Abstract
Description
細胞中に存在する2種の蛋白質の濃度を測定することによって、癌を診断する方 法に関するものである。
の増殖能を損なうことなしに同時に癌細胞の増殖を選択的に阻害する手段が発見
されれば、腫瘍の分化、侵入または転移の程度とは関係なく腫瘍の生育を停止し
得る新しい方法が提供されることになる。癌細胞がどの様に分裂し、正常細胞の
分裂とどの様に異なるのかを理解することは興味深いことである。癌治療の大半
は広範な細胞毒性を有しており、一般に増殖細胞を標的としている。しかしなが
ら、正常組織も増殖する為、細胞毒性剤によって同様に損傷を受けることになる
。標的設定を可能にするか、または、薬剤開発用の特異的な標的を提供し得る癌
特異的マーカーを同定することができれば、腫瘍組織に対して一層特異的な毒性
を発揮し、化学療法による衰弱作用を軽減し得る治療法の開発が可能となる。
系の応答に対してのみ単一遺伝子の発現作用が研究されているが、その理由は、
これらが臨床現場におけるヒト癌の応答に適切なモデル系を提供する為である。
特にこれらは、臨床現場で通常用いられる細胞毒性剤および電離放射線に対する
感受性域を有している。従って、ヒトin vitro細胞系における知見は、癌がどの
程度治療に応答すると期待できるのかについて、臨床上有用な試験へとつながる
可能性が強い。
リンと呼ばれる蛋白質と、サイクリン結合して活性となるサイクリン依存性キナ
ーゼ(CDK)との組合せによって形成されるホロ酵素により決定される。サイク リン/CDKの複合体は、蛋白質p21 WAF1/CIP1(p21)を含むサイクリン依存性キ
ナーゼ阻害剤(CDKI)と呼ばれる蛋白質によって阻害される。
存性キナーゼパートナーであるCDK4およびCDK1について研究されている。サイク
リンD1およびCDK4は、G1期とS-期の間(DNA合成期)の細胞周期チェックポイン トを通じて細胞の進行を制御する。サイクリンB1およびCDK1は、有糸分裂直前の
細胞周期チェックポイントを制御する。一連の16ヒト癌細胞系におけるサイクリ
ンD1蛋白質の発現は、細胞毒性剤CDDPに対する内因性耐性と関連していることが
示されている(Warenius等、1996)が、サイクリンD1蛋白質の濃度は、別の治療
法である放射線感受性とは無関係である。しかしながら、サイクリンD1とCDDP耐
性との関係はそれ自身、臨床上有用な予測試験の根拠となる程、強いものではな
い。

ech12:27‐32、1994)等である。特定の突然変異を調べる為の様々な方法は当 該分野で知られており、例えば、TaqMan試験、オリゴヌクレオチドリガーゼ法、
一重鎖コンホメーション多形性および鋳型核酸のオリゴヌクレオチドアレーへの
ハイブリダイゼーションに基づく試験法が挙げられる。

は、これらの遺伝子のmRNA濃度を測定したとしても、本試験の目的には不充分で
あることを意味している。

担持した液滴には、正電荷または負電荷が与えられる。液滴中に特定の閾値を超
える強度の蛍光を示す細胞が含まれている場合には、この液滴は、一方の極性の
電荷を獲得する。未標識の細胞は逆の極性の電荷を獲得する。次に、荷電した液
滴が電場により偏向し、その表面電荷に応じて個別の容器内に輸送された液滴を
計数したとき、液滴中に1個より多い細胞を含有するときは、個々の細胞よりも 多くの光を散乱するが、これは容易に検出される為、これらは未荷電のままで第

て分類する為に必要である。抗体は、p53蛋白質の既知突然変異型の多くに対し て存在するが、現時点において、p53の全ての突然変異型を、抗体によって検出 することはできない。診断試験は、下記工程を包含する。

瘍の特徴でもある。従って、本発明に到達するに至ったヒトin vitro細胞系にお
ける上記発見は、薬剤発見プログラム用の重要な標的として組込まれ得る可能性
が極めて高い。

vitro細胞系において最も顕著にみられることが当該分野の多数の専門家によっ
て指示されているからである。

,25mM EDTA pH8.0)中でホモジナイズした。0.5%(w/v)ナトリウムラウリルサ
ルコシンおよび8%(v/v)2-メルカプトエタノールを使用直前に添加した。このホ
モジネートを4℃で10分間、8,000rpmで遠心分離(SS34ローター、 Sorvall RC-5B
遠心分離機)して清澄化した後、得られたホモジネ−トを、5.7M塩化セシウム/0
.1M EDTAの緩衝液を介して20℃で20時間、32,000rpmで遠心分離(TST41.14ロータ
ー、Kontron Centrikon T20 60超遠心分離機)し、RNAをペレット化した。RNAの ペレットを0.1%(w/v)SDS中に再懸濁し、-20℃で一晩エタノール中に沈殿させた
後、定量した。

