馬の連続した5つの部分を、その部分が実際にCA注射および損傷領域の場所を含むことに注意しながら調製した。これらの切片の海馬の亜領域(CA1〜CA3)を、海馬のカメラルシダーの図によってトレースした。亜領域全体の長さを、同じ倍率でトレースした1mm標準への比較によって測定した。生存可能な錐体ニューロンを有する(正常な形態を有する)組織の長さおよびニューロンのない(細胞が存在せず、チオニン染色もない)組織の長さを決定した。これらの長さは、各海馬亜領域についてのインタクトなニューロンおよびニューロン喪失を表す。これらの長さを切片毎に平均し、そして標準偏差を決定した。
野生型マウス(c57/Black 6)に麻酔し、そして定位フレーム内に配置した(上記を参照のこと)。次いで、マウスに、単独で、または46μM rt−PAもしくは46μM DSPAα1のいずれかと組み合わせて注射した、50nmol NMDAの片側注射を左側の層に受けさせた。コントロールとして、t−PAおよびDSPAもまた(いずれも、46μMの濃度にて)単独で注射した。この注射の座標は以下の通りであった:ブレグマ−0.4mm、側方中間(midiolateral)2.0mmおよび背腹側2.5mm。溶液(全ての処置について1μl総体積)を、0.2μl/分の速度で5分間の期間にわたって移し、そして液体の逆流を最小にするために、注射後、針をさらに2分間適所に配置した。24時間後、これらのマウスに麻酔をかけ、そして30ml PBS、続いて70mlの4%パラホルムアルデヒド溶液で、心臓を経て灌流し、同じ固定剤中で24時間にわたって事後固定し、続いて30%スクロース中でさらに24時間にわたってインキュベートした。次いで、脳を、凍結ミクロトームで切断(40μm)し、そしてゼラチンをコーティングしたスライドガラスに載せた。
線条体損傷体積の定量を、Callawayら(2000)によって記載された方法を用いて行った。損傷を受けた領域にまたがる10個の連続する冠状縫合切片を調製した。この損傷を受けた領域を、Callaway法を用いて可視化し、そして損傷体積を、マイクロコンピュータ画像化デバイス(MCID,Imaging Research Inc.,Brock University,Ontario,Canada)の使用によって定量した。
免疫組織化学を、標準的な方法論を使用して行った。結腸切片を、3% H2O2および10%メタノールの溶液に5分間浸漬し、続いて5%正常ヤギ血清中で60分間インキュベートした。この切片を、星状細胞検出のための抗GFAP抗体(1:1000;Dako,Carpinteria,CA,USA)、小膠細胞の検出のための抗MAC−1抗体(1:1000,Serotec,Raleigh,NC,USA)またはポリオクローナル抗DSPA抗体(Schering AG,Berlin)のいずれかとともに、一晩インキュベートした。すすいだ後、この切片を適切なビオチン化二次抗体(Vector Laboratories,Burlingame,CA,USA)とともにインキュベートした。これに続いて、アビジン/ビオチン−複合体(Vector Laboratories,Burlingame,CA,USA)とともに60分間最後にインキュベートした後、3,3’−ジアミンベブシジン(diaminebebcidine)/0.03% H2O2で可視化した。次いで、切片を、ゼラチンコーティングしたスライドガラスに載せ、乾燥させ、脱水し、permountとともにカバースリップを載せた。
(1.t−PAまたはDSPAの注入は、t−PA−/−マウスの海馬に分散し、タンパク質分解活性を保持する)
最初の実験を、DSPAおよびt−PAの両方が、注入の7日間にわたって、それらのタンパク質分解活性を保持することを確認するように設計した。この目的のために、t−PAおよびDSPAのアリコート(100nmol)を、水浴中で37℃および30℃にて7日間インキュベートした。タンパク質分解活性を決定するために、プローブの5倍連続希釈物を、非還元条件下でSDS−PAGEに供し、タンパク質活性をザイモグラム分析により評価した。7日間凍結したままで維持したt−PAおよびDSPAのアリコートを、コントロールとして使用した。図1に認められ得るように、30℃または37℃いずれかでのインキュベーションのときに、この期間の間にDSPA活性またはt−PA活性がごくわずかに喪失した。
