本発明の要約
手短にいえば、本発明は、組換え体ウイルスベクター及び広範な病原因子の治療のためのこのようなベクターの使用方法を提供する。本発明の一態様内では、そうでなければ不活性な前駆体を病原因子の活性阻害剤に活性化し得る遺伝子物質の発現を指示するベクター構築物を保有する組換え体ウイルスベクターが提供される。本発明の一態様内では、病原因子はウイルス感染細胞である。別の態様では、遺伝子物質は単純ヘルペスウイルスチミジンキナーゼ又は水疱帯状疱疹ウイルスチミジンキナーゼである。さらに別の態様では、不活性前駆体は AZTである。
本発明を説明する前に、それを理解しやすくするために下記で用語の定義を先ず行うことにする。
上記のように、本発明は組換え体ウイルスベクターを包含する組成物及び方法を提供する。本発明に用いるのに特に好ましい組換え体ウイルスベクターとしては、組換え体レトロウイルスベクター及び組換え体アデノウイルスベクターが挙げられる。組換え体レトロウイルスベクターの構築は、“組換え体レトロウイルス”という表題の出願(U.S.S.N.07/586,603 、1990年9月21日提出)(この記載内容は参照によりホン明細書に含めるものとする)に詳述されている。これらの組換え体レトロウイルスベクターを用いて、それらを適切なパッケージング細胞株に導入することにより、形質導入コンピテントレトロウイルスベクター粒子を生成し得る(U.S.S.N.07/800,921 参照)(この記載内容は参照によりホン明細書に含めるものとする)。同様に、本明細書に開示されたアデノウイルスベクターも容易に調製し、使用し得る(Berkner,Biotechniques 6:616−627,1988 及び Rosenfeld et al.,Science 252:431−434,1991,WO 93/07283,WO 93/06223 及び WO 93/07282 参照)(この記載内容は参照によりホン明細書に含めるものとする)。
本発明の別の態様では、上記のような組換え体ウイルスベクターを製薬上許容可能なキャリアー又は希釈剤と組み合わせて包含する製剤組成物が提供される。このような製剤組成物は、液体溶液として、又は投与前に溶液中に懸濁される固体形態(例えば親液化)として調製し得る。さらに本組成物は、表面投与、注射、経口又は直腸投与に適したキャリアー又は希釈剤とともに調製し得る。
本発明の他の態様において、病原因子が阻害又は破壊されるように、温血動物に上記のような組換え体ウイルスベクターを投与することを包含する、温血動物における病原因子を阻害又は破壊するための方法が提供される。本発明の種々の態様においては、組換え体ウイルスベクターは下記に詳述するように in vivoで又は ex vivoで投与し得る。あるいは本発明の細胞毒性遺伝子、遺伝子物質、ベクター構築物又はウイルスベクターを、種々の方法により患者に投与し得る。代表例としては、種々の物理的方法によるトランスフェクション、例えばリポフェクション(Felgner et al.,Proc.Natl.Acad.Sci.USA 84:7413 7417,1989);直接 DNA注入(Acsadi et al.,Nature 352:815−818,1991);微小発射ボンバード (Williams et al.,PNAS 88:2726 2730,1991);いくつかの型のリポソーム(例えばWang et al.,PNAS 84:7851−7855,1987 参照);CaPO4(Dubensky et al.,PNAS 81:7529−7533,1984); DNA配位子(Wu et al.,J.of Biol.Chem.264:16985−16987,1989);核酸単独投与(WO 90/11092);又は死アデノウイルスと結合させた DNAの投与(Curiel et al.,Hum.Gene Ther.3:147−154,1992)が挙げられる。
ベクターの構築
A.ベクター LTR配列を含有するプラスミドの構築
