Abstract
Squill is macerated with a water-miscible organic solvent such as denatured alcohol, the solution is evaporated and the residue dissolved in water. The aqueous solution is agitated with an organic solvent not miscible with water, e.g. petroleum ether, to dissolve out scillin, fats and oils. The active principles are precipitated from the purified aqueous solution with tannic acid, a salt such as sodium chloride being added to complete the precipitation. The precipitated tannates are decomposed with an inorganic oxide or hydroxide such as zinc oxide. The product is separated into three distinct cardio-active substances by the following procedure. The aqueous solution of the product is agitated with a solvent such as absolute ethyl acetate which dissolves principles I and II. Principle III is recovered from the aqueous solution by precipitation with tannic acid, decomposition of the tannate with a metallic oxide, and recrystallization of the product from absolute ethyl alcohol. The solution of principles I and II is evaporated, redissolved in chloroform, and agitated with dilute aqueous alkaline carbonate solution which dissolves principle II. This solution is neutralized and treated with a non-miscible solvent such as ethyl acetate from which principle II is obtained by evaporation. The chloroform solution of principle I is evaporated and the product recrystallized from ethyl acetate. The combustion analysis and physical properties of the three principles are described.
