Description
-1- La présente invention se rapporte,d'une manière générale, à de
nouveaux
composés chiraux ainsi qu'à leur procédé de préparation.
Ces composés sont particulièrement utiles comme intermédiair Espour la préparation de dérivés de prostaglandine et plus particulièrement des dérivés de prostaglandine décrits dans la demande de brevet N 81 14910
du 31 juillet 1981.
Les nouveaux composés chiraux en question sont des dérivés de cyclopentène qui peuvent être représentés par la formule générae:

Le procédé de préparation de ces Synthons A à F comprend un certain nombre d'étapes originales au départ de la O-cyclohexylidène-3,4 trihydroxy-3,4,5 cyclohexanone ( 3 R,4 S,5 R), ce dernier composé étant obtenu à partir d'acide quinique tel que décrit, par exemple, par CLEOPHAX, LEBOUL, GERO, AKHTAR,
BARNETT, PEARCE dans J Ao C S, 1976, 98, 7110.
Le procédé de l'invention pour la préparation des composés chiraux inter-
médiaires en question peut être résumé comme suit: A Préparation du Synthon A a) Réaction de la O-cyclohexylidène-3,4 trihydroxy-3,4,5 cyclohexanone ( 3 R,4 S,5 R) de formule: OH 0 =

en présence d'éthérate de trifluorure de bore, avec l'éthanedithiol à température ambiante et dans un solvant aprotique tel que par exemple le benzène, le toluène, le chloroforme ou le dichlorométhane, pour obtenir le trihydroxy-3-4,5 cyclohexanone éthylènedithioacétal ( 3 R, 4 S, 5 R) de formule OH k III OH
b) Traitement du dithioacétal III avec le diméthoxy-1,1 cyclohexane en pré-
sence d'un catalyseur acide tel que par exemple l'acide sulfurique ou l'acide p-toluènesulfonique, à température ambiante et dans un solvant aprotique tel que le N,N-diméthylformamide, pour obtenir le 3 -
O-cyclohexylidène-3, 4 trihydroxy-3, 4,5 cyclohexanone éthylènedithio-
acétal ( 3 R, 4 S, 5 R) de formule: OH

E Préparation du Synthon E a) Traitement des diols VI au reflux avec le nickel de Raney (catalyseur au nickel finement divisé obtenu par dissolution de l'aluminium d'un
alliage nickel-aluminium au moyen d'alkali) et dans un solvant appro-
prié par exemple l'éthanol pour donner les dérivés trihydroxy-1,2,3 cyclohexane ( 1 R, 2 S, 3 R) de formule générale:

nate de triphénylbismuth dans un solvant anhydre approprié, par exemple le chloroforme et à température ambiante pour obtenir les dérivés hexanedial de formule générale:

c) Cyclisation des dialdéhydes acryliques XIII in situ et sous atmosphère inerte avec l'acétate de pyrrolidine ou l'acétate de pipéridine dans un solvant approprié, tel que le toluène et à une température allant de -10 C à la température ambiante, de préférence à O C pour obtenir les dérivés formyl-1 cyclopentène-1 de formule générale: -CHO XIV

dans laquelle R a la même signification que précédemment ou Synthon E. i F Préparation du Synthon F On traite le Synthon E ci-dessus avec le propanediol-1,3 à une température entre la température ambiante et 40 C, par exemple 30 C et en présence d'acide p-toluènesulfonique, le traitement étant effectué dans un solvant anhydre, par exemple le benzène ou le toluène, ce qui fournit les dérivés formylpropylèneacétal-1 cyclopentène de formule générale:

EXEMPLE 1
Préparation du formyl-2 (R)-benzyloxy-4 cyclopentèn-2 one éthylènedithio-
acétal ou Synthon A (Composé VIII dans lequel R 1 = phényle) a) Trihydroxy-3,4,5 cyclohexanone éthylènedithioacétal ( 3 R, 4 S, 5 R) (ComposéII A 8 g de 0-cyclohexylidène-3,4 trihydroxy-3,4,5 cyclohexanone ( 3 R, 45, 5 R) (composé II) dissous dans 40 ml de chloroforme anhydre, on ajoute 16 ml d'éthanedithiol et 1,6 ml d'éthérate de trifluorure de bore fraîchement
distillé Après une heure à température ambiante, on effectue une chroma-
tographie sur couche mince (solvant: chloroforme/éther éthylique 3/1) et on constate la disparition du composé de départ Le composé désiré de formule III précipite dès sa formation dans le milieu réactionnel et on le dissout alors par ajout de méthanol On neutralise la solution avec du
bicarbonate de sodium, on filtre et on évapore les solvants.
On dissout à nouveau le solide résultant dans l'acétone chaud et on
filtre les sels insolubles.
De cette manière, on obtient le composé III après cristallisation dans l'acétone. _ 8 _ Rendement: 95 %
P.F: 129-130 C

diméthylformamide, on ajoute 8 ml de diméthoxy-1,1 cydlohexane et 0,5 ml d'acidesulfurique concentré On évapore de temps en temps le méthanol formé au cours de la réaction et ce au moyen d'une trompe à eau de façon
à favoriser le déplacement de l'équilibre conduisant au composé IV attendu.
Au bout de deux jours à température ambiante, la réaction est terminée comme indiqué par chromatographie sur couche mince (solvant: chloroforme/
éther éthylique 3/1) On dilue la solution au dichlorométhane et on neu-
tralise au bicarbonate de sodium Après filtration, on lave à l'eau la phase organique puis on la sèche sur sulfate de sodium et on la concentre
sous pression réduite.
De cette manière, on obtient le composé IV après cristallisation dans
l'éther de pétrole.
Rendement: 95 %

La coloration du milieu réactionnel est alors jaune orange Au bout de 3 heures, la réaction est terminée comrnme l'indique une chromnatographie sur couche mince (solvant: chlroforme/éther éthylique 3/1) On détruit
l'excès d'hydrure par addition de miéthano L et on verse le mélange réac-
tionnel sur de l'eau glacée Après extraction au dichlorométhane, on
lave la phase organique à l'eau et on la sèche sur sulfate de sodium.
Après évaporation des solvants, on recupère une huile jaune.
De cette manière, on obtient le composé V souhait 8 après cristallisation
dans l'éthanol aqueux.

On ajoute 3 ml d'éthylèneglycol pour éliminer le tétraacétate de plomb en excès Lorsque cet excès a réagi, la solution devient claire On dilue alors le milieu réactionnel avec du dichlorométhane et on lave la phase organique d'abord à l'eau puis avec une solution saturée de bicarbonate de sodium et finalement à l'eau jusqu'à neutralité On sèche alors la phase organique sur sulfate de sodium, on filtre et on évapore On obtient ainsi, de manière quantitative, le composé VII désiré sour la forme d'une huile incolore Cette huile acquiert une teinte jaune au cours du temps
et doit par conséquent être utilisée immédiatement.

EXEMPLE 2
Préparation du formylpropylèneacétal-2 (R)-benzyloxy-4 cyclopentèn-2 one éthylènedithioacétal ou Synthon B (composé IX dans lequel R 1 = phényle) Cette réaction est effectuée directement sur l'aldéhyde a,c insaturé obtenu précédemment. Dans 50 ml de toluène sec, on dissout 0,550 g de composé VIII obtenu à l'Exemple 1 f et on ajoute 1,5 ml de propanediol-1, 3 ainsi qu'une trace d'acide p-toluènesulfonique Après 24 heures de réaction, on évapore 3/4 du volume au moyen d'un évaporateur rotatif Après quoi, on ajoute 1,5 ml de propanediol-1,3 et 50 ml de toluène anhydre pour déplacer l'équilibre en faveur du composé désiré et on renouvelle cette opération 24 heures plus tard La réaction est presque terminée après 72 heures comme indiqué par chromatographie sur couche mince (solvant: acétate d'éthyle/éther de
-15 pétrole 3/7) Il subsiste cependant un peu d'aldéhyde de départ ( 5 à 10 %).
On dilue le milieu réactionnel au dichlorométhane et on le neutralise au bicarbonate de sodium Après filtration des sels, on lave la phase
organique à l'eau, on sèche sur sulfate de sodium, on filtre et on éva-
pore On sépare alors le composé obtenu par chromatographie sur couche mince (solvant: acétate diéthyle/éther de pétrole 3/7) On obtient une huile incolore après élution à l'acétate d'éthyle de la bande de Rf
inférieur, filtration des solvants puis évaporation.
De cette manière, on obtient le composé IX souhaité ou Synthon B après
cristallisation dans l'éther de pétrole.
Rendement: 80 %