以下のレトロウイルスベクターはすべて、N2ベクターを基礎にしている(Keller et al.,Nature 318:149−154,1985)。手短にいえば5′及び3′EcoRI LTR断片(それぞれ 2.8及び1.0kb)(Armentano,J.Vir.61:1647,1987;Bglitis,Science 230:1395,1985)を最初にプラスミドSK+ (Stratagene,San Diego,CA)及び pUC31のEcoRIVにサブクローニングする、 pUC31は、ポリリンカーのEcoRI及び SacI間に別の制限部位(XhoI,BglII 及び NcoI)を保有するpUC19 (Stratagene San Diego,CA)の修飾体である。それによりSK+ プラスミドと1.0kb 3′LTR 断片との結紮からプラスミドN2R3+/ を作る。プラスミド p31N2R5+/−及び p31N2R3+/−は、それぞれ pUC31と2.8kb 5′LTR 及びパッケージングシグナル (Ω)又は1.0kb 3′LTR 断片との結紮から構築される。各々の場合、N2はベクター源を示し、Rは断片がEcoRI断片であるという事実を示し、5及び3は、5′又は3′LTRsを示し、そして+又は−は挿入体の配向を示す(LTRサブクローンの例に関しては図1〜6参照)。
HS部位−1チミジンキナーゼ遺伝子(HSVTK)のコード領域及び転写終結シグナルを、pBR322 (ATCC No.31344)中にクローニングしたプラスミド322TK(HS部位−1 (McKnight等)の3.5kb BamHI断片)から1.8kb BglII/PvuII 断片として単離し、そして BglII/ SmaI消化 pUC31中にクローニングした。この構築物を pUCTKと呼ぶ。ターミネーターシグナルの欠失を要する構築物のために、 pUCTKを SmaI及びBamHIで消化し、(A)n シグナルを含有する 0.3kb断片を除去した。以下のオリゴマーから作られる二重鎖オリゴヌクレオチドで残りのコード配列及び付着端BamHIオーバーハングを再構築する:

その結果生じる構築物を pTKΔA(図7)と呼ぶ。
レトロウイルスプロベクター pTK−1,pTK−2及び pTK−3を、本質的には下記のように構築する:
1.5kb XhoI/HindIII 5′LTR 及びプラスミド配列をp31N2R5(+)(図1)から単離する。
1.プラスミドp31N2R5(+)(図1)を XhoI及び HindIIIで切断する。
レトロウイルスベクターpKTVIHAXを本質的に下記のように構築する(図11参照)。
レトロウイルスベクターpKTVIH 5及び pKTVIH5 Neoを本質的に下記のように構築する(図12参照)。
レトロウイルスベクター MHMTK Neoを本質的に下記のように構築する(図14参照)。
a)実施例1AからのプラスミドpN2CR5を FokIで消化して線状化し、クレノウ DNAポリメラーゼを用いて5′オーバーハングをデオキシヌクレオチドトリホスフェートで充填し、 HindIIIリンカーを挿入する。細菌に形質転換後、MLV LTRFokIVに挿入した HindIIIリンカーを有するクローンを、制限酵素分析により同定する。このプラスミドをpN2CR5FHと呼ぶ。
レトロウイルスベクター RRKTVIHを本質的に下記のように構築する(図17参照)。
a)HSVTK遺伝子を1.8kb HincII/PvuII 断片としてベクターSK+ のEcoRV部位にサブクローニングする。この構築物をpSTK (−)(図15)と呼ぶ。
発現ベクター tat及び抗 tatを本質的に下記のように構築する(図18参照)。これらのベクターを擬 HIVとして用いて tat依存性 HSVTKベクターを試験活性化する。
ベクター構築物のパッケージング
これらのレトロウイルスベクターの生物学的特性を以下に記載する。 HIV tat遺伝子 (「tathis」ベクター:図18参照)をマウス PA317細胞中にトランスフェクトする。5つの個々のヒスチジノール耐性サブクローンを生成し、TH1−5 と名付けたが、これらはHIVtatを発現する。これらの細胞はしたがって HIV感染細胞の実験的見積もりである。 次いでベクターKTVIHAX(実施例1Dから)、KTVIH5Neo(実施例1Eから)及びMHMTKNeo (実施例1Fから)をこれらの tat発現細胞株並びに tatを欠いたその親細胞株への感染により導入する。