まず、注入したプロテアーゼが、注入した動物の脳に存在し、それらのタンパク質分解活性もまた保持すると同時に、この区画に存在することを確認しなければならなかった。この点に対処するために、t−PA−/−に、t−PAまたはDSPAのいずれかを7日間にわたり注入した(上記を参照のこと)。次いで。心臓を介してマウスをPBSで灌流し、脳を取り出した。同側海馬領域および対側海馬領域を単離し、同様に、小脳領域もまた単離した(ネガティブコントロールとして)。組織サンプル(20μg)を、方法の節における記載に従って、SDS−PAGEおよびザイモグラム分析に供した。図2に認められ得るように、t−PA活性およびDSPA活性の両方を、海馬の同側領域中で検出したのに対し、いくらかの活性が、対側でも検出された。このことは、注入したプロテアーゼが、脳においてそれらの活性を維持しているのみならず、海馬領域に拡散したこともまた示す。コントロールとして、小脳から調製した抽出物中では活性を検出することはできなかった。
DSPAが、海馬領域に実際に拡散したことをさらに確認するために、t−PA−/−マウスの冠状脳切片を、DSPA注入後に、免疫組織化学的に分析した。DSPA抗原は、海馬領域において検出され、注入部位の領域において最も顕著に染色された。この結果により、注入されたDSPAが可溶性であり、かつ実際に海馬に存在することが確認される。
t−PA−/−マウスは、特徴として、カイニン酸(KA)媒介性ニューロン変性に耐性である。しかし、rt−PAの海馬内注入は、KA媒介性障害に対する感受性を完全に回復する。DSPAが、このモデルにおいてt−PAと置換され得るか否かを決定するために、t−PA−/−マウスの海馬内に、t−PAまたはDSPAのいずれかを、ミニ浸透圧ポンプを使用して注入した。両方の群について、12匹のマウスを試験した。2日後、動物にカイニン酸を注射し、回収するまで放置した。5日後、動物を屠殺し、その脳を取り出し、調製した(上記のように)。コントロールとして、KA処理前に、t−PA−/−マウスに、PBSもまた注入した(N=3)。
t−PA注入によって引き起こされるt−PA−/−マウスのKA感受性の回復はまた、小膠活性化を生じる(Rogoveら、1999)。t−PAまたはDSPA注入および引き続くKA処置後の小膠活性化の程度を評価するために、マウスの冠状切片を、Mac−1抗体を使用する活性化小膠細胞についての免疫組織化学的染色に供した。t−PA注入後のKA感受性回復は、Mac−1ポジティブ細胞の明らかな増加を生じた。このことは、DSPAを注入したマウスにおいては観察されなかった。従って、DSPAの存在は、KA処置後の小膠細胞の活性化を生じない。
注入に使用したt−PAの濃度は、Tsirkaら(1995)によって記載される濃度(0.12mg/mlを100μl[1.85μM])に基づいた。KA傷害実験を、10分の1のt−PA(0.185μM)および10倍高いDSPA(18.5μM)を使用して反復した。より低いt−PA濃度は、なお、KA処置に対する感受性を回復させることができた(n=3)。特に興味深いのは、10倍高いDSPA濃度の注入が、KA処置後のニューロン喪失をほとんど引き起こさないという知見であった。これらのデータは、DSPAが、KAに対する感受性の増大をもたらさないことを強く指摘する。
t−PAおよびDSPAの効果もまた、野生型マウスにおけるニューロン変性のモデルにおいて試験した。これらのマウスの線条におけるt−PAの注入は、グルタミン酸アナログNMDAによって引き起こされるニューロン変性効果の増大をもたらした(Nicoleら、2001)。
