Claims (5)
1. Postupak proizvodnje cirkovirusa, naznačen time, da obuhvaća sljedeće korake:
kalemljenje kulture stanice domaćina s kulturom cirkovirusa;
inkubacija kulture stanice domaćina bez seruma od fetusa mladunca;
nadziranje (i) životne aktivnosti stanica domaćina, i (ii) ekspresije ORF1 i ORF2 putem stanica domaćina; i
sabiranje cirkovirusa iz kulture stanice domaćina kada je ekspresija ORF2 otprilike ista u neprianjajućim stanicama kao i u prianjajućim stanicama od kulture stanice domaćina;
te time, da cirkovirus je PCV2.
2. Postupak prema zahtjevu 1, naznačen time, da je prinos cirkovirusa u kulturi sjemena određen pomoću nadziranja ekspresije cirkovirusnih antigena ORF1 i ORF2 u stanicama domaćina koje su isplivale na površinu, uporabom postupka koji se temelji na sortiranju fluorescentnih stanica protutijela (FACS-fluorescent antibody cell sorting).
3. Postupak prema zahtjevu 1 ili 2, naznačen time, da se ekspresija cirkovirusnih antigena ORF1 i ORF2 u stanici domaćina koja je inficirana virusom, nadzire uporabom postupka koji se temelji na (FACS) sortiranju fluorescentnih stanica protutijela, te time, da se proizvodnja cirkovirusnog antigena odnosi na postotak pozitivnih stanica otvorenog okvira čitanja 1 (ORF1-open reading frame 1) i otvorenog okvira čitanja 2 (ORF2-open reading frame 2).
4. Postupak prema bilo kojem od zahtjeva 1 do 3, naznačen time, da se životna održivost stanica određuje pomoću mjerenja primanja propidijevog jodida uporabom protočne citometrije.
5. Postupak prema bilo kojem od zahtjeva 1 do 4, naznačen time, da stanica domaćina je PK-15.