次に細胞生育能力を種々の濃度の HSVTK特異的細胞毒性剤acyclovir (ACV)で確定する。データはここではLD50 (50%毒性が観察される薬剤濃度)として報告し、生育可能細胞対 ACV濃度のグラフに内挿した。 HSVTKを含有するがしかし tatを欠いた親細胞株 (非 HIV感染モデル)は、試験濃度では ACVによる検出可能な毒性を示さなかった。したがってこれらの細胞は細胞毒性に関しては 100μM又はそれ以上の ACVを要する。これは、ベクターを欠いたこれらの細胞に関して真実である。したがってベクター単独、 ACV単独、又はベクター+ACV でさえも細胞毒性を示さない。しかしながらHIVtatを発現する(HIV感染の実験的表出)細胞株は、 ACVに有効に殺される (図21)。これは、試験した3つのベクター全てに関する程度を変えるのが正しい。これらのデータは、 HIV感染細胞が ACV及び“ポテンシエイター”ベクターの存在下で殺されることを示す。
培地を感染細胞から除去し、10秒間微小遠心分離して、次に68℃に10分間加熱して、内生的ホスファターゼを破壊した。次いで培地を2分間微小遠心分離して、検定のためにアリコート(10〜50μl)を取り出した。緩衝液(1M ジエタノールアミン、pH 9.8;0.5mM MgCl2 ; 10mM L−ホモアルギニン)100μlを加え、その後、 120mM P ニトロフェニルホスフェート(緩衝液中)を20μl加えた。反応混合物のA405を自動プレート読み取り機を用いてモニタリングした。
pBH−1及びpMχ−IFN/TKベクターの構築
HSVTK遺伝子を含有する1.2kb 断片を得るために、実施例1Gからの pRRTKを XhoI及びBamHIで消化する。この断片を、ポリリンカー中に見出される XhoI及びBamHI制限部位でpSP72 (Promega Corp.,Madison,WI)に結紮する。このプラスミドを pRRTK Bと呼ぶ。 pRRTK−Bを XhoI及び ClaIで消化して、 HSVTK遺伝子を含有する1.2kb 断片を得る。次に、 XhoI及び ClaI部位で消化後に、この断片をKT 3 (このベクターの構築物は WO 91/02805 に記載されている)に結紮する。ゲル精製によりKbHIVgag/prot断片を取り出す。このプラスミドを pBH 1と呼ぶ。
DA/TK−3,DA/BH−1,DA Mχ−IFN/TK及びHA Hχ−IFN/TKの構築
プロデューサー細胞株
pTK−3DNA (図9)10μg及びpMLP−G DNA (このベクターの構築物は WO 92/14829 に記載されている)10μgを、標準 CaPO4を用いて293 2 3 (293細胞 (ATCC No.CRL 1573,WO 92/05266)に由来する細胞株)にトランスフェクトする。48時間トランスフェクション後、上清を収集し、0.45μmの円筒濾紙を通して濾過する。この上清は、新たに調製されたDA (D 17 ATCC No.CCL 183,WO 92/05266 由来の細胞株)細胞を感染させるのに用いられるG−擬似型TK−3ベクターを含有する。ウイルス上清付加後24時間目にDA細胞を G−418選択(800μg/ml)下に置く。7〜9日後、 G−418選択非クローン性プールが得られ、これをDA/TK 3と呼ぶ。ベクターが HSVTKを発現する最高力価に関して個々の単離物を限定希釈クローニング及びスクリーニングにより高力価クローンを同定する。これは、 3 T3 TK− 又は Hela TK− のような細胞チミジンキナーゼを欠いた細胞株を形質導入し、細胞をゲネチシン及び HAT培地中に入れるすることにより Neoコロニー形成を測定して達成し得る。これらの条件下で成長するコロニーの数は、 Neo及び HSVTKを発現する形質導入細胞の数を反映する。
TK−3を含有する場合又は含有しない場合のCT26に及ぼす ganciclovirの作用の測定
