Abstract
【解決手段】樹脂からなる被覆層を有する生物活性物質の粒状物であって、被覆層が生物活性物質粒子表面を直接被覆する溶出コントロール層、及び溶出コントロール層の外側に位置するオーバーコート層の少なくとも二層からなり、オーバーコート層が水性エマルジョンを主成分とし無機系コロイドゾル及び無機フィラーの何れか一方または両方を含有する被覆粒状物。

Description
(1)樹脂からなる被覆層を有する生物活性物質の粒状物であって、被覆層が生物活性物質粒子表面を直接被覆する溶出コントロール層、及び溶出コントロール層の外側に位置するオーバーコート層の少なくとも二層からなり、オーバーコート層が水性エマルジョンを主成分とし無機系コロイドゾル及び無機フィラーの何れか一方または両方を含有する被覆粒状物。
(2)オーバーコート層を構成する水性エマルジョンがアクリル樹脂である前記(1)記載の被覆粒状物。
(3)無機系コロイドゾルの固体平均粒子径が10〜200nmである前記(1)または(2)項記載の被覆粒状物。
(4)無機フィラーの比表面積が1〜500m2/gである前記(1)〜(3)項のいずれか1項記載の被覆粒状物。
(5)無機フィラーがシリカ系化合物、ハイドロタルサイト系化合物、リチウム・アルミニウム複合水酸化物塩系化合物から選ばれる少なくとも1種である前記(1)〜(4)項のいずれか1項記載の被覆粒状物。
(6)溶出コントロール層を構成する樹脂がオレフィン系重合体である前記(1)〜(5)項のいずれか1項記載の被覆粒状物。
本発明の被覆粒状物は、樹脂からなる被覆層を有する生物活性物質の粒状物であって、被覆層が、生物活性物質粒子表面を直接被覆する溶出コントロール層及び溶出コントロール層の外側に位置するオーバーコート層の少なくとも二層からなる。オーバーコート層は水性エマルジョンを主成分とし、無機系コロイドゾル及び無機フィラーの何れか一方または両方含有する。主成分とは最も多い成分をいう。
円形度係数={(4π×粒子の投影面積)/(粒子投影図の輪郭の長さ)2}
尚、溶出コントロール層を構成する樹脂が界面活性剤を含む場合、界面活性剤の被膜(溶出コントロール層)材料中の含有割合は、特に限定されるものではないが、樹脂に対して0.01〜10重量%が好ましく、0.1〜5重量%がより好ましい。
尚、親水性と浮上防止性を高めるため、オーバーコート層の外側に更に親水化剤を付着させても良い。

(式中、M2+は2価の金属イオン、mは0≦m<5の範囲にある数、xは0≦x<1の範囲にある数、yは2≦y≦4の範囲にある数を示す。)