Après quoi, on ajoute une goutte d'oxyde de propylène par 50 mg de composé de départ de façon à éliminer toute trace possible d'acide ni- trique Après 20 heures de réaction, une chromatographie sur couche
mince (solvant: acétate d'éthyle /éther de pétrole 3/7) montre la dis-
parition du composé de départ et la formation d'un composé de Rf inférieur.
On dilue légèrement le milieu réactionnel au dichlorométhane, on neutra-
lise au bicarbonate de sodium, on filtre et on sépare par chromatographie sur couche mince (solvant: acétate d'éthyle/éther de pétrole 3/7) La bande de Rf inférieur en lumière U V est éluée à l'acétate d'éthyle,
filtrée et les solvants sont évaporés.
De cette manière,on obtient le composé X souhaité ou Synthon C sous la
forme d'une huile incolore cristallisant dans l'éther de pétrole.
Rendement: 68 %

Préparation du formylpropy lèneacétal-2 (R)-benzyloxy-4 cyclopentèn-2 ol ou Synthon D (composé XI dans lequel R 1 = phényle) A 2, 44 g de composé X obtenu à l'Exemple 3 dissous dans 53 ml de toluène anhydre, on ajoute goutte à goutte, sous atmosphère d'azote et à O C,
13,1 ml d'hydrure de diisobutyl aluminium (solution 1 M dans l'hexane).
Trente minutes plus tard, une chromatographie sur couche mince (solvant chloroforme/éther éthylique 3/1) révèle la disparition totale du composé
de départ On arrête la réaction par addition lente de méthanol à O C.
On verse la solution dans de l'eau glacée saturée en chlorure de sodium puis on reprend au dichlorométhane On sèche la fraction organique sur
sulfate de sodium puis on évapore à sec.
On dissout l'huile obtenue dans un mélange chloroforme/éther éthylique 3/1 On filtre cette solution sur Celite (produit commercial à base de terre d'infusoires, le terme "Celite" étant une marque déposée) puis on évapore à sec On cristallise le produit désiré dans un mélange dichlorométhane/ éther de pétrole En R M N du proton les eaux-mères révèlent la présence
d'un autre produit qui est l'autre isomère.
De cette manière, on obtient le composé XI souhaité ou Synthon D. Analyse élémentaire C 16 H 20 04 P Mol: 276,336 Calculé (%) C: 69,54 H: 7,29 Trouvé (%) C: 69,36; H: 7,30 -16- S.M.

EXEMPLE 5
Preparation du formyl-1 (R)-benzyloxy-3 cyclopentene-1 ou Synthon E (composé XIV dans lequel R 1 = phényle) a) O-Benzyl-3 trihydroxy-1,2 _ 3 cyclohexane ( 1 R, 2 S, 3 R) (comosé XII dans lequel R 1 = phényle)
Dans 80 ml d'éthanol à 95 , on dissout 1,3 g de composé VI obtenu à l'Ex-
emple 1 d Après dissolution complète du produit, on ajoute du nickel de
Raney en large excès et on porte le milieu au reflux pendant 12 heures.
On filtre le mélange réactionnel sur Celite et on rince soigneusement à l'éthanol de façon à éliminer tout le nickel de Raney On reprend dans le chloroforme l'huile obtenue et on filtre sur papier Whatmann Le
produit désiré cristallise dans l'éther de pétrole.
De cette manière, on obtient le composé XII souhaité avec un rendement de 70 % * P.F: 59-60 o C (éther de pétrole) a D = -83 ' (chloroforme, C = 1, 3 P/Vol) Analyse élémentaire C 13 H 18 03 P Mol: 222,287 Calculé (%) C: 70,24; H 8,16 Trouvé (%) C: 70,12; H 8,11 S.M.

