Claims (5)

1. Expressionskasett för produktion av en hirudin-variant i jäst, kännetecknat därav, att den 5 innehäller a) ett första DNA-fragment som kodar för en prekur-sor av hirudinvarianten HV1, vilken prekursor omfattar nämnda hirudinvariant, säsom den i figur 10 angivna hirudinvarianten, och ett extra omräde omfattande mera än tvä 10 aminosyror, vilket har bundits vid hirudinvariantens N- terminal medelst en peptidbindning och vilket innehäller ett enda spjälkningsställe som igenkänns av yscF-proteinas och är dipeptiden Lys-Arg, säsom anges i figur 7, och som bildar det ovan nämnda extra omr&dets C-terminal, 15 b) ett andra DNA-fragment, vilket kodar för en lea- der-peptid, som är pre-pro - eller profragmentet av a-fak-torns pre-pro -prekursor, och vilket andra DNA-fragment bundits vid det första DNA-fragmentets 5-terminal i rätt läsram, och 20 c) ändamälsenliga kontrollomräden för expression av nämnda DNA-fragment, säsom PGK-genpromotorn.
2. Jästcell, kännetecknad därav, att den transformerats med en expressionskasett enligt patentkrav 1. • 25
3. Jästcell enligt patentkrav 2, känne tecknad därav, att expressionskasetten insätts i en plasmid som innehäller en i jäst fungerande replikations-origin.
4. Plasmid, kännetecknad därav, att 30 den innehäller en expressionskasett enligt patentkrav 1 och en i jäst fungerande replikationsorigin.
5. Förfarande för framställning av en hirudinvariant, kännetecknat därav, att en jäst enligt patentkrav 2 eller 3 odlas och hirudinvarianten tas till- 35 vara frän odlingssupernatanten.