t−PA活性の間接的クロモゲン試験を、基質Lys−プラスミノゲン(American Diagnostica)およびspectrocyme PL(American Diagnostica)を用いて、Madisan E.L.,Goldsmith E.J.,Gerard R.D.,Gething M.J.,Sambrook J.F.(1989)Nature 339:721−724;Madison E.LO.,Goldsmith E.J.,Gething M.J.,Sambrook J.F.およびBassel−Duby R.S.(1990)Proc.Natl.Acad.Sci.USA 87,3530−3533、ならびにMadison E.L.,Goldsmith E.J.,Gething M.J.,Sambrook J.F.およびGerard R.D.(1990)J.Biol.Chem.265,21423−21426に従って行った。補因子DESAFIB(American Diagnostica)の存在下および非存在下の両方で試験を行った。DESAFIBは、プロテアーゼバトロキソビンでの非常に純粋なヒトフィブリノゲンの切断によって得られる可溶性フィブリンモノマーの調製物である。バトロキソビンは、フィブリノゲンのAα鎖におけるArg16−Gly17結合を切断し、それによってフィブリノペプチドAを放出する。フィブリンIモノマーを示す、得られたdes−AA−フィブリノゲンは、ペプチドGly−Pro−Arg−Proの非存在下で可溶性である。Lys−プラスミノゲンの濃度を、DESAFIBの存在下では0.0125μM〜0.2μMまで、補因子の非存在下では、0.9μM〜16μMまで変化させた。
間接的クロモゲン標準試験を、上記の刊行物に従って行った。0.25〜1ng酵素、0.2μM Lys−プラスミノゲン、および0.62mM spectrocyme PLを含む、総容積100μlのプローブを使用した。緩衝液、25μg/ml DESAFIB、100μg/ml フィブリノゲンの臭化シアンフラグメント(American Diagnostica)または100μg/mlの刺激性の13アミノ酸のペプチドP368のいずれかの存在下で試験を行った。分析をマイクロタイタープレート中で行い、光学密度を、「Molecular Devices Thermomax」において、1時間の間30秒ごとに、405nm波長で決定した。反応温度は、37℃であった。

- グルタミン酸レセプターアンタゴニストを特徴とする、請求項33に記載の薬学的組成物。
- 競合性アンタゴニストまたは非競合性アンタゴニストを特徴とする、請求項34に記載の薬学的組成物。
- 請求項33に記載の薬学的組成物であって、少なくとも1つのトロンビンインヒビターを特徴とし、該トロンビンインヒビターは、以下の物質:トロンボモジュリン、トロンボモジュリンアナログ、トリアビン、パリジピン、またはソルリンからなる群より優先的に選択される、薬学的組成物。
- 請求項33に記載の薬学的組成物であって、少なくとも1つの抗凝固剤を特徴とし、該抗凝固剤は、以下の抗凝固剤:ヒルジン、ヘパリン、アセチルサリチル酸またはアンクロドからなる群より優先的に選択される、薬学的組成物。

- シチコリンを特徴とする、請求項33に記載の薬学的組成物。
- 請求項1〜19に記載の、請求項36〜45のうちの少なくとも1項に記載の薬学的組成物の使用。
- 1種の非神経毒性プラスミノゲン活性化因子を含む発作の処置のための薬物の製造のための方法であって、該プラスミノゲン活性化因子の改変のために、以下の工程:
チモーゲン三つ組残基の少なくとも一部を導入する工程;
フィブリンの非存在下での触媒活性な立体構造の安定性を減少させるために、Asp194または一致するアスパラギン酸を置換する工程;
自己分解ループまたは該ループと一致するペプチド部分における、疎水性アミノ酸残基を置換する工程;
プラスミンによるチモーゲンの触媒を妨げるために該チモーゲン中に変異を導入する工程;
のうちの少なくとも1つを包含する、方法。 - 請求項42に記載の方法により製造される、薬物。
