Claims (1)
- 【特許請求の範囲】 1. 容器; 微生物の存在又は数について試験するサンプルを固定化するための固定化層; 及び 固定化層と容器の壁の間に配置されたセンサー層であって、当該センサー層の 少なくとも一部分が固定化層の上及び/又は中で固定化された微生物の存在に起 因して検出可能に変化することのできるセンサー層; を含むセンサー器具。 2. さらに、a)固定化層とセンサー層の間、及びb)センサー層と容器の壁 との間の少なくとも1つの間に接着剤層を含む請求項1に記載のセンサー器具。 3. センサー層が不透明である請求項1に記載のセンサー器具。 4. 前記固定化層がゲル化剤を含む請求項1に記載のセンサー器具。 5. 前記ゲル化剤が固体ゲル、半固体ゲル、又は粉末化ゲルの少なくとも1つ である請求項4に記載のセンサー器具。 6. 前記固定化層がさらに微生物の増殖を促進するための増殖成分を含む請求 項1に記載のセンサー器具。 7. 前記容器が固定化層の上方にヘッドスペースを有する密開容器である請求 項1に記載のセンサー器具。 8. さらに前記容器の壁にガス透過性膜を含む請求項7に記載のセンサー器具 。 9. さらに前記ガス透過性膜に隣接して配置された脱着可能なガス不透過性シ ールを含む請求項8に記載のセンサー器具。 10. 前記容器の前記壁が透明又は半透明である請求項1に記載のセンサー器 具。 11. 前記センサー層が紫外線、可視光線、及び/又は赤外線スペクトルにお いて局在性変化をなすことができる請求項1に記載のセンサー器具。 12. 前記局在性変化が前記容器の前記壁を通して検出できる請求項11に記 載のセンサー器具。 13. センサー層が酸素、水素、硫化水素、二酸化炭素、アンモニア、有機酸 、二酸化窒素及びpHのうちの1つ以上における変化に反応して検出可能に変化 する請求項1に記載のセンサー器具。 14. 前記検出可能な変化が赤外線、紫外線若しくは可視光線スペクトルにお いて検出可能である請求項13に記載のセンサー器具。 15. 前記センサー層が画像描出、蛍光又は反射率テクノロジーによって検出 可能な変化を有する指示薬を含む請求項13に記載のセンサー器具。 16. 前記センサー層が約5.0〜11.0のpH範囲に渡って蛍光強度又は 視認できる色の変化を示す請求項15に記載のセンサー器具。 17. 前記固定化層がその中に微生物を含む固定化サンプルを含む請求項1に 記載のセンサー器具。 18. 前記固定化層が単一のゲル化剤又は複数のゲル化剤を含む請求項1に記 載のセンサー器具。 19. 前記ゲル化剤がゴム、寒天、アガロース、カラゲナン、ベントナイト、 アルギン酸塩、コラーゲン、ゼラチン、溶融ケイ酸塩、水溶性デンプン、ポリア クリレート、セルロース、セルロース誘導体、ポリエチレングリコール、ポリエ チレンオキシド、ポリビニルアルコール、デキストラン、ポリアクリルアミド及 び多糖類から選択された1つ以上の物質を含む請求項 18に記載のセンサー器具。 20. 前記固定化層がその表面上に微生物を捕獲するための上層と当該上層を 通して液体サンプルを引き出すためのウィッキング剤として備えられている下層 とを含む請求項3に記載のセンサー器具。 21. 第1ゲル化剤が固定化層の上層に備えられ、さらに第2ゲル化剤が前記 固定化層の下層に備えられる請求項18に記載のセンサー器具。 22. さらに微生物検出能力を向上させるために前記固定化層の近位若しくは その中にコンディショニング成分を含む請求項1に記載のセンサー器具。 23. 前記コンディショニング成分が溶解剤、溶解酵素、界面活性剤及び増殖 阻害剤中和用成分のうちの少なくとも1つを含む請求項22に記載のセンサー器 具。 24. 前記センサー層がシリコーンを含む請求項1に記載のセンサー器具。 25. センサー層が固定化層における微生物コロニーの存在に対応して検出可 能に局在性変化するように構成される請求項1に記載のセンサー器具。 