DA/TK 3で形質導入したCT26細胞 (CT26 TKneo)に Ganciclovirが作用を及ぼすか否かを調べるために、 CT26TKneo細胞を6つの 10cm2プレートに 2.5x106 /プレートで植え付けた。対比した場合、2つの他の細胞型 CT26及びCT26Bgal (この細胞株は大腸菌からの受容体遺伝子βガラクトシダーゼを保有するウイルスで形質導入された)を各々、対照として6つの 10cm2プレートに植え付けた。各細胞型の5つのプレートを1日2回、4連続日間、 100μg/ml,50μg/ml,25μg/ml,12.5μg/ml及び6.25μg/mlの濃度の ganciclovirで処置した。各細胞型の1つのプレートは未処置のままにしておいた。その後、トリプシン処理して各皿から細胞を取り出し、10% FBSを含有するDMEMに再懸濁して、計数した。図22に示したデータは、最低用量の ganciclovirでも CT26TKneo細胞に劇的な細胞毒性作用を及ぼすことを示す。この用量 (6.25μg/ml)又は次に高い用量 (12.5μg/ml)の ganciclovirでもCT26及びCT26Bgal細胞に作用しなかった。しかしながら25μg/mlの用量で開始すると、細胞成長に用量依存性減少が認められたが、しかし CT26TKneo細胞は常に薬剤に対して感受性がより高かった。
CT26TKNeo 細胞を注入されたマウスの治療のための ganciclovir用量の確定
ネズミ腫瘍の in vivo形質導入を用いて疾患を治療し得るか否かを調べるために、形質導入され、 100%形質導入を保証するためにin vitro選択された腫瘍を排除するのに必要な ganciclovirの最適濃度を確定するための実験を実施した。結腸腫瘍26即ちCT26(Brattain,Baylor College of Medicine,Houston,TX)細胞をG擬似型TK−3ベクターて形質導入した。ウイルス上清を付加後24時間目に G−418選択(450μg/ml)下に置いた。10日間インキュベーション後、 G−418選択プールを得て、 CT26TKNeoと名付けた。各3匹12群のマウスに2x105 CT26TKNeo 細胞を注入した。6群のマウスにこれらの細胞を腹腔内(I.P.)注入し、6群のマウスには皮下 (S.C.)注入した。各3匹の他の2群にはI.P.又はS.C.で2x105 個の未修飾CT26細胞(対照として)を注入した。
CT26及び CT26TKNeoにおける細胞毒性の比較
in vivo腫瘍成長
Ganciclovirが in vivoで未修飾CT26腫瘍細胞の成長に作用を及ぼすか否かを調べるために、各7匹の2群に2x106 個の未修飾CT26細胞をS.C.注入し、各7匹の2群には2x105 個の未修飾 CT26TKNeo細胞をS.C.注入した。腫瘍を植え付けて数日後に、CT26注入マウスの1群及び CT26TKNeo注入マウスの1群に、62.5mg/kgでのI.P.ganciclovirの1日2回(午前と午後)レジメンを施した。これらのマウスを12日間、又は CT26TKNeo注入動物が検出可能な腫瘍の苦しみを示さなくなるまで、処置した。3週間に亘って腫瘍成長をモニタリングした。CT26を注入し ganciclovirで処置したマウスはCT26を注入した未処置マウスよりもいくぶん小さい腫瘍を有したが、これは腫瘍成長の HSVTK非依存性阻害が小さいことを示す(図26)。しかしながら CT26TKneo含有マウスを ganciclovirで処置した場合に、そしてこの場合にのみ、腫瘍の苦しみの劇的減少が観察された(図26)。
CT26腫瘍細胞注入マウスへのβ−GAL 直接ベクターの注入
ベクターの直接注入により腫瘍細胞が in vivo形質導入去れたか否かを査定するために、大腸菌β ガラクトシダーゼ遺伝子を発現するリポーターベクター(CB β−gal)を用いた。各2匹の5群に2x105 個のCT26腫瘍細胞をS.C.注入した。各2匹の別の群には、β−gal 染色の対照として、2x105 個のCT26β−gal 発現細胞を注入した。各マウスの注入領域を耐水性マーカーで丸く印を付けた。腫瘍細胞接種2日後、マウスに PBS+ポリブレン(4μg/ml)、ポリブレン含有又は無しの場合のCBβ−gal (5x106 コロニー形成単位(CFU)ml)、あるいはポリブレン含有又は無しの場合の DAhχ−IFN#15を注入した。各群のマウスにそれぞれの接種物を2日毎に耐水性マーカーで印を付けた領域内に注入した。各群は、合計4回の注射を受けた。最終注射の2日後、各群のマウスの腫瘍を取り除き、細かく切って、DMEM+10% FBS及び抗生物質を含有する 10cm2の2つのプレートに植え付けた。これらの腫瘍外植片をin vitroで1週間増殖させた。1週間後、細胞を収穫し、2%ホルムアルデヒドで固定して、 X−galで一夜染色した。この実験の結果を以下に示す。
