Abstract

A substance having the property of inhibiting the growth of tissues is obtained from parathyroid glands by neutralizing an acid extract of the glands to a pH of 4,6 to 5,0, filtering off the insoluble matters precipitated and treating the filtrate to concentrate or purify the hormone therein. The filtrate may be concentrated under reduced pressure the pH being readjusted if necessary to 4,5 to 5,0. The acid liquid may consist of water, an alcohol, e.g. ethyl alcohol, or a ketone, e.g. acetone, and an acid, e.g. sulphuric or hydrochloric acid. The fat may be extracted by treatment of the glands with a fat solvent prior to or simultaneously with treatment with the acid liquid. Examples: (1) Minced glands are shaken with a mixture of water, sulphuric acid, alcohol, and benzene; strained off and the benzene separated. The acid alcoholic liquid is neutralized with caustic soda to pH 4,8. Alcohol is added to precipitate sodium sulphate and the liquid after filtration is concentrated in vacuo, and the pH adjusted to 4,8. (2) Glands are shaken with strong alcohol containing 5 per cent concentrated sulphuric acid, the liquid is neutralized till slightly acid to litmus, filtered, and concentrated by distillation and adjusted to pH=4,8, (3) Glands are treated on a boiling water bath with dilute sulphuric acid, the liquid is cooled and the fat separated and its pH adjusted to pH=9 by addition of sodium hydroxide, strained and centrifuged. The liquid is then neutralized with sulphuric acid to pH=4,8, allowed to stand in the cold and centrifuged. The precipitate so obtained is dissolved in dilute alkali, the pH adjusted to pH=4,8, centrifuged and the filtrate added to that first obtained. Alcohol is added to precipitate sodium sulphate and the filtrate concentrated. (4) Glands are macerated with acetone, and the solid residue is dried, ground and treated with dilute sulphuric acid. The solution obtained is treated in a similar manner to that of example 3.