26. センサー層が微生物の存在下で不透明を維持しながら1つの色から第2 の色に変化する不透明な層である請求項25に記載のセンサー器具。 27. 固定化層とセンサー層の少なくとも1つが不透明である請求項25に記 載のセンサー器具。 28. 器具内の少なくとも1つの層が微生物コロニーの視認に不都合な影響を 及ぼすマトリックスを有する請求項25に記載のセンサー器具。 29. 前記少なくとも1つの層が前記センサー層を包含する請求項28に記載 のセンサー器具。 30. センサー層が固定化層と反対側のセンサー層の側からの試験サンプルの 視認を十分に遮断する請求項25に記載のセンサー器具。 31. センサー層が固定化層と反対側のセンサー層の側からの試験サンプルの 目による視認又は検出器による検出を遮断する請求項25に記載のセンサー器具 。 32. 容器; 微生物の存在又は数について試験するサンプルを固定化するための固定化層; 及び 少なくともセンサー層の一部分が、固定化層の上及び/又は中で固定化された 微生物の存在に起因して検出可能に変化することのできるように、固定化層の近 位に配置されたセンサー層; を含むセンサー器具。 33. センサー層が固定化層における微生物コロニーの存在に対応して検出可 能に局在性変化するように構成される請求項32に記載のセンサー器具。 34. センサー層が不透明な層である請求項33に記載のセンサー器具。 35. 固定化層とセンサー層の少なくとも1つが不透明である請求項32に記 載のセンサー器具。 36. 器具内の少なくとも1つの層が微生物コロニーの視認に不都合な影響を 及ぼすマトリックスを有する請求項32に記載のセンサー器具。 37. 前記少なくとも1つの層が前記センサー層を包含する請求項36に記載 のセンサー器具。 38. センサー層が固定化層と反対側のセンサー層の側からの試験サンプルの 視認を十分に遮断する請求項32に記載のセ ンサー器具。 39. センサー層が固定化層と反対側のセンサー層の側からの試験サンプルの 目による視認又は検出器による検出を遮断する請求項32に記載のセンサー器具 。 40. さらに、a)固定化層とセンサー層の間、及びb)センサー層と容器の 壁の間の少なくとも1つの間に接着剤層を含む請求項32に記載のセンサー器具 。 41. センサー層が微生物の存在下で色の強度を変化させる不透明な層である 請求項32に記載のセンサー器具。 42. 前記固定化層がゲル化剤を含む請求項32に記載のセンサー器具。 43. 前記ゲル化剤が固体ゲル、半固体ゲル、又は粉末化ゲルの少なくとも1 つである請求項42に記載のセンサー器具。 44. 前記固定化層がさらに微生物の増殖を促進するための増殖成分を含む請 求項43に記載のセンサー器具。 45. 前記センサー層が紫外線、可視光線、及び/又は赤外線スペクトルにお いて局在性変化をなすことができる請求項32に記載のセンサー器具。 46. 前記局在性変化が前記容器の前記壁を通して検出でき る請求項45に記載のセンサー器具。 47. センサー層が酸素、水素、硫化水素、二酸化炭素、有機酸、二酸化窒素 、アンモニア及びpHのうちの1つ以上における変化に反応して検出可能に変化 する請求項32に記載のセンサー器具。 48. 前記検出可能な変化が赤外線、紫外線若しくは可視光線スペクトルにお いて検出可能である請求項47に記載のセンサー器具。 49. 前記センサー層が画像描出、蛍光又は反射率テクノロジーによって検出 可能な変化を有する指示薬を含む請求項47に記載のセンサー器具。 50. センサー層が約5.0〜11.0のpH範囲に渡って蛍光強度又は視認 できる色の変化を示す請求項49に記載のセンサー器具。 51. さらに微生物検出能力を向上させるために前記固定化層の近位若しくは その中にコンディショニング層を含む請求項41に記載のセンサー器具。 52. 前記コンディショニング層が溶解剤、溶解酵素、界面活性剤及び増殖阻 害剤中和用成分のうちの少なくとも1つを含 む請求項51に記載のセンサー器具。 