Abstract
A method of preparing a thieno [1,5] -benzodiazepine of the formula: **(See formula)** or an acid addition salt thereof, where R1 and R2 independently represent hydrogen, alkyl, C2-4 alkenyl, C3-6 cycloalkyl, halogen, C1-4 haloalkyl, nitro, amino, C2-4 acylamino, hydroxyl, alkoxy C1-4, C1-4alkylthio or a group of the formula -SO2N (R4) 2 or SO2R4, where R4 is C1-4 alkyl; where (A) R5 is a group of the formula: **(See formula)** where R6 is hydrogen, phenyl optionally substituted by halogen or haloC1-4alkyl C1_4alkyl, cycloalkyl, C3-6, C2_4alkenyl, C1-4alkanoyl, benzyl, C1-4 carboalkoxy or - (CH2) n OX, where n is 2 or 3 and where X is hydrogen or an ester radical, or (B) R5 is a group of the formula: -NH- (CH2) n-Z where n is 2 or 3 and Z is **(See formula)** where R6 is the one defined above, **(See formula)** where R'' and R'''are independently hydrogen or C1-4alkyl or (v) OH and where the group **(See formula)** represents an optionally substituted thiophene ring, fused to the diazepine nucleus; whose procedure consists of: (a) reacting an amine of formula R5-H with a compound of formula (V): **(See formula)** where R1, R2 and R5 are those defined above and where **(See formula)** represents an optionally substituted fused thiophene ring as above and Q represents a radical capable of being cleaved with the hydrogen atom of the amine R5H, (h) if desired, if R5 is **(See formula)** and R6 is C1-4 carboalkoxy, from hydrolysis to the amine where R6 is hydrogen: (c) optionally reacting the product obtained in the previous step of formula (VI): **(See formula)** with an alkylating agent of formula R6X, where R6 is as defined above except for hydrogen and phenyl and where X is a reactive atom.
