Claims (11)

1. Konjugat generičke formule: (Ama-L-X-S)n-Ab, pri čemu je Ama amatoksin, L je spona, X je ostatak koji nastaje spajanjem tiolne skupine na tiol-reaktivnu skupinu, S je atom sumpora cisteinskog aminokiselinskog ostatka, a Ab je sekvenca protutijela, ili fragment koji veže antigen, koji sadrži navedeni cisteinski ostatak, pri čemu je navedeni cisteinski ostatak teški lanac 265Cys prema EU sustavu numeriranja, i pri čemu n označava da je jedna ili više molekula amatoksina vezano na jedno protutijelo ili fragment koji veže antigen.
2. Konjugat prema patentnom zahtjevu 1, pri čemu je spona odabrana između stabilne spone i spone koja se može cijepati.
3. Konjugat prema patentnom zahtjevu 1 ili 2, pri čemu je tiol-reaktivna skupina odabrana između bromoacetamida, jodoacetamida, 4,6-dikloro-1,3,5-triazin-2-ilamina, 4-[5-(metilsulfonil)-1H-tetrazol-1-il]fenila, 2-(metilsulfonil)-benzo[d]tiazol-5-il, 4-[5-(metilsulfonil)-1,3,4-oksadiazol-2-il]-fenil, 2-piridilditio-, 2-(5-nitro-piridiil)ditio-, metiltio-sulfonila i maleimida, posebno maleimida.
4. Konjugat prema bilo kojem od patentnih zahtjeva 1 do 3, pri čemu je n odabran između 1 i vrijednosti između 1 i 4.
5. Konjugat prema patentnom zahtjevu 4, pri čemu je n 2.
6. Metoda za sintezu konjugata generičke formule: (Ama-L-X-S)n-Ab reakcijom spoja Ama - L - X’ s protutijelom ili fragmentom koji veže antigen, Ab - SH, pri čemu je Ama amatoksin, L je spona, X je ostatak koji nastaje spajanjem tiolne skupine na tiol-reaktivnu skupinu X', skupina - SH je tiol cisteinskog aminokiselinskog ostatka, a Ab je sekvenca protutijela ili sekvenca fragmenta koji veže antigen, koji sadrži navedeni cisteinski ostatak, pri čemu je navedeni cisteinski ostatak teški lanac 265Cys prema EU sustavu numeriranja, i pri čemu n označava da je jedna ili više molekula amatoksina vezano na jedno protutijelo ili fragment koji veže antigen.
7. Metoda prema patentnom zahtjevu 6, pri čemu je n odabran između 1 i vrijednosti između 1 i 4.
8. Metoda prema patentnom zahtjevu 7, pri čemu je n 2.
9. Komplet koji sadrži (i) spoj Ama - L - X’, pri čemu je Ama amatoksin, L je spona, X’ je tiol-reaktivna skupina, i (ii) protutijelo ili fragment koji veže antigen, Ab - SH, pri čemu je skupina -SH tiol cisteinskog aminokiselinskog ostatka, a Ab je sekvenca protutijela ili sekvenca fragmenta koji veže antigen, koji sadrži navedeni cisteinski ostatak, pri čemu je navedeni cisteinski ostatak teški lanac 265Cys prema EU sustavu numeriranja.
10. Farmaceutski pripravak, koji sadrži konjugat prema bilo kojem od patentnih zahtjeva 1 do 5.
11. Konjugat prema bilo kojem od patentnih zahtjeva 1 do 5 za upotrebu u liječenju bolesti povezane sa stanicama koje predstavljaju metu, pri čemu je navedeno protutijelo, ili funkcionalni fragment protutijela, specifičan za navedenu metu.