53. 前記固定化層が溶解剤、溶解酵素、界面活性剤及び増殖阻害剤中和用成 分のうちの少なくとも1つを含む請求項41に記載のセンサー器具。 54. 前記センサー層がフルオレセイン、クマリン、フェノールフタレイン、 チモールフタレイン、ブロモチモールブルー、チモールブルー、キシレノールブ ルー、オルトクレゾールフタレイン及びα−ナフトールベンゼインから選択され た少なくとも1種の指示薬を含む請求項32に記載のセンサー器具。 55. 前記センサー層が微生物代謝に起因するガス成分の増加又は減少に反応 して変化する請求項32に記載のセンサー器具。 56. 前記センサー層がセンサー層を着色するための少なくとも1種の顔料を 含む請求項32に記載のセンサー器具。 57. 前記顔料が金属酸化物である請求項56に記載のセンサー器具。 58. 前記顔料がチタン、亜鉛、マグネシウム又は鉄の酸化物である請求項5 7に記載のセンサー器具。 59. 前記センサー層がポリマー材料を含む請求項49に記 載のセンサー器具。 60. 前記容器が実質的に扁平な基体、一端で開放している保持器具、断面が 長方形の閉鎖型箱、又はボトルである請求項32に記載のセンサー器具。 61. 前記固定化層が試験サンプルを吸収及び固定化するための吸収性材料を 含む請求項32に記載のセンサー器具。 62. 前記吸収性材料が濾紙、スポンジ、ガラス繊維、及びセルロースから選 択される請求項61に記載のセンサー器具。 63. さらに、前記固定化層の上又は中に溶解剤を含んでおり、前記溶解剤が サポニン、ジギトニン、トゥイーン及びポリソルビタンモノラウレートから選択 される請求項61に記載のセンサー器具。 64. 前記ゲル化剤が固体ゲルであり、前記固体ゲルが脱水されたゲルを含む 請求項43に記載のセンサー器具。 65. 前記ゲル化剤が半固体ゲルであり、前記半固体ゲルが部分的に脱水され た半固体ゲルを含む請求項43に記載のセンサー器具。 66. 前記ゲル化剤が粉末である請求項43に記載のセンサー器具。 67. さらに、微生物増殖阻害剤を中和するための成分を含む請求項32に記 載のセンサー器具。 68. 前記中和成分が樹脂、エコソルブ(ecosorb)及び活性炭のうち の少なくとも1つを含む請求項67に記載のセンサー器具。 69. 微生物の検出方法であって、 試験されるサンプルが請求項32に記載のセンサー器具の固定化層の上又は中 に固定化されるように前記サンプルを前記センサー器具に添加するステップ; 微生物コロニーの増殖を可能にする一定時間に渡りセンサー器具をインキュベ ートするステップ;及び 固定化層の上又は中における微生物コロニーの存在に相関するセンサー器具の センサー層における何らかの局在性変化を測定するステップ; を含む方法。 70. サンプルが裸眼による微生物コロニーの視認を妨害するように十分に不 透明である請求項69に記載の方法。 71. サンプルが蛍光性である請求項69に記載の方法。 72. サンプルが電磁領域において特性を有し、さらに前記 局在性変化が前記電磁領域での前記センサー層における変化を検出する検出器に より決定される請求項69に記載の方法。 73. サンプルが蛍光性であり、センサー層が前記固定化層における微生物の 存在若しくは増殖に起因するガス成分における変化に反応して検出可能に変化す る請求項72に記載の方法。 74. サンプルが着色されている及び/又は蛍光性であり、センサー層が蛍光 若しくは可視pH指示薬を含む請求項69に記載の方法。 75. サンプルが血液サンプルである請求項74に記載の方法。 76. 固定化層が粉末化ゲル層であり、サンプルが、固定化されてセンサー層 に付着するように粉末化ゲル及びゲル類と混合する液体サンプルである請求項6 9に記載の方法。 77. 前記サンプルが食品サンプルである請求項69に記載の方法。 78. 前記サンプルが滅菌体液サンプルである請求項69に記載の方法。