TK−3直接ベクターによるCT26腫瘍細胞の in vivo形質導入
TK−3ベクターが in vivoで標的細胞に HSVTK遺伝子を供給し、 ganciclovirの存在下で腫瘍成長を阻害し得ることを調べるために、本実験を計画した。要するに、各10匹の6群に1x105 CT26腫瘍細胞をS.C.注入した。さらに各10匹の1群に、対照として1x105 CT26TKNeo 細胞をS.C.注入した。S.C.注入領域を耐水性マーカーで丸く印を付けた。腫瘍接種24時間後、ポリブレン(4μg/ml)とともに処方したTK−3又はβ−gal ウイルス上清を耐水性マーカーで印を付けた領域内に注入した。1日1回のベクター投与を4連続日間継続した。各ベクター用量は、2x105CFU/mlを含有した。最終ベクター処置の24時間後、これらのマウスに8日間、1日2回(午前と午後)、62.5mg/kgで ganciclovirをI.P.注入した。最後に、実験終了まで、62.5mg/kgの ganciclovirをマウスに1回投与した。4週間に亘って腫瘍成長を測定した(図27)。実験を下記の表Bに要約する。
耐性マーカー以外の随伴遺伝子を含有する HSVTKベクターの構築
HSVTKのような条件致死遺伝子を含有する不全−安全能力を伴うレトロウイルスベクターを設計する目的は、患者に投与された後に、治療遺伝子の発現を制御させることである。単純ヘルペスチミジンキナーゼ遺伝子を発現する細胞は ganciclovirに感受性に鳴り、一方正常細胞は影響を受けない(Moolten et al.,Cancer Res.46:5276,1986)。治療遺伝子が発現される時間の長さは、修飾細胞が有害になった場合には限定されるか打ち切られる。
先ずTK−2を XhoIで消化し、その後クレノウ処理して、子牛腸アルカリ性ホスファターゼで脱リン酸化することにより、レトロウイルスプロベクター pKT1−TKを構築した。処置線状化TK−2を次にリン酸化 ClaIリンカーと結紮し、その後細菌を形質転換した。制限酵素分析により正しい個々のクローンを同定し、TK−2と名付けた(ClaI(図10))。TKプロモーター及びTKコード配列の両方を含有する 2.0kb ClaI断片を次に手 K−2から単離した(ClaI)。 ClaIで消化し、その後子牛腸アルカリ性ホスファターゼで処理して、1%アガロースゲル上でゲル精製して、 HIV IIIBenvを含有するKT1レトロウイルスベクター (RecombinantRetrovirusesPatent Application #586,603)を調製した。TK 2からの 2.0kb単離 ClaI断片を次に前処理KT1ベクターと結紮し、細菌中に形質転換して、制限酵素分析により正しい個々のクローンを同定した。その結果生じたレトロウイルスベクターをKT1−TKと名付けた (図28参照)。もちろん他のプロモーター、例えば CMV直前プロモーターを HSVTKプロモーターの代わりに用い得る。この性質のベクターを用いて、HIVenvに対する免疫反応を誘発し得る一方、 ganciclovirの投与により何時でも形質導入細胞を破壊し得る。
pKT1−HBc「Neoless」プロベクターDNA(1993年8月4日提出の米国特許出願第08/102,132 号参照)を ClaIで切断してレトロウイルスプロベクター pKT−HBc/TKを生成した。次にこれをpTK−2 (ClaI)からの 2.0kb ClaI断片と結紮し、細菌の形質転換に用いて、個々のクローンを正しい配向に関してスクリーニングした(図29参照)。
レトロウイルスプロベクターpmχTK及びphχTKを、以下の工程で生成した。
TKベクター及び ganciclovirで処置したマウスにおける免疫反応の誘発
A.TKレトロウイルスベクター構築物の構築