(式中、M2+は2価の金属イオン、An―はn価の陰イオンであり、mは0≦m<5の範囲にある数、xは0.01≦x<1の範囲にある数、nは1≦n≦3の範囲にある数を示す。)
(式中、M2+はMg2+、Ca2+、及びZn2+の中から選ばれた少なくとも1種の2価金属イオンを示し、A1 n−はn価のアニオン、xは0<x<0.5の範囲にある数、mは0≦m≦2の範囲にある数、nは1≦n≦3の範囲にある数を示す。)
親水化剤のコート層の外側表面への付着処理時の温度等は特に限定するものではないが、付着を効率よく行うためには、オーバーコート層の融着等が起こらない範囲でオーバーコート層の表面温度を上げた状態で付着処理を行う方法が推奨される。この際の温度は特に限定されるものではないが、オーバーコート層に用いた樹脂の融点または軟化点の5℃以下の温度である。
界面活性剤のHLBは6〜20が好ましく、9〜16が更に好ましい。この界面活性剤は非イオン性界面活性剤、イオン性界面活性剤の何れも用いることができる。非イオン性の界面活性剤としては、ポリオキシエチレンアルキルエーテル類、ポリオキシエチレンフェニルエーテル類、ポリエチレングリコールと脂肪酸のエステル類、ポリオキシエチレンソルビタン脂肪酸モノエステル類、ポリオキシエチレンアミン類等が挙げられる。また、イオン性界面活性剤としては、脂肪酸塩類、高級アルコールサルフェート類、アルキルベンゼンスルホン酸塩類等を挙げることができる。
(a)初期浮上率の測定
被覆粒状物100粒をシャーレに入れ、純水をシャーレの壁に沿わせて10ml/minで注水した。50ml注水後水面に浮上している粒子数を初期浮上率とした。
目開き2mmの篩で篩い、105℃で3日間乾燥した黒ボク土100gと被覆粒状物100粒子を混合した後500mlポリ瓶に入れ、純水300mlを加えて攪拌し混合した後、40℃90日間恒温水槽で保存した。その後、目開き2mmの篩で篩い、105℃で3日間乾燥した黒ボク土500gの上に全量取り出し、水を除き、土壌ごと30℃で7日間熱風乾燥した。その後1200mlの水をゆっくり注水し、このとき水面に浮上している粒子状の被覆材の数を求めて経時後浮上率とした。
被覆粒状物の表面を走査型電子顕微鏡で観察し、表面に無機系コロイドゾルまたは無機フィラーの少なくとも一部が露出しているか否かを観察した。この評価基準は次の通りである。
○:被覆粒状物の表面に撥水防止剤が露出している。
×:被覆粒状物の表面に撥水防止剤が露出していない。
被覆粒状物100gを300ml容の三角フラスコに入れ振とう機で1時間振とうした後、被覆粒状物を排出する。三角フラスコの内壁の付着状況を肉眼で観察した。この評価基準は次の通りである。付着が少ないほど、被膜と親水化剤との密着性が良好である。
○:オーバーコート層の剥離がほとんど無く、フラスコ壁面も透明のまま。
×:オーバーコート層の剥離が見られ、フラスコ壁面に濁りあり。
1)一次粒子の製造
尿素を加熱可能な容器に投入し130℃で加熱溶融し尿素溶融液を得た。該尿素溶融液を1時間撹拌後、直径3mmのコック付ガラス管から、容量5Lのステンレス容器中で50℃に加温され液深さ20cmの流動パラフィン液面に滴下し粒状物を得た。流動パラフィン液面からガラス管出口までの高さは20cmであった。この時、得られた尿素粒子が2〜3.5mm程度になるように流量を調節した。次いで該粒状物をヘキサンで洗浄し、乾燥後に粒径2〜3.5mmの篩にかけ一次粒子を得た。
高温熱風が下部から上部に向けて流入し、噴流塔の上部に設置されている排ガス用出口から排出されるように循環している、塔径250mm、高さ2000mm、空気噴出口径50mm、円錘角50度の形状を有する噴流塔の内部に、上記で得た一次粒子を噴流塔の側面に設置されている投入口から投入し、一次粒子を噴流状態にする。この際、粒子温度が70℃±2℃になるように、排気温度を測定しながら熱風温度を調節した。熱風流量は流量計で測定しながら調節した。別途、溶解槽において尿素を融解し造粒用尿素融解液を作成した。
二次粒子を回転円盤式整粒機(不二パウダル株式会社製、マルメライザーQJ400)に供給し、円形度係数が0.7以上になるまで平滑化処理を行った。処理後、二次粒子を、熱風循環乾燥機を用いて50℃で3日間乾燥し、次いで75℃で4時間乾燥した後に目開き1.0〜4.0mmの篩にかけ分級し三次粒子(生物活性物質粒子)を得た。
図1に示す製造装置を用いて次の方法により溶出コントロール層を形成した。
熱風が噴流塔1の下部から上部に向けて流れ集塵機6を通過し、コンデンサー7でガスを冷却し、溶媒を凝縮回収する。コンデンサー7を通過したガスはブロワー8からヒーター12を通過して加熱され熱風として再度噴流塔1へ導かれるように循環している。塔径450mm、高さ6000mm、空気噴出口径100mm、円錘角50度の形状を有する噴流塔1の内部に、上記で得た三次粒子(生物活性物質粒子)を噴流塔1の側面に設置されている投入口から投入し、三次粒子を噴流状態にする。この際、粒子温度が70℃±2℃になるように、熱風流量及び熱風温度を調節した。熱風流量はブロワー9と整流缶11の間に設置した流量計で測定しながら調節し、熱風温度は、粒子温度や排気温度(噴流塔1上部温度)を測定しながら調節した。他方、溶解槽9に被膜材料組成としてポリエチレン(低密度ポリエチレン、密度0.923g/cm3(JIS K 6760)、メルトフローレート(MFR)0.3g/10min.(JIS K 6760))50重量部、コーンスターチ5重量部、タルク(平均粒経10μm)45重量部、鉄アセチルアセトナート錯体0.01重量部の各成分とテトラクロロエチレン1900重量部を投入し、100℃±2℃で1時間混合撹拌することによって樹脂を溶解し、濃度5重量%の均一な被膜材料溶解液を調製した。

1)塗工液の作製
塗工液に用いられるアクリル樹脂は、表1に記載のモノマー配合組成に従い、溶媒としてMEK(メチルエチルケトン)を用い、開始剤としてベンゾイルパーオキサイドをモノマー合計量に対し1重量%使用して、70℃で3時間、更にベンゾイルパーオキサイドを0.5重量%追加して70℃で3時間反応を行って得られた。尚、溶媒中のアクリル樹脂濃度は40重量%になるように調整した。上記の原料の合計重量に重合開始剤を1重量%別途添加した。
2.項で得られた溶出コントロール層を有する生物活性物質粒子(被覆粒状物1)10kgを図2に示した直径60cmの糖衣機の回転容器4に入れ、ブロワー7、ヒーター8、熱風吹込管6より60±5℃の空気を吹き込み該粒子の温度を50±3℃に加温した。30rpmの回転速度で該被覆粒状物1を転動させながら、前記1)で調整しておいた塗工液(オーバーコート層用樹脂含有)をポンプ3を通してスプレーノズル5から0.3kg/minで該粒状物に向けて噴霧し、熱風吹込管6より60±5℃の空気を300m3/hで吹き込み、表2記載の被覆率になるまで乾燥し、塗膜を形成して比較例1〜9、実施例1〜15の被覆粒状物を得た。

- 樹脂からなる被覆層を有する生物活性物質の粒状物であって、被覆層が生物活性物質粒子表面を直接被覆する溶出コントロール層、及び溶出コントロール層の外側に位置するオーバーコート層の少なくとも二層からなり、オーバーコート層が水性エマルジョンを主成分とし無機系コロイドゾル及び無機フィラーの何れか一方または両方を含有する被覆粒状物。
- オーバーコート層を構成する水性エマルジョンがアクリル樹脂である請求項1記載の被覆粒状物。
- 無機系コロイドゾルの固体平均粒子径が10〜200nmである請求項1または2記載の被覆粒状物。
- 無機フィラーの比表面積が1〜500m2/gである請求項1〜3のいずれか1項記載の被覆粒状物。
- 無機フィラーがシリカ系化合物、ハイドロタルサイト系化合物、リチウム・アルミニウム複合水酸化物塩系化合物から選ばれる少なくとも1種である請求項1〜4のいずれか1項記載の被覆粒状物。
