ワクチン調製:ワクチンとして使用すべきポックスウイルスの産出および増幅は、以上で詳細に説明してきた。ポックスウイルスは任意に精製される。本発明による調合物は、以上で定義したジサッカライド、ポリマーおよび任意に緩衝液、L−グルタミン酸および任意にさらなる添加物を、ポックスウイルス調製品に加えることによって得る。
試料の凍結は、溶液中で成分を固定化し、それによって、圧力を低下させた時の生成物の浮き上がりを予防する。凍結は、水をまず核とし、続いて氷結晶が発達し、結果として氷と溶媒濃縮物の混合物となる、二段階工程である。温度を減少させ、冷却した懸濁液を撹拌することによって氷核化が促進される。核化と対照的に、温度を上昇させ、それによって懸濁液粘度を減少させることによって、氷の発達が促進される。正確な凍結パターンにかかわらず、培地全体での氷の増殖により、結果として、溶媒濃縮物が増加する。溶液中または懸濁液中のバイオポリマー類は、これらの溶媒の濃度の増加への曝露によってダメージを受けるか、不活性化する。急速な冷却によって、生物生成物の濃縮物への曝露が最小化される。臨界温度(ガラス伝達温度)以上で、重量粘度が、ガラスが軟化および歪曲するのに十分な程減少しうる。これを乾燥させ、バイアル内で、無型棒状残余物が形成される。歪曲化の温度は、崩壊温度と言われる。より特別には、崩壊温度は、マトリックスの間質領域中の水の移動度が顕著になる温度として定義される。歪曲化をさけるために、凍結温度は、水性調合物の崩壊温度より低くなるべきである。崩壊温度は、例えば異なる温度解析によってのような、当業者に公知の方法によって決定可能である(Jennings,T.A.,「凍結乾燥、紹介と基本的原理(Lyophilization,Introduction and Basic Principles.)」Interpharm Press,Denver,CO,US,1999,ISBN 1−57491−081−7、ページ132−134)。
第一乾燥(昇華):
第一乾燥は、溶媒(氷)の凍結マトリックスからの昇華を促進する凍結乾燥工程の部分である。第一乾燥工程は、凍結乾燥器が、必要なコンデンサー温度およびチャンバー圧に達した後に開始する。チャンバー内の圧力は通常1mbarより低く、好ましくは0.2mbarより低い。典型的に、圧力は、0.04〜0.12mbarの範囲である。本明細書において、これらの条件は時折、「低圧(low pressure)」と呼ばれる。
第二乾燥(吸収):
第二乾燥の間、水蒸気を、第二乾燥の間に形成したケーキの表面より吸収する。これは、水がケーキ表面より吸収されるように、チャンバーがまだ定圧であるまま、温度をあげることによって達成される。
生成物密封および除去:
全ての生成物含有バイアルを、当業者に公知の方法にしたがって密封する。バイアルを非常に低圧(例えば0.04〜2.56mbar)にて、直接凍結乾燥器内で密封可能である。また、窒素またはヘリウムのような化学的に不活性な基体を使用することで、通常の圧力以上または以下でバイアルを密封することも可能である。典型的には、バイアルを、900mbarの圧力で、窒素大気下で密封してよい。バイアルを、好ましくはブチルゴムストッパーを用いてしめる。いったん生成物を密封したならば、系を大気圧に戻すことが可能であり、棚を開放可能である。その後、バイアルをさらに長期間の保存のために、アルミニウムキャップで密封してよい。
ワクチン保存:
凍結乾燥生成物を、室温(25℃)にて保存可能であり、少なくとも18週間、好ましくは少なくとも20週間以上、好ましくは少なくとも22週間、この温度で維持する。「特定の時間の間、特定の温度で安定である(stable at a certain temperature for a certain period of time)」は、この時間の間、この温度でのウイルスタイターの欠損が0.5ログ以下であることを意味する。しかしながら、冷却が可能である場合、凍結乾燥生成物を4℃のようなより低温で保存することが好ましい。好ましくは、生成物を暗所で保存する。不可能な場合、保存のために、着色ガラスまたは任意の他のバイアルを使用することが好まく、これにより光への不利益な曝露を避ける。
再構成:
凍結乾燥生成物の再構成のために、適切な量の溶媒を、凍結乾燥生成物に加え、結果として、薬学的に許容な調合物を、ヒトまたは動物に投与可能にする。溶媒は好ましくは水である。通常、溶媒を、凍結乾燥工程の間、溶媒欠損量に十分に相当する量で、調合物に加える。