b) (R)-Benzyloxy-2 hexanedial (composé XIII dans lequel R 1 = phényle) Dans 120 ml de chloroforme anhydre on dissout 2,34 g de composé XII obtenu précédemment et on ajoute, par petites fractions, 5,7 g de tétraacétate de plomb On effectue la réaction à l'abri de la lumière
et on la contr 8 le par chromatographie sur couche mince (solvant: acé-
tate d'éthyle/ éther de pétrole).
Quatre-vingt dix minutes plus tard, la réaction est terminée et on ajoute
-de 1 ' éthylèneglycol pour éliminer le tétraacétate de plomb en excès.

Lorsque la solution est devenue claire, on la reprend dans du dichloro-
méthane, on lave à l'eau, puis au bicarbonate de sodium et de nouveau à
l'eau On sèche la phase chloroformique sur sulfate de sodium et on éva-
pore à sec.
De cette manière, on obtient le composé XIII souhaité que l'on met direc-
tement en réaction dans l'étape suivante.
R.M N des 2 protons aldéhyde à 60 Mz: 9,6 ppm -1 Spectre I R: CHO à 1720 cm c) Formyl-1 l(R)-benzyloxy-3 cyclope:itene-1 (óomposé XIV dans lequel R = phényle) Dans 100 ml de toluène sec, oa dissout le composé XIII obtenu précédemment et on le met en réaction, sous atmosphère d'azote et à O C, avec I ml d'une
solution benzénique 2 N en acétate de pyrrolidine.
On abandonne le milieu réactiomiel durant environ 8 heures à O C puis on le verse dans l'eau glacée On reprend par le d 1 ohloromttanle, on -sèche la phase organique et on la concentre sous pression réduite L'aldéhyde
a, P insaturé ainsi obtenu, sous form Ie d? me huile jaune clair, est rela-
tivement instable ce qui nécessite son utilisation sans purification dans
l'étape suivante.
De cette manière, or obtient le comnlposé XIV souhaite ou Synthon E. R.M N à 60 M Hz : c(ppm) -H: * 9,8 aldéhyde : 7,3 phényle:

EXEMPLE 6
Préparation du formylpropylèneacétal-1 (R)-benzyloxy-3 cyclopentène ou Synthon F (composé XV dans lequel R _phnyle) Au composé XIII obtenu à l'Exemple 5 b dissous dans 100 ml de toluène sec,
on ajoute 10 ml de propanediol-1,3 et me trace d'acide ptoluènesulfonique.
On agite le mélange à 30 C pendant une heure et on le rmaintient sous 18- agitation pendant 24 heures supplémentaires Lorsque la réaction est terminée, on dilue le milieu réactionnel au dichlorométhane et on le
neutralise au bicarbonate de sodium.
Après 30 minutes d'agitation à température ambiante, on filtre la solution et on l'extrait au dichlorométhane Après lavage à l'eau, on sèche la phase organique sur sulfate de sodium et on évapore sous pression réduite pour obtenir une huile foncée que l'on purifie par chromatographie
sur couche mince (solvant: acétate d'éthyle/éther de pétrole 1/2).
De cette manière, on obtient le composé XV souhaité ou Synthon F sous
forme d'une huile incolore.
Rendement: 55 % (à partir du composé XII) -D = + 74 (chloroforme, C = 1, 4 P/vol) Analyse élémentaire C 16 H 2003 P Mol: 260,333 Calculé (%) C: 73, 81; H : 7,74 Trouvé (%) C: 73,81; H : 7,71 S.M.
