Claims (1)
- 【特許請求の範囲】 1. 抗体に特異的に結合することができる少なくとも2種の異なるレセプター R1およびR2と試料とをインキュベートすることによる免疫グロブリンMクラス の抗原特異的抗体の決定方法であって、R1は固相に結合しているか、または固 相に結合可能であり、R2が標識を有するものであり、R1の必須成分が決定対象 の抗体によって特異的に認識される多量体型の結合パートナーであり、かつ試料 中に存在する同じ特異性を有するIgG分子による干渉を単量体型の結合パート ナーを添加することにより低減させる、免疫グロブリンMクラスの抗原特異的抗 体の決定方法。 2. R1およびR2の必須成分が決定対象の抗体により特異的に認識される多量 体型の結合パートナーであり、かつ試料中に存在する同じ特異性を有するIgG 分子による干渉を単量体型の結合パートナーを添加することにより減少させる、 請求項1記載の方法。 3. ビオチン/アビジン、ビオチン/ストレプトアビジン、ビオチン/アンチ ビオチン、ハプテン/アンチハプテン、抗体のFc断片/該Fc断片に対する抗 体または炭水化物/レクチンを特異的結合系として用いてR1と固相とを結合さ せる、請求項1または2記載の方法。 4. 該レセプターR2を、化学発光性、蛍光性もしくは放射活性の物質により 、または酵素もしくは他の生体分子により標識する、請求項1〜3いずれか記載 の方法。 5. 該試料を、R1およびR2と、干渉を減少させるために用いられる単量体 型の結合パートナーとを同時にインキュベートする、請求項1〜4いずれか記載 の方法。 6. レセプターR2の標識に特異的に結合し、R2の標識に特異的なレセプター と標識とのコンジュゲートである別のレセプターと共に試験混合物をインキュベ ートし、ついで該標識を決定する、請求項1〜5いずれか記載の方法。 7. 干渉を減少させるために用いられる単量体型の結合パートナーを、R1お よびR2の多量体型の結合パートナー上にあるエピトープの濃度に比較して、1 0倍〜10,000倍過剰の濃度で添加する、請求項1〜6いずれか記載の方法 。 8. 免疫アッセイのための慣用の試験添加剤に加えて、単量体型の結合パート ナーと、固相に結合可能である、決定対象の抗体に結合可能なレセプターR1と を含み、かつその必須成分が多量体型の結合パートナーである、前記請求項のい ずれか記載の免疫グロブリンMクラスの抗原特異的抗体を決定するための試薬。 9. 免疫アッセイのための慣用の試験添加剤に加えて、単量体型の結合パート ナーと、決定対象の抗体に結合可能な2つのレセプターR1およびR2とを含み、 その必須成分が多量体型の結合パートナーであり、かつR1が固相に結合可能で あり、R2が標識を有する、前記請求項のいずれか記載の免疫グロブリンMクラ スの抗原特異的抗体を決定するための試薬。 10. 抗原特異的IgM抗体の決定において、IgG抗体および/またはリウ マチ因子による干渉を減少させるための単量体型の結合パートナーの使用。