を30サイクル行った。PCR産物を0.8%(w/v)アガロースゲル上で確認した後、Wiz
ardミニカラム(Promega)を用いて精製し、直接配列決定反応に付した。
総RNA(5μg)、ヒト胎盤リボヌクレアーゼ阻害剤(HPRI)20U、および1μgのオリゴ
(dT)を70℃で10分間加熱した後、氷冷し、これを、1×第1鎖緩衝液(50mMTris-HC
l,pH8.3,75mM塩化カリウムおよび3mM塩化マグネシウム)、0.01M DTT、dNTP(各 デオキシリボヌクレオシド3リン酸につき0.5mM)、スーパースクリプト逆トラン
スクリプターゼ(Superscript Reverse Transcriptase)(Gibco)400Uに添加し、
37℃で1時間インキュベートした。エクソン9〜11のPCRは前述の通り、PCR反応溶
液50μL中で上記インキュベーション液5μlを用いて行った。

この様にして同定した突然変異型の推定配列の全てについて、アンチセンス方向
のエクソン配列決定を更に行うと共に、PCRを反復し、細胞系の配列決定を行っ て確認した。
センス突然変異以外、従来より報告されているものである。これとH417のコドン
298におけるGからTへの変異は、p53遺伝子が最も高度に維持された領域内には存
在しない。OAW42卵巣癌細胞系においては、CGAからCGGへの1塩基ミスセンス突 然変異はサイレントであり、突然変異3塩基はなお、野生型p53(wtp53)蛋白質中
に存在するものと同様のアミノ酸(Arg)をコードしていた。即ち、正常p53蛋白質
は細胞系の半数で発現していた。細胞系の残り半数のmRNAは、異常p53蛋白質を コードしていた。RPMI7951およびH417は停止突然変異を有しており、夫々、165 および297アミノ酸で蛋白質を切断していた。COLO320およびH322は夫々独立して
、ミスセンスのG:CからA:Tへの突然変異を同じ部位で示しており、その結果、
ArgからTrypへのアミノ酸置換が生じていた。RT112とHT29/5もまた、Argにおけ る(夫々、GlyおよびHisへの)変化をコードする突然変異を有していた。COLO32

変異系は、ヘテロ接合度を有しているという証拠を示していた。HT29/5およびRP

為に、ブロットを、ヒトCDK1に対するsc-054マウスモノクローナル抗体(Santa
Cruz Biotechnology, CA)と共にインキュベートし、次に、1/1000のウサギ抗マ ウスコンジュゲート抗体(Dako UK)と共にインキュベートした後、ニトロブルー テトラゾリウムおよび5-ブロモ-4-クロロ-3-インドイルホスフェート(Sigma, Po
ole, Dorset,UK)(ジメチルホルムアミド中50mg/mL)を含有するアルカリホスフ ァターゼ緩衝液で発色させた。CDK4蛋白質の発現濃度を測定する為に、ブロット
を、ヒトCDK4に対するsc-260ウサギポリクローナル抗体(Santa Cruz Biotechno
logy, CA)と共にインキュベートし、次に、1/1000のウサギ抗マウスコンジュゲ ート抗体(Dako UK)と共にインキュベートした後、ニトロブルーテトラゾリウム および5-ブロモ-4-クロロ-3-インドイルホスフェート(Sigma, Poole, Dorset,UK
)(ジメチルホルムアミド中50mg/mL)を含有するアルカリホスファターゼ緩衝液 で発色させた。

により、光学密度(OD)の直線関係は、細胞系を通じてCDK1蛋白質およびCDK4蛋白
質に対して見出された範囲に渡って得られることが、従来より知られている(War
enius等,1994,Browning 1997)。様々なCDK1蛋白質およびCDK4蛋白質の濃度を細 胞系間で比較する為に、全ての系の平均OD値を計算し、個々の細胞系各々におけ
るCDK1蛋白質およびCDK4蛋白質に対する相対的OD値を平均OD値で規格化し、任意
値5.0を掛けた。
観察されている。多くの上記系に対するp53突然変異状態に関するDNA配列決定デ
ータを用いると、CDK1/CDK4の関連性はp53突然変異を有する細胞系で特に顕著に
見られることが更に観察された(図5および図6を参照)。
関係は、対応するmRNAでは見られなかった。また、一の遺伝子における蛋白質発
現と、他の遺伝子のmRNAにおける蛋白質発現との間にも、相互相関は見られなか
った。これらの結果により、CDK1蛋白質およびCDK4蛋白質が同時に上昇すること
は、転写制御レベルの段階ではなく、翻訳または翻訳後の段階で生じることが示
唆される。即ち、蛋白質自身が、何らかの方法で相互に影響し合っているか、或
いは、双方が未知の因子の影響を受けていると考えられる。
相関関係に関する上記知見は、ラット正常筋上皮細胞系RAMA37へのトランスフェ
クション試験によって指示される。RAMA37を用いたプロット試験により、無条件
プロモーター制御下において、CDK4がうまくトランスフェクションしてCDK4蛋白
質が過剰に発現すると、CDK1の構成的発現が同時に上昇することが示唆された。
