Abstract
A sex hormone of polypeptide type may be obtained from an extract of male sex glands by precipitation by means of picric acid, and decomposition of the precipitated picrate with alcoholic hydrochloric acid to yield the hydrochloride which is then precipitated in crystalline form by addition of acetone.ALSO:Hormones of polypeptide type which are synergistic to the action of the male hormones of the sterol group are prepared from testicles, seminal vesicles and prostate glands by extraction in known manner followed by precipitation at the iso-electric point. p Fresh, or acetone dried, glands may be extracted with aqueous alcohol or acetone containing sodium hydroxide or ammonia and the extract acidified to pH 5,9 to 6,2, according to the particular glands treated, and allowed to stand in an ice box to throw down the active substance as a precipitate which is then taken up in N/10 NaOH and reprecipitated. A crystalline hydrochloride may be obtained by cooling a solution of the product in acid alcohol or by decomposing a picric of the active substance. Active substance remaining in the initial extract after iso-electric precipitation may be precipitated with picrate acid, the picrate decomposed with alcoholic hydrochloric acid and the solution obtained treated with acetone to precipitate a crystalline hydrochloride. Substances obtained by picric acid precipitation are further purified by iso-electric precipitation. The initial extraction may be done under acid or neutral conditions and water may, less favourably, be employed in place of the aqueous organic solvent.