単純ヘルペス1型ウイルスのチミジンキナーゼをコードする原遺伝子は、プラスミド 322TKからの 1.8kb BglII/PvuII 断片であった (McKnight et al)(実施例1参照)。5′プロモーター及び3′ポリアデニル化シグナルを取り出し、遺伝子をさらに修飾して5′ XhoI部位並びに3′ ClaI部位を有した。この新規の修飾HSV−TK遺伝子を次に、 gag発現を阻害するために ATTに対して突然変異を起こさせた gag発現に対する正常 ATGメチオニン イニシエーターコドンを有するMoloney Murine LeukemiaVirusから得たN2ベースのベクター骨格の XhoI及び ClaI部位にクローニングした (BHI−TK構築物)。レトロウイルス LTRはHSV−TK遺伝子の発現を制御し、下流SV40プロモーターはネオマイシン耐性マーカーの発現を駆動する。これらのベクターを、本質的には実施例4と同様に、パッケージング細胞(DA及びHX。 WO 92/14829 参照)に導入した。
ネズミ脾臓エフェクター細胞に関する 51Crm放出細胞毒性検定を以下のように実施した。要するに、CT26TKを注入し、 ganciclovirで処置したマウスにおける完全腫瘍切除後種々の時間に、マウスを屠殺し、脾臓細胞(3x106 /ml)を未修飾照射CT26細胞(6x104 /ml)とともに T−25cm2フラスコ (Corning,Corning,NY)中で invitro培養した。培地は RPMI−1640、5%加熱不活性化ウシ胎児血清(Hyclone,Logan,UT)、2mM グルタミン、1mM ピルビン酸ナトリウム、1x 非必須アミノ酸、10mM HEPES、ゲンタマイシン (50mg/ml)、及び2−メルカプトエタノール(10−6M)で構成された。エフェクター細胞を4〜7日後に収穫し、幾つかのエフェクター:標的比(E:T 100:1,30:1,10:1,3:1及び1:1.各比率は3通りウエルに用意した)を96ウエル微小滴定プレートに入れて標準4−6時間検定に用いて試験した。検定は、最終容積 200ml中にNa251 CrO4 (Amersham,Arlingtyon Heights,IL)−標識化CT26又はCT26TK標的細胞(1x104 細胞/ウエル)を用いた。インキュベーション後、検定培養上清を採集し、特異的51クロミウム放出(計数/分)に関して Beckmanガンマ分光計を用いて検定した。標的1培地からの1分当たりの計数として自発性放出を測定し、典型的には最大放出の10〜20%であった。最大放出値を標的+1M HClからの1分当たりの計数として測定した。パーセント標的細胞溶解を(実験放出−自発的放出)/(最大放出−自発的放出)x 100として算出した。
CT26TK細胞を注入し、 ganciclovirで処置したマウスにおける完全腫瘍切除後に、それぞれの脾臓細胞を収穫し、in vitroで再刺激して、その結果生じたエフェクター細胞を、未修飾CT26又はCT26TK細胞を溶解するその能力に関して分析した。CT26及びCT26TK細胞の両方に対する有意の細胞毒性反応が、CT26TK腫瘍を ganciclovirにより取り除かれたマウスから得た脾臓細胞で観察された(図32参照)、一方CT26腫瘍保有マウスからの脾臓細胞をCT26で再刺激すると、極わずかの溶解を示した(図3)。これらの結果は、TKプロドラッグベクター、その後のプロドラッグ自体の導入による腫瘍細胞の除去は、未修飾腫瘍細胞に対する有効且つ強力な CTL反応を生じた。

- ヒト細胞に感染するレトロウイルスベクターであって、該レトロウイルスベクターは、制御配列に作動可能に連結された、第1の組換え遺伝子および第2の組換え遺伝子を含み、該第1の組換え遺伝子は、ほとんどまたは全く細胞毒性を持たない化合物(プロドラッグ)を毒性物質へと活性化する生成物をコードする遺伝子であり、該第2の組換え遺伝子は、ヒトサイトカインをコードする、ベクター。
- 請求項1に記載のベクターであって、前記プロドラッグは、プリンベースの薬物またはピリミジンベースの薬物である、ベクター。
- 請求項2に記載のベクターであって、前記第1の組換え遺伝子の生成物は、チミジンキナーゼ、シトシンデアミナーゼ、および大腸菌グアニンホスホリボシルトランスフェラーゼからなる群より選択される、ベクター。