図1は31℃の温度での、MVA含有凍結乾燥調合物の安定性を示している。試験した調合物 はGT23(実施例項および表6を参照のこと)である。本発明による水性調合物におけるウイルスタイターを、凍結乾燥前に測定する。凍結乾燥は、T23に関して記述したように作製した(実施例および表6を参照のこと)。凍結乾燥後、調合物を31℃にて保存した。示した時間点で、凍結緩衝調合物 を再構成し、ウイルスタイターを再び測定した。
[実施例]
本実施例において、MVAを含む本発明による調合物を、異なる条件下で凍結乾燥した。凍結乾燥生成物を、異なる温度で保存した。調製におけるMVAの安定性を、凍結乾燥品の再構成の後にMVAのタイターを測定し、凍結乾燥前のMVAタイターと比較することによって解析した。ウイルスタイターにおける異なる保存時間の影響を決定した。MVA含有調合物のタイターの決定のためのプロトコールを実施例2で示す。
1.実験設定:
ワクチン調製/調合物
本発明による凍結乾燥工程を試験するために、いくつかのMVA調製品を使用した。凍結乾燥実験(表6中、GT1〜4、6〜10、および13〜15行目)のために、修飾ワクシニアウイルス株アンカラ(MVA)を使用した。ウイルスを、36%および40%スクロースクッション遠心、続いてpH9での1mM Tris−緩衝液中の再懸濁によって精製した。
試料凍結
試料を、凍結乾燥器(クリスト(Christ)凍結乾燥器、アルファ2〜4型)内で凍結させた。スクロース(DSG)を含む調合物に関しては、異なる凍結温度(−30℃〜−45℃)の比較により、完全なケーキ構造を得るために、懸濁液を、本調合物 の崩壊温度以下である、−40℃まで凍結させなければならないことが示された。凍結乾燥器内で凍結する場合、約3.5〜4.5時間以内に−40℃に達した(約20℃から開始)。
第一乾燥
スクロース(DSG)を含む調合物に関し、約−30℃〜−37℃の崩壊温度は、スクロースの崩壊温度(−31℃)を考慮に入れて仮定された。したがって、生成物温度を、−37℃〜−41℃の範囲の値に調整し、完全な凍結マトリックスを確かにした。0.04〜0.12mbarの圧力(水相ダイアグラム中、−50℃および−40℃)を使用した。
第二乾燥
第二乾燥のための温度を、生成物の安定性によって決定した。また、調合物の残存する湿度は、生成物の要求に依存する。最適な生成物安定性を得るために、時折より高い湿度、また時折低い湿度が必要である。最適な残存する湿度ならびにそれに達するまでの時間は、実験的に測定しなければならない。第二乾燥を、生成物が0℃以上の温度に達した時に開始するので、第二乾燥は2段階で実施した。第一段階において、棚温度を、生成物温度が0℃以上(4〜6℃の範囲)である様式で、数時間(4〜7時間の範囲)調節した。そのような様式で、第一乾燥の後に残っている、すべての可能性のある存在する氷が融解した。第二乾燥を開始するために、そのような穏やかな条件を使用することによって、生成物に対するダメージが最小化されうる。その後、第二段階を、生成物温度を、20〜30時間、18〜21℃の範囲の値まで上昇させることによって開始した。第二段階のための時間は、凍結乾燥生成物の必要とする残存する湿度に非常に依存する。異なる残存する湿度を得るために、異なる時間を使用した。凍結乾燥物質の残存する湿度を測定するためのアッセイとして、電量分析Karl−Fischer滴定を使用した(Jennings,T.A.,「凍結乾燥、紹介と基本的原理Lyophilization,Introduction and Basic Principles.)」Interpharm Press,Denver,CO,US,1999,ISBN 1−57491−081−7、415−418頁)。
生成物密封および除去
処理が進行する間に産出されたすべての生成物を、凍結乾燥器内で直接、非常に低圧(0.04〜2.56mbar)下で密封した。バイアルを、ブチルゴムストッパーを用いて閉じた。いったん生成物を密封し、系を室温に戻し、棚を開放した。その後、バイアルを、長期間保存のために、アルミニウムキャップで密封した。
ワクチン保存
調合物実施の重要な観点は、棚安定なワクチンを産出することである。安定性に影響を与える因子には、残存する湿度、密封環境組成、および温度、湿度および光を含む保存条件が含まれる。
再構成
凍結乾燥試料を、オートクレーブMilli−Q水にて再構成した。より特別には、水(1.2ml)を、シリンジを用いて試料に添加した。懸濁液を、穏やかに撹拌して混合した。再構成はただ数秒間だけかかった。再構成生成物のウイルスタイターを測定し、凍結乾燥前のウイルスタイターと比較した。
加速された安定性試験
調合物GT23(表6を参照のこと)の安定性を、31℃、37℃および45℃で査定した。ウイルスタイターを、定期的にモニタした。結果を図1〜3に示している。
2.結果と結論:
MVAを、異なる添加物を使用するか、使用せずに凍結乾燥した。添加物無しの調合物は、不安定であるということが示された(表6を参照のこと)。この文中において、試料は、タイターが0.5ログ以上落ちなかった場合に安定であると考えられる。したがって、用語「特定の時間間隔で、特定の温度において安定(stable at a certain temperature for a certain period of time)」は、この温度でのウイルスタイターの欠損が、この時間間隔で、0.5ログ以下であることを意味している。示唆した温度での示唆した時間間隔の、ウイルスタイターの欠損が0.5ログ以上である場合に、調合物は「不合格である(fail)」。デキストランおよびグルコースを含む調合物 は、室温で、非常に低い安定性を示した。異なる濃度のスクロースおよびデキストランを含む、本発明による調合物が安定であることが証明された。本発明による調合物 内のMVAの安定性は、4℃および室温にて少なくとも25週間である。
修飾ワクシニアウイルスアンカラ(MVA)の滴定を96ウェルフォーマットで、10倍希釈を使用する、TCID50に基づくアッセイで実施する。アッセイのエンドポイントにて、感染細胞を、抗ワクシニアウイルス抗体および適切な染色溶液を用いて視覚化する。

Claims (23)
- (i)オルトポックスウイルス類、パラポックスウイルス類、アビポックスウイルス類、カプリポックスウイルス類およびスイポックスウイルス類からなる群より選択された、精製または部分的に精製されたポックスウイルス、(ii)ジサッカライド、および(iii)薬学的に許容可能なポリマーを含む調合物。
- さらに緩衝液を含む、請求項1に記載の調合物。
- 前記ポックスウイルスがワクシニアウイルスであることを特徴とする請求項1又は2記載の調合物。
- 前記ワクシニアウイルスが、株Elstreeおよび修飾ワクシニアウイルス株アンカラ(MVA)から選択される、請求項1〜3のいずれかに記載の調合物。
- ポックスウイルスが組換え体ポックスウイルスであることを特徴とする、請求項1〜4のいずれかに記載の調合物。
- 前記ジサッカライドが、スクロース、ラクトースおよびトレハロースのからなる群より選択されることを特徴とする、請求項1〜5のいずれかに記載の調合物。
- 前記ジサッカライドの濃度が、10〜100g/lの範囲であることを特徴とする、請求項1〜6のいずれかに記載の調合物。
- 薬学的に許容可能なポリマーが、デキストランおよびポリビニルピロリドン(PVP)から選択されることを特徴とする、請求項1〜7のいずれかに記載の調合物。
- デキストランが、30,000〜70,000の範囲の分子量、および1〜50g/lの濃度を持つことを特徴とする、請求項8に記載の調合物。
- リン酸緩衝液が、緩衝液としてのぞかれることを特徴とする、請求項2〜9のいずれかに記載の調合物。
- 調合物がさらに、グルタミン酸を含む、請求項1〜10のいずれかに記載の調合物。
- 前記調合物の崩壊温度が、−37℃〜−30℃の範囲であることを特徴とする、請求項1〜11のいずれかに記載の調合物。

- 以下の、
(i)請求項1〜13のいずれかにて定義したような調合物を、前記調合物 の崩壊温度以下の温度まで凍結し、凍結生成物マトリックスを得ること、
(ii)低圧および生成物温度で、凍結生成物 の第一乾燥をして、生成物マトリックス内の氷の昇華を可能にすること、この際、前記生成物温度は、調合物 の崩壊温度より低い、
(iii)生成物の残余水分が5%以下になるまで、低圧および0℃〜30℃の範囲内での生成物温度にて第二乾燥させること、
の段階を含む、請求項16に記載の方法。 - 請求項16〜17のいずれかに記載の方法によって入手可能な、凍結乾燥生成物。
- (i)ポックスウイルス、(ii)ジサッカライド、(iii)薬学的に許容可能なポリマー、および任意に(iv)緩衝液を含む、凍結乾燥生成物であって、前記ポックスウイルス、前記ジサッカライド、前記ポリマーおよび前記緩衝液が、請求項1〜6、8、10〜11のいずれかにて定義されたようなものである、凍結乾燥生成物。
- 残余水分含量が、1〜3%の範囲であることを特徴とする、請求項18〜19のいずれかに記載の、凍結乾燥生成物。
- ワクチンの調製のための、請求項18〜20のいずれかに記載の凍結乾燥生成物の使用方法。
- 前記生成物を、適切な量の薬学的に許容可能な溶媒中に溶解することを特徴とする、請求項18〜20のいずれかに記載の凍結乾燥生成物の再構成のための方法。
- 請求項1〜13のいずれかに記載の調合物、請求項14に記載のワクチン、または請求項22に記載の方法によって得た再構成生成物を用いて必要な、ヒトを含む動物のワクチン接種方法。
